Increased Expression of aac(3)II by Tn3 in Gentamicin - Resistant Bacteria Isolated from Hospital Sewage (병원하수로부터 분리한 Gentamicin 저항성 세균에서 Tn3에 의한 aac(3)II의 발현 증가)

  • We tested gentamicin - resistant bacteria isolated from hospital sewage to confirm the presence of aac(3)II encoding aminoglycoside- (3)-N- acetyltransferase by dot-blot hybridization. A probe from the internal fragment of aac(3)II was hybridized to DNA from 41 % (39/95) of gentamicin resistant isolates. PCR was performed with primers from aac(3)II and Tn3. Of 39 strains, 13 strains had Tn3-aac(3)II structure. Minimal inhibitory concentration (MIC) test demonstrated that 18 strains containing Tn3-aac(3)II showed higher resistance to gentamicin than those of other strains. Thirteen strains were identified as 5 Escherichia coli, 3 Acinetobacter johnsonii, 2 Enterobacter agglomerans, 2 Micrococcus luteus, and 1 Pseudomonas facilis. These results suggest that gentamicin-resistant determinant of Tn3-aac(3)II structure was widely distributed in the gentamicin-resistant bacteria.

A study on the Perceptual Model for MPEG II AAC Encoder (MPEG-II AAC Encoder의 perceptual Model에 관한 연구)

  • Currently, the most important technology is the compression methods in the multimedia society. Audio files are rapidly propagated through internet. MP-3 is offered to CD tone quality in 128Kbps, but 64Kbps below tone quality is abruptly down and high bitrate. on the other hand, MPEG-II AAC (Advanced Audio Coding) is not compatible with MPEG-I, but AAC has a high compression ratio 1.4 better than MP-3. Especially, AAC has max. 7.1 channel and 96KHz sampling rate. In this paper, the perceptual model is dealt with 44.1KHz sampling rate for SMR(Signal to Masking Ratio)

Spreading of Aminoglycoside-Modifying Enzymes among Escherichia coli Isolated from Clinical Specimens in Chungcheong Province (충청지역의 임상검체로부터 분리된 대장균에 Aminoglycoside-Modifying Enzymes 확산)

  • The antimicrobial resistance rate in bacteria has increased over the last several decades. The transfer of antimicrobial resistant determinants on mobile genetic elements could cause the accelerated emergence and spread of multidrug resistant bacteria. This study investigated the aminoglycoside resistance determinants transferred by mobile genetic elements in a total of 33 aminoglycoside non-susceptible E. coli isolated from clinical specimens in Chungcheong province. 16S ribosomal RNA methyl-transferases (RMTases) and aminoglycoside-modifying enzyme (AME) genes were detected via PCR and DNA sequencing. The most common AME genes were aac(3')-II gene (54.5%), followed by aph(3')-Ia (18.2%) and aac(6')-Ib (15.2%). None of the evaluated RMTase genes were detected in the 33 isolates. Seventeen of the 18 isolates harboring aac(3')-II gene were resistant to gentamicin, and 16 of them were resistant to tobramycin. The 5 isolates harboring aac(6')-Ib gene were all resistant to tobramycin. In this study, we confirmed that one of the important mechanisms of aminoglycoside resistance in E. coli isolated from human is the acquisition of AME genes. Continuing investigations of antimicrobial resistant determinants in bacteria isolated from human may be required to prevent dissemination of antimicrobial resistant bacteria.

Molecular Biological Detection of the Genes Encoding Aminoglycosise Acetyltransferases and Aerolysin in Water Samples from Juam Lake (주암호에서 Aminoglycoside Acetyltransferases와 Aerolysin 유전자의 분자생물학적 검출)

  • The aacCl, aacC2, aacC3, and aacC4 genes, which encode aminoglycoside acetyltransferase AAC(3)-I, AA(3)-II, AAC(3)-III, and AAC(3)-IV, respectively, and aerolysin genes in water samples from Juam lake were surveyed by polymerase chain reaction. Surface water was collected from January 1996 to December 1998, and then bacterial DNA was extracted from the water. Twelve samples were tested by PCR to servey the genes for aminoglycoside acetyltransferase and aerolysin in the lake water. The aacC2 gene was detected in 9 of 12 DNA samples. Among 9 samples showing aacC2 positive, 7 samples were associated with Tn3 sequence. However, none of the twelve samples amplified the expected DNA fragment for aacC1, aacC3, and aacC4 genes. PCR primer to detect the aerolysin gene was designed using the conserved region of the genes for aerolysin and hemolysin of Aeromonas spp. This primer set successfully amplified the expected 414 bp PCR product with the DNA samples from the lake water. The aerolysin gene was detected in 7 of 12 DNA samples. When Southern hybridization of the gel with probe was performed, the aerolysin gene was detected in 10 of 12 DNA samples. However, the seasonal fluctuation of these genes was not found.

  • Cellulose grafted with acrylic acid(AAc) was prepared by radiation grafting technique. The effects of reaction temperature. reaction time, monomer concentration, and the crosslinkers on the AAc grafting reaction on cellulose were examined. The amount of AAc grafted on the cellulose reached maximum at the concentration of 0.75vol% difunctional crosslinker and 1.0vol% trifunctional crosslinker, respectively. In the presence of acid, the amount of AAc grafted on the cellulose was decreased when reaction solution contains difunctional crosslinker, while that was increased when reaction solution contains trifunctional crosslinker. In the grafting reaction of cellulose with AAc and TMETA, mixture containing ferrous sulfate and acid enhanced further AAc grafting yield than mixture containing ferrous sulfate only.

Hydrogen sulfide ameliorates abdominal aorta coarctation-induced myocardial fibrosis by inhibiting pyroptosis through regulating eukaryotic translation initiation factor 2α phosphorylation and activating PI3K/AKT1 pathway

  • This study aimed to assess the effects of exogenous hydrogen sulfide (H2S) on abdominal aorta coarctation (AAC) induced myocardial fibrosis (MF) and autophagy in rats. Forty-four Sprague-Dawley rats were randomly divided into control group, AAC group, AAC + H2S group, and H2S control group. After a model of rats with AAC was built surgically, AAC + H2S group and H2S group were injected intraperitoneally with H2S (100 µmol/kg) daily. The rats in the control group and the AAC group were injected with the same amount of PBS. We observed that H2S can improve left ventricular function and the deposition of myocardial collagen fibers, inhibit pyroptosis, down-regulate the expression of P-eif2α in myocardial tissue, and inhibit cell autophagy by activating the phosphatidylinositol 3-kinase (PI3K)/AKT1 signaling pathway (p < 0.05). In addition, angiotensin II (1 µM) H9c2 cardiomyocytes were injured in vitro experiments, and it was also observed that pyroptosis was inhibited after H2S (400 µmol/kg) intervention, the expression of P-eif2α in cardiomyocytes was significantly down-regulated, and the PI3K/AKT1 signaling pathway was activated at the same time. Therefore, increasing the expression of P-eif2α reverses the activation of the PI3K/AKT1 signaling pathway by H2S. In conclusion, these findings suggest that exogenous H2S can ameliorate MF in rats with AAC by inhibiting pyroptosis, and the mechanism may be associated with inhibiting the phosphorylation of eif2α and activating the PI3K/AKT1 signaling pathway to inhibit excessive cell autophagy.

In this paper, we designed the optimized MDCT (Modified Discrete Cosine Transform) that could decrease the capacity of enormous computation and could increase the processing speed in the MPEG-2 AAC encoder.

  • Currently, the compression is one of the most important technology in multimedia society. Audio files arc rapidly propagated throughout internet Among them, the most famous one is MP-3(MPEC-1 Laver3) which can obtain CD tone from 128Kbps, but tone quality is abruptly down below 64Kbps. MPEC-II AAC(Advanccd Audio Coding) is not compatible with MPEG 1, but it has high compression of 1.4 times than MP 3, has max. 7.1 and 96KHz sampling rate. In this paper, we propose an algorithm that decreased the capacity of AAC encoding computation but increased the processing speed by optimizing psychoacoustic model which has enormous amount of computation in MPEG 2 AAC encoder. The optimized psychoacoustic model algorithm was implemented by C++ language. The experiment shows that the psychoacoustic model carries out FFT(Fast Fourier Transform) computation of 3048 point with 44.1 KHz sampling rate for SMR(Signal to Masking Ratio), and each entropy value is inputted to the subband filters for the control of encoder block. The proposed psychoacoustic model is operated with high speed because of optimization of unpredictable value. Also, when we transform unpredictable value into a tonality index, the speed of operation process is increased by a tonality index optimized in high frequency range.

Kinetic Modeling for Biosorption of Metylene Blue onto H3PO4 Activated Acacia arabica

  • Batch sorption experiments were carried out for the removal of metylene blue from its aqueous solution using $H_3PO_4$ activated Acacia arabica carbon (AAC). The prepared activated carbon was characterized and was found as an effective adsorbent material. The operating variables studied were initial metylene blue concentration, AAC concentration and solution pH. AAC activated carbon posses a maximum sorption capacity for the range of initial dye concentrations studied (60~100 mg $L^{-1}$). The sorption kinetics were analyzed using reversible first order kinetics, second order, reversible first order, pseudo-first order, and pseudo-second order model. The sorption data tend to fit very well in pseudo-second order model for the entire sorption time. The average pseudo-second order rate constant, $K_{II}$ and regression coefficient value were determined to be 0.0174 mg $g^{-1}$ $min^{-1}$ and 0.9977. The biosorption process also fit well to reversible I order kinetics with a regression coefficient of 0.9878.

Preliminary Products of Precise Orbit Determination Using Satellite Laser Ranging Observations for ILRS AAC

  • In this study, we present preliminary results of precise orbit determination (POD) using satellite laser ranging (SLR) observations for International Laser Ranging Service (ILRS) Associate Analysis Center (AAC). Using SLR normal point observations of LAGEOS-1, LAGEOS-2, ETALON-1, and ETALON-2, the NASA/GSFC GEODYN II software are utilized for POD. Weekly-based orbit determination strategy is applied to process SLR observations and the post-fit residuals check, and external orbit comparison are performed for orbit accuracy assessment. The root mean square (RMS) value of differences between observations and computations after final iteration of estimation process is used for post-fit residuals check. The result of ILRS consolidated prediction format (CPF) is used for external orbit comparison. Additionally, we performed the precision analysis of each ILRS station by post-fit residuals. The post-fit residuals results show that the precisions of the orbits of LAGEOS-1 and LAGEOS-2 are 0.9 and 1.3 cm, and those of ETALON-1 and ETALON-2 are 2.5 and 1.9 cm, respectively. The orbit assessment results by ILRS CPF show that the radial accuracies of LAGEOS-1 and LAGEOS-2 are 4.0 cm and 5.3 cm, and the radial accuracies of ETALON-1 and ETALON-2 are 30.7 cm and 7.2 cm. These results of station precision analysis confirm that the result of this study is reasonable to have implications as preliminary results for administrating ILRS AAC.
