Administration of Phytoceramide Enhances Memory and Up-regulates the Expression of pCREB and BDNF in Hippocampus of Mice

  • This study was aimed at investigating the possible effects of phytoceramide (Pcer) on learning and memory and their underlying mechanisms. Phytoceramide was orally administered to ICR mice for 7 days. Memory performances were assessed using the passive avoidance test and Y-maze task. The expressions of phosphorylated cAMP response element binding protein (pCREB), brain-derived neurotrophic factor (BDNF) were measured with immunoblot. The incorporation of 5-bromo-2-deoxyuridine (BrdU) in hippocampal regions was investigated by using immunohistochemical methods. Treatment of Pcer enhanced cognitive performances in the passive avoidance test and Y-maze task. Immunoblotting studies revealed that the phosphorylated CREB and BDNF were significantly increased on hippocampus in the Pcer-treated mice. Immunohistochemical studies showed that the number of immunopositive cells to BrdU was significantly increased in the hippocampal dentate gyrus regions after Pcer-treatment for 7 days. These results suggest that Pcer contribute to enhancing memory and BDNF expression and it could be secondary to the elevation of neurogenesis.

포공영 기원식물의 mouse 기억력 개선효과 비교 (A Comparative Study of Memory Improving Effects of Taraxaci herba on Scopolamine-induced Amnesia in Mouse)

  • Objectives : The purpose of this study was to characterize the effect of the fraction of Taraxacum officinale and T.coreanum on the learning and memory impairments induced by scopolamine. Methods : The cognition-enhancing effect of Taraxacum officinale and T.coreanum was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : The results showed that the Aug harvested T.offiicinale extract-treated group (200 mg/kg, p.o.) and the tacrine-treated group (10 mg/kg, p.o.) significantly ameliorated scopolamine-induced amnesia based on the Passive avoidance Y-maze test and Watermaze test. And these results are same manner in DPPH radical scavenger effect and Acetylcholineseterase inhibition effect. Conclusions : These results suggest that Taraxacum officinale extract maybe a useful cognitive impairment treatment, and its beneficial effects are depending on the collecting time and origin plants. As a result, Taraxacum officinale harvested in August improve memory most.

Dynamic Programming을 적용한 트리구조 미로내의 목표물 탐색 알고리즘 (Target Object Search Algorithm under Dynamic Programming in the Tree-Type Maze)

  • 본 논문에서는 Dynamic Programming(DP)을 적용한 트리구조 미로내의 목표물 탐색 알고리즘을 구현한다. DP는 큰 문제를 이루는 작은 문제들을 먼저 해결하고 작은 문제들의 최적해를 이용하여 순환적으로 큰 문제를 해결한다. 먼저 실험을 위해 적외선 센서론 부착한 소형 이동 로봇과, 'Y'형태로 갈라진 길을 연결한 트리 구조의 미로 환경을 구성한다. 실험에서는 두 개의 서로 다른 알고리즘 - 좌수법, DP - 을 사용하여 목표물 탐색을 시도한다. 마지막으로 위 실험을 통해 DP를 미로 탐색문제에 적용했을 때의 성능을 검증한다.

승모판막 질환과 동반된 심방세동에 대한 Cox-Maze 술식 후 좌심방 크기 및 기계적 수축력 변화: 중.단기 경과 관찰 분석 (Change of the Left Atrial Dimension and Transport Function after the Cox-Maze Procedure for Treating Atrial Fibrillation Associated with Mitral Valve Disease: the Short-term and Mid-Term Results)

  • 배경: Cox-Maze 술식은 심방세동의 동율동 전환율은 높지만, 좌심방의 크기 및 기계적 수축력 회복 측면에 대한 효과는 아직 확립되어 있지 않다. 본 연구는 Cox-Maze 술식 후 좌심방 크기 및 기계적 수축력 변화를 알아보기 위해 수행되었다. 대상 및 방법: 1997년 7월부터 2008년 7월까지 승모판막 질환과 동반된 심방세동으로 총 647명의 환자에게 Cox-Maze 술식이 이루어졌다. 이 중, (1) 수술 후 경과 관찰 기간이 2년 이상인 경우, (2) 수술 후 항부정맥약의 복용 여부와 상관없이, 경과 관찰 기간도중 시행한 검사상 동율동이 지속된 경우, 그리고 (3) 경과 관찰 기간 도중 시행한 경흉부 심초음파 검사상, 판막의 Grade III 이상의 폐쇄부전증 또는 중등도 이상의 협착증이 없는 경우 등의 3가지 조건을 충족하는 211명을 연구 대상으로 하였다. 결과: 술 후 1년과 그 이후 추적 관찰기간의 경흉부심초음파 검사상 좌심방 크기는 증가하였으며, 기계적 수축력(E/A ratio)은 감소하였다. 추적 관찰기간이 길어질수록 통계적으로 유의하게 좌심방의 크기 증가와 기계적 수축력(E/A ratio) 감소의 소견을 보였다. 결론: 술 후 초기에 보여지는 회복된 좌심방 크기와 기계적 수축력은 시간이 경과함에 따라 각각 크기 승가와 기계적 수축력 감소를 보인다. 따라서 동율동으로 심박동 전환된 환자도 장기적인 추적관찰을 통해 꾸준한 좌심방 기능 평가가 필요하다고 생각된다.

Assessment of long-term working memory by a delayed nonmatch-to-place task using a T-maze

  • Long-term working memory (LTWM) is a subdivision concept of working memory and indicates the enhancement of performance in a working memory task. LTWM has been shown in humans who have been engaged in a specific task requiring working memory over a long time. However, there is very little understanding of the exact mechanism of LTWM because of limitations of experimental methods in human studies. We have modified the standard T-maze task, which is used to test working memory in mice, to demonstrate LTWM in an animal model. We observed an enhancement of performance by repeated experience with the same working memory load in mice, which can be regarded as an LTWM. This effect seems to depend on the condition wherein a delay was given. This task may be a good experimental protocol to assess LTWM in animal studies.

A Design and Implementation Mobile Game Based on Kinect Sensor

  • In this paper, we design and implement a mobile game based on Kinect sensor. This game is a motion recognition maze game based on Kinect sensor using XNA Game Studio. The game consists of three stages. Each maze has different size and clear time limit. A player can move to the next stage only if the player finds the exit within a limited time. However, if the exit is not found within the time limit, the game ends. In addition, two kinds of mini games are included in the game. The first game is a fruit catch game using motion recognition tracking of the Kinect sensor, and player have to pick up a certain number of randomly falling fruits. If a player acquire a certain number of fruits at this time, the movement speed of the player is increased. However, if a player takes a skeleton that appears randomly, the movement speed will decrease. The second game is a Quiz game using the speech recognition function of the Kinect sensor, and a question from random genres of common sense, nonsense, ancient creature, capital, constellation, etc. are issued. If a player correctly answers more than 7 of 10 questions, the player gets useful items to use in finding the maze. This item is a navigator fairy that helps the player to escape the forest.

우황청심원(牛黃淸心元)이 NOS inhibitor에 의한 흰쥐의 학습(學習) 및 기억장애(記憶障碍)에 미치는 영향(影響) (The effect of Woohwangchungsimwon on the learning and memory in NOS inhibitor treated rats in Morris water maze.)

  • This study was conducted to find out the effects of Woohwangchungsimwon on learning and memory in the NOS inhibitor treated rats. The Morris water maze was used in evaluating them. The result of the study was summarized as follows. 1. In the learning test, three groups have showed a gradual improvement of learning ability by repeating the trials in Morris water maze. WHCS group have showed statistical improvement than control group at 4,5,6 trial(p<0.05, p<0.01, p<0.01). 2. In the memory test, the first latency of WHCH group was statistically shortened than that of control group(p<0.05). 3. In the memory test, there was no statistical difference in the entry number between WHCH group and control. 4. In the memory test, there was no statistical difference in the memory score between WHCH group and control. The result of this experimental study presents that Woohwangchungsimwon has the improving effect on impaired learning and memory in NOS inhibitor treated rats, and implies that Woohwangchungsimwon may be one of the useful prescription for the treatment of vascular dementia after cerebral ischemia.

식이내 아미노산의 조성과 환경이 흰쥐의 두뇌중 RNA 단백질함량 및 학습능력에 미치는 영향 (Effects of Amino Acid Composition of Diet and Environment on RNA, Protein Content in Brain and Learning Ability in Rats)

  • This study was performed to investigate the influence of the amino acid composition of diet and environment on RNA, protein content in brain and learning ability in rats. Forty-two Sprague-Dawley male rats were divided into six groups according to type of diet, casein, soybean meal, or corn gluten and rearing condition, isolated or enriched. They were fed foods ad libitum for 6 weeks. A water maze was used to test behavioral performance for 3 weeks from 4th week. The rats were sacrificed at 6th week and their whole brains were taken and frozen for assay of RNA and protein. The results were summarized as follows : 1) The body weight gain for the experimental periods of corn gluten group was significantly lower than the casein and the soybean meal group. 2) The brain weight of the corn gluten group was significantly lower than the casein and the soybean meal group and the environmental enrichment slightly increased it among rats fed the corn gluten diet. 3) The total RNA contents were the greatest in the environmentally enriched casein group. The brain protein contents of the isolated corngluten group was the smallest. However, the contents of the enriched corn gluten group were similar to those of the others. 4) In the water maze test, the isolated corngluten group spent significantly more time than the others. Environmental enrichment could decrease time to perform the task of the maze.

갈근해주탕(葛根解酒湯) 합(合) 단주환(斷酒丸)과 해주지갈탕(解酒枳葛湯)이 실험적(實驗的) Alcoholism 생쥐의 행동(行動)에 미치는 영향(影響) (The Effects of Galgunhaejutang + Danjuhwan and Haejujigaltang on the Experimental Behaviors of Alcohol-Intoxicated Mice)

  • This experiment was designed to investigate the effects of Galgunhaeiutang + Danjuhwan and Haejujigaltang on the experimental behaviors of alcohol-inticxicated mice. ICR mice were used as subjects for alcohol intoxication in this experiment and sample group received Herb Medicine. The foot-step and rearing score were measured in mice. Also Rota-Rod treadmill & number of successful trials swimming time in the T-maze were done. And the following results were observed; 1. The food-step score was decreased in Galgunhaejutang + Danjuhwan group at the 8th hour, the difference was statistically significant. But Haejujigaltang group showed no significant difference. 2. The number of rearing was decreased in Galgunhaejutang + Danjuhwan group at the 3rd and 8th hour, the difference was statistical significant, while Haejujigaltang group showed no significant differnce. 3. The motor incoordination measered by the Rota-Rod treadmill was decreased in Galgunhaejutang + Danjuhwan group at the 8th hour, but Haejujigaltang group showed no significant difference. 4. The number of successful trials in T-maze showed no significant difference in both groups. 5. The passing time in T-maze was increased in Galgunhaejutang +Danjuhwan group at the 8th hour, but Haejujigaltang group showed no significant difference.

정상 동물모델에서 다시마(Saccharina japonica) 발효물의 기억력 개선 효과 (Memory-improving Effects of Fermented Sea Tangle Saccharina japonica in Normal Mice)

  • Marine organisms are sources of many bioactive compounds, such as essential fatty acids, essential amino acids, vitamins, and minerals, making them useful candidates for the production of safe bioactive substances. They also synthesize glutamic acid, which can be used to produce γ-aminobutyric acid (GABA), the major inhibitory neurotransmitter in the central nervous system (CNS), via fermentation with Lactobacillus brevis BJ-20. This study investigated the degree to which fermented sea tangle (FST) inhibits enzymes such as acetylcholine esterase (AChE) and prolyl endopeptidase (PEP) and affects memory of normal mice using the T-maze test. FST inhibited more than 90% of AChE at 1 mg/mL and 50% of PEP at 8 mg/mL. Oral FST (100 mg/kg) significantly improved performance of normal mice on the T-maze. Therefore, sea tangle fermented with L. brevis BJ20 effectively contributes to memory improvement and might be a useful functional food ingredient.
