가압조건에서 생강 유래 6-shogaol 변환을 위한 가열 조건 최적화 (Optimization Study for the Production of 6-Shogaol-rich Ginger (Zingiber officinale Roscoe) under Conditions of Mild Pressure and High Temperature)

  • 본 연구는 다진 생강에 다량 함유된 6-gingerol을 뇌질환 개선 등의 기능성이 보고된 6-shogaol로 변환시키기 위하여 $CO_2$를 이용한 6.4MPa의 가압조건에서 가열 온도와 시간을 변수로 두고 중심합성설계법을 이용한 최적반응표면분석법을 적용하였다. 적용 온도 $70-130^{\circ}C$ ($X_1$)와 적용 시간 95-265분($X_2$)을 독립변수로, 6-gingerol함량($Y_1$, mg/g dried ginger)와 6-shogaol함량($Y_2$, mg/g dried ginger)을 종속변수로 설정하였다. 그 결과 온도와 시간이 대체적으로 증가할수록 6-gingerol은 감소하였으며 반대로 6-shogaol은 증가하였다. 도출된 반응표면 설계를 분석한 결과 압력이 6.4 MPa일 경우 적용 온도($X_1$)는 $130^{\circ}C$, 적용 시간($X_2$)는 204분이었다. 최적반응표면분석 결과 예측된 6-gingerol과 6-shogaol 함량은 각각 건조시킨 생강을 기준으로 0.13, 3.85 mg/g이며, 이때 만족도는 0.991이었다. 본 실험 결과를 통하여 도출된 가압조건에서의 열처리 방법은 스팀 등을 이용한 물리적 열처리 기술에 비해 소요 시간과 소모되는 에너지를 줄일 수 있었고, 6-gingerol에서 6-shogaol로의 변환율이 탁월하기 때문에 친환경적이고 효율적인 6-shogaol을 얻을 수 있는 기술임을 확인할 수 있었다.

  • [6]-Gingerol, a major pungent ingredient of ginger (Zingiber officinale Roscoe, Zingiberaceae) has a wide array of pharmacologic effects. Our previous studies have demonstrated that [6]-gingerol inhibits mouse skin tumor promotion and anchorage-independent growth of cultured mouse epidermal cells stimulated with epidermal growth factor. 생강 추출물 투여가 마우스 면역세포 활성에 미치는 영향 (Effect of Zingiber Officinale Roscoe Extracts on Mice Immune Cell Activation)

  • Ginger (Zingiber officinale Roscoe) has been used as a raw material in many traditional preparations since the ancient time. As a component of traditional health products, Ginger is known to be effective as appetite enhancer, anticold and anti-inflammation. This study was performed to investigate the immunomodulative effects of Ginger in mouse, using in vitro and ex vivo experiments. In vitro experiment, the mice splenocytes proliferation and three kinds of cytokines (IL-1 $\beta$, IL-6, and TNF-$\alpha$) prodution by peritoneal macrophages cultured with ethanol and water extracts of Ginger were used to indicate the immunomodulative effect. In order to elucidate the immunomodulative effects of Ginger ex vivo, water extract of Ginger was orally administrated into mice, and isolated splencytes and macrophages were used as experimental model. Ex vivo experiment, six to seven week old mice were fed ad libitum on a chow diet, and water extract of finger was orally administrated every other day for four weeks at two different concentractions (50 and 500 mg/kg B.W./day). In vitro study, the splenocytes proliferation was increased when water extract was supplemented in the range of 50-500 $\mu$l/ml concentration. In case of cytokines production, IL-1 $\beta$, IL-6 and TNF-$\alpha$ released by activated peritoneal macrophages were augmented by the supplementation of water extract of the Ginger. Ex vivo experiment, the highest proliferation of splenocytes and production of cytokines by activated peritoneal macrophages were seen in the mice orally administrated at the concentration of 500 mg/kg B.W./day. In conclusion, this study suggests that Ginger extracts may enhance the immune function by regulating the splenocytes proliferation and enhancing the cytokine prodution capacity by activated macrophages in mice.

  • Background : Ginger has been extensively used in foods and traditional medicines in Asian countries. Despite its frequent consumption in daily life, the mechanism of potential interactions between ginger components-drug has not been examined. To elucidate the mechanism of governing the effects of 6-shogaol, a primary constituent of dried ginger, on human cytochrome P450 (CYP) isoenzymes an incubation studies were carried out using pooled human liver microsome (HLM). Methods and Results : CYP isoenzyme specific substrate was incubated with multiple concentrations of inhibitor, HLM and cofactors. 6-shogaol showed a potent inhibitory effect on CYP2C9, CYP1A2 and CYP2C19 with half maximal inhibitory concentration ($IC_{50}$) values of 29.20, 20.68 and $18.78{\mu}M$ respectively. To estimate the value of the inhibition constant ($K_i$) and the mode of inhibition, an incubation study with varying concentrations of each CYP isoenzyme-specific probe was performed. 6-shogaol inhibited CYP2C9 and CYP2C19 noncompetitively ($K_i=29.02$ and $19.26{\mu}M$ respectively), in contrast, the inhibition of CYP1A2 was best explained by competitive inhibition ($K_i=6.33{\mu}M$). Conclusions : These findings suggest that 6-shogaol may possess inhibitory effects on metabolic activities mediated by CYP1A2, CYP2C9 and CYP2C19 in humans.

  • 이상의 실험결과를 종합해보면 6-gingerol의 최적추출조건은 $30^{\circ}C$에서 80% 에탄올을 이용하여 추출함이 바람직하다고 여겨진다. 또한 용매만을 이용해서 추출하는 것보다 초음파를 도입하여 추출하는 것이 7% 정도 높은 결과를 얻는 것을 확인할 수 있었다. 6-gingerol의 경우 분취크로마토그래피를 이용했을 때 비교적 높은 순도의 gingerol을 얻을 수 있었으며, F9 분획에서 $0.53\;mg/m{\ell}$의 6-gingerol를 얻었다. 6-gingerol의 항산화 활성은 아스코르브산의 95% 정도로 매우 높게 나타났다. 또한 항산화 활성을 나타내는 다른 물질이 포함되어 있다는 것을 확인할 수 있었다.

약용식물 추출물로부터 항산화활성 탐색 (Screening of Antioxidant Activity from Medicinal Plant Extracts)

  • This study was carried out to search the natural antioxidants from the 36 medicinal plants. $IC_{50}$ values of linoleic acid peroxidation inhibition to the BHA, BHT and ${\alpha}-tocopherol$ showed $7.2{\mu}g/ml$, $7.3{\mu}g/ml$, and $26.6{\mu}g/ml$, but hexane extract of Zingiber officinale showed $13.2{\mu}g/ml$, EtOAc extracts of Phellinus linteus, Morus alba, Zingiber officinale and butanol extract of Diospyros kaki showed $15.9{\mu}g/ml$, $16.9{\mu}g/ml$, $20.1{\mu}g/ml$, and $21.5{\mu}g/ml$ respectively. $IC_{50}$ values of DPPH radical scavenging activity of BHA, BHT and $\alpha$-tocopherol showed $14.9{\sim}27.7{\mu}g/ml$, but EtOAc extract of Paeonia suffruticosa showed the lowest $6.5{\mu}g/ml$ than those of other extracts, EtOAc extracts of Camellia sinensis, Salvia miltiorrhiz, and Phellinus linteus showed $8.0{\mu}g/ml$, $11.5{\mu}g/ml$, and $13.0{\mu}g/ml$ respectively.

생강(生薑) 추출물의 위장관 기능개선 및 체온저하 억제효과에 대한 실험적 연구 (Effect on Promoting Gastrointestinal Function and Inhibiting of Decreasing Body Temperature of Ginger Extracts(Zingiber Officinale))

  • This study was performed to investigate the effect of promoting gastrointestinal function and inhibiting of decreasing body temperature of ginger extract(Zingiber officinale) in rats. In order to elucidate the gastrointestinal function and inhibiting effect of body temperature of native ginger and improved ginger, water extracts of ginger were orally administrated into rats. The results are as follows: The gastrointestinal transit time was significantly decreased in native ginger(7.66hrs) and improved ginger(7.72hrs) extract administrated groups compare to control group(8.44hrs). The mean red faecal weight was increased in native ginger(30.6%) and improved ginger(31.1%) extract administrated groups compare to control group(24.9%) for 24hrs. Inhibiting effect of decreasing body temperature induced by serotonin was increased in native ginger($1.116^{\circ}C$) and improved ginger($1.416^{\circ}C$) extract administrated groups compare to positive control group($0.384^{\circ}C$) during 40 minutes. Gastrin and CGRP immunoreactive density was more strongly expressed in native ginger and improved ginger extract administrated groups compare to control group. Serotonin immunoreactive density was more weakly expressed in native ginger and improved ginger extract administrated groups compare to control group. These results suggest that ginger extracts may enhance physiological activity such as gastrointestinal motility, protection of mucosa and gastric acid secretion in gastrointestinal tracts, and inhibits decreasing body temperature
