Early Bronchoconstriction After Allergen Challenge of Nonanesthetized Guinea Pigs (Ovalbumin으로 감작된 기니픽에서 Allergen 흡입으로 인한 즉시형 기관지 수축반응에 대한 비침습적 측정)

  • Background: Bronchial asthma is a complex disease, which is characterized by spontaneous exacerbations of airway obstruction and persistent bronchial hyperresponsiveness. Animal models have fallen short of reproducing the human disease, particularly in mimicking the spontaneous and persistent airflow obstruction that characterized in asthma. In animals, airflow obstruction is usually assessed by measuring airflow resistance during tidal breathing under such invasive technique as tracheostomy and anesthesia. A noninvasive technique for measuring pulmonary function in small animals is needed to evaluate long-term changes in lung function during the course of experimentally produced disease without sacrificing the animal. Purpose: The purpose of this study was to evaluate early bronchoconstrcition after allergen challenge and airway responsiveness (AR) to inhaled methacholine in nonanethetized, unrestrained guinea pigs. Method: Guinea pig model of asthma was sensitized by subcutaneous injection with ovalbumin and challenged by inhalation of aerosolized ovalbumin(1% wt/vol ovlabumin). Airflow obstruction of conscious guinea pig was measured as specific airway resistance (airway resistance $\times$ thoracic gas volume). Airway resistance and thoracic gas volume of conscious guinea pig were assessed by body plethysmography before challenge and at regular intervals for as long as 30 minutes after challenge. AR to aerosolized methacholine of asthma group was compared with that of control group in body plethysmography. Result: Asthma model<> developed in 13 (65%) among 20 guinea pigs, in which early responses occurred in the airways after the exposure to inhalation with ovalbumin. Airway challenge with ovalbumin caused increase in specific airway resistance, which peaked at 6 minutes and amounted to a $231.5{\pm}30.4%$ increase from baseline. AR to aerosolized methacholine of asthma model increased significantly compared with control group. Conclusion: These results have showed a useful animal model to evaluate early bronchoconstrcition after allergen challenge and airway responsiveness in nonanethetized, unrestrained guinea pigs.

Effects of Different Sources of Dietary Chromium and Copper on Growth Performances, Nutrients Digestibility, Fecal Cr, Cu and Zn Excretion in Growing Pigs (크롬과 구리의 형태별 병용급여가 육성돈의 육성성적, 소화율 및 분의 Cr, Cu, Zn 배출량에 미치는 영향 미치는 영향)

  • This study was carried out to evaluate effects of either organic or inorganic sources of both chromium and copper on growth performances, nutrients digestibility and fecal Cr, Cu, and Zn excretion in growing pigs. A total of 36 growing pigs((Landrace×Yorkshire)×Duroc, weighing 61.2kg in average) were allotted to 4 treatments with 3 replicates and 3 pigs per replicate. Four treatments were designated by supplemental sources of both chromium and copper as follows: ①200ppb Cr as Cr-methionine chelate(CrMet) and 200ppm Cuas copper methionine chelate(CuMet), ②200ppb Cr as CrMet and 200ppm Cu as copper sulfate(CuSO4), ③200ppb Cr as chromium chloride(CrCl3) and 200ppm Cu as CuMet, ④200ppb Cr as CrCl3 and 200ppm Cu as CuSO4. Growth performance was highest(p<0.05) in CrMet and CuMet supplemented diet treatment. Nutrients digestibility of diets was lowest(p<0.05) in CrMet and CuSO4 supplemented diet treatment, and highest(p<0.05) in CrMet and CuMet supplemented diet treatment. Fecal copper, zinc and chromium excretion was highest(p<0.05) in CrCl3 and CuSO4 supplementation treatment and lowest(p<0.05) in CrMet and CuMet supplementation treatment. This study showed a relatively high degree of utilization of Cr and Cu as well as Zn by supplementation of CrMet and CuMet compared with those of the inorganic sources.

Development of High Meat Quality Using Microsatellite Markers in Berkshire Pigs (Microsatellite Marker를 이용한 육질 우수 버크셔 계통 조성에 관한 연구)

  • In this study, the efficiency of microsatellite (MS) markers for pork quality was examined and further, their suitability to domestic pork industry also was verified, by measuring meat quality parameters of Berkshire breeds. A total of 323 pigs of Berkshire breeds were slaughtered and subjected to meat quality evaluation. In addition, the genomic DNAs from blood samples of slaughtered pigs were used for genotyping analysis of 50 MS markers. The results revealed that Berkshire breeds have excellent meat quality, compared with the popular domestic breeds such as Duroc, Yorkshire, and Landrace. Noticeably, the Berkshire breeds exhibited a significant post-mortem pH24hr ($5.88{\pm}0.01$) and fat content ($2.878{\pm}0.06$). Through the linkage analysis between MS markers, 14 MS markers showed significant association with meat quality traits (p<0.05). Maximum significant differences of 0.55 pH24hr value and 2.04% fat content were observed between the highest and lowest allele populations. If these 14 MS markers are applied to the pork quality diagnosis kit, the synergistic effect can be expected in meat quality parameters such as meat color, fat content, pH 24 hr, cooking loss, drip loss and water-holding capacity.

  • This study aimed to visualize a pig character targeting the golden pig in Geuknakjeon of Bulguksa Temple. As for a pig, it is included in this list of the 12 Chinese zodiac animals and controls Haeshinjang. Today, the characters market is enjoying rapid growth due to diversification of platforms. However, we do not see many characters made out of Korean traditional cultures and customs. This study first looks into significance of pigs that are often observed in Korean traditional culture and then offers possibilities of their being transformed into contemporary characters. In order to achieve the research goals, the study shed new light on the golden pig in Bulguksa Temple. As making contemporary applications to , the study proceeded with work of visualization. As for a face of a character, the study focused on that of the pig but regarding a body, the study applied a man's body. After all, the study came up with this SD character in a second-proportioned figure. This study discuses formative characteristics of existing Buddhist cultural assets and uses the character for improving understanding on Korean traditional culture. In the future, the study will propose ideas on animation contents development targeting infants and children.

  • The first outbreak of Aujeszky's disease(AD) was identified from piggery located at the southern part of Korea in July, 1987. This piggery suffered from a significant economic loss caused by unexpected piglet mortality and reproductive failure. Etiologic viral agents were isolated from tonsil and spleen of the infected piglets, and the isolates produced a typical cytopathic effects of herpesvirus with giant cell formation when inoculated in many different cells. Subsequently the field isolates were characterized as suid herpesvirus I by cross-neutralization test and indirect fluorescence assay utilizing specific monoclonal antibody, and were proved to be a pathogenic strain of AD virus(ADV).

  • The small and the large intestine of swine represent the organs that extract nutrients from feedstuffs through digestion and fermentation and that allow their absorption and incorporation into the blood circulation. Special attention is directed towards the small intestine of young pigs since the transition to a solid diet at weaning exerts major impacts on the structural and functional integrity of the small intestine. Dietary factors involved in postweaning changes of gut morphology and biochemistry such as removal of bioactive compounds in sows milk at weaning, anti-nutritional factors in weaner diets, dietary fiber and the role of voluntary feed intake will be elucidated. The microbial function of the large intestine which is carried out by a diverse population of microorganisms is dependent on substrate availability. Short chain fatty acids as main fermentation products contribute to the energy supply of the host but they are also important for the maintenance of the morphological and functional integrity of the epithelium in the colon. As a result of bacterial nitrogen assimilation in the large intestine, nitrogen is shifted from the urinary to the fecal excretion route thus saving metabolic energy to the pig because less ammonia would become available for conversion to urea.

  • The reactions of [Ni(L)(H₂O)₂]Cl₂ (L = 2,5,9,12-tetramethyl-1,4,8,11-tetraazacyclotetradecane) with terephtalate (tp) and 2,5-pyridinedicarboxylate (pdc) generate one-dimensional nickel(II) complexes, [Ni(L)(H₂O)₂](tp) · ₄H₂O (1) and [Ni(L)(H₂O)₂](pdc)·₄H₂O (2). The structures have been characterized by X-ray crystallography, magnetic susceptibility and spectroscopy. The crystal structures of 1 and 2 show a distorted octahedral coordination geometry around the Ni(II) ion, with secondary amines of the macrocycle and two water molecules at the trans position. Complexes 1 and 2 display the one-dimensional hydrogen-bonded infinite chains. The magnetic behavior of all compounds exhibits weak interchain antiferromagnetic interactions with J values of -1.09(3) for 1 and -1.14(2) cm-1 for 2.

  • Porcine pancreas development is not well studied at the molecular level despite being a therapeutic resource for diabetic patients. In this study, we investigated expression of lineage markers and performed proteomic analysis. Expression of the early lineage markers Pdx1 and Ptf1a was developmentally conserved between mice and pigs, whereas expression of the islet differentiation marker Pax4 was delayed in porcine compared with murine pancreas development. Proteomic analysis found that expression levels of chymotrypsinogen were down-regulated during porcine pancreas development while those of digestive enzymes like lipases, elastase and serine protease were up-regulated. In addition, specific isoforms of protein folding assistants such as protein disulfide isomerase and prefoldin were expressed at specific stages during the maturation of digestive enzymes. Taken together, these results show that development of the porcine pancreas is regulated by a concerted interplay of pancreas lineage marker proteins and other specified proteins, resulting in a functional endocrine and exocrine organ.

Prevalence and Antimicrobial Susceptibility of Erythromycin-Resistant Campylobacter jejuni and Campylobacter coli Isolated from Swine

  • Campylobacter species are known to the high optimum growth temperature ($42^{\circ}C$) and the cause of enteritis in people. Erythromycin has a curative effect for enteritis caused by the bacteria. However, the rate of erythromycin-resistant bacteria was not well known until recently in Korea. Swine are one of sources of the infection with a Campylobacter species which cause the symptom of a high temperature. In this study, we cultured rectum fecal specimens of 100 pigs in an area of Buan-gun, Jeonbuk Province during July 2009. As a result, the detection rate of C. jejuni and C. coli and the rate of erythromycin-resistant bacteria for the separated Campylobacter species on the condition of high temperature were investigated. The possession or not of hipO and glyA gene and ciprofloxacin-resistant gene gyrA was also reviewed with biochemical characteristics and PCR.

Rapid and Easy Detection of Porcine Epidemic Diarrhea Virus (PEDV) by in situ Hybridization (In Situ Hybridization에 의한 돼지 유행성설사증 (Porcine Epidemic Diarrhea)의 진단)

  • Molecular diagnostic techniques have been used to identify porcine epidemic diarrhea virus (PEDV), a causative agent of acute enteritis in swine, but they were difficult to be petformed and time-consuming. To detect PEDV in a rapid and easy way, we developed biotinylated cDNA probe for N gene encoding the nucleoproteins of PEDV. Formalin-fixed and paraffin-embedded tissues from 24 naturally infected pigs were used for the experiment. The ISH produced a positive reaction in all cases. When intestinal tissues were hybridized with PEDV probe, strong signals were seen in the villus enterocytes of the jejunum and ileum. Hybridization signals were also found in the duodenum from one pig and in colon from dnother. In conclusion, ISH with a biotinylated cDNA probe was provided to be a useful diagnostic method for detecting PEDV effectively in routinely processed tissue sections.
