Microbial cellulose has many potential applications due to its excellent physical properties. The production of cellulose from Acetobacter xylinum in submerged culture is, however, beset with numerous problems. The most difficult one has been the appearance of negative mutants under shaking culture conditions, which is deficient of cellulose producing ability. Thus genetic instability of Acetobacter xylinum under shaking culture condition made developing a stable mutant major research interest in recent years. To find a proper type of bioreactor for the production of microbial cellulose, several production systems were developed. Using a reactor system with planar type impeller with bottoms sparging system, it was possible to produce 5 g/L microbial cellulose without generating cellulose minus mutants, which is comparable to that of static culture system.
One strain of cellulose-producing Acetobacter was isolated from the traditionally fermen- ted grape vinegar in Korea. The isolated strain, designated as KI strain was identified as the Acetobacter xylinum with respect to physiological and biochemical characteristics. KI produced acetic acid from ethanol, and then decomposed acetate to CO$_{2}$ and H$_{2}$O. When the isolated strain was cultivated statically in broth culture, a thick cellulose pellicle was formed. KI was tolerance of 8% ethanol and 30% glucose, and the isolate was positive in ketogenesis from glycerol, $\gamma$-pyrone from glucose and fructose, and 2-ketogluconic acid from glucose. KI strain possessed straight-chain C$_{18:1}$, C$_{16:0}$, and C$_{14:0}$ fatty acid, and contained ubiquinone Q$_{9}$ and Q$_{10}$ as isoprenoid quinone. DNA base composition of KI strain was 57.6% G+C.

Productivity of bacterial cellulose by Acetobacter xylinum GS11 was investigated in the several culture conditions. In various carbon sources, others with the exception of glucose were not found to be effective for cellulose production, and 2% was better in yield than other concentration of glucose. Yeast extract and soytone among several organic nitrogens were effective, but inorganic nitrogen sources tested were not efficient for cellulose production by A. xylinum GS11. The effects of various inorganic salts, amino acids and vitamins were also investigated: $MgSO_4$, phenylalanine and $\alpha$-tocopherol gave the cellulose yield of 1.5, 1.4 and 1.4 fold, respectively, compared with basal medium. In our experiment, cellulose production by A. xylinum GS11 added with 10% coconut milk and 0.5% lignosulfonate in basal medium, was the most efficient among the several material sources employed here, and these were 2.2 and 2.1 fold, respectively.
A complex medium was developed for the production of microbial cellulose by Acetobacter xylinum ATCC 23769. The optimum concentration of each nutrient for the production of microbial cellulose was determined to be 10g peptone, 20g yeast extract, 5g glucose, 1.56g Na2HPO4, 1.8g KH2PO4, 0.05g MgSO4, 0.002g FeCl3, 5g citric acid and 10 mL ethanol per liter. With synergistic effects of citric acid and ethanol, cellulose productivity achieved in developed medium was 0.446 gram of cellulose per gram glucose for static culture, which is much higher than reported values. Cell growth and the cellulose production in the developed medium under static culture was also investigated.
Eight strains producing bacterial cellulose (BC) were isolated from rotten fruits and traditionally fermented vinegars. One of the isolated strains from the rotten grape in Gwangju, Korea, maintained a relatively stable BC production in shaking cultures. This isolated strain proved to be Acetobacter xylinum, based on several biochemical and morphological tests. It was shown that the slant-baffled flask was more efficient than the conventional flask for the BC production in shaking cultures. To determine the most suitable carbon and nitrogen sources for the production of BC, various compounds were examined. Fructose was found to be the most effective carbon source with an optimal concentration of 2%. Mixed carbon source (glucose:fructose=1:3) was also better than glucose or fructose alone. Optimal nitrogen source, when basal medium was used, was 10% (v/v) com steep liquor (CSL). When com steep liquor was used with a mixed carbon source (glucose:fructose=1 :3),4% CSL exhibited the best BC production. Based on these results, a defined medium was developed for the BC production by Acetobacter xylinum KJ-1. When this medium was used under optimal culture conditions, the BC production was 7.2 g/1, which was approximately 3 times higher than that with the traditional HS medium.
A screening was performed to isolate the cellulose-producing microorganisms from vinegar in Korea. The isolated strain was identified as Acetobacter sp. with respect to physiological and biochemical characteristics and designated as Acetobacter CBI-2. Cellulose production of Acetobacter CBI-2 was equal with the well known cellulose-producing bacteria, A. xylinum. The result of separation on thin layer chromatography(TLC) was consistent with the degradation product of native cellulose. The presence of genes required for the cellulose biosynthesis in Acetobacter CBI-2 was confirmed by Southern hybridization.
GDP-mannosyltransferase (GMT) is an enzyme responsible for the addition of a mannose to glucose ($\alpha$[1$\rightarrow$3]) during biosynthesis of the water-soluble branched polysaccharide acetan in Acefobacter species. In an effort to obtain a cellulose-overproducing bacterium, a mutant defective in GMT of Acetobacter xylinum KCCM 10100 was constructed by single crossover homologous recombination using part of the aceA gene encoding GMT amplified by polymerase chain reaction. The GMT-disrupted mutant produced 23% more cellulose, but 16% less water-soluble polysaccharide than those of the wild-type strain. Analysis of the sugar composition by gel permeation chromatography revealed that water-soluble polysaccharides produced by the GMT-defective mutant contained no mannose molecule.
Four mutants of Acetobacter xylinum BRC5 defective in gluconic acid production were isolated from UV-irradiated cells. The gluconic acid-negative mutants did not show glucose oxidase activity. The mutants were also defective in cellulose production. A randomly selected mutant grown in the Hestrin-Schramm medium (pH 6.0) supplemented with gluconic acid, however, was found to synthesize cellulose. The mutant grown in Hestrin-Schramm medium whose pH was adjusted to 5.0 with HC1 and contained no gluconic acid also produced cellulose. Wild-type cells grown under the same condition synthesized cellulose more rapidly than those grown in the pH 6.0 medium.
The characteristics of a strain that contaminates the manufacturing of rice vinegar by Acetobacter pasteruianus was invetigated. Conditions for inhibiting pellicle formation and growth of the contaminant, which occurs during static culture and storage, were also observed. Examining the morphological, cultural, and physiological characteristics and measuring the amount of cellulose production during static culture for 14 day, we found that the strain was known to be Acetobacter xylinum. No growth was observed below $10^{\circ}C$ as well as over $40^{\circ}C$. Also, the extent of growth was limited when the concentrations of ethanol, NaCl, and acetic acid were more than 10%, 1.5%, and 7%, respectively.
