Pie-establishment of Microwave-Assisted Extraction Conditions for Antioxidative Extracts from Cabbage (양배추의 항산화성 추출물 제조를 위한 마이크로웨브 추출조건 설정)

  • Microwave-assisted extraction (50 W, 2,450 MHz, MAE) with properties of selective heating and subsequent extraction for certain phytochemicals from natural materials was applied to pre-establish the extraction conditions for total yield total phenolics, and electron donating ability (EDA) from Brossica oleacea. The experiments with $50\%$ EtOH solvent showed that 20 mesh in particle size of cabbage flake $(moisture\;4.5\%)$ and 1:10 (g/mL) in the sample to solvent ratio for both raw $(moisture\;90.2\%)$ and flake cabbages were optimal for MAE efficiency. Under these conditions, total yield increased with extraction tim, which was highest for raw cabbage extract in $50\%\;EtOH$ solvent followed by $100\%\;EtOH$ and water. While that of flake cabbage extracts was highest in $50\%\;EtOH$ followed by water and $100\%\;EtOH$. The contents of total phenolics and EDA in extracts gradually increased after 3 min of MAE, which were highest when using $100\%\;EtOH$ solvent followed by $50\%\;EtOH$ and water in raw cabbage and $50\%\;EtOH$ followed by water and $100\%\;EtOH$ in flake cabbage, respectively.

Optimization of Extraction Conditions for Ethanol Extracts from Citrus unshiu Peel by Response Surface Methodology (반응표면분석에 의한 진피 에탄올 추출물의 추출조건 최적화)

  • This study was conducted to monitor the quality characteristics Citrus unshiu ethanolic extracts by a response surface methodology. In extraction conditions based on the central composite design with variations in ethanol concentration (0~100%), extraction temperature($35\sim95^{\circ}C$), and ratio of solvent to sample(20 mL/g). The maximum value of yield was 36.31% at $93.18^{\circ}C$, 67.13% of ethanol concentration. The extraction yield and total polyphenol content were improved with the increase of ethanol concentration than extraction temperature. Total flavonoid content was improved with the increase of extraction temperature than ethanol concentration. The coefficients of determinations($R^2$) were 0.8646(p<0.1) and 0.9153(p<0.05) in electron donating ability and hesperidin content, respectively. Estimated conditions for the maximized extraction including yield, total polyphenol content, total flavonoid content, electron donating ability, and hesperidin content were 55~73% in ethanol concentration, $89\sim95^{\circ}C$ in extraction temperature, and 20 mL/g in ratio of solvent to sample.

Effect of Light with Different Wavelengths on Nostoc flagelliforme Cells in Liquid Culture

  • The effects of lights with different wavelengths on the growth and the yield of extracellular polysaccharides of Nostoc flagelliforme cells were investigated in a liquid cultivation. N. flagelliforme cells were cultured for 16 days in 500 ml conical flasks containing BG11 culture medium under $27{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ of light intensity and $25^{\circ}C$ on a rotary shaker (140 rpm). The chlorophyll a, phycocyanin, allophycocyanin, and phycoerythrin contents in N. flagelliforme cells under the lights of different wavelengths were also measured. It was found that the cell biomass and the yield of polysaccharide changed with different wavelengths of light. The biomass and the yield of extracellular polysaccharides under the red or violet light were higher than those under other light colors. Chlorophyll a, phycocyanin, and allophycocyanin are the main pigments in N. flagelliforme cells. The results showed that N. flagelliforme, like other cyanobacteria, has the ability of adjusting the contents and relative ratio of its pigments with the light quality. As a conclusion, N. flagelliforme cells favor red and violet lights and perform the complementary chromatic adaptation ability to acclimate to the changes of the light quality in the environment.

  • Room and high temperature deformation behaviors of Cu-Zr-Ti-Pd bulk metallic glasses produced by copper mold casting were investigated. The addition of Pd was shown to enhance the glass forming ability and thermal stability of Cu-Zr-Ti base bulk metallic glass. The compressive strength of $Cu_{55}Zr_{30}Ti_{10}Pd_5$ bulk metallic glass was 2230 MPa with 1.8 plastic strain. The stress overshoot and yield drop phenomenon were observed below $487^{\circ}C$ and a drastic decrease in the flow stress was observed at $487^{\circ}C$. The stress overshoot is thought to be associated with stress-induced structural relaxation.

Functional Properties of Pine Needle Extract and Its Antioxidant Effect on Soybean Oil (솔잎 추출물의 기능적 특성과 대두유 산화억제 효과에 관한 연구)

  • To investigate the functional properties and the antioxidant effect of pine needle(Pinus densiflora), the pine needle extract was obtained with methanol and its functionality was measured by spectrophotometric method, and the antioxidant experiment on soybean oil was carried out by the active oxygen method. The extraction yield of pine needle with 99% methanol was about 19%, the total flavonoid content of the pine needle-methanol extract was 11.32 mg/g on dry basis and the superoxide dismutase-like activity was 94.3%. The nitrite scavenging ability of the extract was pH dependent with the values of 77.44% at pH 1.2, 48.45% at pH 3.0 and 11.04% at pH 6.0, respectively. The peroxide value was 92.6 meq/kg at 5% dosage, 138.4 meq/kg at 2% dosage of the extract on 8 oxidation days. The period of the peroxide value to be 100 mg/kg was 4.9, 6.3 and 8.5 days at control, 2% and 5% dosage of extract, respectively. And the relative antioxidant effectiveness of the extract was 27.9% and 72.3% increase at 2% and 5% dosage, respectively, compared to control. The thiobarbituric acid value showed few differences within 4 oxidation days, but with the dosage of the extract it fairly decreased with considerable antioxidant effect to control above 4 days.

  • The existing conjugated blue emitting material polymer which has been used for the two-wavelength method white-emission has good stability and low operating voltage as merits, but the imbalanced carrier transport has been indicated as problem area. We have introduced a novel blue emitting material having perylene moiety unit with hole transporting ability and blue emitting property and triazine moiety unit with electron transporting ability into the same host chain. We have synthesized N-[p-(perylen-3-y1)pheny1]methacry1 amide (PPMA) monomer and [N-(2,4-dipheny1-1,3,5-triazine)pheny1 methacry1 amide] (DTPM) monomer having blue light-emitting unit and electron transport unit, respectively by three steps. A novel non-conjugated blue emitting material Poly[N -[p­(perylene-3-y1) pheny1] methacry1 amide-co-N-[P-(4,6-dipheny1-1,3,5-triazine-2-y1]pheny1]methacry1 amide] (PPPMA-co-DTPM) copolymer having electron transporting unit was synthesized by the solution polymerization of PPMA and DTPM monomers with an AIBN initiator and showed high yield of $75{\%}$. It was very soluble in common organic solvents, and the fabrication of the thin film using a spin coating method was very simple. The PPPMA exhibited a good thermal stability.

Breeding of a New Silkworm Variety, Chugangjam, with a Sex-Limited Larval Marking and High Silk Yielding for Summer-Autumn Rearing Season

  • A new silkworm variety, Chugangjam, with a sex-limited larval marking and high silk yielding for summer-autumn rearing season is $F_1$ hybrid between Jam 147, a Japanese race bred from P8503/8453, and Jam 148, a Chinese race from M8306/Jam 130. Jam 147, Japanese parent of the Chugangjam showed a high GCA (general combining ability) in pupation percentage and Jam 148, Chinese parent, showed a high GCA in pupation percentage and single cocoon weight. In the local adaptability test performed at 8 local areas in autumn of 2001, Chugangjam was 5% higher in cocoon yield and 8% in raw silk productivity in the 10,000 of the 3rd molted larvae, respectively, than the check variety Daesungjam.

Physiological Activities of Roots Extracts from Calystegia japonica

  • This study was investigated to evaluate the contents of polyphenols and flavonoids, and physiological activities of various extracts from Calystegia japonica roots for making good use of their functional materials. The roots of C. japonica were extracted with water (WE), ethanol (EE) and hot water (HWE) by different methods. Among these extracts, the highest extracting yield was 30.30% of HWE, while the highest contents of total polyphenols and flavonoids were 40.85% and 6.40% of WE, respectively. The nitrite scavenging abilities were ranged from 31.31% (HWE) to 37.46% (EE) at pH 1.2 and 1.0 mg/ml concentration. In the measurements of electron donating abilities, EE showed the highest effect as 91.83% at 0.3 mg/ml assay concentration, and the electron donating ability was decreased as the extract concentration was increased. In the results of superoxide dismutase (SOD)-like activity, HWE showed the highest effect as 7.15% at 1.0 mg/ml. The tyrosinase inhibition activities of WE and EE were 15.28% and 14.97%, respectively. The xanthine oxidase inhibitory effects were ranged from 97.50 to 99.28% at 1.0 mg/ml. These results indicate that C. japonica extract has a good antioxidant effects and could be useful for developing functional products.

An Improved Method of Parthenogenetic Development and Analysis of Combining Ability in Bivoltine Breeds of the Silkworm, Bombyx mori L.

  • Parthenogenesis, the development of unfertilized ovum opens new perspectives in silkworm breeding in the development of homozygous breeds. In order to improve induction of artificial parthenogenesis in the excised unfertilized eggs of different breeds of the silkworm, Bombyx mori L., a new method was devised and the results were compared with the routine method. General and specific combining abilities and hybrid vigour of newly developed bivoltine breeds were analyzed utilizing bivoltine breeds viz., $CSR_2,\;CSR_4,\;CSR_{17}\;and\;NB_4D_2$ Estimation of GCA revealed superiority of the breeds, $DNB_1$ for eight characters followed by $DNB_4$ for five characters. Among the testers, $CSR_2$ was found good general combiner for seven characters followed by $CSR_{17}$ for four characters. A great deal of variations was observed among the hybrids studied. Five hybrids namely, $DNB_1{\times}CSR_2,\;DNB_4{\times}CSR_4,\;DNB_4{\times}NB_4D_2,\;DNB_6{\times}CSR_2\;and\;DNB_7{\times}CSR_2$ showed significant SCA effects for 5-6 characters. The hybrid, $DNB_4{\times}CSR_4$ showed its superiority by expressing significant hybrid vigour over BPV for 7 characters. Majority of the hybrids exhibited significant hybrid vigour for survival rate, yield/10,000 larvae by weight, cocoon weight, cocoon shell weight, filament length and denier.

Analysis of Pheasant Carcass and Sensory Characteristics of Pheasant meat Products (꿩의 도체분석 및 꿩고기 가공제품의 관능 특성)

  • To develop processed meat products of pheasant, cut-up parts of pheasant carcass was analyzed, and eight different pheasant meat products were prepared and evaluated for sensory qualities. The average live weight of pheasant was 1,089.2g, and the ratio of carcass to live weight was 75.6%. The cut-up part ratios of breast, leg, neck, back and wings to carcass weight were 33.4%, 22.5%, 5.0%, 4% and 9.5%, respectively. The chemical compositions of breast and leg meat were shown to be moisture of 73.72% and 75.58%, protein of 25.31% and 22.69%, fat of 0.28% and 0.83%, and ash of 0.84% and 0.90%, respectively. Sensory evaluation of eight different meat products of pheasant revealed that all products of pheasant meat, except frankfurt sausage, received equal or better taste score compared with products of chicken or pork, and flavor score except pressed ham and salad. Color, binding ability and particle perception scores of pheasant meat products were equal or superior to those of respective meat products of chicken or pork. The present results suggest that pheasant meat has a potential to be utilized for various value-added products and that the meat bun is the most promising product of pheasant meat. A reinforcement of color, binding ability and particle perception of meat products of other species could also be expected by addition of pheasant meat to them.
