Microbiological Characteristics and Volatile Components of Deastringent Persimmon Vinegar

  • Acetic acid producing bacteria were isolated from deastringent persimmon vinegar and the major bacterium was identified using morphological and biochemical tests. Acetobacter sp. AH-1 was motile, gram negative rod with catalase positive and oxidase negative. The strain can grow up to 5 % ethanol and 2% NaCl as well as 25% glucose. Optimum temperature and pH for growth were 3$0^{\circ}C$ and 5.0, respectively. Volatile constituents of persimmon vinegar were analyzed by purge and trap sampling . Acetic acid adn alcohol were the largest volatile compounds quantitiatively in persimmon vinegar. Among alcohols, 20methyl-1-propanol, isoamyl alcohol and amyl alcohol were detected. Isovaleradehyde and benzaldehyde for aldehyde, isoamyl acteate, ethyl formate, propyl aceetate, and ethyl acetate for esters were likely to contribute to persimmon vivegar flavor.

Purification and Characterization of Alcohol Dehydrogenase from Acetobacter sp. KM (Acetobater sp.KM Alcohol Dehydrogenase의 분리 및 특성)

  • Membrane-bound alcohol dehydrogenase(ADH) was purified to homogeneity from the acetic acid producing bacteria, Acetobacter sp. KM. The enzyme was solubilized and extracted with Triton X-100 and purified using the Mono-Q ion exchange chromatography and Superose 12 gel filtration chromatography. The enzyme was purified to 12-fold with a yield of 30%. The molecular weight of the purified enzyme was to be 335 KDa. SDS-PAGE of the enzyme showed two subunits with molecular weights of 79 KDa and 49 KDa. It indicated that the enzyme consisted of three subunits of the 79 KDa and two subunits of the 49 KDa. The purified .ADH preferentially oxidized straight chain aliphatic alcohol except methanol. Formaldehyde, acetaldehyde and glutaraldehyde were also oxidized. The apparent Km for ethanol was 1.04 mM and the optimum pH and temperature were 5.0∼6.0 and 32$^{\circ}C$, respectively. V2O5 and divalent cation such as ZnCl2 and NiCl2 inhibited enzymatic activity.

  • Cephalexin synthesizing enzyme (${\alpha}$ amino acid ester hydrolase) was partially purified from the culture broth of Acetobacter turbidans ATCC9325 through ammonium sulfate fractionation, DEAE, CM, and Sephacryl S-200 gel filtration. The enzyme has optimum pH 6.0 and temperature, 40$^{\circ}C$ respectively. From the analysis of reaction mixtures by thin layer chromatographic and high performance liquid chromatographic techniques, it was confirmed this enzyme catalyzed simultaneously the following reactions : 1) Synthesis of cephalexin from D-${\alpha}$-phenylglycine methylester (PGM) and 7-amino 3-deacetoxy-cetoxycephalosporanic acid (7-ADCA) 2) Hydrolysis of cephalexin to form 7-ADCA and phenylglycine (PG) 3) Hydrolysis of PGM to form PG and methanol. Base on the above experimental observations, the reaction model of this enzyme was identical with that of the enzyme from Xanthomonas citri.

  • The characteristics of cell growth and cellulose production by Acetobcter xylinum BRC5 were studied in shaking flasks and jar fermentors. Of the substrates tested, the highest cellulose yield was obtained from the fructose medium. Some inductive cultivation was observed and then cellulose was produced with cell growth. When glucose alone or mixture of glucose and fructose was used as the carbon source, cellulose could be biosynthesized under the glucose limitation. Corn steep liquor (CSL), as a low-cost nitrogen source, was a good substitute for yeast extract. In a jar fermentor experiment, 4.14 g/l of disintegrated cellulose was obtained from 8% CSL- medium containing 0.5% glucose and 1.5% fructose.

  • In addition to catalyzing the synthesis of glucan from sucrose as a primary reaction, glucansucrase also catalyzes the transfer of glucose from sucrose to other carbohydrates that are present or are added to the reaction digest. Using dextransucrase and altemansucrase, prepared from Leuconostoc mesenteroides B-742CBM and B-1355C, respectively, we modified the bacterial cellulose in Acetobacter xylinum ATCC10821 culture, and then produced a characteristic cellulose that is soluble and has a new structure. There were also some partially modified insoluble cellulose and oligosaccharides in the modification culture. After methylation and following acid hydrolysis of both the soluble and insoluble glucans, there were ($1{\rightarrow}4$) as well as ($1{\rightarrow}6$) and ($1{\rightarrow}3$) glycosidic linkages in the soluble glucan.

Comparison of Bacterial Cellulose Production in a Jar Fermentor Between Acetobacter xylinum BPR2001 and its Mutant, Acetan-Nonproducing Strain EP1

  • The bacterial cellulose (BC) production by a wild­strain Acetobacter xylinum BPR2001 and that by its acetan­nonproducing mutant, EPI, were compared in a jar fermentor. EPI produced about $28\%$ less BC than the wild-strain. The apparent difference in the cultivation of the two strains was the viscosity increase in the culture broth that was closely associated with acetan production. Increasing the viscosity of the culture broth of EPI by adding agar led to the formation of relatively small and uniform BC pellets, and BC production consequently became two-fold higher than that in the absence of agar and was almost equal to that by BPR2001. Therefore, acetan has an important role in BC production by inducing physical changes in the culture broth of the wild-type strain.

  • Footpad swelling having the relationship with arthus-reaction of antibody-mediated hypersensitivity and delayed type hypersensitivity was recovered to the almost normal level. The nethemoglobin induced by aniline showed no significant deviation. PFC/spleen cell and PFC/106 spleen cell were increased slightly, but not in case of RFC. Hemagglutination value was increased slightly, but hemolytic value was not changed significantly. Mice in the administration of the polysaccharide does not show any significant stress factor in the cage for mice admlrustered plasma corticosterone.   • Dose-dependent responses indicated by the increase of leukocyte, peritoneal exudate cell and weights of immunorgans revealed the improvement of immunity. In the effect of macrophage on phagocytes, there were not substantial differences in the phagocytic and corrected phagocytic index. The administered group being compared with the controlled group, there were no significant changes in SGOT, S-GPT, alaline phosphatase, total protein, albumin, globulin, cholesterol, triglyceride, blood urea nitrogen and glucose.   • 미생물학적 물질변환에 대해서는 인류 초기에서부터 효모를 이용하여 빵, 유제품, 알코올, 음료 등의 생산에 이용하여 왔으며, 주로 농업분야 또는 식품 분야에 국한되어 왔다. 1862년 Pasteur에 의해 bacterium xylinum의 순수 배양균주를 사용하여 알코올로부터 초산을 만드는데 응용한 것이 본격적인 시발점으로 보아 무방하겠다. 그 후 acetobacter aceti에 의한 포도당으로부터 gluconic acid 생산과 acetobacter sp.에서의 sorbitol로부터 sorbose 생산 등이 이루어졌고 정통적인 유기합성 방법에 의해 쉽게 만들 수 없는 반응들에 응용되기 시작하였다. 인류 초기의 혼합배양에 의한 물질변환에의 응용과는 달리 순수배양으로 미생물, 식물세포, 혹은 정제된 효소들에 의해 반응이 이루어지게 되었고 순수한 특정 물질에 대한 새로운 순수물질로의 선별적인 수식도 가능해지게 되었다. 특히 발효와 bioconversion의 차이는 racemates의 분리, 비슷한 반응성을 갖는 여러 기들로부터 특정기능을 갖는 기만의 선별적인 수식, 입체 이성체(chiral center)의 제작, 특정 비활성화된 탄소의 기능 등이 bioconversion에서만 수행할 수 있는 독특한 영역으로서 정밀화학분야, energy 분야, 환경오염분야에서의 특히 미래의 관심 기술로 대두되고 있다.
