Angiotensin Converting Enzyme Inhibitory Activity in Enzymatic Hydrolysates of Anchovy Muscle Protein (멸치육 효소 가수분해물의 Angiotensin 전환효소 저해작용)

  • To develop functional food material with angiotensin converting enzyme (ACE) inhibitory peptides, muscle protein of anchovy, Engraulis japonica was hydrolyzed during 48 hrs by digestive pretenses such as pepsin, trypsin, $\alpha$-chymotrypsin, and commercial proteases such as papain, bromelain, complex enzyme, Elavourzyme, Novozym, Neutrase, Protamex and Alcalase. The only $50\%$ ethanol soluble hydrolysates were tested for inhibitory activity against ACE and yield of $50\%$ ethanol soluble peptide-nitrogen ($ESPN_{50}$). ACE inhibition effects and yield of $ESPN_{50}$ occurred as hydrolysis time increased to 8 hrs, Among those pretenses tested, hydrolysates by Alcalase and $\alpha$-chymohypsin had greater ACE inhibitory activity (80 and $74\%$, reipectively) with eletated levels of $ESPN_{50}$ (48 and 58 mg/ml, respectively), while Protamex hydrolysates had greater ACE inhibitory activities ($73\%$) with reduced levels of $ESPN_{50}$ (7.2mg/ml) than others. Amino acid compositions of $50\%$ ethanol solubles obtained from those hydrolysates were rich in glutamic acid, aspartic acid, cysteine and leucine.

Antioxidant and ACE Inhibitory Activities of Styela clava according to Harvesting Time (채취시기에 따른 미더덕의 부위별 항산화 활성 및 ACE 저해 활성)

  • The antioxidant and angiotensin converting enzyme (ACE) inhibitory harvesting times were evaluated. During February and July of 2008, Styela clava were harvested once per month, and divided into flesh and tunic parts. Each collected part was extracted with water and 70% ethanol. DPPH radical scavenging activity (RSA) for flesh part was higher than that of tunic part, and water extract of flesh harvested at April showed the highest value (53.02% at 10 mg/mL). The highest ABTS RSA was found at water extract of flesh part harvested at March. Water extracts of flesh parts harvested from March to May showed relatively higher ACE inhibitory activity, and freezing did not affect ACE inhibitory activity. The results indicated that antioxidant and ACE inhibitory activity of S. clava were variable depending on harvesting time and parts.

  • ACE2000 MPLS 시스템은 대용량 국가 교환시스템으로 사용하기 위해 개발한 ACE2000 ATM 교환시스템에 Ships-in-the-night 모드 형태로 MPLS 기능을 추가함으로써 사용자들에게 차별화 된 서비스를 제공할 수 있는 시스템이다. MPLS 시스템에서는 채널 사용의 유연성과 채널 자원의 효율적인 사용을 위해 VC 머징 기능을 요구하고 있다. ACE2000 MPLS 시스템에서는 고성능 SAR 소자를 이용하여 VC 머징 모듈을 개발하여 사용하고 있다. VC 머징 기능을 위해서는 고성능 SAR로 구성되어 있는 입력부로부터 패킷을 받아들여 채널 연결설정 정보에 따른 VC 머징 제어기의 패킷 처리 후 또 하나의 고성능 SAR로 구성되어 있는 출력부로 전달하여야 만다. VC 머징 제어기에서는 머징 수신부로부터 UTOPIA-2 인터페이스를 통해 패킷을 받아들여 일반 AAL-5 데이터 패킷, 포워딩 엔진 제어 및 PPP 제어 등틀 위한 EFC 패킷 그리고, 순수 ATA 셀로 구분/저리한 후 UTOPIA-2 인터페이스를 통해 머징 송신부로 보내고 있다. 본 논문에서는 ACE2000 MPLS 시스템에 도입된 VC 머징 기술에 대한 내용으로서 고성능 SAR 소자를 이용한 VC Merger의 핵심 기술인 VC머징 제어기 설계기술에 대하여 언급하였다.

The Novel Angiotensin I Converting Enzyme Inhibitory Peptide from Rainbow Trout Muscle Hydrolysate

  • The purpose of this study was the purification and characterization of an angiotensin I converting enzyme (ACE) inhibitory peptide purified from enzymatic hydrolysates of rainbow trout Oncorhynchus mykiss muscle. After removal of lipid, the approximate composition analysis of the rainbow trout revealed 24.4%, 1.7%, and 68.3% for protein, lipid, and moisture, respectively. Among six hydrolysates, the peptic hydrolysate exhibited the highest ACE inhibitory activity. We attempted to purify ACE inhibitory peptides from peptic hydrolysate using high performance liquid chromatography on an ODS column. The $IC_{50}$ value of purified ACE inhibitory peptide was $63.9{\mu}M$. The amino acid sequence of the peptide was identified as Lys-Val-Asn-Gly-Pro-Ala-Met-Ser-Pro-Asn-Ala-Asn, with a molecular weight of 1,220 Da, and the Lineweaver-Burk plots suggested that they act as a competitive inhibitor against ACE. Our study suggested that novel ACE inhibitory peptides purified from rainbow trout muscle protein may be beneficial as anti-hypertension compounds in functional foods.

Effects of Albizziae Cortex Extracts on the Elastase Activity and DPPH and NO Scavenging Activities

  • Elastic fibers are found in the skin, lungs, arteries, veins and other structures. Elastases destroy the elastic fibers and cause the emphysema and pulmonary hypertension. Oxidative stress is needed for these pathologic changes. Accordingly, present study was designed to investigate the effect of Albizziae Cortex extracts (ACE) on elastase activity and anti-oxidative effects of ACE. The in vitro inhibitory effects on elastase and di(phenyl)-(2,4,6-trinitrophenyl)iminoazanium (DPPH) and nitric oxide (NO) free radical scavenging activities of ACE were measured. The elastase activity was significantly inhibited by ACE. DPPH and NO free radicals were significantly scavenged as well. ACE showed the elastase-inhibiting effects and anti-oxidative activities in vitro. These results suggest that ACE may have potential roles in the treatment of pulmonary emphysema and pulmonary hypertension.

Isolation of Angiotensin Converting enzyme inhibitors from Ripe Cucurbita moschata Duch

  • Angiotensin converting enzyme (ACE) inhibitor acts on the inhibition of ACE and causes a decrease in blood pressure. There have been several reports on screening of ACE inhibitors from natural food products and protein hydrolysates of various food sources. Ripe Cucurbita moschata Duch has been used as an oriental medicine in Korea. To isolate ACE inhibitors, crude water extracts of the edible portion of ripe Cucurbita moschata Duch were obtained after heating in water at 95$^{\circ}C$ for 2 h. Crude extracts were then filtered using PM-10 and YM-1 membranes. The membrane-filtered solution was loaded onto Sephadex G-15 column equlibrated with a phosphate buffer. Among the four major fractions of gel permeation chromatography, the second fraction had the highest inhibitory activity of 65%. Further purification of the fraction using reversed-phase HPLC with a $C_{18}$ column produced ACE inhibitors, which were identified as a mixture having molecular mass of 222 and 273 by Tandem mass spectrometry.

  • For any non-negative real number ${\epsilon}_0$, we shall introduce a concept of the ${\epsilon}_0$-dense subset of $R^m$. Applying this concept, for any sequence {${\epsilon}_n$} of positive real numbers, we also introduce the concept of the {${\epsilon}_n$}-attainable sequence and of the points of {${\epsilon}_n$}-attainable ace in the open subset of $R^m$. We also study the characteristics of those sequences and of the points of {${\epsilon}_n$}-dense ace. And we research the conditions that an {${\epsilon}_n$}-attainable sequence has no {${\epsilon}_n$}-attainable ace. We hope to reconsider the social consideration on the ace in social life by referring to these concepts about the aces.

Yeast copper-dependent transcription factor ACE1 enhanced copper stress tolerance in Arabidopsis

  • Copper is essential but toxic in excess for aerobic organisms. Yeast transcription factor ACE1 functions as a sensor for copper and an inducer for the transcription of CUP1. In addition, ACE1 can activate the transcription of superoxide dismutase gene (sod1) in response to copper. In this study, we introduced the yeast ACE1 into Arabidopsis and analyzed its function in plant. Under high copper stress, the transgenic plants over-expressing ACE1 showed higher survival rate than the wild-type. We also found that over-expression of ACE1 in Arabidopsis increased the activities of SOD and POD, which were beneficial to the cell in copper buffering. Excess copper would suppress the expression of chlorophyll biosynthetic genes in Arabidopsis, RT-PCR analysis revealed that over-expression of ACE1 decrease the suppression. Together, our results indicate that ACE1 may play an important role in response to copper stress in Arabidopsis.

Physiological Functionality of Various Extracts from Danmemil and Legumes (단메밀과 콩 추출물들의 생리 기능성)

  • Physiological functionalities of various extracts from Danmemil and legumes were determined and its optimal extraction conditions were also investigated. Angiotensin I-converting enzyme (ACE) inhibitory activity and tyrosinase inhibitory activity of Danmemil were higher in water extracts (53%, 58%) than those of ethanol extracts. However, its electron-donating ability was the highest in ethanol extracts (72%). ACE inhibitory activity and electron-donating ability of Black bean No. 1 and Taekwangkong(one of bean) were higher in water extracts than those of ethanol extracts, whereas SOD-like activity was the highest in ethanol extracts. ACE inhibitor and tyrosinase inhibitor of Danmemil were maximally extracted when it were treated with 20 times of distilled water at 35$^{\circ}C$ for 24 h and 36 h, respectively. Its electron donating compound was maximally extracted by treatment of 50$^{\circ}C$ for 18 h. ACE inhibitor of Black bean No. 1 was extracted maximally when it was treated with distilled water (1 :20) at 20$^{\circ}C$ for 24 h, whereas the other functional compounds were maximally extracted at 20$^{\circ}C$ for 18 h.

Purification of ACE Inhibitor from Soybean Paste (된장으로부터 Angiotensin Converting Enzyme 활성 저해물질의 정제)

  • Seven commercial soybean paste were tested for ACE inhibition effect. In purification of ACE inhibitor from No. 2 soybean paste, acetone fraction $(50{\sim}80%)$ had 57% protein yield with 92.8% ACE inhibition effect. Inhibitor was purified from acetone fraction of soybean paste by Sephadex G-25, Sephadex LH-20 and ODS column chromatography and HPLC. $IC_{50}$ value of the purified inhibitor was 0.6 mg/ml. The inhibitor showed the competitive inhibition patterns on ACE. Amino acid analysis showed that the peptides consist of Ala, Phe, Leu, Glu, Gly, Ser, and Asp.
