Pathogenicity of the fish nodavirus causing viral nervous necrosis of sevenband grouper, Epinephelus septemfasciatus (능성어, Epinephelus septemfasciatus의 바이러스성 신경괴사증 바이러스의 병원성 연구)

  • The pathogenicity of the fish nodavirus causing viral nervous necrosis (VNN) of sevenband grouper, Epinephelus septemfasciatus was examined in sevenband grouper and other marine fish by intramuscular injection. Sevenband groupers of 27~104 g in body weight were highly susceptible to the fish nodavirus, but yellowtail (537 g in body weight), red seabream (207 g), rock bream (43 g), flounder (41 g), tiger puffer (27 g) and rockfish (94 g) of the sizes used to this experiment were not. The pathogenicity of the viral agent to the sevenband grouper was high without regard to fish sizes at rearing water temperature over $20^{\circ}C$, but not at $15^{\circ}C$. Therefore, susceptibility of sevenband grouper to the viral agent causing VNN was thought to be water temperature-dependent rather than fish size (age-dependent). Compared to the infectivity of the viral agent to sevenband grouper with artificial infection methods, fish were successfully affected by intramuscular, intraperitoneal, oral, dipping and cohabitation administrations although there were slight differences in mortalities among infection methods. And survival sevenband grouper after infection with the fish nodavirus was resistant to the reinfection for a long time.

  • The purpose of this study was to understand the arrangement of the religious food and religious diet in Kang-Rung region which was recogniged as cultural region. Investtigation was done by interview person to person, and the results are as follows. 1. The arrangement of religious food and regious diet in Kang-neung region were differed from home to home. 2. In Kang-neung region, Chu-suk religious ceremonies and religious diet were setted much religious diet than memorial service which was held on an anniversary of their's death. 3. At first line, they have arranged Hynbab and Kuk, or Papbaab and Miyegkuk or Paogikuk. On the other hand, Songpyen was placed in traditional standard setting. At second line, they have arranged Songpyen, Kijung, Aegeok, Yukgeok, Nurumgeok, DonBaegigeok, Memilgeok, and cooked octopus(muneo) on the Aegeok. They have also arranged fishes which were the cooked Alaska pollack(rnyongtai), codfish(daeku), flatfish (kajami), yellowtail(bangeo), salmon(yeoneo), trout(songeo) and Saegsunjeon, Dubujeon, Wanjajeon, Kokumajeon. At third line, they have arranged Yuktang, Sotang, Eotang, Altang which is boiled eggs. Altang was arranged instead of steamed chicken. At fourth line, they have arranged Bukeopo, Cuttlefish(dried Ojingeo), Gajamisikhe, Myungtaeposikhe. On the other hand, Sikhe was necessarily placed on the setting of traditional standard service for the ancestor. They have arranged also Chinese bellflower(Doragi), Fernbrake(Kosali), Spinach, Osmund (Kobi), Greenbean Sprouts, Leopard plant(Komchwi), Gourd(Pak), and Lentinus edodes (Pyokjo), but Kimchi was omitted. At fifth line, they have arranged Date, Chestnut, Persimmon, the fruit of Actinidia arguta(Dare), Heron, Banana, Kwajeul and Kangiung were considered as dishes. Dasik and biscut were also used for setting dishes.

Residues of Antibiotics in Wild and Cultured Fishes Collected from Coast of Korea (시중 유통 자연산 및 양식산 활어의 항생제 잔류)

  • Wild and cultured fish including olive flounder, sea bass, rock bream, yellowtail, gray mullet, gizzard shad, black rockfish, red seabream and squid were collected from a fish market located on the coast of Korea, and the antibiotic content of their muscle was investigated. Tetracycline group antibiotics were not detected in the 108 individuals of 9 species of wild fish. However, oxytetracycline (OTC) and tetracycline(TC) were detected in some samples of the 111 individuals in 7 cultured live fish species. The detected ranges of OTC and TC were ND~ 0.06 and ND~ 0.03, respectively. Five different fluoroquinolone antibiotics were also tested for, but were not detected in the wild fish species. Only small amount of criprofloxacin(ND~0.029 mg/kg) were detected in a few cultured fish samples. Oxolinic acid was not detected in either wild and cultured fish samples. Results showed that even very low levels of antibiotics could be detected by the testing methods used. Antibiotics were identified in a few fish samples but levels were far below the maximum allowable limits of the Korean Food Code, and the safety of fish being sold in markets, with regard to antibiotic levels, was confirmed.

Phylogenetic analysis of marine birnavirus (MABV) isolated from cultured starry flounder Platichthys stellatus and olive flounder Paralichthys olivaceus in Korea (양식 강도다리, Platichthys stellatus 및 넙치, Paralichthys olivaceus에서 분리한 marine birnavirus (MABV)의 phylogenetic 분석)

  • In this study, we have compared the genome of marine birnavirus (MABV) detected from starry flounder Platichthys stellatus and olive flounder Paralichthys olivaceus. A molecular analysis based on the nucleotide sequence (433 bases) of VP2-NS-VP3 region revealed that MABV (08-KU) from starry flounder showed 98% similarity with MABV Y6 isolated from Yellowtail Seriola quinqueradita in Japan (Accession no: AY283781) and with other aquabirnaviruses identify more than 76%. Comparison with MABV strains (06-KP, 08-KC) from olive flounder and MABV Y6 strain showed 97-98% sequence identities. Phylogenetic analysis was performed in order to examine the relationship among previously determined aquatic birnaviruses isolates showed that MABV and IPNV strains were classified into seven clusters. Three isolates from starry flounder and olive flounder in this study, belong to the genogroup VII including MABV Y6 strain and other aquabirnaviruses isolated from marine fish and molluscan shellfish in Japan. This report is the first description of a MABV from starry flounder in Korea.

Infection of Parasitic Anisakis Type Larvae (Nematoda) from Some Rockfishes, Sebastes spp. (볼락류 Sebastes spp.에 있어서 선충류 아니사키스 유충의 감염)

  • From February through March 2002, sixteen rockfishes representing three species were collected from Oregon Coast Aquarium and Newport fish market and examined far nematodes. During this study, a total of 157 Anisakis type larvae were recovered from Sebastes spp. and parasitized for an average infection prevalence of 50.0%. The yellowtail rockfish, S. flavidus, darkblotched rockfish, S. crameri, and canary rockfish, S. pinniger examined were infected with 110 Anisakis simplex larva, 30 Contracaecum spp. larva and 17 Pseudoterranova decipiens larva. Of the rockfishes examined, 75.0% carried Sebastes crameri, and 12.5% S. flavidus and S. crameri, respectively. The sites of infection far Anisakis type larvae were the intestine, omentum, and stomach walls. A. simplex larvae infected 60.0% on S. crameri and 33.3% on S.flavidus and S. pinniger respectively. The prevalence of Contracaecum sp. larvae and Pseudoterranova decipiens larvae on S. pinniger was 33.3%. The intensities of A. simplex larvae ranged from 1 to 87 in S. flavidus, S. crameri, and S. pinniger. The intensity of Contracaecum spp. larvae and P. decipiens larvae ranged from 0 to 30 in S. pinniger.

  • Marine fish aquaculture has been introduced on the East sea since 1964. The production volume of the marine aquaculture has been increasing drastically since 1990 with the technological improvements and the introduction of new species. However, the increasing costs of feed, seedlings and medical supplies and the decreasing prices due to the increase in the imports and domestic production have decreased the profits in the aquaculture business. Futhermore, the damage from the natural disasters such as red tides and typhoons has accelerated the exit from the business. Even though managing abilities to overcome the business crisis caused by the above factors are required, business managers based on the small scale family businesses do not have enough managing abilities to deal with the difficulties in the business. On the other hand, advanced countries in the marine aquaculture such as Norway have reached the stage where the managing abilities of marine aquaculture are similar to those in the manufacturing industry. And the number of large scale aquaculture farms with developed technologies and advanced marketing strategies in those countries is increasing. Considering that the marine aquaculture in Japan under the similar fishery systems of Korea has developed the state-of-the-art management skills or lead to large scale management, it is difficult to expect the decrease in the production costs under the small scale family business in Korea and this will lead to the decreasing competitive advantage over the imported seafood. Therefore marine aquaculture in Korea needs to increase the economy of scale to acquire the competitive advantage. This study addresses the possibility of introducing the large scale aquaculture system in Korea by analysing the factors and process of leading to large scale businesses in the aquaculture in Japan.

Functional characterization and expression analysis of c-type and g-like-type lysozymes in yellowtail clownfish (Amphiprion clarkii)

  • Lysozymes are well-known antibacterial enzymes that mainly target the peptidoglycan layer of the bacterial cell wall. Animal lysozymes are mainly categorized as g-type, c-type, and i-type based on protein sequence and structural differences. In this study, c-type (AcLysC) and g-like-type (AcLysG-like) lysozymes from Amphiprion clarkii were characterized in silico via expressional and functional approaches. According to in silico analysis, open reading frames of AcLysC and AcLysG-like were 429 bp and 570 bp, respectively, encoding the corresponding polypeptide chains with 142 and 189 amino acids. Elevated expression levels of AcLysC and AcLysG-like were observed in the liver and the heart tissues, respectively, as evidenced by quantitative real-time polymerase chain reaction assays. AcLysC and AcLysG-like transcript levels were upregulated in gills, head kidney, and blood cells following experimental immune stimulation. Recombinant AcLysC exhibited potent lytic activity against Vibrio anguillarum, whereas recombinant AcLysG-like showed remarkable antibacterial activity against Vibrio harveyi and Streptococcus parauberis, which was further evidenced by scanning electron microscopic imaging of destructed bacterial cell walls. The findings of this study collectively suggest the potential roles of AcLysC and AcLysG-like in host immune defense.

Pilot Study on the Introduction of Stationary Fishery in Coastal Waters of Ulleungdo Island, thd East Sea of Korea (울릉도 해역의 정치성 구획어업 도입을 위한 시험 연구)

  • In this study, pilot study on the introduction of stationary fishery was performed to solve the problem of fisheries resource reduction. The Fyke net, which is a test fishing net was selected considering the environment, operation and management costs of Ulleungdo, conditions that can be operated by small fishing vessels and personnel. As a result of 11 times survey using Fyke net from April to May 2017, 2,735 individuals and 983.4 kg caught and the dominant species were red seabream, yellowtail, olive flounder, mitra squid, horse mackerel, filefish, etc. In conclusion, if the production of squid, which is one of the major fisheries resources of Ulleungdo, is continuously decreased, it is considered that introduction of small-scale stationary fishery such as Fyke net would be useful as a means replace income of fishermen.

Screening and prevention of the mutagenicity for fishes accordind to cookery and storage (어류의 가열조리 및 보존에 의해 생성되는 변이원성 물질의 정량적 해석과 제어법)

  • This study was performed to measure the mutagenicity of fish by cooking and storage. Mutagenicity of the fish extract was measured by Ames test(Salmonella typhimurium reversion assay with TA 100) in vitro and by micro-nucleus test in vivo. The fish samples screened in this study were white fish(Trichiurus, Croaker, Salted Croaker) and red fish(Saury pike, Mackerel, Yellowtail, Salmon). The number of revertants of red fish were significantly higher than that of white fish. And the mutagenicity of mackerel was higher than other red fish, so followed experiment was made by using the extract of mackerel. Mutagenicity of the samples cooked on microwave oven was the lowest, whereas there was no significant difference between the samples cooked on gas grill and the ones on electric grill. In the presence of S9 mixture, the methanol extract of mackerel showed 2∼4 times high values of mutagenicity in comparison with the extract without S9. The extract of mackerel cooked with various vegetable juices showed inhibitory effects on the mutagenicity in the order of green tea, ginger, and radish. Also, the number of revertants was increased in the stored samples. Mutagenicity of the samples stored in the refrigerator was higher than that of the freezer. In micronucleus test, the methanol extract treated with vegetable juice inhibited micro-nucleus formation in bone marrow by cyclophosphamide in the order of ginger, green tea, and radish. In TBA test, there was a tendency that TBA values were increased as the storage time increased. Also, the rancidity of sample were stored in the refrigerator was higher value than sample stored in the freezer. Samples cooked on microwave oven showed the highest value in rancidity. When the antioxidant effect of vegetable juice was measured by electron donating ability(EDA) of mackerel cooked with vegetable juice to DPPH, the samples treated with onion showed the highest value of EDA(%), and the samples treated with green tea, ginger and cabbage also showed the antioxidant effect.

A Study on Red Tide Control with Loess Suspension (부유황토에 의한 적종방제 연구)

  • As one of the red tide control method, montmorillonite was used to eliminate the causative organisms in Korea and Japan. We assayed the loess to replace the montmorillonite because it distribute in large quantity and nearby the red tide occurrence in South Coast of Korea. By using the mixture of loess and coal ashes, we examined the decreasing level of nutrients such as ammonia and phosphate, the elimination of causative organisms as a chlorophyll a content, and the harmful effect on aquaculture orgarnisms in cage culture farms. Half of the ammonium and phosphate was adsorbed by the loess particles, but only $25\%$ of ammonium was adsorbed by the coal ashes particles. In water column test, the particles of loess and coal ashes were settled down by $80\%$ in 20 minutes, the red tide organisms was eliminated by $80\%$ after 2 hours in 1,000 ppm of loess suspension, but the organisms were eliminated only $30\%$ by the same concentration of coal ashes. The harmful test of fishes and invertbrates, we observed any other negative effects of test animals than a tint deceleration in yellowtail.
