Effects of Enhanced Light Transmission Rate During the Early Growth Stage on Plant Growth, Photosynthetic Ability and Disease Incidence of Above Ground in Panax ginseng (생육 초기에 투광량 증가가 인삼생육 및 지상부 병 발생에 미치는 영향)

  • This study was performed to investigate the effects of enhanced light transmission on plant growth, photosynthetic ability, and disease tolerance to leaf blight, anthracnose in ginseng (Panax ginseng C. A. Meyer, Araliacease family) during the early growth stage (April to June). The photosynthetic ratio, stomatal conductance, and stem diameter of plants grown under a shade net with 15% light transmission rate showed an increasing trend compared to the control plants (5% light transmission rate) although the growth of the aerial parts were not influenced significantly. Plant height, stem length, and leaf length of treated plants were not significantly different from those of the control plants. Root parameters, such as root length, diameter, and weight of treated plants increased significantly compared to the control. Yield performance ($187.4kg{\cdot}10a^{-1}$) of treated plants was 55.5% higher than that of the control ($150.4kg{\cdot}10a^{-1}$). Additionally, disease severity scores of treated plants were lower than those of the control plants, revealing higher survival rates. To retain high yield potential and enhance the level of disease tolerance in ginseng, we suggest the increase of light transmission rate during the early growth stage.

  • A new high yielding mulberry genotype, Victory-1 (V-1) was recommended to the field for irrigated areas by Central Sericultural Research and Training Institute (CSRTI), Mysore during 1996-1997. As K-2 and S-36 mulberry genotypes are already in the field, a study on the comparative performance of V-1, 5-36 and K-2 varieties, in terms of leaf yield, yield attributes, leaf moisture and moisture retaining capacity and their impact on silkworm rearing was carried out under Telangana conditions of Andhra Pradesh during 1999-2001. Among the three genotypes the leaf yield was found to be significantly (P < 0.01) higher in V-1 (64,130 kg/ha/yr) followed by S-36 (44,064 kg/ha/yr) and K-2 (23,990 kg/ha/yr). The maximum leaf moisture was also observed in V-1 (71.84%) compared to S-36 (66.86%) and K-2 (64%). The moisture retaining ability was more in V-1 (70.17%) followed by S-36 (66.20%) and K-2 (60.08%). The yield attributes were found to be significantly (P < 0.01) enhanced in V-1 over S-36 and K-2. Silkworm rearing results (bioassay) indicated a significant (P < 0.01) reduction in larval duration and improvement in larval weight, effective rate of rearing by number and weight, single cocoon and shell weights and shell ratio in the silkworm lots fed on V-1 variety followed by S-36 and K-2. Hence, in this study, V-1 was found to be superior among the three varieties studied.

  • The present studies were carried out to obtain some basic informations of the breeding of tobacco varieties. Genetically divergent 8 varieties, 3 flue-cured, 2 burley and 3 sun-cured tobaccos, were used in half diallel cross. In order to analyze the heterosis, combining abilities, modes of inheritance and correlations for some agronomic and chemical characters, 8 parents and 28 $F_1$ were tested. The results obtained were summarized as follows: The percentages of heterosis for stem diameter, internode length and total sugar content in $F_1$ hybrid were 3.6%, 3.1% and 10.6%, whereas these for days to flower, total alkaloids and leaves per plant were -6.3%, -6.9% and -5.0%, respectively. Yield had significant positive genotypic correlations with plant height, days to flower and leaf length, but negative with internode length and total sugar content. It also had significant prositive phenotypic correlations with plant height, days to flower, leaves per plant, leaf length, leaf width and leaf shape index (Leaf length/leaf width). General (GCA) and specific combining abilities (SCA) for all characters of $F_1$ hybrid were significant. The effects of GCA were positive on yield, plant height, stem diameter, leaves per plant and days to flower of Burley 21. And those were positive on yield, leaf shape index and plant height, but negative on leaves per plant and total nitrogen of Hicks. The effects of SCA for yield and leaves per plant were greater than those of others on the combinations of Coker 139 and Burley type, respectively.

Characteristics in Tissue Cultured Plants of Chinese foxglove(Rehmannia glutinosa) (지황 조직배양주의 수량성과 성분함량 특성)

  • Chinese foxglove(Rehmannia glutinosa) is receiving much attention as one of the principal medicinal crops and the crud drug. This study was conducted to obtain the basic breeding information of Chinese foxglove derived from tissue culture. To compare plant characteristics between local variety and tissue cultured seedlings, plant growth and root yield has been investigated. In addition, catalpol and free sugar contents were also analized. The ability for the storage of root stock originated from tissue culture seedlings were better than that of local variety. The growth and root yield of in vitro propagated plants were superior to those of conventionally propagated plants. Root yield of 1-year-old and 2-year-old seedlings investigated 112% and 246% respectively than Seacheon local shows 384 kg/10 a. Although there were no significant different between tissue culture seedlings and conventionally propagated one, slight decline of component contents in tissue culture plants were still existed. Investigated sugar content among Seocheon local, 1-year-old tissue culture seedlings, and 2-year-old tissue culture seedlings were 1.86%, 1.21%, and 1.10% respectively. Catalpol content as one of standard materials indicates 0.46% in Seocheon local and 0.40% in tissue cultured ones.

Effects of Higher Dietary Cation with or without Protected Fat and Niacin on the Milk Yield and Thermoregulatory Ability in Holsteins During Summer Heat Stress (여름철 고온스트레스 기간에 고 양이온 사료와 반추위 보호지방과 나이아신의 추가공급시 착유우의 유생산 및 체온조절에 미치는 영향)

  • The objective of this study was to evaluate the effects of dietary cation-anion difference (DCAD) with or without ruminally protected fat and niacin on the thermoregulatory ability, milk yield and milk composition of lactating dairy cows during summer in Korea. Thirty mid-lactating Holstein cows (134±12.4 DIM and 23.4±2.3kg/d of milk yield) were divided into three groups (10 animals/group). Cows were housed in a free-stall barn and were provided with forced- air ventilation (wind velocity = 4 m/s) using 41 cm diameter fans. Diet one was formulated to contain low DCAD (+15 DCAD) while the remaining two diets were higher in DCAD (+30 DCAD). One higher DCAD diet was formulated to contain by-pass fat and the second higher DCAD diet contained the niacin along with by-pass fat. The maximum ambient temperature during July was 28.5℃ which could be seen as a period of mild heat stress. As summer progressed, August was characterized as a severe heat stress condition with maximum ambient temperature (32.4℃) and THI (74.0). Dry matter, crude protein and total digestible nutrients intake was not affected by the DCAD level and supplementation of ruminally protected fat or niacin. Milk production was higher in cows fed diets supplemented with fat and niacin than those fed un-supplemented diet. No difference in milk yield was observed in cows fed diets supplemented with fat or niacin plus fat. Milk fat and rectal temperature were not affected by the DCAD level and supplementation of ruminally protected fat or niacin. However, respiration rate was decreased in cows fed diets supplemented with either fat or fat and niacin compared to those fed. The results of the present study indicated that higher DCAD (+30) and supplementation of fat along with niacin can somehow mitigate the negative effects of heat stress on milk yield and physiology of lactating Holsteins during July and August in Korea. In present study reduced respiration rate and increased milk yield in lactating cows may be attributed to the cooling effect of supplemented fat along with vasodilatory functions of niacin. Prediction of the Optimum Conditions for Microwave-Assisted Extraction of the Total Phenolic Content and Antioxidative and Nitrite-scavenging Abilities of Grape Seed (포도씨의 총페놀 성분, 항산화능 및 아질산염소거능에 대한 마이크로웨이브 추출조건 예측)

  • Response surface methodology (RSM) was used for the microwave-assisted extraction (MAE) of the effective components of grape seed, such as its antioxidative and nitrite-scavenging abilities. Microwave power (2,450 MHz, 0-160W), ethanol concentration (0-100%), and MAE time (1-5 min) were used as independent variables (Xi) for the central composite design to yield 16 different MAE conditions. The optimum MAE conditions were predicted for the dependent variables of the extracts, such as the total phenolic content ($Y_1$) antioxidative ability ($Y_2$), and nitrite-scavenging ability ($Y_3$), depending on different microwave powers, ethanol concentrations, and MAE times. The determination coefficients ($R^2$) of the regression equations for the dependent variables ranged from 0.8024 to 0.9498. The maximal values of each dependent variable were predicted at different MAE conditions, as follows: 3.19% total phenolic content at 142.32W, 44.30% ethanol, and 4.36 min, and 1.22 antioxidative ability at 84.44W, 56.60% ethanol, and 3.28 min. More than 99.5% nitrite-scavenging ability was predicted at pH 1.2-3.0, 30.80-106.58W, 49.32-55.18% ethanol, and 3.72-4.58min, respectively. The results indicated that the total phenolic content and anti oxidative ability showed a higher correlation with each other in that they were more influenced by microwave power than by the other variables, while the nitrite-scavenging ability was largely influenced by the ethanol concentration.

Antioxidative ability of ethanol extract from the leaves of Leucaena leucocephala (Lam.) de Wit (백연수(Leucaena leucocephala (Lam.) de Wit) 잎 에탄올 추출물의 항산화능)

  • Antioxidative ability of 50% ethanol extract from leaves of Leucaena leucocephala (Lam.) de Wit was investigated. The extraction yield, total polyphenols and total flavonoids of the ethanol extract were $18.3{\pm}0.40%$, $121.3{\pm}11.04{\mu}g$ gallic acid equivalents/mg and $105.2{\pm}2.77{\mu}g$ quercetin equivalents/mg, respectively. The ethanol extract possessed the antioxidative activities such as free radical, cation radical, and nitrite scavenging activities as well as reducing power and lipid peroxidation inhibitory activity. These antioxidative activities of the ethanol extract increased in proportion to the extract concentration. $EC_{50}$ values for free radical scavenging, cation radical scavenging nitrite scavenging, and reducing power were $133.51{\mu}g/mL$, $667.22{\mu}g/mL$, 2.58 mg/mL, and $250.00{\mu}g/mL$, respectively. Polyphenols and flavonoids are suggested to be the key contributors for the antioxidative ability of the ethanol extract from Leucaena leucocephala leaves.

Biological Activities of Maca (Lepidium meyenii) Extracts (마카 추출액의 생리활성 효과)

  • This study was conducted to determine the optimal extraction conditions for maca by comparing the yields, total polyphenol contents, superoxide dismutase (SOD)-like activity and the nitrite scavenging ability. The proximate composition analysis showed 6.57% moisture, 12.83% crude protein, 1.05% crude fat, 4.80% ash and 74.75% carbohydrate. Maca was extracted with 7 different solvents (water, methanol, ethanol, acetone, ethyl acetate, chloroform and hexane) and the extracts were tested for biological activities. The extraction yields of water, methanol and ethanol extracts were 46.2%, 21.4% and 16.8%, respectively. Acetone, ethyl acetate, chloroform and hexane exhibited very low extraction yield, ranging from 0.2 to 1.0%. Total polyphenol contents and the nitrite scavenging ability were the highest in water extract. Electron donating ability and the SOD-like activity were the highest in methanol extract. When water extract was drawn out at different extraction temperatures (30, 70, $100^{\circ}C$) and time (1, 3, 5 hr), the improved biological activities (total polyphenol contents, electron donating ability, SOD-like activity and nitrite scavenging ability) were found in extracts treated at $100^{\circ}C$ for 3 or 5 hrs.

A Study on the Physicochemical Properties and the Nitrite Scavenging Ability of Japanese Sumac Extracts (붉나무 추출물의 이화학적 특성과 아질산염 소거능에 관한 연구)

  • Japanese sumac (Rhus javanica) is one of the common herbaceous plants growing over the country. This study was conducted to investigate the extraction characteristics and physiological activities inluding nitrite scavenging ability of the water extracts from leaf, fruit and bark of Japanease sumac. Extraction yield was 6.62~13.84%, free amino acids were detected as 24 kinds with 37.9 mg/100g in leaf extract, 23 kinds with 27.0 mg/100g in fruit extract and 27 kinds with 39.0 mg/100g in bark extract, respectively, and seven kinds essential amino acids were detected. Total contents of flavonoids equivalent to naringin were 587.2 mg/100g in bark extract, 557.3 mg/100g in fruit extract and 379.9 mg/100g in leaf extract, respectively. Total contents of phenolics equivalent to gallic acid were 111.2 mg/100g in leaf extract, 108.4 mg/100g in fruit extract and 80.4 mg/100g in bark extract, respectively. The nitrite scavenging ability of extracts was order of 61.93% in bark extract>57.38% in fruit extract>55.49% in leaf extract, and was 78.1~100% of those of BHT (butylated hydroxytoluene) equivalents at pH 1.2. The electron donating ability was order of 47.38% in fruit extract>43.06% in leaf extract>38.55% in bark extract, and was compared to 65.6%, 58.8% and 53.6% of those of BHT equivalents, respectively. The reduction power was evaluated to 37% higher in leaf extract, 43% higher in fruit extract and 46% higher in bark extract than those of BHT equivalents. The metal chelating ability of extracts was considerably low and was order of 27.3% in bark extract>20.6% in leaf extract>11.2% in fruit extract.

Gene Amplification of aceA and aceB in Lysine-producing Corynebacterium glutamicum ssp. lactofermentum ATCC21799

  • The role of glyoxylate bypass in lysine production by Corynebacterium glutamicum ssp. lactofermentum ATCC21799 was analyzed by using cloned aceA and aceB genes which encode enzymes catalyzing the bypass. Introduction of a plasmid carrying aceA and aceB to the strain increased enzyme activities of the bypass to approximately 5 fold on acetate minimal medium. The strain with amplified glyoxylate bypass excreted 25% more lysine to the growth medium than the parental strain, apparently due to the increased availability of intracellular oxaloacetate. The final cell yield was lower in the strain with amplified glyoxylate bypass. These changes were specific to the lysine-producing C. glutamicum ssp. lactofermentum ATCC21799, since the lysine-nonproducing wild type Corynebacterium glutamicum strain grew faster and achieved higher cell yield when the glyoxylate bypass was amplified. These findings suggest that the lysine producing C. glutamicum ssp. lactofermentum ATCC21799 has the ability to efficiently channel oxaloacetate, the TCA cycle intermediate, to the lysine biosynthesis pathway whereas lysine-nonproducing strains do not. Our results show that amplification of the glyoxylate bypass efficiently increases the intracellular oxaloacetate in lysine producing Corynebacterium species and thus results in increased lysine production.
