Studies of Plant Tumor Induction (Pat 2) On the Study of Peroxidase Activities of Tumor Tissues Developed on Tomato Stem in Outdor Conditions. (식물의 암종유발에 관한 연구 2 (제 2 ) 에서 도마도 줄기에 유발된 의 Peroxidase Activitiy 에 대하여)

  • The relationships between tumor score and peroxidase activities of tomato stems infected with Agrobacterium tumefaciens strain A6Kl, B6, T372 11BNV6, 11BV7 and wounded stem as a control were examined in relation to crown gall tumor development on purpose to study the lignification of tumor tissue which is affected to the development of crown gall tumor. As the previous paper has been mentioned the fact that the induction of tumor tissues were inhibited or limited in the lignified stem of host plant. It was presumed that the activities of peroxidase related to the development of lignification were decreased during the period of tumor development. But the experimental result in this experiment shows that the peroxidase activities of crown gall tumor-tissues infected with the A. tumefaciens strains which are already known as virulent are increasing during four weeks, however, in the strain 11BNV6 and wound the peroxidase activities are decreasing on the second week after the inoculation of the bacteria strains. These results could be explained on the basis of that possible regulatory agents of lignification which were accumulated in tumor tissues, IAA, ascorbic acid, glutathion(GSH) and caffeic acid esters, were postulated to act as antioxidants which has been suggested by Stafford. Total nitrogen contents in relation to crown gall tumor development were determined for the detection of protein synthesis related to the enzyme activities which are increasing in the time of plant growth. Generally six groups are contained the largest amount of nitrogen on the second week after the inoculation of the bacterium. Comparing to the tumor score, it is presumed that the all of enzyme activities including peroxidase in tumor tissues are increasing from the second week through the third week after the inoculation of bacterium and the protein synthesis is stimulated under the most appropriated temperature during the above periods.

  • Chemical analysis and enzymatic hydrolysis of certain Korean acorns were performed in order to make use of the local acorns. Exclusion of tannin from acorns is aided in the processing. Studies of these were undertaken and the results obtained are summarized as follows; 1. Several Korean acorns were used to analyze their proximate composition, tannin and major inorganic principles. The content of acorn tannin was found to be 6.5 to 7.5%. 2. An Attempt was made to screen suitable strains in order to make acorn-tannin-hydrolyzing enzyme accumulated in the culture broth, and Aspergillus flavus and Asp. sp. AN-11, which showed in their culture broths, were obtained from the contaminated acorns. 3. Cultural measures for the strains of Aspergillus flavus and Asp. sp. AN-11 for an improved tannase production and optimal conditions were determined.

The Screening of Aflatoxin Producing Fungi from Commercial Meju and Soy Bean Paste in Western Gyeongnam by Immunoassay (면역분석기법을 이용한 서부경남 시판 메주 및 된장에서의 Aflatoxin 생성균 검색)

  • Generally, non-aflatoxigenic fungi, such as Aspergillus oryzae, and Aspergillus are main microflora in Korean traditional fermented foods including Meju and soybean paste, but sometimes, Aspergillus flavus and Aspergillus parasiticus can be contaminated and accumulated aflatoxins during fermentation and storage. So the screening of aflatoxigenic strains in fermented traditional food is very important to improve the sanitary quality of those foods. In this work, we screened aflatoxin producing fungi from commercial Meju and soybean paste in Western Gyeongnam by immunoassay. Samples were randomly purchased from market of the commercial Meju(10 EA) and soybean paste(20 EA) in nine areas of Western Gyeongnam. Of the samples collected,24 strains and 22 strains of Aspergillus sp. were isolated from Meju and soybean paste, respectively. The isolated strains were cultured on SLS media at $25^{\circ}C$ for 15 days. The cultured broth were extracted with ethyl acetate and were analysed to determine aflatoxin B$_1$(AFB$_1$) by direct competitive ELISA(DC-ELISA). Six strains(25%) isolated from Meju, and 2 strains(9%) isolated from saybean paste, were confined as aflatoxin producing strains. The average range of aflatoxin productivity of isolates from Meju was 54.6 $\pm$ 38.7 ng/ml and that from soybean paste was 11.1 $\pm$ 8.6 ng/ml, respectively. Among them, isolated strain No. M-5-4 produced a high level of AFBl and showed 98.26 ng/ml of AFB$_1$. Every isolates were also re-confined their AFB$_1$productivity by thin layer chromatography(TLC). The TLC results also showed same trend as DC-ELISA results. As the above results, the screening of hazard mycotoxigenic fungi from traditional fermented foods should be necessary for the safety and the application of HACCP system in the food manufactory in Korea.

Site-specific Disruption of Glyoxylate Bypass and Its Effect in Lysine-producing Corynebacterium lactofermentum Strain

  • The role of glyoxylate bypass in a lysine-producing Corynebacterium lactofermentum strain was analyzed. Unlike the wild type, the strain expressed enzymes of glyoxylate bypass during growth in the fermentation broth containing glucose as the carbon source. To evaluate the importance of glyoxylate bypass in the strain, we disrupted chromosomal aceA by using a cloned fragment of the gene. Site-specific disruption of aceA which codes for the isocitrate lyase, the first enzyme of the bypass, was confirmed by Southern blot analysis. The aceA mutant strain completely lost isocitrate lyase activity and ability to grow in a minimal medium containing acetate as the sole carbon source. The mutant strain was similar to its parental strain in growth characteristics and produced comparable amounts of lysine in shake flasks containing glucose as the carbon source. The amount of oxaloacetate accumulated in the fermentation medium was similar for both strains, suggesting that expression of glyoxylate bypass does not necessarily lead to the increase in intracellular oxaloacetate. These data clearly demonstrate that glyoxylate bypass does not function as one of the routes of carbon supply for lysine production in the strain. It appears that the leakiness of the glyoxylate bypass in the strain might be the result of a secondary mutation which arose during previous strain development by random mutagenesis.

  • The 7.1 Mdal Xbaf fragment of Bacillus pasteurii ATCC 11859 containing gene for urease was inserted into the Xbal site of bifunctional plasmid pGR71, and its urease gene was cloned and expressed in E. coil RRI. But the cloned gene was not expressed in Bacillus subtilis BR151 in consequence of deletion of inserted DNA fragment. The recombinant plasmid thus formed was named pGU66. The restriction map of the plasmid pGU66 was determined, and the size of the plasmid was estimated to be 12.6 Mdal by double digestion of restriction enzymes of the plasmid. The urease of the cloned strain was accumulated in periplasmic space and very similiar to that of donor strains in their enzymatic properties.

  • The supra molecular weight poly(〔R〕-3-hydroxybutyrate) (PH B), having a molecular weight greater than 2 million Da, has recently been found to possess improved mechanical properties compared with the normal molecular weight PHB, which has a molecular weight of less than 1 million Da. However, applications for this PHB have been hampered due to the difficulty of its production. Reported here, is the development of a new metabolically engineered Escherichia coli strain and its fermentation for high level production of supra molecular weight PHB. Recombinant E. coli strains, harboring plasm ids of different copy numbers containing the Alcaligenes latus PHB biosynthesis genes, were cultured and the molecular weights of the accumulated PHB were compared. When the recombinant E. coli XL1-Blue, harboring a medium-copy-number pJC2 containing the A. latus PHB biosynthesis genes, was cultivated by fed-batch culture at pH 6.0, supra molecular weight PHB could be produced at up to 89.8 g/L with a productivity of 2.07 g PHB/L-h. The molecular weight of PHB obtained under these conditions was as high as 22 MDa, exceeding by an order of magnitude the molecular weight of PHB typically produced in Ralstonia eutropha or recombinant E. coli.

Earthquake resistance of structural walls confined by conventional tie hoops and steel fiber reinforced concrete

  • In the present study, the seismic performance of structural walls with boundary elements confined by conventional tie hoops and steel fiber concrete (SFC) was investigated. Cyclic lateral loading tests on four wall specimens under constant axial load were performed. The primary test parameters considered were the spacing of boundary element transverse reinforcement and the use of steel fiber concrete. Test results showed that the wall specimen with boundary elements complying with ACI 318-11 21.9.6 failed at a high drift ratio of 4.5% due to concrete crushing and re-bar buckling. For the specimens where SFC was selectively used in the plastic hinge region, the spalling and crushing of concrete were substantially alleviated. However, sliding shear failure occurred at the interface of SFC and plain concrete at a moderate drift ratio of 3.0% as tensile plastic strains of longitudinal bars were accumulated during cyclic loading. The behaviors of wall specimens were examined through nonlinear section analysis adopting the stress-strain relationships of confined concrete and SFC.

Elevated Temperature Design of KALIMER Reactor Internals Accounting for Creep and Stress-Rupture Effects

  • In most LMFBR(Liquid Metal Fast Breed Reactor) design, the operating temperature is very high and the time-dependent creep and stress-rupture effects become so important in reactor structural design. Therefore, unlike with conventional PWR, the normal operating conditions can be basically dominant design loading because the hold time at elevated temperature condition is so long and enough to result in severe total creep ratcheting strains during total service lifetime. In this paper, elevated temperature design of the conceptually designed baffle annulus regions of KALIMER(Korea Advanced Liquid MEtal Reactor) reactor internal strictures is carried out for normal operating conditions which have the operating temperature 53$0^{\circ}C$ and the total service lifetime of 30 years. For the elevated temperature design of reactor internal structures, the ASME Code Case N-201-4 is used. Using this code, the time-dependent stress limits, the accumulated total inelastic strain during service lifetime, and the creep-fatigue damages are evaluated with the calculation results by the elastic analysis under conservative assumptions. The application procedures of elevated temperature design of the reactor internal structures using ASME Code Case N-201-4 with the elastic analysis method are described step by step in detail. This paper will be useful guide for actual application of elevated temperature design of various reactor types accounting for creep and stress-rupture effects.

Round robin analysis to investigate sensitivity of analysis results to finite element elastic-plastic analysis variables for nuclear safety class 1 components under severe seismic load

  • As a part of round robin analysis to develop a finite element elastic-plastic seismic analysis procedure for nuclear safety class 1 components, a series of parametric analyses was carried out on the simulated pressurizer surge line system model to investigate sensitivity of the analysis results to finite element analysis variables. The analysis on the surge line system model considered dynamic effect due to the seismic load corresponding to PGA 0.6 g and elastic-plastic material behavior based on the Chaboche combined hardening model. From the parametric analysis results, it was found that strains such as accumulated equivalent plastic strain and equivalent plastic strain are more sensitive to the analysis variables than von Mises effect stress. The parametric analysis results also identified that finite element density and ovalization option in the elbow elements have more significant effect on the analysis results than the other variables.

Molecular Cloning and Expression of Heat-stable Enterotoxin Gene from Swine Enterotoxigenic Escherichia coli (돼지에서 유래한 병원성 대장균의 내열성 장독소 생산유전자의 Cloning 및 발현)

  • Enterotoxigenic E. coli is one of the major causative agents of the infantile diarrhea and traveler's diarrhea. The heat-stable enterotoxin(ST) is thought to be a virulence factor in the pathogenesis of the diarrhea and to be a maker for identification of the enterotoxingeic E. coli from non pathogenic E. coli. The isolate of enterotoxigenlc E. coli was isolated from swine during 1989 year(from 5 to 10 month) in the Kyong-gi and Chung-Cheong provinces, and three strains(KM-4, KM-7 and KM-12) was selected from 189 isolates of ST producing E. coli. The detection of a ST produced of the isolated E. coli was performed by the infant mouse assay(IMA). This study was designed to know optimal conditions for the production of the ST and the molecular properties of plasmids of the enterotoxigenic E. coli. Amount of ST produced were the most at initial pH 8.5~9.0 of succinate salts medium culture. The cultural time of the same medium was accumulated the highest level of ST was at the 14 to 16 hours, and then stationary phase was at the 20 hours. From this experiment the KM-7 strain was selected among ST producing strains by IMA. Partial plasmid-curing experiment was done to select plasmid encoding for ST among other plasmids and then comparing the plasmid pattern of ST producing strain(KM-7) with those of other ST non-producing strains, it is found that ST gene exists on the about 80 Kbp plasmid. Each fragment of this plasmid digested with EcoRl was ligated to vector pBR 322 and transformed into E. coli K-12. A clone producing ST(eKT 53) was selected by IMA. The EcoRl digestion pattern of the isolated plasmid(pKD 37) from the ST producing clone it is indicated that the size of the inserted fragment in eKT 53 strain is 16 Kbp. The cultured supernatant of eKT 53 strain was positive result of ST production in IMA.
