Molecular Characterization of 170 New gDNA-SSR Markers for Genetic Diversity in Button Mushroom (Agaricus bisporus)

  • We designed 170 new simple sequence repeat (SSR) markers based on the whole-genome sequence data of button mushroom (Agaricus bisporus), and selected 121 polymorphic markers. A total of 121 polymorphic markers, the average major allele frequency (MAF) and the average number of alleles (NA) were 0.50 and 5.47, respectively. The average number of genotypes (NG), observed heterozygosity (HO), expected heterozygosity (HE), and polymorphic information content (PIC) were 6.177, 0.227, 0.619, and 0.569, respectively. Pearson's correlation coefficient showed that MAF was negatively correlated with NG (-0.683), NA (-0.600), HO (-0.584), and PIC (-0.941). NG, NA, HO, and PIC were positively correlated with other polymorphic parameters except for MAF. UPGMA clustering showed that 26 A. bisporus accessions were classified into 3 groups, and each accession was differentiated. The 121 SSR markers should facilitate the use of molecular markers in button mushroom breeding and genetic studies.

Use of Near-Infrared Spectroscopy for Estimating Lignan Glucosides Contents in Intact Sesame Seeds

  • Near-infrared spectroscopy(NIRS) was used to develop a rapid and efficient method to determine lignan glucosides in intact seeds of sesame(Sesamum indicum L.) germplasm accessions in Korea. A total of 93 samples(about 2 g of intact seeds) were scanned in the reflectance mode of a scanning monochromator, and the reference values for lignan glucosides contents were measured by high performance liquid chromatography. Calibration equations for sesaminol triglucoside, sesaminol($1{\rightarrow}2$) diglucoside, sesamolinol diglucoside, sesaminol($1{\rightarrow}6$) diglucoside, and total amount of lignan glucosides were developed using modified partial least square regression with internal cross validation(n=63), which exhibited lower SECV(standard errors of cross-validation), higher $R^2$(coefficient of determination in calibration), and higher 1-VR(ratio of unexplained variance divided by variance) values. Prediction of an external validation set(n=30) showed a significant correlation between reference values and NIRS estimated values based on the SEP(standard error of prediction), $r^2$(coefficient of determination in prediction), and the ratio of standard deviation(SD) of reference data to SEP, as factors used to evaluate the accuracy of equations. The models for each glucoside content had relatively higher values of SD/SEP(C) and $r^2$(more than 2.0 and 0.80, respectively), thereby characterizing those equations as having good quantitative information, while those of sesaminol($1{\rightarrow}2$) diglucoside showing a minor quantity had the lowest SD/SEP(C) and $r^2$ values(1.7 and 0.74, respectively), indicating a poor correlation between reference values and NIRS estimated values. The results indicated that NIRS could be used to rapidly determine lignan glucosides content in sesame seeds in the breeding programs for high quality sesame varieties.

Identification of New Source of Resistance to Powdery Mildew of Indian Mustard and Studying Its Inheritance

  • Powdery mildew of Indian mustard (Brassica juncea), caused by Erysiphe cruciferarum, is emerging as major problem in India. All the Indian mustard cultivars presently grown in India are highly susceptible to powdery mildew and so far no resistance source has been reported. In this study, with an aim to identify resistant source, 1,020 Indian mustard accessions were evaluated against E. cruciferarum PMN isolate, at Wellington, The Nilgiris, Tamil Nadu, India under natural hot spot conditions. The study identified one accession (RDV 29) with complete resistance against E. cruciferarum PMN isolate for the first time, which was consistent in five independent evaluations. Genetic analysis of F1, F2 and backcross populations obtained from the cross RSEJ 775 (highly susceptible) × RDV 29 (highly resistant) for two season revealed that the resistance is governed by two genes with semi-dominant and gene dosage effect. Further, a new disease rating system using six scales (0, 1, 2, 3, 4, and 5) has also been proposed in this study to score powdery mildew based on progress of fungal growth in different plant parts of the F2 population. The outcome of this study viz. newly identified powdery mildew-resistant Indian mustard accession (RDV 29), information on inheritance of resistance and the newly developed disease rating scale will provide the base for development of powdery mildew-resistant cultivars of Indian mustard.

배추 유전자원의 항산화 효과 (Antioxidant Activity of Chinese Cabbage (Brassica rapa L. ssp. pekinensis) using Different Genotypes)

  • This experiment was conducted to assess the high antioxidant activity varieties of Chinese cabbage (Brassica rapa L. ssp. pekinensis) from the 55 accessions. The antioxidant activity of Chinese cabbage were determined by the TPC, TFC, DPPH, ABTS, and chlorophyll, carotenoid contents. The TPC and TFC showed a range of 1.21~4.61 mg GAE/g DW, 0.18~3.09 mg CE/g DW. The DPPH and ABTS assay were in the range of 0.65~4.36 and 1.42~6.91 mg ascorbic acid equivalent (ASCE)/g DW, respectively. The UPLC analysis was performed quantitatively to identify chlorophyll and carotenoid in the Chinese cabbage extract. The levels of the total chlorophyll and total carotenoid were 86.60~1,235.91, and 75.86~490.11 ㎍/g, respectively. The comprehensive differences in the total and individual chlorophyll contents have also been observed among different varieties. These results will be valuable as basic data for the standardization of Chinese cabbage.

Phytonutrient Profile of Purple Perilla (Perilla frutescens var. crispa) Seeds

  • To characterize phytonutrients, the seeds of 12 purple Perilla (Perilla frutescens var. crispa) accessions collected from Korea and Japan were used for quantitative analysis of tocopherols, phytosterols, squalene and fatty acids. The average tocopherol, squalene and phytosterols contents were 12.2 mg $100g^{-1}$, 3.99 mg $100g^{-1}$ and 77.20 mg $100g^{-1}$, respectively. Among 4 tocopherol (T) isomers (${\alpha}$-T, ${\beta}$-T, ${\gamma}$-T, and ${\delta}$-T), ${\gamma}$-T was present in the highest quantity (11.03 mg $100g^{-1}$) with the least variation (CV = 13.7%), while ${\beta}$-T was present in lowest quantity (0.25 mg 1$100g^{-1}$). Compared to campesterol (4.36 mg $100g^{-1}$) and stigmasterol (13.32 mg $100g^{-1}$), ${\beta}$-sitosterol exhibited higher quantity (59.51 mg $100g^{-1}$) with 9.5% of variation. The major fatty acids were unsaturated fatty acids such as linolenic (61.5%), linoleic (17.3%), and oleic (9.9%) acids compared to saturated ones: palmitic (7.6%) and stearic (3.7%) acids. When Korean and Japanese accession were compared, almost no difference in content could be observed, while more variation as evaluated by CV (%) could be observed in Japanese accession in most phytonutrients suggesting wider genetic variation of purple Perilla in Japan. Presence of all above-mentioned phytonutrient compounds strongly suggested health beneficial value of purple Perilla seeds.

Isoform-Specific Responses of Superoxide Dismutase to Oxidative Stresses and Hormones in Parquat-Tolerant Rehmannia glutinosa

  • All accessions of Rehmannia glutinosa show the unique characteristic of intrinsic tolerance to paraquat. The higher level of endogenous superoxide dismutase(SOD) activity and its increase upon paraquat treatment indicated the involvement of SOD in the tolerance mechanism to paraquat in R. glutinosa. In this study, we examined the isoform-specific response of SOD to oxidative stresses and hormones. Six SOD isoforms were found in the leaf, and they were identified as two MnSODs(named MnSOD I and MnSOD II, in order of increasing mobility), one FeSOD and three Cu/ZnSODs(named Cu/ZnSOD I, Cu/ZnSOD II, and Cu/ZnSOD III, in order of increasing mobility). MnSOD I, MnSOD II, FeSOD, Cu/ZnSOD I, Cu/ZnSOD II, and Cu/ZnSOD III, contributed to 4, 11, 7, 15, 30, and 32% of the total SOD activity, respectively. Total SOD activity levels in the leaf were increased by 4, 24, and 21% by paraquat, salicylic acid(SA), and yeast extract(YE), respectively, but little by ethephon. Six SOD isoforms responded differentially to these stresses and hormones. The activities of all the isoforms were increased by YE and SA except that of MnSOD I which was decreased by SA. The activities of MnSOD I, FeSOD, and CuZnSOD I were increased by paraquat. These results suggest that amelioration of oxidative stresses by SOD is fine-tuned by the differential expression of isoforms in R. glutinosa.

Utilization of Elite Korean Japonica Rice Varieties for Association Mapping of Heading Time, Culm Length, and Amylose and Protein Content

  • Association mapping is widely used in rice and other crops to identify genes underlying important agronomic traits. Most association mapping studies use diversity panels comprising accessions with various geographical origins to exploit their wide genetic variation. While locally adapted breeding lines are rarely used in association mapping owing to limited genetic diversity, genes/alleles identified from elite germplasm are practically valuable as they can be directly utilized in breeding programs. In this study, we analyzed genetic diversity of 179 rice varieties (161 japonica and 18 Tongil-type) released in Korea from 1970 to 2006 using 192 microsatellite markers evenly distributed across the genome. The 161 japonica rice varieties were genetically very close to each other with limited diversity as they were developed mainly through elite-by-elite crosses to meet the specific local demands for high quality japonica rice in Korea. Despite the narrow genetic background, abundant phenotypic variation was observed in heading time, culm length, and amylose and protein content in the 161 japonica rice varieties. Using these varieties in association mapping, we identified six, seven, ten, and four loci significantly associated with heading time, culm length, and amylose and protein content, respectively. The sums of allelic effects of these loci showed highly significant positive correlation with the observed phenotypic values for each trait, indicating that the allelic variation at these loci can be useful when designing cross combinations and predicting progeny performance in local breeding programs.

Evaluation of QTL Related SSR Marker Universality in Korean Rice Breeding Populations

  • If a quantitative trait loci (QTL) marker identified in a population is applicable to different populations (marker universality), this will not only reduce the labor and cost in marker assisted selection (MAS), but accelerate the application of molecular markers to real breeding programs. Present study aims to evaluate the defined QTL related markers from a population to a different breeding population for the MAS. Four rice breeding populations were subjected to seventy-five simple sequence repeat (SSR) markers which were already identified for their polymorphism information content (PIC) in the parents of the crossings. Among them, eight markers were evaluated for their correlation between presence of marker alleles and phenotypic expression in breeding populations. A reasonable level of polymorphism for the mapped markers originated from any sources of rice accessions was observed between crosses of any sources (marker repeatability). However, correlation between presence of markers and expression of the traits in rice breeding populations was not significant except for minor portion of traits and markers examined (failure of marker universality). In the present study, various strategies were discussed to develop new markers with universality of breeding application.

RAPD를 이용한 고추(Capsicum annuum) 유전자원의 분류 (Classification of Capsicum annuum Germplasm Using Random Amplified Polymorphic DNA)

  • 본 연구는 RAPD표지를 이용하여 국내외에서 수집된 고추 유전자원들간의 유전적관계를 평가하고자 수행되었다. Random primer를 이용한 고추의 PCR반응은 $MgCl_2$ 3mM, Taq. DNA polymerase 1.5U, 주형 DNA 10ng, dNTPs $200{\mu}M$, random primer 200nM 그리고 $42^{\circ}C$의 annealing 온도조건으로 최적화하였다. 80개의 random primer로부터 높은 밴드선명도와 재현성을 보이는 16개가 선발되었으며 70%의 GC함량을 갖는 primer들이 GC함량이 60%인 것보다 DNA증폭에 있어 효과적이었다. 31개의 고추품종및 계통들에 대해 71개의 polymorphic밴드와 22개의 monomorphic밴드를 포함하는 총 93개의 DNA밴드가 선발된 16개의 random primer들로부터 형성되었다. Primer당 약 4.4개의 polymorphic밴드가 형성되었다. 이들 71개의 polymorphic밴드를 이용하여 유사도지수가 구해졌으며 이를 근거로 31고추 계통 또는 품종들을 뚜렷이 구분하는 dendrogram이 작성되었다.

Changes in Cell Ca2+ Distribution in Loquat Leaves and Its Effects on Cold Tolerance

  • Calcium has been associated with improved cold tolerance in many crops. The aim of this study was to investigate the changes in leaf cell $Ca^{2+}$ distribution and cell organelle ultrastructure of loquat (Eriobotrya japonica Lindl.) plants in response to cold stress at $-3^{\circ}C$, using transmission electron microscopy (TEM). Two loquat accessions, Zaozhong 6 (a commercial cultivar) and oakleaf loquat (a wild relative) were used. Cold tolerance, as measured by leaf browning rate, was higher in oakleaf plants, and calcium treatment improved cold tolerance in both species. Cold stress first induced inward transport of $Ca^{2+}$ from the intracellular space. Then, the imported $Ca^{2+}$ was aggregated around the chloroplast membrane, finally entering the chloroplast. This pattern of $Ca^{2+}$ distribution in leaf cells occurred earlier in Zaozhong 6 than in the wild loquat. With increasing time of cold exposure, the chloroplast membranes of Zaozhong 6 leaves were damaged, blurred and even disappeared, while those of wild oakleaf loquat leaves maintained their structure longer. In Zaozhong 6, cold stress induced a clear cavity between poorly structured granal thylakoids and vesicles appearing inside the chloroplast, while in oakleaf leaves cold stress had little effect on the ultrastructure of chloroplasts (although chloroplast membranes looked blurred). Loquat leaves accumulated free calcium ions around chloroplasts in response to cold stress, with earlier calcium accumulation occurring in the cold-sensitive cultivar Zaozhong 6 than in wild oakleaf loquat. These results demonstrate that these two loquat species have differences in both cold tolerance and calcium accumulation dynamics.
