Production of Salidroside in Rhodiloa sachalinensis A. Bor Callus by the Elicitation and Precursor (참돌꽃 (Rhodiola sachalinensis A. Bor) 캘러스에서 elicitor와 전구체에 의한 Salidroside 생산성의 변화)

  • The effect of elicitor and precursor on salidroside production from Rhodiola sachalinensis A.Bor callus cultures was investigated. Callus cultures were treated with yeast extract, soft-ferrite ceramics powder, methyl jasmonate, ascorbic acid, jasmonic acid and $CuCl_2$/$CdCl_2$ as an elicitor. When callus cultures were treated with $0.2g/\ell$ of yeast extract, salidroside production from callus treated with yeast extract is 3.45 times higher than that of the controlled group. Among of them, callus cultures treated with yeast extract produced the highest salidroside. Callus cultures were treated with L-phenylalanine and L-tyrosine as a precursor for 4 days. The result of salidroside content analysis showed that all feeding of precursors not affected salidroside production from callus cultures. In case of L-tyrosine fed into callus cultures, both callus growth and salidroside production decreased at all concentrations.

Elicitor-induced Phenylalanine-Ammonia Lyase, Cinnamic Acid 4-Hydroxylase and $rho-Coumaroyl$ transferase Activity in Ephedra Distachya Cultures

  • Ephedra olistachya cultures have been known to accumulate $rho-coumaroylamino$ acids by elicitor treatment. Based on their chemical structures, the biosynthetic pathway of$rho-coumaroylamino$acids was postulated and phenylalanine ammonia-lyase (PAL), cinnamic acid 4-hydroxylase (4-CH) and p-coumaroyl CoA: D-Ala p-coumaroyltransferase ($rho-CT$) were supposed to be involved in the pathway. The time course inductions of these enzymes were investigated after treatment of yeast extract, yeast-derived mannan glycopeptide and D-Ala. They were detectable at only 4 hours and reached to their maximum level at 9 hours after onset of elicitor treatment. The activities of PAL and 4-CH were almost disappeared within 24 hours, however, that of $rho-CT$was remained up to 48 hours irrespective of the kind of elicitors. $rho-CT$ showed substrate specificity to D-Ala at crude enzyme extract level.

  • Effects of various elictitors were investigated to enhance the production of berberine in plant cell suspension cultures of Thalicrtrum rugosum. Treatments of yeast elicitor, 15 different types of abiotic elicitors, 16 kinds of fungal elicitors from three species of fungi were performed. Cell growth and berberine production were examined and compared for both normal and elicitor treated cultures. No distinguished increases in berberine yield by the addition of elicitors could be attained.

Characteristics of Growth and Oil Production of Peppermint Cells in an Air-bubble Bioreactor (기포 생물반응기에서 페퍼민트 세포의 생육 및 정유 생산 특성)

  • To investigate the characteristics of growth and oil production of peppermint cells during a batch culture, cells derived from peppermint callus was cultivated in an air bubble reactor. During the batch culture, effects of inoculum size, abiotic stress, yeast elicitor, and two stage culture on the cell growth, the productivity of oleolesin, and the formation of flavor components were determined and also the sugar concentrations and kinetics of cell growth were analyzed. Among the various sizes of inoculum, the culture with 2.0% packed cell volume inoculum showed the optimum condition for cell growth in the proposed bioreactor, and the cell yield and essential oil production reached to 5.7g/1 and 0.109g/1, respectively. When the abiotic stress of daily 8hr dark and $10^{\circ}C$ cold treatments were given to the culture cell growth decreased but essential oil production increased to 0.546g/l. In a modified Lin-Staba medium in which 100mg/l yeast extract as an elicitor was added to the culture, the cell growth and oil production increased, and menthol content was 22.5% of oil. In the two stage culture, in which the basic culture conditions of 27$^{\circ}C$, light, and without elicitor were employed during the first six days followed by the second stage with daily 8hr treatment of cold and dark condition, and also with yeast extract as an elicitor, cell growth decreased after eight days, essential oil production was not increased, and menthol was not detected. Dry cell yield was 0.38g dry cell/g sugar and specific growth rate was 0.25 day-1. The major terpenoid in the oil was not the menthol but pulegone and piperitone, precursors of menthol were accumulated. However, when yeast elicitor was added, menthol was produced to the level of 22.5% which was the highest value in the peppermint cell culture reported so far.

Elicitation and In Situ Recovery of Alkaloids in Suspension Cultures of California Poppy

  • Large and rapid increases in benzophenanthridine alkaloid production occured in suspension cultures of Eschscholtzia californica cells treated with elicitors. Response to different biotic elicitors showed that elicitors prepared from yeast extract, Collectotrichum lindemuthianum and Verticillium dahliae induced alkaloid formation. Highest alkaloid accumulation was obtained with $60\;\mu\textrm{g}$ of yeast extract elicitor per gram of fresh cell weight. In time course performance after elicitor addition, more than 40 hours were required to obtain saturated alkaloid accumulation. Compounded silicone fluid, an ideal accumulation phase for two-phase culture of E. californica, accumulated a large amount of alkaloids produced in a specific manner. Elicitation in two-phase culture clearly increased net alkaloid production as well as their concentrations in the accumulation phase.

  • The accumulation of benzophenanthridine alkaloids, sanguinarine, chelerythrine, chelirubine and macarpine occurred in suspension cultures of Eschscholtzia californica. To increase alkaloid production, feeding experiments with the biosynthetic precursors, tyrosine, tyramine, L-dopa, dopamine with and without elicitation were studied. In feeding experiments with various precursors, the total alkaloid production was slightly increased. The precursor feeding with elicitation, however, increased total alkaloid production several times.

Elicitors which Induce the Accumulation of p-Coumaroylamino Acids in Ephedra distachya Cultures

  • Some ammonium oxalate soluble pectic fragments prepared from cultured cell wall of Ephycla distrahya elicited the accumulation of p-coumarocylamino acids (p-CAA) in E. distachya cultures while water soluble and alkali soluble fractions had no activity. Partial purification of the pectic fragments fraction using DEAE-cellulose chromatography afforded two active fractions (PS-I and PS-II) which were composed of mainly uronic acids (98-99 w/w %). They elicited the accumulation of p-CAA in an amount of 52-60 nmol per gram fresh weight of cultures. The acidic sugar compositions of PS-I and PS-II were found to be galacturonic acid and glucuronic acid by TLC analysis. They were supposed to act as endogenous elicitors of p-CAA accumulation. In order to investigate the effect of ethylene on p-CAA accumulation, Ethrel, which is known as ethylene generator, and ACC(1-aminocyclopropane-1-carboxylic acid), a direct precusor of ethylene biosynthesis, were added to the culture. However, they did not glycopeptide elicitor [(Con A-II)], either. Consequently, no relationships between ethylene and p-CAA accumulation were recognized. Several tentative elicitors were teted for their activity. Commercial yeast glucan, $CuCl_2$, laminarin and laminariheptaose had slight activity whereas ${\alpha}$-methylmannopyranoside and commercial yeast mannan had no elicitor activity. ${\alpha}$-methylmannopyranoside which has been known as a tentative inhibitor of glucan elicitor in Glycine max did not affect on the elicitor activity of Con A-II.

  • Benzophenanthridine alkaloids - sanguinarine, chelirubine, macarpine, and chelerythrine are produced from Eschscholtzia californica (Californica Poppy, used as a sedative by Native Americans) and most of them are derived from dihydrosanguinarine. The properties of sanguinarine are the basis of its antimicrobial activity and its use in chemosurgery and skin cancer excision. For overproduction of sanguinarine from E. californica, yeast extract was used as elicitor and the elicited cell's metabolites were checked. Sanguinarine production was increased intracelluarly about 8 times in the cell and 5 times extracelluarly. We have peformed proteomic analysis of proteins sequentially extracted from E. califormica suspended cells which were cultured with elicitor, an increase of spot intensity was seen at 24 hours following elicitation. These proteins were separated by two-dimensional electrophoresis (2-DE). We found several spots that were expected to be related to benzophenanthridine alkaloids production by comparing the production profiles of metabolites such as sanguinarine. These results demonstrate the use of metabolite analysis as a tool for detecting target proteins related to metabolites production pathway.

Effects of Several Elicitors and Amino Acids on Production of Taxane Derivatives in Cultured Cells (수종의 Elicitor 및 Amino Acid가 배양세포내 Taxane 유도체의 생산에 미치는 영향)

  • To develop new elicitors inducing the high productivity of taxane derivatives, plant growth inhibitors, namely, maleic acid hydrazide, N-phosphomethyl glycine and succinic acid 2.2-dimethyl hydrazide, coconut milk and yeast extract were administrated in the cell suspension culture system of Taxus cuspidata, and the production of baccatin III were analysed. The effects of amino acid related with the biosynthesis of baccatin III were also examined in these culture system. As the results, a remarkable enhancement of baccatin III production was observed in the cultivation with coconut water and with maleic acid hydrazide.
