Enzymatic Assessment of $2-Hydroxyethyl-{\beta}-undecenate$ Purified from Cumin (Cuminum cymium L.) Seed for Anti-periodontitis (Cumin(Cuminum cymium L.) seed로부터 정제한 $2-hydroxyethyl-{\beta}-undecenate$의 항치주염 효과의 효소학적 평가)

  • The present study was conducted to explore the anti-inflammatory action of $2-hydroxyethyl-{\beta}-undecenate$ (HPS) purified from Cumin (Cuminum cymium L.) seed against periodontitis. From the study in which human leukocyte was employed to detect the inhibiting effects of 5-lipokygenase and cyclooxygenase, enzymes generating carriers of infection like $LTB_4$ and PGs, as well as of collagenase and elastase, organ-destroying enzymes, following conclusions could be drawn: HPS was found to inhibit leukotrien $B_4$ biosynthesis by stimulating more than 97% of human polymorphonuclear leukocyte (PMNL) with addition of $5\;{\times}\;10^{-2}\;M$ when $IC_{50}$ was set at $2\;{\times}\;10^{-4}\;M$. Ninety-two percent of enzyme activation turned out to be inhibited when $5\;{\times}\;10^{-2}\;M$ was added in a test to prove inhibiting effects of HPS against activation of PMNL 5-lipoxygenase from homogeneous humans and purified 5-lipoxygenase on the market. Besides, $IC_{50}$ for enzyme activation was valued at $2.5\;{\times}\;10^{-4}\;M$, while the value of $IC_{50}$ for purified 5-lipoxygenase was $2.3\;{\times}\;10^{-4}\;M$. The $IC_{50}$ values of COX-activated leukocyte and purified collagenase were $5.1\;{\times}\;10^{-4}\;M$ and $2.3\;{\times}\;10^{-4}\;M$, respectively. Moreover, the value of $IC_{50}$ for activation of leukocyte collagenase was $2\;{\times}\;10^{-3}\;M$, whereas that for purified collagenase was $5\;{\times}\;10^{-2}\;M$. In case of leukocyte elastase, addition of $5\;{\times}\;10^{-2}\;M$ inhibited its activation by 66%. In case of purified one, however, activation of enzyme was inhibited by 25% with addition of $5\;{\times}\;10^{-2}\;M$. Furthermore, the $IC_{50}$ value for activation of leukocyte elastase was revealed to be $7.5\;{\times}\;10^{-3}\;M$. From the virulence test with human gingiva cell, it was shown that, on the second day of cultivation, 47.83% of the cell had been activated when HPS was added by $5\;{\times}\;10^{-2}\;M$. Even the addition of HPS by $1\;{\times}\;10^{-2}\;M$ featured 68.53% of cell activation, suggesting relatively strong toxicity of the substance against gingiva cell.

  • Automatic counting of yeast cells in baker's yeast culture was tried using a conventional light microscope equipped with a pc camera. Relatively good binary image was obtained by using white LED as microscope light source, but uneven brightness distribution in original image hindered counting accuracy. A block binarization method using local thresholds proportional to local brightnesses was used to get improved binary images. The brightnesses of the blocks were expressed as the value component in HSV color model. Good quality binary images were obtained by binarization on $8{\times}6$ blocks of original images and connected-component labelling of the binarized images produced reliable counting results in the concentration range $1.4{\times}10^5/mL{\sim}1.4{\times}10^7\;cells/mL$.

Bactericidal Efficacy of a Fumigation Disinfectant with Ortho-phenylphenol as an Active Ingredient Against Pseudomonas Aeruginosa and Enterococcus Hirae (Ortho-phenylphenol을 주성분을 하는 훈증소독제의 Pseudomonas aeruginosa와 Enterococcus hirae에 대한 살균효과)

  • This test was performed to evaluate the bactericidal efficacy of a fumigation disinfectant containing 20% ortho-phenylphenol against Pseudomonas aeruginosa (P. aeruginosa) and Enterococcus hirae (E. hirae). In preliminary tests, P. aeruginosa and E. hirae working culture suspension number (N value) were $2.8{\times}10^8$ and $4.0{\times}10^8CFU/mL$, respectively. And all the colony numbers on the carriers exposed to the fumigant (n1, n2, n3) were higher than 0.5N1 (the number of bacterial test suspentions by pour plate method), 0.5N2 (the number of bacterial test suspentions by filter membrane method) and 0.5N1, respectively. In addition, the mean number of P. aeruginosa and E. hirae recovered on the control-carriers (T value) was $2.8{\times}10^8$ and $3.4{\times}10^6CFU/mL$, respectively. In the bactericidal effect of the fumigant, the reduction number of $2.8{\times}10^8$ (d value) was 6.46 and 5.19 logCFU/mL, respectively. According to the French standard for the fumigant, the d value for the effective bactericidal fumigant should be over than 5 logCFU/mL. With the results from this study, the fumigation disinfectant containing 20% ortho-phenylphenol has an effective bactericidal activity, then the fumigant can be applied to disinfect food materials and kitchen appliances contaminated with the pathogenic bacteria.

  • In this work, we consider a finite difference operator $L^2_N$ corresponding to $$Lu:=-(u_{xx}+u_{yy})\;in\;{\Omega},\;u=0\;on\;{\partial}{\Omega}$$, in $S_{h^2,1}$. We derive the relation between the absolute value of the bilinear form $l_{N^2}$(u, v) on $S_{h^2,1}{\times}S_{h^2,1}$ and Sobolev $H^1$ norms.

Physiochemical Properties of Repeated Deep-frying Oil and Odor Pattern Analysis by Electronic Nose System (재가열 튀김유의 이화학적 특성과 전자코에 의한 향기 패턴 분석)

  • Chemical characteristics of soybean oil after deep-frying with potato sticks (200 g, 10% w/w of soybean oil) were studied according to the 34 deep-frying times. After consecutive 34 deep-frying, total polyunsaturated FA contents was gradually decreased while the total saturated FA and trans FA were increased. Acid value and peroxide value were increased while iodine value decreased, respectively. The Hunter $L^{\ast}$ value decreased while each $a^{\ast}\;and \;b^{\ast}b$ value were gradually increased. Electronic nose equipped with 12 metal oxide sensors was used for the discrimination of odor pattern of frying oils against the times of deep-trying. The proportions of 1st and 2nd principal component analysis showed 75.97% and 21.23%, respectively. While 6 among total 12 sensors well responded to discrimination of odor in the repented frying oils, suggesting that the odor pattern of each oil after deep-frying would be discriminated against fresh soybean oil, especially after 14 times. From the results, electronic nose could differentiate the degree of quality deterioration of the repeated deep-frying oils.

Spatio-temporal Distributions of Phytoplankton Community in the Coastal Waters of Central South Sea (CWoCSS), Korea (남해 중앙부 연안해역 식물플랑크톤 군집의 시·공간적 분포특성)

  • This paper described the spatio-temporal distributions in the phytoplankton community, such as species composition, standing crops, and dominant species, from July 2012 to April 2013 in the Coastal Waters of Central South Sea (CWoCSS) of Korea. A total of 87 species of phytoplankton belonging to 52 genera were identified. In particular, diatoms and phytoflagellates comprised more than 62.1% and 37.9% of the total species, respectively. The phytoplankton cell density fluctuated with an annual mean of $7.9{\times}10^4cells{\cdot}L^{-1}$ between the lowest value of $1.0{\times}10^3cells{\cdot}L^{-1}$ in spring and the highest value of $4.5{\times}10^5cells{\cdot}L^{-1}$ in winter. The seasonal succession of the dominant species were Chaetoceros curvisetus, Ch. debilis in summer, Eucampia zodiacus in autumn, E. zodiacus, Thalassiosira curviseriata in winter and Skeletonema costatum -ls (like species), Leptocylindrus danicus in spring. According to principal component analysis, the phytoplankton community of the CWoCSS was characterized by the mixing rate between the freshwater inflow from Somjin River and the seawater of the South Sea, Korea.

A Study on the Characteristics of Alluvial Clay in Yangsan-Mulgum (양산-물금 충적점토의 토질특성에 관한 연구)

  • Experiments both in laboratory and field were performed to compare and analyze the characteristics of alluvial clay. The alluvial clay was sampled in test site in which large-scaled tests for the part of the site are under process to suggest the rational method for alluvial clay and the criterion for ground settlement monitoring system. The followings were observed through the experiments : 1. Natural water content, plastic limit, and liquid limit of alluvial clay composed of highly fine grains were 40~80%, 10~20%, and 30~55%, respectively. The values of these properties were relatively small at the ground surface, while the values showed maximum at G.L.- l0m and gradually decreased below the level. 2. Shear strength of alluvial clay was proportionally increased to the depth. Unconfined and triaxial compressive strengths were 0.2~0.6kgf/$cm^2$ and 0.1~0.3kgf/$cm^2$, respectively. 3. Compression index and secondary compression index showed maximum values at G.L.-l0m and gradually decreased below the level. The value of consolidation coefficient was relatively large at the ground surface, constant with decreasing the depth, and incresed when G.L. was below -20m. 4. Piezocone test appeared that alluvial clay with N value of 2~4 was uniformly distributed with 20~ 30m thickness from the ground surface, sand seam was nonuniformly distributed, and penetration pore pressure was 0.8 ~ 1 times of the hydrostatic pressure. Undrained shear strength and consolidation coefficient were 0.04 ~ 0.76kgf / $cm^2$ and $2.88{\times} 10{^-4}~1.3{\times} 10{^-2} cm^2/s$ respectively.

Study on Inheritance and Transgressions at Cocoon and Shell Weight Characters between Genetically Sex-limited Silkworm Bombyx mori L. Lines

  • Inheritance of cocoon and shell weight characters in silkworm crosses between sex-limited at larva stage lines, including three (NIG$_{1}$ $\times$ TV$_{3/2}$ ,NIG$_{1}$ $\times$ TBV$_{2/24}$ and NIG$_{1}$ $\times$ B$_{2/6}$ ) from Japanese type and three (NIG$_{2}$ $\times$ B$_{2/6}$ ,TV$_{3/2}$ $\times$ NIG$_{1}$ and NIG$_{2}$ $\times$ TBV$_{2/24}$ ) from Chinese type was studied at Regional Centre for Scientific­Applied Service - Vratza during the period of 2000 ­2002. Inheritance of tested characters in F I was deter­mined through genetic parameters, additiveness (a), dominance (d) and their ratio (d/a). Degree and frequency of transgressions in F$_{2}$ were calculated also. Over dominance (d/a(>) 1) with bigger contribution of the parent with higher value was determined at inheritance tested characters. Transgressions obtained at cocoon and shell weight characters were positive and varied in wide limits. The productivity of parent's lines had defined the variability of genetic parameters for degree and frequency of transgressions. Combination of positive transgressions at cocoon and shell weight characters in F2 hybrid populations from sex-limited at larva stage allow us to select individuals with high productive potential for further use in selection programs.

Changes of Physicochemical Properties during Fermentation of Peach Wine and Quality Improvement by Ultrafiltration (복숭아주 발효시 이화학적 특성변화와 한외여과에 의한 품질 향상)

  • Peach wine was fermented at $25^{\circ}C$ for 2 weeks using Saccharomyces cerevisiae KCCM 12224, aged at 15$^{\circ}C$ for 14 weeks, and its physicochemical and microbiological changes were investigated. The viable bacterial cell numbers, 1.4$\times$10$^3$ CFU/mL at the beginning of fermentation, increased to 2.8$\times$10$^{6}$ CFU/mL after 2 weeks, but decreased to 7.0$\times$10$^3$ CFU/mL after 14 weeks. The viable yeast cell numbers were changed from 3.4$\times$10$^2$ CFU/mL to 2.4$\times$10$^{7}$ CFU/mL during fermentation, and decreased to 4.0$\times$10$^4$ CFU/mL after aging. Turbidity total sugar content, reducing sugar content, solid content and b value of peach wine decreased during fermentation but acidity, alcohol content, L and a value increased. Most physicochemical properties except alcohol content and reducing sugar content were not changed significantly during aging. When peach wine was filtered through 0.45 ${\mu}{\textrm}{m}$ nitrocellulose membrane followed by various ultrafiltration membranes with different molecular weight cut-off values, Biomax 100K membrane, with 79 liter/$m^2$/h (LMH) of initial flux, was suitable for ultrafiltration process of peach wine. These membrane filtration treatments resulted in complete removal of microorganisms and decrease in turbidity and alcohol content without changes in other chemical properties. The physicochemical properties of peach wine were not changed and any microorganisms were not found during the storage at 3$0^{\circ}C$ for 12 Weeks.

Susceptometry Application of Portable HTS SQUID-Based System

  • A portable RF HTS SQUID-based susceptometer was used for small size magnetized sample testing in weak DC (up to 200A/m) and AC (up to 4 A/m) magnetic fields. The system resolution for the magnetic moment is of the order of $1.6{\times}10^{-10} A.m^2$. The measured DC susceptibility of a tested sample agrees well with the value obtained by using a commercial liquid helium susceptometer.
