감마선 조사가 칡의 생리 활성과 색상 변화에 미치는 영향 (Effect of Gamma Irradiation on the Biological Activity and Color Change of Puerariae radix)

  • 칡 추출물에 감마선을 조사하여 색상 개선, 항산화성 및 항균성에 미치는 영향을 검토하였다. 칡 추출물은 methanol 과 acetone으로 추출하였으며, 감마선은 10, 20 그리고 30 kGy로 조사하였다. 칡 추출물은 감마선 조사 선량이 증가할수록 명도(Hunter color L-value)가 증가하였으며, 적색도(a값) 와 황색도(b값)은 감소하는 경향을 보여줬다. 칡 추출물의 항산화력(TBARS)은 acetone 추출물보다 methanol 추출물이 더 높았다. 전자 공여능 측정 결과는 methanol 추출의 경우 비조사구와 조사구 모두 비슷한 활성을 보여줬으나 acetone 추출의 경우 감소하는 경향을 보여줬다. 칡 추출물의 용매별 및 감마선 조사 선량에 따른 항균효과는 acetone 추출의 경우, B. subtilis, B, natto, B, megaterium, S, aureus, Sal, typhymurium and E coli.등 모든 균주에 대체적으로 높은 항균 활성을 나타냈으나, methanol 추출의 경우에서는 Sal. typhymurium과 E. coli을 제외하고 항균 활성을 나타내었다. 또한 칡 추출물은 30 kGy까지 감마선을 조사했을 때, 추출물의 생리활성이 감소하는 경향은 나타나지 않았다. 따라서 칡 추출물을 감마선 조사 후, 식품 보존제 및 화장품 원료로서 가능성이 있을 것으로 생각된다.

솔잎과 쑥 추출물의 기능성 점토 (Studies on the Physiological Functionality of Pine Needle and Mugwort Extracts)

  • 솔잎과 쑥의 기능성 검토를 위한 연구의 일환으로 열수 및 70% acetone으로 처리하여 얻은 추출물의 항변이원성, angiotensin I converting enzyme(ACE) 저해작용, 전자 공여작용, polyphenol oxidase(PPO)에 대한 저해작용, 아질산염 소거작용, 항산화 작용을 조사하였다. 솔잎의 열수 및 70% acetone 추출물 모두 총 polyphenol 성분 중 flavanol형 tannin의 함량이 60% 이상이었고, 간접변이원인 Trp-P-1을 이용한 항변이원성 실험결과 70% acetone 추출물은 처리구는 82.2%로 열수 추출물보다 높은 억제 활성을 나타내었으나 ACE 저해작용은 열수 추출물이 61%로 70% acetone 추출물의 50%보다 저해능이 높았다. 전자공여작용은 두가지 추출물 모두 80% 이상의 높은 효과를 나타내었다. 미나리에서 분리한 PPO 조효소에 대한 70% acetone 추출물의 저해효과는43%로 나타났고 또한 아질산염 소거작용은 산성 부근(pH 3.0)에서 각 추출물 모두 $80{\sim}90%$의 높은 효과를 보였다. 한편 쑥추출물의 경우 항변이원성은 70% acetone 추출물이 84.1%로 솔잎과 유사하였으나 ACE 저해작용은 열수 추출물이 75.1%로 acetone 추출물의 57.6%보다 높았고, 전자공여 작용은 두 처리구 모두 45% 정도였으며, 아질산염 소거작용은 솔잎에 비해 $20{\sim}30%$ 분해능이 떨어지는 것으로 판명되었다 또한 솔잎 열수 추출물을 chloroform, ethyl acetate, butanol을 이용하여 얻은 분획물을 대두유에 첨가하여 저장기간 중 항산화성을 조사한 결과 ethyl acetate 분획물이 가장 효과가 있었다.

무 추출물이 식품관련 미생물의 증식에 미치는 영향 (Effect of Radish Extract on the Growth of Food-born Microorganisms)

  • To understand the effect of radish on growth of food born microorganisms, mashedflesh radishes were extracted by using acetone and distilled water. Their effect was assayed by measuring the optical density of cultural broth of food born microorganisms. In the experiment, seven strains of yeast were used as the test organism. Acetone extract inhibited growth of the cells of L. plantarum, L. sake and Danmuji film yeast. Growth of the film yeast was drastically inhibited in the concomitant presence of 0.03% extract, while other microbes such as L. faecalis, P. pentosaceus, B/ subtilis and E. coli grew by succeeding cultivation for 4 to 8 hour after addition of the extract. Water extract, on contrast to acetone extract, at he concentrations of 0.1∼1.5% stimulated the growth of lactic acid bacteria. Culture of L. faecalis and L. sake showed an optical density higher than that of control by 40∼50 times. The effect was not so apparent against E. coli, S. aureus and Danmuji film yeast.

두충잎 아세톤 추출물의 in vitro 항암 및 항산화 효과 (In vitro anticancer and antioxidant effects of acetone extract of Eucommia ulmoides oliver leaves)

  • 두충(Eucommia ulmoides Oliver)잎 아세톤 추출물의 in vitro 항암 및 항산화 활성을 조사하였다. 두충잎 아세톤 추출물의 수율은 $1.13{\pm}0.033%$ (w/w)이었고, 총 페놀성 화합물 함량은 $36.7{\pm}1.96mg$ gallic acid equivalents/g-추출물로 나타났다. 세포증식을 절반 억제하는 추출물의 농도인 $GI_{50}$값은 사람의 암세포인 비소세포폐암세포(A549)에서 $53.4{\mu}g/mL$, 결장암세포(SNU-C4)에서 $53.8{\mu}g/mL$ 및 자궁경부암세포(HeLa)에서 $88.3{\mu}g/mL$이었고, 사람의 정상세포인 배아 폐 상피세포(L132)에서는 $153.9{\mu}g/mL$로 나타났다. 두충잎 아세톤 추출물은 사람의 정상세포에는 낮은 독성을 나타내면서 농도에 비례하여 사람의 비소세포폐암세포(A549)와 결장암세포(SNU-C4)의 증식을 효과적으로 억제하였다. DPPH 유리라디칼을 절반 소거하는 추출물의 농도인 $EC_{50}$값은 2 mg/mL 정도로 높게 나타났고, 환원력의 $EC_{50}$값은 $275.8{\mu}g/mL$, 지질과산화를 절반 저해하는 농도인 $EC_{50}$값은 $257.9{\mu}g/mL$이었다. 유근피 아세톤 추출물은 농도에 비례하여 우수한 환원력 및 지질과산화 억제활성을 보여주었다.

향미유 제품에 대한 추출 용매별 로즈마리 추출물의 저장 안정성 향상 효과 (Increasing Effect on Storage Stability of Rosemary Extracts used for Various Solvent System on Seasoning Oils)

  • RPSO, SO, PNO의 3종 향미유에 대하여 로즈마리 유기 용매 추출물과 비교군으로 토코페롤을 각각 1,000ppm씩 동량으로 적용하여 60${\pm}$2$^{\circ}C$의 항온기에 저장하여 AV, POV 및 카르보닐 화합물 생성량의 변화 양상을 측정하였다. 이들 향미유의 AV, POV측정 과정에서는 고유의 색상으로 인하여 상대적으로 측정 오차가 크게 발생하였으며, 토코페롤은 AV, POV, 카르보닐 화합물 생성량 등 각종 측정 항목에서 항산화제로서의 역할보다는 오히려 산화 촉진제로 작용하는 것으로 밝혀졌다. 전체적으로 chloroform/MeOH 추출물 처리의 저장 안정성의 향상 효과가 가장 우수하였으며, 측정 항목 및 추출물별 저장 안점성의 향상 효과는 다음과 같았다. 즉, AV는 chloroform/ MeOH 추출물>ethyl alcohol 추출물>열수 추출물>ethyl acetate 추출물>acetone 추출물>무처리군>토코페롤 처리군이었으며, POV는 chloroform/MeOH 추출물>ethyl alcohol추출물${\geq}$ethyl acetate 추출물>acetone 추출물${\geq}$열수 추출물>무처리군>토코페롤 처리군이었고, 카르보닐 화합물 생성량은 chloroform/ MeOH 추출물>ethyl acetate추출물>ethyl alcohol 추출물>열수 추출물>acetone 추출물>무처리군>토코페롤 처리군의 순이었다.

Antioxidant Activities and Tyrosinase Inhibitory Effects of Different Extracts from Pleurotus ostreatus Fruiting Bodies

  • We evaluated the antioxidant activity and tyrosinase inhibitory effects of Pleurotus ostreatus fruiting bodies extracted with acetone, methanol, and hot water. The antioxidant activities were tested against $\beta$-carotene-linoleic acid, reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity, and ferrous chelating ability. Furthermore, phenolic acid and flavonoid contents were also analyzed. The methanol extract showed the strongest $\beta$-carotene-linoleic acid inhibition as compared to the other exracts. The acetone extract (8 mg/mL) showed a significantly high reducing power of 1.54 than the other extracts. The acetone extract was more effective than other extracts for scavenging on 1,1-diphenyl-2-picrylhydrazyl radicals. The strongest chelating effect (85.66%) was obtained from the acetone extract at 1.0 mg/mL. The antioxidant activities of the extracts from the P. ostreatus fruiting bodies increased with increasing concentration. A high performance liquid chromatography analysis detected seven phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, naringenin, hesperetin, formononetin, and biochanin-A in an acetonitrile and 0.1 N hydrochloric acid (5 : 1) solvent extract. The total phenolic compound concentration was $188{\mu}g$/g. Tyrosinase inhibition of the acetone, methanol, and hot water P. ostreatus extracts increased with increasing concentration. The results revealed that the methanol extract had good tyrosinase inhibitory ability, whereas the acetone and hot water extracts showed moderate activity at the concentrations tested. The results suggested that P. ostreatus may have potential as a natural antioxidant.

유채박의 이화학적 특성 및 항산화 효과 (Physicochemical Properties and Antioxidative Activities of Rapeseed Meal)

  • This research was to investigate physicochemical properties and antioxidative activities of rapeseed meal for the development of functional cosmetic material. Seventeen kinds of amino acid at rapeseed meal were found and glutamic acid concentration was significantly the highest (28.4 mg/g), followed by glycine, proline, arginine, isoleucine, and aspartic acid. Among various vitamins, cloline content was the highest (459.1 mg/kg), followed by niacin, tocopherol, and pantothenic acid. Among various fatty acids of rapeseed meal, oleic acid was the highest (36.7%), followed by linoleic acid and linolenic acid. DPPH radical scavenging activities of methanol and acetone extract of rapeseed meal at 2.0 mg/mL were 80.4 and 78.9%, respectively. The methanol and acetone extracts of rapeseed meal were a stable at the range of pH 3-9 on DPPH radical scavenging activity. The maximum reducing powers of methanol and acetone extract of rapeseed meal at 4.0 mg/mL were 0.7 and 0.68 OD 700 nm, respectively. The maximum superoxide inhibition activities of hot water, acetone, and methanol extract of rapeseed meal were 70.2, 75.2, and 81.4%, respectively. These results showed that the methanol and acetone extract of rapeseed meal can be used as a new source of functional cosmetic material.

Phenolic constituents and biological activities of leaf extracts of traditional medicinal plant Plectranthus amboinicus Benth (Lamiaceae)

  • Plectranthus amboinicus Benth (Lamiaceae) is a medicinal plant native to India, and its leaves are widely used in several traditional medicinal preparations. The purpose of this study was to detect and quantify phenolics present in ethyl acetate and acetone extracts of P. amboinicus leaves, and evaluate their antioxidant, antibacterial, antimutagenic and anticancer activities. The HPLC chromatograms of crude leaf extracts indicated the presence of phenolics like caffeic acid, coumaric acid, rutin, quercetin and gallic acid, which were present in the range of 0.01 - 1.41 mg/g in ethyl acetate and 0.03 - 1.93 mg/g in the acetone extract. The acetone extract showed statistically (p < 0.05) higher antioxidant activity ($IC_{50}$, 99.59 ${\mu}g/ml$) than ethyl acetate extract ($IC_{50}$, 149.96 ${\mu}g/ml$). Statistically (p < 0.05) higher antimutagenicity was shown by acetone extract (46.16%) as compare to ethyl acetate extract (12.16%) at 500 ${\mu}g/plate$ concentration. The acetone extract showed higher antibacterial activity than ethyl acetate extract, and both the extracts showed highest activity against B. cereus (375 and 625 ${\mu}g/ml$, respectively) and lowest activity against Y. enterocolitica (1000 and 1125 ${\mu}g/ml$, respectively). Both the extracts also showed inhibitory effect on cancer cell lines HCT-15 and MCF-7. These results suggest that the leaves of P. amboinicus possess various biological activities, and validate the traditional use of the leaves of P. amboinicus against cold, infection and ulceration.

수원 5호 메밀의 유리라디칼 소거 및 Xanthine oxidase 활성 저해 (Free radical Scavenging Activities and Inhibitory Effects on Xanthine oxidase of Buckwheat (Suwon No. 5))

  • To examine the free radical scavenging activities from buckwheat, Suwon No. 5 was extracted with various solvents. The seed of Suwon No. 5 had a high contents of carbohydrate, but the vegetable of that had a 31.4% of protein, 28.6% of ash and 25.7% carbohydrate as moisture free basis. Phenols contents of vegetable extracts had higher than those of seed extracts. Acetone extract of vegetable showed the greatest electron donating ability. Butanol and acetone extracts of vegetable showed high inhibitory effect on lipid peroxidation. Acetone extract of vegetable has also excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effects of extracts on xanthine oxidase were measured. Acetone extract had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was 2.2$\mu\textrm{g}$.

싸리나무 줄기 추출물의 추출 용매에 따른 항산화활성과 Nitric Oxide 생성 억제 활성 (Antioxidative and Nitric Oxide Production Inhibitory Activities of Lespedeza bicolor Stem Extracts Depending on Solvents)

  • In this study, we investigated on antioxidative activity and nitric oxide production inhibitory activity of various solvent extracts of Lespedeza bicolor. The total polyphenol content of the methanol extract was 192.6 mg/g and flavonoid content of the acetone extract was 40.6 mg/g, as the highest content. In DPPH radical scavenging ability, $SC_{50}$ values of the ethanol and methanol extract were exhibited $0.69mg/m{\ell}$ and $0.89mg/m{\ell}$, respectively. However, in nitric oxide(NO) scavenging ability, $SC_{50}$ values of the acetone was exhibited $0.72mg/m{\ell}$ as the highest activity. Moreover, the acetone extract showed strong NO production inhibitory effect in lipopolysaccharide(LPS)-stimulated Raw 264.7 cell. In the cytotoxicity measurement by MTT assay, the extracts were exhibited Raw 264.7 cell viabilities of 92.57~129.04% as nontoxic result in concentration of $65{\sim}650{\mu}g/m{\ell}$. As a result, the acetone extract of L. bicolor could be applicable to functional materials for anti-inflammatory related fields.
