유동성 복합 레진을 적용한 예방적 레진 수복물의 미세 누출 양상에 관한 비교 연구 (A COMPARATIVE STUDY ON THE MICROLEAKAGE OF PRVENTIVE RESIN RESTORATION USING FLOWABLE COMPOSITE RESIN)

  • 성인에 비해 치료를 견별 수 있는 시간이 상대적으로 짧은 소아 환자에 있어 시술 시간은 치료 결과의 중요한 변수로 작용하게 된다. 최근 소개된 유동성 복합 레진을 예방적 레진 수복술에 사용하게 될 경우 전색재 도포 과정을 생략할 수 있어 시술 시간의 단축 편의성 등의 장점이 인정되나 미세누출 등에 관한 연구 결과는 미흡한 실정이다. 본 연구의 목적은 최근 소개된 유동성 복합 레진을 사용한 예방적 레진 수복물과 기존의 복합 레진과 전색재를 사용한 예방적 레진 수복물 간의 미세누출 및 기포발생 정도를 비교하고자 함이었다. 교합면이 건전한 상악 소구치 60개를 대상으로 고속용 1/2 round bur를 이용하여 교합면 열구를 따라 길이$3.0\times$$0.7\times$깊이2.0mm의 I급 와동을 형성하고 각 군 당 20개씩 무작위로 시편을 배분하고, 제조자의 지침에 따라 본딩재인 Scotchbond $Multipurpose^{TM}$(3M dental product, U.S.A.)를 적용하고 유동성 복합 레진 수복군인 I, II군에는 각각 $Revolution^{TM}$(Kerr, U.S.A.)과 Arabesk Flow(VOCO, Germany), 대조군인 III군(통상적 예방적 레진 수복군)에는 Restorative $Z-100^{TM}$(3M dental product, U.S.A.)과 $Concise^{TM}$(3M dental product, U.S.A.)를 제조자의 지시에 따라 충전, 적용하였다. 3,000회의 열 순환 및 색소 침투 후 시편을 절단하여 컴퓨터에 영상을 입력하고 색소의 침투 길이 및 침투 비율을 계산하고 미세누출 평가 기준을 이용하여 각 시편에 대한 미세누출 정도를 평가하였다. 또한, 각 군별로 특징적으로 나타난 기포의 발생 양상을 별도의 평가기준에 따라 평가하였다. 미세누출은 수복재와 법랑질의 계면부에 국한되어 관찰되었고, 와동 내부까지 침투된 경우는 없었으며, 유동성 복합레진 수복군(I, II군)이 기존의 예방적 레진 수복군(III군)에 비해 낮은 미세 누출 정도를 나타냈다(p<0.05). 미세누출 결과 외에, 대부분의 충전물 내에서 다양한 형태의 기포가 발견되었으며, I, II군 표본에서 III군에 비해 크기가 큰 기포가 더욱 많이 관찰되었다.

Fatty Acid Profiles of Supraspinatus, Longissimus lumborum and Semitendinosus Muscles and Serum in Kacang Goats Supplemented with Inorganic Selenium and Iodine

  • Fat and fatty acids in muscle and adipose tissues are among the major factors influencing meat quality particularly nutritional value and palatability. The present study was carried out to examine the effects of supplementing inorganic selenium (Se), iodine (I) and a combination of both on fatty acid compositions in serum, and supraspinatus (SS), longissimus lumborum (LL), and semitendinosus (ST) muscles in goats. Twenty-four, 7 to 8 months old, Kacang male goats with a mean live weight of $22.00{\pm}1.17kg$ were individually and randomly assigned into four groups of six animals each for 100 d of feeding prior to slaughter. The animals were offered the same concentrate (basal) diet as 1% of body weight with ad libitum amount of fresh guinea grass. The four groups were as follows: T1 (control) - basal diet without supplementation; T2 - basal diet with 0.6 mg Se/kg DM; T3 - basal diet with 0.6 mg I/kg DM; T4 - basal diet with combination of 0.6 mg Se/kg DM and 0.6 mg I/kg DM. The major fatty acids (FAs) detected in the serum were palmitic (C16:0), stearic (C18:0), oleic (C18:1n9) and linoleic (C18:2n-6), while the major FAs in the selected muscles were C16:0, C18:0 and C18:1n9 acids. The main polyunsaturated fatty acids (PUFA) detected in muscles and serum were (CI8:2n-6), linolenic acid (C18:3n-3), and arachidonic acid (C20:4n-6). No significant differences (p>0.05) were observed in the concentration of total saturated fatty acids (SFA) among the four groups. PUFA concentrations in the goats supplemented with Se (T2) were significantly higher (p<0.05) than the goats of the control group (T1). The PUFA: SFA ratio was significantly higher in the animals supplemented with dietary Se (T2) than those of control ones (T1). It is concluded that dietary supplementation of inorganic Se increased the unsaturated fatty acids in muscle. The supplementation of iodine with or without Se had negligible effects on muscle fatty acid content of Kacang crossbred male goats.

Sol-Gel Synthesis, Crystal Structure, Magnetic and Optical Properties in ZnCo2O3 Oxide

  • Synthesis of ZnCo2O3 oxide is performed by sol-gel method via nitrate-citrate route. Powder X-ray diffraction (XRD) study shows monoclinic unit cell having lattice parameters: a = 5.721(1) Å, b = 8.073(2) Å, c = 5.670(1) Å, β = 93.221(8)°, space group P2/m and Z = 4. Average crystallite sizes determined by Scherrer equation are the range ~14-32 nm, whereas SEM micrographs show nano-micro meter size particles formed in ZnCo2O3. Endothermic peak at ~798 K in the Differential scanning calorimetric (DSC) trace without weight loss could be due to structural transformation and the endothermic peak ~1143 K with weight loss is due to reversible loss of O2 in air atmosphere. Energy Dispersive X-ray (EDX) analysis profile shows the presence of elements Zn, Co and O which indicates the purity of the sample. Magnetic measurements in the range of +12 kOe to -12 kOe at 10 K, 77 K, 120 K and at 300 K by PPMS-II Physical Property Measurement System (PPMS) shows hysteresis loops having very low values of the coercivity and retentivity which indicates the weakly ferromagnetic nature of the oxide. Observed X-band EPR isotropic lineshapes at 300 K and 77 K show positive g-shift at giso ~2.230 and giso ~2.217, respectively which is in agreement with the presence of paramagnetic site Co2+(3d7) in the oxide. DC conductivity value of 2.875 ×10-8 S/cm indicates very weakly semiconducting nature of ZnCo2O3 at 300 K. DRS absorption bands ~357 nm, ~572 nm, ~619 nm and ~654 nm are due to the d-d transitions 4T1g(4F)→2Eg(2G), 4T1g(4F)→4T1g(4P), 4T1g(4F)→4A2g(4F), 4T1g(4F)→4T2g(4F), respectively in octahedral ligand field around Co2+ ions. Direct band gap energy, Eg~ 1.5 eV in the oxide is obtained by extrapolating the linear part of the Tauc plot to the energy axis indicates fairly strong semiconducting nature of ZnCo2O3.

Association between HSP70 Genotypes and Oocytes Development on In vitro Maturation/Fertilization in Pig

  • This study was performed to clarify whether the variation of stress related heat shock protein 70 (HSP70) (GenBank X68213) gene was associated with the nuclear morphological change of in vitro maturation and in vitro capacitation in oocytes of pig ovaries obtained at the slaughterhouse. The nucleic acid substitution of C to G at the 483rd position was found out in HSP70 K1 (290-512) from X68213. The ovaries were categorized into CC, CG, and GG genotypes using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) (BsiHKA I). After the second in vitro maturation of immature fresh oocytes, the relation of nuclear morphological change in oocytes with the genotype of HSP70 K1 gene was such that the MII ratios of the genotype GG and CG (46.93% and 42.20%, respectively) were significantly higher than that of the CC genotype (10.71%) (p<0.05). With respect to in vitro maturation of frozen-thawed oocytes by an open pulled straw (OPS) method, the percentage of oocytes matured to MII stage of the CG genotype showed a higher trend than CC and GG genotypes. After the in vitro maturation of immature fresh oocytes and frozen-thawed oocytes by the OPS method, the relation of the pronuclei change in oocytes matured in vitro with HSP70 genotype was assessed, and the result showed that the enlarged sperm heads (ESH) of matured fresh oocytes and frozen-thawed oocytes were 80.0% and 60.0% in the CC genotype, respectively. The CC genotype group had a significantly higher rate of ESH than the CG and the GG genotype group (p<0.05). The ratios of polyspermic invasion were not different among HSP70 of the three genotypes. It was considered that the rate of in vitro maturation of fertilized oocytes was expected to differ according to genotype of the stress related gene.

Milk Conjugated Linoleic Acid (CLA) Profile and Metabolic Responses of Dairy Cows Fed with High-temperature-micro-time (HTMT) Treated Diets Containing High Quantity Extruded Soybean (ESB)

  • A feeding trial was conducted to examine the effect of high-temperature-micro-time (HTMT) processing of diets containing extruded soybean (ESB) in high quantity on milk fat production, metabolic responses, and the formation of conjugated linoleic acid (CLA) and trans-vaccenic acid (TVA). Twenty-one multiparous Holstein cows in mid-lactation were blocked according to milk yield in the previous lactation. Cows within each block were randomly assigned to either normal concentrate or HTMT treated diets containing ESB (7.5% HTMT-ESB and 15% HTMT-ESB). It was hypothesized that the HTMT-ESB would affect the undegradable fatty acids in the rumen and, thus, would modify the fatty acid profile of milk fat. Both 7.5% and 15% HTMT-ESB did not affect milk yield, fat, protein, lactose and solid-not-fat (SNF), but the proportion of cis-9, trans-11 CLA in milk fat was significantly increased by these treatments. Content of TVA in milk fat was not affected by HTMT-ESB. The HTMT-ESB influenced the fatty acid profile in milk fat, but there was little difference between 7.5% and 15% of supplementation. HTMT-ESB feeding significantly decreased the concentration of plasma insulin and glucose, while plasma growth hormone (GH), triglyceride (TG), non-esterified fatty acid (NEFA) and HDLcholesterol were increased by 7.5% and 15% ESB-HTMT supplementation in comparison to the control group (p<0.05). However, no significant difference was observed in plasma LDL-cholesterol, insulin like growth factor (IGF)-1, T3, T4, and leptin concentrations among treatments (p>0.05). The present results showed that cis-9, trans-11 CLA production was increased by HTMT treatment of dietary ESB without reduction of milk fat, and the unchanged milk fat and yield was assumed to be associated with the constant level of thyroid hormones, leptin, and IGF-1.

돼지 액상정액 보존을 위한 Lactose-Egg Yolk와 $B\ddot{u}tschwiler$ 희석액의 비교 (Comparision of Preservation of Liquid Boar Semen between Lactose-Egg Yolk and $B\ddot{u}tschwiler$ Diluents)

  • 본 연구는 돼지 액상정액을 인공수정용 100ml 플라스틱 병에 보존하면서 Lactose-Egg yolk 희석액과 B tschwiler 희석액 간에 보존 온도별 차이를 조사하고, Lactose-Egg yolk 희석액에서의 글리세롤 농도의 효과를 조사하여 돼지 액상정액을 좀더 장기간 사용할 수 있는 방법을 찾고자 실시하였다. 돼지 액상정액을 5$^{\circ}C$ 냉장고에 보존하면서조사한 바에 의하면, 37$^{\circ}C$에서 0.5 및 2시간 배양 후의 정자 운동성은 전체 보존기간동안 Lactose-Egg yolk 희석액이 B tschwiler 희석액보다 유의하게 (P,0.05) 높게 나타났고, 정상첨체비율은 두 희석액간에 차이가 없었다. 돼지 액상정액을 15$^{\circ}C$에 보존하면서 조사한 바에 의하면, 3일부터 7일 보존시까지 정자 운동성과 정상첨체비율에 있어서 B tschwiler 희석액이 Lactose-Egg yolk 희석액보다 유의하데 높게 나타났다. Lactose-Egg yolk 희석액을 이용한 돼지 액상정액의 글리세롤 농도의 효과에 있어서는 최종 글리세롤 농도가 0, 1, 3 및 5%보다 2%일 때 가장 높은 정자 운동성과 정상첨체비유을 나타내었다. 분만율, 복당 생존자돈수 그리고 출생시 평균 생시체중은 Lactose-Egg yolk 희석액과 B tschwiler 희석액간에 차이가 없었다. 이상의 연구 결과를 종합해 볼 때 Lactose-Egg yolk 희석액은 5$^{\circ}C$에서 B tschwiler 희석액은 15$^{\circ}C$에서 6~7일 동안 돼지 액상정액을 보존할 수 있었다.

코어 형태와 시멘트 종류에 따른 전부주조금관의 유지력에 관한 연구 (A STUDY OF CORE TYPE AND LUTING CEMENTS ON COMPLETE CAST CROWN RETENTION)

  • The purpose of this study was to compare the retention of complete cast crown over amalgam ores, composite resin cores, and cast gold cores when cemented with three different luting agents. Eighteen core specimens each of amalgam(Bestaloy, Dong Myung, Seoul, Korea), composite resin (Z100, 3M Dental product, st. Paul, Minn) and type IV gold alloy (Ba-4, Heesung Engelhard Corp., Korea) were made in a customized milling stainless steel die. A wax pattern with a loop attached to occlusal surface was made for each core and a type II gold alloy casting was fabricated. The castings which had clinically acceptable marginal fit were used as test samples. The following luting cements were used to cement cast crowns on each core material : (1) zinc phosphate cement (Confi-dental Products Co., USA) (2) glass-ionomer cement (Fuji Plus, GC Industrial Corp., Tokyo, Japan) (3) resin cement (Panavia 21, Kuraray Co., USA). All cements were mixed according to manufacturers' instructions. A static load of 5kg was then applied for 10 minutes on the crowns. All specimens were stored in saline solution for 24 hours at $37^{\circ}C$ and thermocycled for 500 cycles. After storage and cycling, the tensile bond strengths were measured by using a universal testing machine (Instron Corp., Canton, Mass.) at a crosshead speed of 0.5mm/min. The results were as follows 1. The retentive strength of resin cement was the highest of alt three types of cement for resin core (p<0.05). 2. There was no statistical difference among the retentive strengths of three cements for amalgam core (p>0.05). 3. The retentive strength of resin cement was higher than that of zinc phosphate for cast core, but there was no difference between the retentive strength of glass ionomer cement and those of rein and zinc phosphate cement. 4. The retentive strength of the zinc phosphate cement for amalgam core was the highest of all type of cores.

속양(速釀)간장 제조(製造)에 관(關)한 연구(硏究) (Studies on the Quick Ripening Process for Soysauce)

  • 간장의 원료이용률(原料利用率)을 향상(向上)시킴과 아울러 담금기간(期間)을 단축(短縮)시키기 위한 속양(速釀)간장 제조법(製造法)을 개발(開發)하기 위하여 2종(種)의 국균(麴菌)(Asp. sojae, Asp. niger)을 번식(繁殖)시킨 부국추출(부麴抽出) 효소액(酵素液)을 이용(利用)하여 콩 Koji 및 증자원료(蒸煮原料) 그대로 4배(倍)의 물을 넣은 것을 식염(食鹽)을 첨가(添加)하지 않고 $40^{\circ}C$ pH$4{\sim}6$의 조건에서 8시간분해(時間分解)시킨후(後) 4가지의 서로 다른 방법(方法)으로 간장을 담근후(後) 내염성(耐鹽性) 효모(酵母)(Zygosaccharomyces sojae) 및 유산균(乳酸菌)(Pediococcus sojae)를 첨가(添加)하여 후숙(後熟)을 1개월간(個月間) 촉진(促進) 숙성(熟成)시키면서 상법(常法)으로 담근 Control구(區)와 각성분변화(各成分變化)를 비교(比較)함과 아울러 식미시험(食味試驗)을 한 결과(結果)를 요약(要約)하면 다음과 같다. 1) 국법(麴法)이 증자소화법(蒸煮消化法)보다 Asp. niger enzyme 처리구(處理區)가 Asp. sojae enzyme 처리구(處理區)보다 일반성분(一般成分) 용출량(溶出量)이 많았다. 2) 국법(麴法)으로 Asp. niger enzyme 처리구(處理區)는 1개월간(個月間) 숙성(熟成)으로 상법(常法)간장의 $5{\sim}6$개월(個月) 숙성결과(熟成結果)와 비슷하였다. 3) 식미시험결과(食味試驗結果)는 국법(麴法)이 증자소화법(蒸煮消化法)보다 Asp. sojae enzyme 처리구(處理區)가 Asp. niger enzyme 처리구(處理區)보다 맛이 우량(優良)하였고 상법(常法)으로 6개월근(個月間) 숙성(熟成)시킨 것에 비(比)하면 일반성분(一般成分) 용출량(溶出量)은 비슷하지만 식미(食味)는 약간 떨어지나 상법(常法)으로 1개월(個月) 숙성(熟成)시킨것에 비(比)하면 매우 우량한 편이었다. 4) 각처리구중(各處理區中) 일반성분(一般成分) 용출량(溶出量)과 식미(食味)를 종합(綜合)해 볼때 국법(麴法)으로서 Asp. sojae enzyme 처리구(處理區)가 속양(速釀)간장 제조법(製造法)으로서 가장 우량하였다.

Melatonin protects endothelial progenitor cells against AGE-induced apoptosis via autophagy flux stimulation and promotes wound healing in diabetic mice

  • Wound healing is delayed in diabetic patients. Increased apoptosis and endothelial progenitor cell (EPC) dysfunction are implicated in delayed diabetic wound healing. Melatonin, a major secretory product of the pineal gland, promotes diabetic wound healing; however, its mechanism of action remains unclear. Here, EPCs were isolated from the bone marrow of mice. Treatment of EPCs with melatonin alleviated advanced glycation end product (AGE)-induced apoptosis and cellular dysfunction. We further examined autophagy flux after melatonin treatment and found increased light chain 3 (LC3) and p62 protein levels in AGE-treated EPCs. However, lysosome-associated membrane protein 2 expression was decreased, indicating that autophagy flux was impaired in EPCs treated with AGEs. We then evaluated autophagy flux after melatonin treatment and found that melatonin increased the LC3 levels, but attenuated the accumulation of p62, suggesting a stimulatory effect of melatonin on autophagy flux. Blockage of autophagy flux by chloroquine partially abolished the protective effects of melatonin, indicating that autophagy flux is involved in the protective effects of melatonin. Furthermore, we found that the AMPK/mTOR signaling pathway is involved in autophagy flux stimulation by melatonin. An in vivo study also illustrated that melatonin treatment ameliorated impaired wound healing in a streptozotocin-induced diabetic wound healing model. Thus, our study shows that melatonin protects EPCs against apoptosis and dysfunction via autophagy flux stimulation and ameliorates impaired wound healing in vivo, providing insight into its mechanism of action in diabetic wound healing.

반구형 플라스틱 구조체 성형을 위한 프리폼 몰드 사출성형공정 최적화 (Optimization of preform mold injection molding process for hemispheric plastic structure fabrication)

  • Traditional cell culture(2-dimensional) is the method that provide a nutrient and environment on a flat surface to cultivate cells into a single layer. Since the cell characteristics of 2D culture method is different from the characteristics of the cells cultured in the body, attempts to cultivate the cells in an environment similar to the body environment are actively proceeding in the industry, academy, and research institutes. In this study, we will develop a technology to fabricate micro-structures capable of culturing cells on surfaces with various curvatures, surface shapes, and characteristics. In order to fabricate the hemispheric plastic structure(thickness $50{\mu}m$), plastic preform mold (hereinafter as "preform mold") corresponding to the hemisphere was first prepared by injection molding in order to fabricate a two - layer structure to be combined with a flat plastic film. Then, thermoplastic polymer dissolved in an organic solvent was solidified on a preform mold. As a preliminary study, we proposed injection molding conditions that can minimize X/Y/Z axis deflection value. The effects of the following conditions on the preform mold were analyzed through injection molding CAE, [(1) coolant inlet temperature, (2) injection time, (3) packing pressure, (4) volume-pressure (V/P). As a result, the injection molding process conditions (cooling water inlet temperature, injection time, holding pressure condition (V / P conversion point and holding pressure size)) which can minimize the deformation amount of the preform mold were derived through CAE without applying the experimental design method. Also, the derived injection molding process conditions were applied during actual injection molding and the degree of deformation of the formed preform mold was compared with the analysis results. It is expected that plastic film having various shapes in addition to hemispherical shape using the preform mold produced through this study will be useful for the molding preform molding technology and cast molding technology.
