Production of Poly(3-hydroxybutyrate) by Cupriavidus necator at Various Concentrations of Carbon Dioxide (Cupriavidus necator를 이용한 Poly(3-hydroxybutyrate) 생산에 이산화탄소의 농도가 미치는 영향)

  • Polyhydroxyalkanoates (PHAs) are synthesized by numerous bacteria as carbon and energy storage compounds and are raw materials for biocompatible plastics. In this paper, the effect of $CO_2$ concentrations on the growth of C. necator and the accumulation of Poly(3-hydroxybutyrate) (P(3HB)) are investigated by increasing the $CO_2$ concentration in the substrate gas mixture. During 6 d cultivation in a nitrogen-present mineral medium, the $CO_2$ concentration did not affect the growth of the cells, while the Poly(3- hydroxybutyrate) (P(3HB)) content decreased with increasing $CO_2$ concentrations from 1% to 20%. During 4 d cultivation in the nitrogen-limited medium, the P(3HB) accumulation was the greatest at 3% $CO_2$; however, the total amount of accumulated P(3HB) was the greatest at 1% $CO_2$, which decreased with increasing $CO_2$ concentrations. The results indicate that the gas mixture with 1% $CO_2$ is the most effective in both growing the cells and accumulating P(3HB) under our experimental conditions.

  • This study was designed to assess the quality and quantity of food resources such as the chlorophyll a concentration (Chl-a), the particulate organic carbon (POC) and the particulate organic nitrogen (PON), on blood cocke (Tegillarca granosa Linnaeus) farms from May to November 2017 in the northwestern area of Yeoja Bay, Korea. The values of Chl-a, POC and PON were $1.69-7.68{\mu}g{\cdot}L^{-1}$ (average: $3.48{\mu}g{\cdot}L^{-1}$ in the surface), 0.88-2.58 mM (average: 1.97 mM) and 0.17-0.90 mM (average: 0.54 mM), respectively, and these values were higher in the spring and summer and lower in the autumn. Furthermore, Chl-a and POC had higher values on the bottom layer than those vales on the surface, while PON had higher values on the surface than those values on the bottom. The POC/PON ratio and POC/Chl-a ratio were 1.56-7.88 (average: 3.71 on the surface of the water) and 216-967 (average: 700 on the bottom of the water), respectively, with most of the carbon sources being sediment-accumulated particle organic matter, and the contribution by phytoplankton was assessed as being low. These results show that the food source of the blood cockle farms in the northwest area of Yeoja Bay seems to be abundant in quantity, but this is considered to be very poor in quality.

  • We experimented on growth in light and production of hydrogen and organic matters in dark fermentation by using C. reinhardtii. In the light, growth rate of C. reinhardtii following $CO_2$ fixation was proportional to consumption rate of nitrogen source. And the starch in cell was accumulated more when the period of culture was lengthened more. But the accumulation rate of starch in cell was decreased when the growth rate of cell become dull. In the dark fermentation, the production volume and production rate of hydrogen were the highest value in the mid exponential state among other states. The utilization efficiency of substrate was better in the early exponential state than other states. In production of organic matters, acetic acid didn't change remarkably and ethanol showed the highest value in early exponential state.

Distribution and Cyclings of Nutrients in Phragmites communis Communities of a Coastal Salt Marsh (해안염습지 갈대 군락의 무기영양소 순환과 분배)

  • The aboveground production, nutrient distribution and nutrient cyclings were compared between two Phragmites communis communities growing in the different salt contents of soil in a coastal salt marsh. Inorganic nutrient contents of soil for plant growth were greater at the low salt stand than at the high salt stand except for sodium(Na). Maximum aboveground biomass of the plant at the low and the high salt stands were 2,533 and 1,719 g dw/$m^2$, respectively, in August. Seasonal changes of nutrient content of biomass in dry weight decreased with growth except for Na. Nutrient contents in biomass per unit land area increased continuously as biomass increases, although the amount of potassium(K) reached the maximum content in July and thereafter decreased. Vertical distributions of total nitrogen(T-N) and phosphorus(P) increased with plant height, but Na showed the reverse trend. That of K was similar to the patterns for T-N and P in the leaves, and to the pattern of Na in the stems. The Na was greatly accumulated in underground biomass but transported scarcely to aboveground. At the low and the high salt stands, the ratios of the inorganic nutrients contained in the plant were 100 : 66 for T-N, 100 : 61 for P, 100 : 62 for K and 100 : 97 for Na. the ratios of the amounts of nutrients retrieved to soil were 100 : 242 for T-N, 100 : 408 for P, 100 : 127 for K and 100 : 269 for Na, respectively. Turnover times of the T-N, P, K and Na in the communities were 56, 1, 15 and 174 years at the low salt stand, and 75, 2, 24 and 323 years at the high salt stand, respectively. In nutrient cyclings, all of the nutrients retrieving to soil were less than uptake by plant. Among the nutrient, especially P is expected to be exhausted from soil, sooner or later, because of the harvest by men.

Effects of garlic on the cadmium accumulation in the tissue on the hematological picture in dogs (마늘이 개의 조직내 카드뮴축적 및 혈액상에 미치는 영향)

  • The environmental pollution by heavy metals has become a significant hygenic problem. Recently it is said the garlic supplementation in diet have protective effect on the heavy metal accumulation in tissue of rat by a researcher. In this study two experiments were undertaken. Firstly, dogs were fed diet supplemented with cadmium of cadmium plus 7% of raw garlic for 50 days. on the 50th day of experiment all dogs were euthanasied and the cadmium content of various tissue were measured by atomic absorption spectrophotometeric method. Secondly, in order to find out the toxic effect of garlic dogs were fed diet supplemented with 7% of raw garlic for 50 days and hematological or hematochemical observations were undertaken at 10 day intervals. The results obtained from these experiments were summarized as follows; 1. In dogs treated with cadmium and cadmium plus garlic cadmium was accumulated in renal cortex, liver, and muscle. However the concentrations of cadmium showed no significant between the two group. 2. In dogs fed diets supplemented with 7% of raw garlic significant decrease of erythrocyte count, packed cell volume and hemoglobin content and significant increase of Heinz body erythrocyte and reticulocyte count were recognized. 3. The garlic-treated dogs and control dogs had normal asparate aminotransferase, alanine aminotransferase, alkaline phosphotase, blood urea nitrogen value. The value showed no significant difference between the two group. 4. From these results it may be concluded that the garlic have protective effect for cadmium accmulation in tissue and excess garlic in diet could induce hemolytic anemia in dogs.

Nutrient production from Korean poultry and loading estimations for cropland

  • Background: Poultry breeding has increased by 306% in Korea, inevitably increasing the production of manure which may contribute to environmental pollution. The nutrients (NP) in the manure are essential for crop cultivation and soil fertility when applied as compost. Excess nutrients from manure can be accumulated on the land and can lead to eutrophication. Therefore, a nutrient load on the finite land should be calculated. Methods: This study calculates the nutrient production from Korean poultry by investigating 11 broiler and 16 laying hen farms. The broiler manure was composted using deep litter composting while for layer deep litter composting, drying, and simple static pile were in practice. The effect of weight reduction and storing period during composting was checked. Three weight reduction cases of compost were constructed to calculate nutrient loading coefficients (NLCs) using data from; i) farm investigation, ii) theoretical P changes (${\Delta}P=0$), and iii) dry basis. Results: During farm investigation of broiler and layer with deep litter composting, there was a 68 and 21% N loss whereas 77 and 33% P loss was found, respectively. In case of layer composting, a loss of 10-56% N and a 52% P loss was observed. Drying manure increased the P concentrations therefore NLCs calculated using dry basis that showed quite higher reductions (67% N; 53% P). Nutrient loss from farm investigation was much higher than reported by Korean Ministry of Environment (ME). Conclusions: Nutrients in manure are decreased when undergo storing or composting process due to microbial action, drying, and leaching. The nutrient load applied to soil is less than the fresh manure, hence the livestock manure management and conservation of environment would be facilitated.

The Relation between Fertilization Practices and Functional Metabolites of Crops: A Review

  • Various researches on the effects of fertilization levels on functional metabolites in crop have been conducted. This review summarizes the previous studies on the relation between fertilization supply and accumulation of metabolites (phenolics, carotenoids, ascorbic acid and glucosinolates) which function as antioxidants in crop. The accumulation of phenolic compounds is related to the activation of phenylalanine ammonia lyase (PAL) in phenylpropanoid pathway. Most of the previous studies discuss that low nitrogen (N) supply activates PAL, thereby increasing the synthesis of phenolics. Similarly, high N supply leads to a decrease in ascorbic acid because of the shading effect derived from the accelerated vegetative growth under high N level. Unlike the phenolics and ascorbic acid, carotenoids are accumulated with increasing N supply. In this regard, the previous studies explain that N is a main element closely associated with formation of key enzyme for the synthesis of carotenoids. Glucosinolates are generally increased under decreasing N supply and increasing S supply. Although the previous studies show similar trends about the accumulation of metabolites by nutrient level, they also suggest that many other factors including crop types, cultivars, cultural environment (water, temperature, light, etc.) influence the accumulation of functional metabolites in crop.

Property Changes of Vertical Soil Layers Planted with Zelkova serrata(Thunberg) Makino on the Reclaimed Land from the Sea in the Gwangyang Bay, Korea (광양만 임해 매립지 느티나무 식재지 토양의 수직적 특성 변화)

  • This study was carried out to elucidate the vertical characteristics of soil properties at six planted sites of land reclaimed from the sea, in Gwangyang Bay, Jeollanam-do Province, Korea. Based on the types of planting site, the chemical properties of the vertical soil layers varied. The vertical variation was great in the planting sites $Z_1\;and\;Z_2$, but less varied in the mounded planting sites $Z_3,\;Z_5,\;and\;Z_6$. Major reasons for the vertical variation in soil chemical properties included differences in the accumulation of organic matter, soil disturbance by heavy construction equipment, and heterogeneity of soil properties between soil horizons. As soil depths increased, soil salts varied. The electrical conductivity (ECe) increased in the lower areas of planting sites $Z_1\;and\;Z_2$, and the disturbed, saline planting site $Z_3$, but decreased in the lower areas of $Z_3,\;Z_5,\;and\;Z_6$. These tendencies did not coincided with exchange cation concentrations $(Na^+,\;K^+,\;Mg^{++},\;Ca^{++})$. Both total carbon (T-C) and total nitrogen (T-N) accumulated more in the lower areas of planting sites than in the higher areas, and levels were higher closer to the surface than in the soil depths. It is supposed that these tendencies are related to the accumulation of fallen leaves or other organic matter at the soil surface, and the soil chemicals then slowly move downward from the surface. Impediments to tree growth included soil hardiness, high soil salinity and exchangeable cation concentration, low soil moisture content, acidic or alkaline soil, low organic matter, heterogeneity of soil texture and establishment of soil stratification.

Biological Perchlorate Reduction in Municipal Sewage (도시하수에서 생물학적 퍼클로레이트의 환원)

  • This research was done to evaluate the potential for destruction of perchlorate in municipal sewage. Laboratory experiments were conducted in flasks containing 3 liters of raw sewage. Sewage was mixed with defined amount of perchlorate and various additives. Perchlorate reduction in sewage did occur, but was quite variable, ranging from 0 to 72% over 72 hour. Addition of even a small amount of perchlorate acclimated biomass (167 mg/L SS) significantly reduced the lag and resulted in complete perchlorate removal. Perchlorate reduction in sewage-brine mixtures was inhibited when the dissolved oxygen level was greater than 2 mg/L, and when the mixture salinity was relatively high (conductivity = 14 mS with equivalent TDS = 8 g/L). When nitrate ($NO_3{^-}$) was present with perchlorate in the laboratory flask tests of sewage-brine mixtures, nitrate reduction proceeded first. A significant amount of nitrite ($NO_2{^-}$) accumulated in the sewage-brine mixtures, accounting for about 66% of initial nitrate nitrogen ($NO_3$-N).

Thiamin Requirements for Vegetative Growth and Fruit Body Formation of Lentinula edodes

  • The effects of thiamin on vegetative mycelial growth and fruit body formation of Lentinuia edodes were investigated in basal peptone-glucose liquid medium in relation to the uptake of thiamin. Thiamin was essential for fruit body formation, and the minimum requirements for thiamin were estimated to be approximately 10 ${\mu}g$/L. The vegetative mycelial growth was little influenced by the addition of thiamin in the range of 1.5 ${\mu}g$~1.5 mg/L. While the mycelium was successively transferred to fresh peptone-glucose-agar medium three times, the repression of mycelial growth was not significant. Even in cases using vitamin-free casamino acid or glutamic acid as a nitrogen source instead of peptone, a thiamin deficiency for mycelial growth did not occur as a result of transferring the mycelia to fresh media. Almost all of the thiamin contained in the media accumulated in the mycelia during the first 3 weeks of a 9-week incubation. These results suggest that only trace amounts of thiamin are required for vegetative mycelial growth in Lentinula edodes and that almost all thiamin added to a basal medium will be used for fruit body formation.
