Quantitative analysis and validation of naproxen tablets by using transmission raman spectroscopy

  • A transmission Raman spectroscopy-based quantitative model, which can analyze the content of a drug product containing naproxen sodium as its active pharmaceutical ingredient (API), was developed. Compared with the existing analytical method, i.e., high-performance liquid chromatography (HPLC), Raman spectroscopy exhibits high test efficiency owing to its shorter sample pre-treatment and measurement time. Raman spectroscopy is environmentally friendly since samples can be tested rapidly via a nondestructive method without sample preparation using solvent. Through this analysis method, rapid on-site analysis was possible and it could prevent the production of defective tablets with potency problems. The developed method was applied to the assays of the naproxen sodium of coated tablets that were manufactured in commercial scale and the content of naproxen sodium was accurately predicted by Raman spectroscopy and compared with the reference analytical method such as HPLC. The method validation of the new approach was also performed. Further, the specificity, linearity, accuracy, precision, and robustness tests were conducted, and all the results were within the criteria. The standard error of cross-validation and standard error of prediction values were determined as 0.949 % and 0.724 %, respectively.

Effect of Dang Gui Bo Hyul-tang on fatigue types of Mibyeong (피로형 미병에 대한 당귀보혈탕의 효과)

  • Objectives : Mibyeong is a korea medicine have original concept of the disease. However, no previous report has effect of mibyeong herbal medicine in fatigue types of mibyeong. This study investigated the question of whether Dang Gui Bo Hyul-tang(DGBHT) of effect on fatigue types of Mibyeong. Methods : C57BL/6 mice were randomaly divided into three group (n=10). The mice were then group (1) Nocontrol, (2) Restraunt stress(veh), (3) Dang Gui Bo Hyul-tang 200mg/kg. The administered Dang Gui Bo Hyul-tang 200mg/kg or distilled water (orally) 1 hr prior to daily exposure to repeated restraint stress (2 h) for 15 days. The performed behavior test (Mechanical hyperalgesia test,, Open-field test, Forced swimming test, Sucrose preference test and immunostaining and biochemical measured in serum. Results : Stress fatigue induced mices significantly increased lethargic, hyperalgesia through behavior test (Mechanical hyperalgesia test (decrease 43%), Open-field test ($4,809{\pm} 226.13cm$ vs. $3121{\pm}226.64cm$), Forced swimming test ($11.45{\pm}3.96$ vs. $79.10{\pm}8.12sec$), Sucrose preference test (decrease 58%)). In addition, chronic fatigue model evidently increased corticosterone level ($122.54{\pm}18.88$ vs. $186.94{\pm}18.26ng/ml$), AST level ($46.22{\pm}3.23$ vs. $31.40{\pm}3.86U/L$), ALT level ($38.78{\pm}5.72$ vs. $17.60{\pm}1.30$), liver necrosis, lateral ventricle size. These alterations were significantly ameliorated by DGBHT. DGBHT significantly attend the elevated serum concentrations of corticosterone ($155.90{\pm}6.29ng/ml$), AST ($31.40{\pm}3.86U/L$), ALT ($17.60{\pm}1.30U/L$). Moreover, DGBHT improved lethargic, hyperalgesia when compared the stress fatigue (Mechanical hyperalgesia test (improve 28%), Open-field test ($4,038{\pm}615.81cm$), Forced swimming test ($7.56{\pm}1.88sec$), Sucrose preference test (increase 21%) Conclusions : Theses result suggest that DGBHT have improved lethargic, hyperalgesia and fatigue-associated hormone and liver protective on stress fatigue model. It will be necessary to research to present evidences on benefits and effects of Korean medical treatment for Mibyeong through clinical researches based on benefits and effects of those animal models.

Physicochemical Characterization and Dissolution Properties of CS-891 with Different Crystallinity

  • Ground CS-891 (N-[1-(4-methoxyphenyl)-1-methylethyl]-3-oxo-4-aza-5a-androst-1-ene-$17{\beta}$-carboxamide) of poorly water soluble drug was obtained using a Heiko Seisakusho model TI-100 vibration mill, and samples with different crystallinity were prepared at mixture ratios of 10:0, 7:3, 5:5, 3:7 and 0:10 (intact;ground CS-891). Physicochemical characterizations were obtained using qualitative and quantitative X-ray diffractometry, different scanning calorimetry (DSC), scanning electron microscopy (SEM), Quantasorb surface area analyzer, and controlled atmosphere microbalance. With increase of amorphous CS-891 in mixture ratios, the intensities of X-ray diffraction peaks of crystalline CS-891 were decreased, whereas surface area, water absorption, and exothermic peaks in DSC were increased. The apparent solubility of ground CS-891 was $4.4\;{\mu}g/ml$ and the solubility of intact CS-891 was $3.1\;{\mu}g/ml$ at $37{\pm}1^{\circ}C$. The apparent precipitation rates of CS-891 in a supersaturated solution during the solubility test were increased with an increase of amorphous CS-891, and a crystalline form of CS-891 transformed from amorphous CS-891 after the solubility test was found by X-ray diffraction analysis, DSC and SEM. The dissolution profiles of CS-891 with different crystallinity at $37{\pm}1^{\circ}C$ by the USP paddle method were investigated, and the apparent dissolution rate constant of ground CS-891 was about 5.9-fold higher than that of intact CS-891. A linear relationships between the crystallinity of CS-891 and the apparent dissolution rate constant (r>0.96) were obtained.

Research on the Residential Environment of New Town & Surrounding Area (대규모 신도시 및 주변지역 거주환경 비교분석 연구)

  • During the 1990's, five new towns were constructed around Seoul such as Bun-Dang and Il-San. These new towns are very popular since they were created based on systemic urban planning and they provide various residential amenities. Many people consider them livable cities. Taking such advantages, other small-scale new public and private developments continued around those new towns and consequently created 'agglomerated region with new development'. The most representative area is Bun-Dang and north-western Yong-In area which are formed along the Seoul-Busan expressway. This area is a mega-community having more than one million residents in 2005.Judging from recent development trends in the Seoul metropolitan area, it is expected that the formation of other 'agglomerated region with new development' would be continued for the time being. This study is aimed to analyze the residential environment of Bun-Dang and north-western Yong-In and find implications to the management of 'agglomerated region with new development'. The relevant existing studies have their limits, only focusing on analyzing current conditions and pointing out current problems. This study could be differentiated from the existing analyses in that it conducted comprehensive analyses on various factors that constitute urban residential environment. The conclusion of this study was derived from diverse analyses of current development condition, residential sites, regional transportation, awareness of residents, and oversee cases.

Fire Risk Assessment of Temple Components in Young-dong Areas using Fire Loads (영동지역 사찰의 구성요소별 화재하중을 통한 화재위험성 평가)

  • In this study, we have assessed the fire risk of Buddhist temples located in Youngdong area by means of fire loads of components. First, we categorized the components into Buddhist temples (Dae-Ung-Jeon, Pal-Sang-Jeon), Sam-Seong-Gak (Chil-Seong-Gak, San-Sin-Gak), a Buddhist temple dormitory (Sim-Geom-Dang, Seol-Seon-Dang), Lu, Il-Ju-Mun and then carried out a field survey. Then, we examined the area of each room, types as well as quantity and dimension of combustibles. The fire loads calculated in this way were 446.96 $kg/m^2$ for Buddhist temples, 331.71 $kg/m^2$ for Sam-Seong-Gak, 164.14 $kg/m^2$ for the Buddhist temple dormitory, 463.91 $kg/m^2$ for Lu and 1042.14 $kg/m^2$ for Il-Ju-Mun, thus showing Il-Ju-Mun with the biggest fire load. We speculate that this is because construction materials were similar in size and quantity to others albeit the area of Il-Ju-Mun is smallest.

Correlation Analysis Between Narrowing Disc Space of Lumbar X-ray and HIVD of L-spine CT in Lumbosacral Strain Patients (좌섬요통환자에서 X-ray상 narrowing과 CT상 HIVD와의 연관성 분석)

  • Objective : This study was designed to analyze the of correlation between narrowing disc space of lumbar X-ray and HIVD of L-spine CT in lumbosacral strain patients. Methods : 63 cases of lumbosacral strain patients who visited Bul-kyooh Oriental Hospital from June 13, 2000 to August 9, 2001 were selected. We performed the radiography by lumbar X-ray and L-spine CT for all cases. Results : 28 of 63 cases revealed narrowing disc space in Lumbar X-ray and these 28 cases were diagnosed as HIVD in L-spine CT at all. 35 of 63 cases revealed normal in Lumbar X-ray and 13 of these 35 cases were diagnosed as HIVD in L-spine CT. Conclusions : We found that narrowing disc space in lumbar X-ray has significant relevance to HIVD in L-spine CT(P-value = 0.001). For accurate diagnosis and treatment of lumbosacral strain patients. combination of clinical symptoms, physical examination and radiography of X-ray and HIVD is needed.

An Analysis of the Style of Kogufo Costume -Trousers- (고구려 복식의 양식 분석 -고구려 바지를 중심으로-)

  • The purpose of this study is to analyze the style of trousers of Koguryo(高句麗). Trousers of Koguryo were called Go(袴: trousers) and those had various forms according to the shape of gusset(dang, 糖), width and length of the trousers. Commonly the trousers that had worn by Koguryo people had gausset in those bottom not to be shown hip. The gausset was cut triangle or square so if a person wear the trouser with the triangular gausset, the trouser has a pointed hip. Sometimes the trousers had non pointed hips were dicovered in the wall-paintings of the Koguryo tombs, they are thought the trousers that had no gaussets or square one. And also there were another trousers that had wide/narrow legs and long/short legs. Trousers of wide legs were called Gwango(寬袴) and narrow legs Sego(細袴). Gwango(寬袴) grew wider over time and in the late age, the trousers were called Daegugo(大口袴) with wide ends were worn. Trousers were also decorated elaborately with patches on the edges, leg ties, vents, etc.

Effects of Dietary Bong-ip(Morus alba L.), Gam-chei(Glycyrrhizae glabra), Sol-ip(Pinus densiflora) and Dang-gi(Angelica gigas) on Serum Composition in Rats (뽕잎, 감초, 솔잎 및 당귀분말이 흰쥐의 혈청조성에 미치는 영향)

  • The purpose of this study was to investigate the effect of dietary Morus alba L.(Bong-ip, B), Glycyrrhizae glabra(Gam-chei, C), Pinus densiflora(Sol-lp, S) and Angelica gigas(Dang-gi, D)powder on serum composition in rats(Sprague-Dawley male rats, 100-110g). Serum TG(triglyceride, p<0.01), total cholesterol, glucose, total protein, albumin, GGT$({\gamma}-glutamyl$ transferase, p<0.05) were significantly increased D group than that of nomal and other groups, but UA(uric acid, p<0.05) was significantly decreased, and C group(p<0.05) was significantly increased. but C group of urine(p<0.05) was significantly decreased. Also, B and S groups(p<0.05) of BUN(blood urea nitrogen), S group(p<0.05) of ALP(alkaline phosphokinase, Band C(p<0.05) of CPK(creatinine phosphokinsae, p<0.05) were significantly increased. B, S and C groups were better than D group for lipid metabolism, and pretection to liver. Also, B and C groups of glucose were same as normal diet, so Morus alba L. was good food for lipid metabolism and hypoglycemic effect.
