Genetic Analysis of 5 Mountain Cultivated Ginseng and Wild Ginseng in Korea (국내 5개 지역의 장뇌삼과 산삼의 유전 분석)

  • ISSR PCR technique was applied to investigate genetic relationship among 5 Mountain cultivated ginseng populations (Jinan, Hongcheon, Punggi, Andong and Yeongju) and cDNA libraries of wild ginseng roots were constructed and analyzed functional genes related to morphogenesis via EST. Twenty four ISSR markers tested produced 127 polymorphic loci from 5 regional Mountain cultivated ginseng. Among the regional samples, Yeongju was made 18 polymorphic loci that were the highest level of variations among the cultivated regions. The range of similarity coefficient was 0.46~0.58 and the regional samples of Punggi and Hongcheon, Jinan and Andong were classified to similar groups respectively, whereas Yeongju was shown to be separate group with high level of genetic variation in UPGMA cluster analysis. As a result, there was no relationship according to geographical distance and genetic similarity. Eleven cDNA clones were consisted of 9 known genes and 2 unknown genes analyzed by BLAST program of NCBI. To recognize expression pattern of Homeodomain transcription factor related genes, Northern Blot analysis was performed for wild ginseng's leaf and root. As a result, the gene was only expressed by Mountain wild ginseng root.

Physicochemical Characteristics of Chokong and Soaking Solution on Soaking Period (담금기간에 따른 초콩과 담금액의 이화학적 특성)

  • Effects of soaking period on physicochemical characteristics of chokong (black soybean picked in brown rice vinegar) and soaking solution were investigated. The pH and soluble solid contents in soaking solution increased rapidly whereas total acidity decreased during the first 4 days of soaking. The sudden drop of color 'L' and 'b' values in soaking solution occurred one day after start of soaking; in contrast, 'a' value increased by 1 day, and then decreased for soaking periods. The strength and hardness of chokong showed a decreasing trend as soaking for a long time, and the values of chokong were lower than those of control (black soybean soaked in water and freeze dried). The protein bands of chokong for soaking appeared only under 24,000 Da of molecular weight by electrophoresis. The content of total amino acids of chokong pickled for 7 days was 86.86 mg%, about 3 times for 28.36 mg% of raw black soybean. The contents of essential amino acids, especially, leucine and phenylalanine increased greatly in chokong. In vitro digestibility for protein was 62% in raw black soybean and 84% in chokong pickled for 7 days. Therefore, these results may assumed that chokong will be good source of amino acids.

Effect of Salting Methods on Component and Quality Characteristics of Eggplants (가지의 절임방법에 따른 성분변화)

  • This study was carried out to analyze the physicochemical properties according to salting methods of eggplants exported mainly to Japan. Moisture contents of Chukyang and Shikibu were decreased after 3 days of salting regardless of salting methods. The decrease of moisture content in mixed treatment of rice husk and salt was higher than that of salt treatment after 7 days of salting. Ash contents of Chukyang and Shikibu were increased according to extended salting time regardless of salting methods. The Brix of Chukyang and Shikibu without salting were $4.0^{\circ}brix\;and\;5.0^{\circ}brix$, respectively, and were increased according to extended salting time regardless of kinds and salting methods. pH of Chukyang and Shikibu without salting were 6.04 and 6.00, respectively. Extended the salting time from 0 to 7 days did not result in a pH change. Reducing sugar contents of Chukyang and Shikibu were increased according to extended salting time, the increase of reducing sugar content in mixed treatment of rice husk and salt was higher than that of salt treatment during salting. Soluble protein contents of Chukyang and Shikibu without salting were 1.40 g and 1.85 g, respectively. Extended the salting time from 0 to 7 days resulted in gradual increase in soluble protein content of Chukyang and Shikibu.

Effects of Colchicine on Pulmonary Injury Induced by Paraquat (Colchicine이 Paraquat에 의한 폐상해에 미치는 영향)

  • This study was undertaken to investigate paraquat-induced pulmonary injuries and effects of colchicine on pulmonary fibrosis by paraquat. Fifteen Sprague-Dawley rats were intraperitoneally injected 10 mg/kg of paraquat and repeatedly with 2 days interval. Another 15 rats were injected paraquat as same manner and simultaneously injected 10 mg/kg of colchicine in a week. Five rats in each group were sacrificed 1, 2, and 4 weeks after initial injections, and lungs extracted were observed by light and electron microscopes. On light microscopy, there was mild infiltration of neutrophils, macrophages, and lymphocytes in alveolar spaces and walls at 1 week after paraquat injection. The cellularity of alveolar wall was increased with time. However, the cellularity was not so prominent in paraquat and colchicine simultaneously injected group. On electron microscopy, there was marked swelling or excoriation of type I epithelial cells and alveolar capillary endothelium with infiltration of neutrophils, macrophages and monocytes, and lymphocytes in alveolar walls. Such findings were persisted with time. In addition, fibroblastic proliferation and deposition of collagen fibers were prominent at 4 weeks after paraquat injection. Fibrosis also occurred at 4 weeks after paraquat and colchicine simultaneous injection. It was not proninent than that of paraquat injected group. According to the above result, it would be concluded that the type I pneumocytes and alveolar capillary endothelial cells are most vulnerable on paraquat poisoning, and that the colchicine is effective on inhibition of paraquat-induced pulmonary fibrosis.

Grape Pruning Stem Extract (GPSE) Suppresses Allergy and Skin Proliferation Inhibition against UVB Induced Skin Damage (포도전정가지 추출물이 UVB에 유도된 피부 손상에 대한 알레르기와 피부증식 억제효과)

  • This study examined the effects of polyphenols in grape pruning stem extracts (GPSE) using grape stems discarded after harvest. The inhibitory effects on allergy, proliferation, and apoptosis in UVB-induced HR-1 hairless mice were analyzed. The applicability as a material for functional food and functional cosmetics was evaluated. The contents of the active ingredients of GPSE were analyzed by HPLC. After UVB irradiation on the dermis of HR-1 hairless mice, the number of mast cells was determined by toluidine blue staining to confirm that the skin allergic reaction was caused by GPSE. The results were similar to the normal group in the group applied GPSE, and there was no allergic reaction in the GPSE application group and a significant decreased compared to the sun cream control. PCNA immunohistochemical staining of the epidermal proliferation factor had an inhibitory effect on epidermal epithelial cell proliferation in all concentrations of GPSE and serum base mixture as an average of 42% compared to the control group. The mixture of GPSE and serum base suppressed the apoptosis inhibition rate by 27% on average compared to the control. These results confirmed the inhibitory effects GPSE on the allergic, proliferation, and apoptosis activities by with a serum base on UVB-induced skin damage. GPSE is a functional ingredient with potential skin protection effects, and has a high utilization as an ingredient for functional cosmetics.

Antimutagenic Study on Acanthopanax Koreanum Nakai (섬오가피에 대한 항돌연변이원 시험)

  • This study was undertaken to investigate the mutagenicity and antimutagenicity of Acanthopanax koreanum Nakai. Antimutagenic study on extract of A. koreanum was studied using the test with Salmonella typhimurium TA100, TA98. And mutagenicity study was studied using the test with S. typhimurium TA100, TA98, TA1535, TA1537 and Escherichia coli WP2 uvr A. A. koreanum was negative in Ames test with S. typhimurium and E. coli with or without S-9 mixture. Test substances of $5000\;{\mu}g/{\mu}l$, $2500\;{\mu}g/{\mu}l$ and $600\;{\mu}g/{\mu}l$ of A. koreanum extracts were chosen via toxicity test. Ames test was performed on positive control group, experimental group and negative control group in the presence of the metabolic activation system and metabolic non-activation system. As a result, there was no coherent increase and reverse mutation in all concentrations. Therefore, A. koreanum does not cause reverse mutation. In addition, A. koreanum showed strong antimutagenic activities in S. typhimurim TA100 and TA98. In conclusion, A. koreanum root may be an excellent antimutagenic agent.

A Survey of Consumer's Consumption Characteristics of Environmental-Friendly Agricultural Products (EFAP) -Focused on Consumers in Discount Stores- (친환경농산물 소비행태 조사 -대형할인마트 고객을 중심으로-)

  • We investigated the consumption inclinations of consumers for environmentally-friendly agricultural products (EFAP) for better production and distribution of EFAP. Consumer inclinations in large supermarkets from April 7th to 30th, 2008 were surveyed. Data were analyzed using the SPSSWIN 12.0 program. Results showed that 90.4% of consumers recognized EFAP and 76.4% of those who recognized the products had actually bought the products. Members of the "have purchased group" bought EFAP once a week because they believed these products were safe from hazards, such as pesticide. The amount spent on EFAP was less than 20% of the total food budget and the major consumed items were green vegetables and fruits. The sale location was mainly large supermarkets and agricultural cooperation outlets. Although many people were satisfied with EFAP from the viewpoint of nutritional and sensory qualities and safety compared with general agricultural products, they were notsatisfied with the price. Many consumers trusted that EFAP were safe and nutritional. Members of the "not purchased group" also trusted the nutritional and sensory qualities of EFAP and recognized the safety issue, but they were not satisfied with the price, compared with general agricultural products. Consequently, to succeed in the market, EFAP should maintain the confidence of the consumer while decreasing in price.

Effect of Feeding Broiler Litter and Bakery By-product Ration on Production, Economy and Meat Quality of Growing Hanwoo Steers (육계분-제과부산물 혼합사료 급여가 육성 거세 한우의 생산성, 경제성 및 육 특성에 미치는 영향)

  • This study was conducted to determine the effect of feeding a mixture of broiler litter and bakery by-product on production, economy and meat characteristics of growing ‘Hanwoo’ steers. A control diet (conventional formulated feed and rice straw) and a treatment diet(90% mixture of broiler litter 64% and bakery by-product 36% and 10% rice straw on DM basis) were prepared on an isoenergetic basis and fed to 20 steers for 12 weeks. Palatability of TMR by ‘Hanwoo’ was fair. Compared with feeding a control diet, feeding a treatment diet resulted in increased(P〈0.05) DM intake, similar OM intake, similar daily weight gain(0.75 vs 0.64 kg/d) and feed efficiency of DM(10.1 vs 12.5) and OM(9.0 vs 10.6), considerably(45%) reduced(P〈0.05) feed cost/gain, and similar meat characteristics. These results show that a mixed ration manufactured with broiler litter and bakery by-product may be potentially used for growing ‘Hanwoo’ steers.

Allopurinol Decreases Liver Damage Induced by Dermal Scald Burn Injury (피부 화상으로 유도된 간 손상에서 Allopurinol의 효과)

  • In order to investigate a pathogenesis of liver damage induced by skin burn, thermal injury was induced by scald burn on entirely dorsal surface in rats (total burn surface area $20\sim25\%$) except for inhalated injury. At 5 and 24 h after scald burn, biochemical assay and morphological changes in serum and liver tissue were examined. Skin burn increased liver weight (% of body weight, p<0.05) and the activity of serum aniline amino-transferase (ALT, p<0.05), in addition, the activity of xanthine oxidase (XO), an enzyme of oxygen free radical generating system, was elevated (p<0.01) in serum, but not in skin and in liver. Postburn treatment of allopurinol intraperitoneally decreased liver weight, serum ALT activity and serum XO activity. Scald burn induced ultrastructurally swelling of endoplasmic reticulum, ribosome detachment, accumulation of lipid, dilatation of bile canaliculi and intercellular space, neutrophil infiltration, activation of Kupffer's cells and degeneration of hepatocytic microvilli. Futhermore , thermal injury decreased not only the protein concentration in plasma but also the number of intravascular leukocytes, that indicates induction of edema formation with protein exudation and inflammation by neutrophil infiltration into the internal organs. However allopurinol injection after burn inhibited post burn ultrastructural changes. These data suggest that acute dermal scald burn injury leads to liver damage, that is related to elevation of xanthine oxidase activity in serum. Xanthine oxidase may be a key role in the pathogenesis of liver damage induced by skin burn.

Economic Evaluation System for Deteriorated Military Facilities (노후 군시설물의 경제성 평가 시스템)

  • Given both structural safety and economic benefits of aging facilities, remodelling of the existing facilities is preferable to reconstruction. This recognition provides an opportunity to reduce the commitment of resources and national budget. However, when a subordinate troop asks for remodeling or reconstruction of a deteriorated facility, it is difficult to ensure the consistency and objectivity in the process of decision making for the alternatives due to the absence of systematic and quantitative rating methodology. Their economic evaluation methodology only exists in a manual format. Thus, further research is required for converting the methodology into an automated system in view of practicality such as rapid and accurate data processing. The contributions of this study are as follows: 1) Literature review found out a representative economic evaluation model focused on military facilities, and comparative analysis with a similar study identified the strength and weakness. 2) this study presented how to convert the theoretical framework which enables to solve a specific subject matter into an automated system. 3) it developed a user friendly interfaces which consist of four functional modules by considering the usability and accessibility of the system user. 4) the developed system was verified by a case study in terms of four kinds of performance indicators.
