Synthesis of Fe­Garnet for tile Immobilization of High Level Radioactive Waste (고준위 방사성폐기물의 고정화를 위한 Fe­석류석 합성 연구)

  • Garnet has been considered as a possible matrix for the immobilization of radioactive actinides. It is expected that Fe­based garnet be able to have the high substitution ability of actinide elements because ionic radius of Fe in tetrahedral site is larger than that of Si of Si­based garnet. Accordingly, we synthesized Fe­garnet with the batch composition of $Ca_{2,5}$C $e_{0.5}$Z $r_2$F $e_3$ $O_{12}$ and $Ca_2$CeZrFeF $e_3$ $O_{12}$ and studied their phase relations and properties. Mixed samples were fabricated in pellet forms under the pressure of 400 kg/$\textrm{cm}^2$ and were sintered in the temperature range of 1100∼140$0^{\circ}C$ in atmospheric conditions. Phase identification and chemical composition of synthesized samples were analyzed by XRD and SEM/EDS. In results, where the compounds were sintered at 130$0^{\circ}C$, we optimally obtained Fe­garnets as the main phase, even though some minor phases like perovskite were included. The compositions of Fe­garnets synthesized from the batch compositions of $Ca_{2,5}$C $e_{0.5}$Z $r_2$F $e_3$ $O_{12}$ and $Ca_2$CeZrFeF $e_3$ $O_{12}$, are $Ca_{2.5­3.2}$C $e_{0.3­0.7}$Z $r_{1.8­2.8}$F $e_{1.9­3.2}$ $O_{12}$ and $Ca_{2.2­2.5}$C $e_{0.8­1.0}$Z $r_{1.3­1.6}$ F $e_{0.4­.07}$ F $e_{3­3.2}$ $O_{12}$, respectively. Ca contents were exceeded and Ce contents were exceeded or depleted in 8­coodinated site, comparing to the initial batch composition. These results were caused by the compensation of the difference of ionic radius between Ca and Ce.

Fluid Inclusions in Amethyst from the Korea Amethyst Deposit, Uljin, Gyeongbuk (경북 울진 코리아 광상의 자수정에 대한 유체포유물 연구)

  • Three distinct types of fluid inclusions in amethyst and quartz crystals are associated with metamorphic events in the Korea Amethyst deposit from Uljin-Gun, Gyeongbuk Province. The amethyst displays bimodal grain size distribution in fine-grained, strain-free equigranular quartz with coarse-grained quartz grains with kink bands and undulose extinction. Type I inclusions are liquid-rich and salinity is 0~7 wt% NaCl and the homogenization temperatures ($T_h$) $91{\sim}231^{\circ}C$ with eutectic temperatures ($T_e$) $-52{\sim}-20^{\circ}C$. Type II inclusions are vapor-rich (80~90 vol%). The salinity and $T_h$ ranges 3~6 wt% NaCl and $230{\sim}278^{\circ}C$, respectively with $T_e$ $-56{\sim}-23^{\circ}C$. Type III inclusions contain a daughter mineral other than NaCl. The salinity ranges 32~36 wt% NaCl and $T_h$ $210{\sim}271^{\circ}C$. The textural and fluid inclusion evidences suggest that the host Buncheon granite gneiss and Amethyst pegmatite experienced dynamic recrystallization and the studied fluid inclusions are metamorphic in origin. The metamorphic event possibly occurred at higher temperature than $271{\sim}278^{\circ}C$. The amethysts from Uljin Korea Amethyst can be distinguished from the synthetic amethyst on basis of the distinctive two and three-phases fluid inclusions. Furthermore, it is noticeable that Korea amethyst do not contain NaCl-bearing and $CO_2$-rich fluid inclusions unlike those compared to those from Eonyang and Samcheonpo deposits related to unmetamorphosed granitic rocks.

Size Dependent Analysis of Phytoplankton Community Structure during Low Water Temperature Periods in the Coastal Waters of East Sea, Korea (저수온기 동해연안의 식물플랑크톤 크기에 따른 군집구조)

  • In order to understand the phytoplankton community structure based on their cell size duringlow water temperature periods, we studied 10 stations in the East Sea, Korea on March, 2012. The minimum standing crops of total phytoplankton were $3.4{\times}10^6cells\;L^{-1}$ at the station 5. The maximum values were $7.6{\times}10^6cells\;L^{-1}$ at the station 8, which is two times the amount of the minimum. The carbon mass at the station 4 ($6.3{\times}10^8pg\;L^{-1}$) was more than forty times higher compared with station 5 ($0.08{\times}10^8pg\;L^{-1}$). From these results, we found a significant difference between standing crops and carbon mass which might have caused due to their differences in community structure and cell size. Therefore, we considered the types of plankton biomass to estimate the primary product in the specific location and/or time. The phytoplankton communities were classified in 3 types: microplankton (> $20{\mu}m$), nanoplankton (< $20{\mu}m$) and picoplankton (< $2{\mu}m$). In the case of picoplankton, various morphological types were observed during the study period. These various picoplankton species were further classified as S (spherical), SF (spherical&flagella), O (oval), OF (oval&flagella) or R (rod) type, and we analyzed their community structure based on these categories. The picoplankton was found to be the most dominant type at 8 stations and S type as the most popular. The picoplankton seems to be the significant organism in the marine ecology during low water temperature periods in the coastal waters of East Sea. Therefore, picoplankton \;-with scientific surveys can be considered as the database for their identification. In conclusion, we suggest that cell size of the phytoplankton would be the best criteria to accurately analyze their community structure and to reveal groups having more ecological influence.

Identification of Raw Materials in Processed Meat Products by PCR Using Species-Specific Primer (종 특이 프라이머를 이용한 식육가공품의 사용원료 판별법)

  • In this study, a method was developed using molecular biological technique to distinguish an authenticity of meats for processed meat products. The genes for distinction of species about meats targeted at 12S or 16S genes in mitochondrial DNA and the species-specific primers were designed by that PCR products' size was around 200bp for applying to processed products. The target materials were 10 species of livestock products and it checked whether expected PCR products were created or not by electrophoresis after PCR using species-specific primers. The results of PCR for beef, pork, goat meat, mutton, venison, and horse meat were 131, 138, 168, 144, 191, and 142 bp each. The expected PCR products were confirmed at 281, 186, 174, and 238 bp for chicken, duck, turkeymeat, and ostrich. Also, non-specific PCR products were not detected in similar species by species-specific primers. The method using primers developed in this study confirm to be applicable for composite seasoning including beefs and processed meat products including pork and chicken. Therefore, this method may apply to distinguish an authenticity of meats for various processed products.

Effect of Planting Date and Hybrid on Forage Yield and Quality of Corn for Silage. II. Fiber component and digestibility of ear, stover and whole plant (파종시기 및 품종이 사일리지용 옥수수의 수량과 사료가치에 미치는 영향 II. 경엽, 암이삭 및 전식물체의 섬유소 및 소화율)

  • Dry matter yield and ear percent are commonly considered the most important factors for evaluating silage corn(Zea mays L.) hybrids for silage production. But quality of stover as well as forage production in important in identification of hybrids. The objective of this study was to evaluate nutritional value of stover, ear and whole plant from eight corn hybrids for silage at two dates of planting. In content of fiber components(ADF, NDF, ADL, hemicellulose and cellulose) from stover, ear and whole plant samples, optimum planting corn was lower than late planting one(P<0.01). Differences(P<0.01) in the content of fiber components in stover and whole plant were observed among the corn hybrids. Differences between different planting dates in TDN and $NE_1$ were detected in the stover and whole plant samples(P<0.05). Differences among the corn hybrids in TDN and $NE_1$ were also observed in stover, ear and whole plant samples. TDN and $NE_1$ of 'G4624', 'P3352' and 'P3394' for whole plant were higher than those of the other corn hybrids. ln vitro dry matter digestibility(IVDMD) at optimum planting date was higher than that of late(P<0.05), and there also were significant differences among the corn hybrids. Correlation coefficients for whole plant IVDMD with NDF, ADF, hemicellulose, and cellulose were $-0.82^{**}$, $-0.71^{**}$, $-0.63^{**}$, and $-0.69^{**}$, respectively. Results of this study indicate that optimum planting of corn resulted in increased quality of silage corn. Differences among corn hybrid, particularly, in nutritive value of stover and whole plant were also observed. Therefore, the nutritional value of stover and whole plant may be important in assessing the corn hybrids for silage.

Reflection and perspective of the geomorphology in Korea (한국 지형학의 50년 회고와 전망)

  • In Korea, modern geomorphology has developed as one of main subjects in geography, such as in Europe. Geomorphology is one of the most advanced specialties in the geography dicipline, since foundation of Korean Geographical Society in 1945. Untill 1960's study, first generation of the Korean geomorphologists trained the younger ones, together with energetic research activities. Their great works in education and research established the base of ulterier development of the geomorphology in Korea. Since 1970s, research manpower and quality has incresed rapidly, partly due to the various international activities and cooperations of Korean geomorphologists. Owing to above development, Korean geomorphologist was able to found "The Geomorphological Association of Korea" in 1990 and publish "Journal of GAK", since 1994. Furthermore, geomorphologists are playing important roles in interdisciplinary academic societies, such as "The Korean Quaternary Assocition". Still 1960s, our research had focused on the identification and interpretation of erosional surfaces in Korea Peninsular. Of course, W.M. Davis's "Geographical Cycle Theory" and L.C. King's "Pedimentation Theory" had a great influence on the Koerans' works. After 1970s, the study of erosional surface played the important role in setting up the morphoclimatic viewpoint and methodology. Research scope tend to be notably broad and various than it was untill 1960's. Disposotion of the scientific methods and techniques become more and more apparent. These trends of research has settled precise descreption and interpretation of actual landforms, based on the careful field works, scientific measuring, and analisis, rather than methodology focused on the particular master theories. Recent geomorphological researches show the scope from climatic geomorphology and Quaternary geomorphology to granite and limestone weathering, pedo-geomorphogenic environment and periglacial landforms, focused on the small-to-medium scales. And then there have been new trying to interprete erosional surfaces such as hillslopes and terraces. Also, studies of coastal and plain landforms have been successfully developed. Recent new trends show the quantitative and analytic modelling using field measurement and laboratory work, and study on the human impacts on the natural landforms.y on the human impacts on the natural landforms.

Molecular Identification of Trichogramma (Hymenoptera: Trichogrammatidae) Egg Parasitoids of the Asian Corn Borer Ostrinia furnacalis, Based on ITS2 rDNA Sequence Analysis (ITS2 rDNA 염기서열 분석을 통한 Trichogramma 속(벌목: 알벌과)의 조명나방 알기생벌에 대한 종 추정)

  • To identify the species of Trichogramma occurring in the corn fields of Korea as egg parasitoids of Ostrinia furnacalis, we sequenced the full-length of ITS2 nuclear rDNA from 112 parasitoids collected during this study. As a reference to distinguish species, we also retrieved full-length ITS2 sequences of 60 Trichogramma species from the NCBI GenBank database. On the basis of the size and 3'terminal sequence pattern of the ITS2 sequences, the Trichogramma samples collected in this study were divided into three groups (K-1, -2, and -3). Evolutionary distances (d) within and between groups based on ITS2 sequences were estimated to be ${\leq}0.005$ and ${\geq}0.080$, respectively. In the net average distance between groups or species, the d value between K-1 and T. ostriniae, K-2 and T. dendrolimi, and K-3 and T. confusum was the lowest, with values of 0.016, 0.001, and 0.002, respectively. In the phylogenetic tree, K-1 and K-2 were clustered with T. ostriniae and T. dendrolimi, respectively. However, K-3 was clustered with three different species, namely, T. confusum, T. chilonis, and T. bilingensis. NCBI BLAST results revealed that parasitoids belonging to K-1 and K-2 showed 99% identity with T. ostriniae and T. dendrolimi, respectively. Parasitoids in K-3 collected from Hongcheon showed 99-100% identity with T. confusum and T. chilonis, and one parasitoid in K-3 collected from Gochang had 98% identity with T. bilingensis, T. confusum, and T. chilonis. On the basis of these results, we infer that the species of Trichogramma collected in this study are closely related to T. ostriniae (K-1) and T. dendrolimi (K-2). However, it was not possible to distinguish species of K-3 using the ITS2 sequence alone.

Effect of Irradiation, Packaging and Storage on the Oxidation of Cholesterol and Lipid in Pork Longissimus Meat (돈육 등심의 콜레스테롤 및 지질 산화에 전자선 조사, 포장 및 저장기간이 미치는 영향)

  • Pork loins that retailed in market were used as experimental samples. Some pork samples in raw state were packaged with PVDC in either aerobic or vacuum condition. The other pork samples were cooked until core temperature reached at 70$\^{C}$ and then packaged immediately in the same way with the raw samples. After these samples were irradiated by electron beam 6 kGy, the samples were stored in a refrigerator (2∼4$\^{C}$). Identification and quantification of cholesterol oxides were performed at 0 and 7 days. The results were following. During the early stage of storage, cholesterol oxides were not produced from the raw meat samples, but with the passage of storage time,7 $\alpha$-hydroxycholesterol, 7$\beta$-hydroxycholesterol, 7-ketocholesterol, 20 $\alpha$-hydroxycholesterol, $\beta$-epoxide and $\alpha$-epoxide, which were not produced during the early stage of storage, were produced. The production of cholesterol and lipid oxidation products were significantly higher (P<0.05) in the meats with aerobic packaging than those with vacuum packaging, Cooked meat after irradiation showed 7 $\alpha$-hydroxycholesterol, 7 $\beta$-hydroxycholesterol, $\alpha$-epoxide and cholestanetriol on the 7th day of storage, although those chemicals were not produced during the early stage of storage. Production of cholesterol oxides was significantly increased (P<0.05) with the passage of storage time for all treatments, and showed significantly lower value (P<0.05) with the vacuum packaging than these for aerobic packaging. Species of cholesterol oxides from irradiated meat after cooking were similar to those from cooked meat after irradiation. Collectively, it was found that the production of cholesterol oxides was more easily affected by packaging condition than irradiation.

Clinical Utility of Amplified Mycobacterium Tuberculosis Direct Test in the Diagnosis of Pulmonary Tuberculosis (폐결핵 잔단에서 Amplified Mycobacterium Tuberculosis Direct Test의 임상적 유용성)

  • Background: Acid-fast stain and cultures for diagnosis of pulmonary tuberculosis are primary and essential method, but have their limitation : low sensitivity and time consuming. The objective of this study is comparison of amplified Mycobacterium tuberculosis direct test(MTD) by the conventional AFB smears and cultures in the detection of Mycobacterium tuberculosis in respiratory specimens. Methods: During the period between November, 1997 and May, 1998 a total of 267 respiratory specimens (sputum 173, bronchial washing 94) from 187 patients suspected pulmonary tuberculosis were subjected to AFB smears, cultures and MID test. MID is based on nucleic acid amplification. We compared the MID with 3% Ogawa culture method. In positive AFB smear and negative MID specimen, positive culture identification between nontuberculous mycobacterium and M.tuberculosis was assesed by using Accuprobe M.tuberculosis complex probe. In negative AFB smear and negative AFB culture, MTD results are assessed by clinical follow-up. Results : 1) Compared with culture in sputum and bronchial fluid specimens, sensitivity and specificity of MTD in positive AFB smear is 79.7% and 20.0%, sensitivity and specificity of MTD in negative AFB smear specimens is 75.0% and 79.7%. 2) Discrepant analysis is assessed by clinical follow-up and other specimen results beyond study. Culture negative but MTD positive specimens were proved to be true positive and gave MTD sensitivity 79.2%, specificity of 84.4%, positive predictive value 80.5% and negative predictive value 83.2%. 3) 14 out of 31 specimens in negative AFB smear, negative AFB culture and positive MTD showed pulmonary tuberculosis diagnosed on clinical follow-up and sensitivity is 45.2%. 4) 2 out of 13 specimens in positive AFB smear, positive AFB culture and negative MID diagnosed as non tuberculous mycobacterium by Accuprobe culture. Conclusion: This study suggested that MID in respiratory specimens is simple and rapid diagnostic method, but considered adjuvant method rather than replace the conventional AFB smear and culture.

Improved Preservation Methods for Big and Old Tress in South Korea (우리 나라의 노거수자원(老巨樹資源) 보호관리실태(保護管理室態) 및 개선방안(改善方案))

  • This study was conducted in order to provide essential data and relevant management proposal to conserve and maintain big and old trees in a rational way. For the field survey, 77 big and old trees preserved by the laws in Chollabuk-do, Korea were investigated. The study results are summarized as follows : 1. To conserve and manage big and old trees, the valuable trees have been designated as natural monument trees and protection-needed trees. There are 141 individuals of 37 species designated as natural monuments and 10,049 individuals of 102 species designated as protection-needed trees. 2. Management budget for natural monument trees was devoted at 70% from the national expenditure, but that for protection-needed trees was devoted at 98% from the local expenditure. 3. Standardized sign boards and sign stones for natural monument trees were well placed and other protection facilities such as fences, branch supports and branch holdings were established. On the other hand, management of protection-needed trees was deficient overall. 4. Problems for designation process and management of protection-needed trees could include items such as insufficient management budget, various development activities, land ownership, misjudgement of tree age and species identification, unsatisfaction of sign board placement, insufficient surgery for damaged trees, pavement around tree root system and environmental pollution around the trees. 5. In order to improve the existing management methods of big and old trees, the following schemes were suggested : the development of practical criteria for natural monument and protection-needed trees, nationwide surveys of big and old tree resources, the security of national budget, securing sufficient spaces for the tree growth, specialization of management systems, extended practices of tree form management, establishment of permanent standard signs and consideration of opinions of village residents.
