Effects of Fructus Schisandrae Water Extract on Cultured Mouse Myocardial Cells Induced by Xanthine Oxidase/Hypoxanthine (오미자 추출물이 산소지유기에 의하여 손상된 생쥐의 배양 심근세포에 미치는 영향)

  • The purpose of this study was to elucidate protective effect of Fructus Schsandrae(FS) water extract against xanthine oxidase/hypoxanthine(XO/HX)-induced cardiotoxicity in myocardial cells this experiment was performed. Cardiotoxicity of XO/HX was examined by MTT(MTT [3-(4,5-dimethylthiazol-2-yl)-2.5,-diphenyl tetrazolium bromide) assay. XO/HX induced the decrease of cell viability. Also XO/HX induced the increase of LDH activity and the decrease of beating rate on cultured myocardial cells in a dose-dependent manner. To investigate cardioprotective effect of FS water extract cultures were preincubated with FS water extract for 3 hours. Cultures were then exposed to XO/HX for 72 hours. FS water extract have an efficacy in decreaasing LDH activity and increasing heart beating rate on cultured myocardial cells damaged by XO/HX. From the results it is suggested that XO/HX may show toxic effect in cultured myocardial cells derived from neonatal mouse and FS water extract is effective in the prevention of XO/HX-induced cardiotoxicity.

  • This study was undertaken to observe the effect of xanthine such as caffeine and aminophylline on the activity of carbonic anhydrase in the kidney and stomach of the mouse. Carbonic anhydrase activities were measured by Philpot & Philpot method (1936). The results of this experiment were as follows: 1. The activity of carbonic anhydrase in the kidney of the mouse was silightly inhibited by the administration of caffeine (0.1 mg/gm, B.W.) or aminophylline (0.08 mg/gm, B.W.). The inhibition was more pronounced by the administration of aminophylline than that of caffeine. 2. In the stomach, there was no significant change in the activity of the carbonic anhydrase after the administration of either caffeine or aminophylline.

An Effect of Ethionine Treatment on the Activity of Hepatic Xanthine Oxidase in Rats (흰쥐의 간 Xanthine Oxidase 활성에 미치는 Ethionine의 영향)

  • Liver and serum xanthine oxidase(XOD) activity were determined in rats treated with DL-ethionine. Concomitantly, the enzyme activity was compared with that of rats treated with CCl$_{4}$, actinomycin D and those fed a protein depleted diet. The activity of XOD in liver was inhibited by injection of ethionine to the rats. But, there were no differences in activity of serum XOD between control group and ethionine-treated rats. And the pattern of changes in enzyme activity of serum and liver in ethionine-treated rats, was similar with actinomycin D treated rats or those fed a protein depleted diet. On the other hand, the activity of XOD was rather elevated both in serum and liver by injection of CCl$_{4}$ to rats.

Estimation of Death Time by Changes of Postmortem Xanthine Oxidase Activity in Rats

  • To evaluate the postmortem changes in activities of oxygen free radical metabolizing enzymes, the rats were sacrificed with cervical dislocation and were kept in an incubator at $25^{\circ}C$, 70% of humidity for 12 hours. The activities of aniline hydroxylase, catalase, glutathione-S-transferase and superoxlde dismutase were decreased with the time. On the other hand, the activity and type conversion ratio (type D ${\to}$type O) of hepatic xanthine oxidase (XO) were gradually increased. From these changes of XO, the estimation of death time (mathematical equation) could be determined with the least square method. To clarify the cause of increasing XO activity, enzyme kinetics were examined. The Km values of XO were decreased with the time. In conclusion, the determination of liver XO activity might be used for the estimation of death time in the early postmortem period.

Effect of Water Extract from Fugu xanthopterus on the Hyperuricemia in Alcohol-treated Rats (복어추출물이 Alcohol성 고요산혈증에 미치는 영향)

  • The present study was undertaken to investigate the effect of the water extract of the puffer fish Fugu xanthopterus(FXH) on the alcohol induced hyperuricemia. The normal group and the FXH treated group showed no sigbificant changes in the levels of blood uric acid but, the blood uric acid significantly decreased in the FXh treated rats with 100mg/kg for two weeks compared to the ethanol treated group. There were no significant changes in the activities of uricase, adenosine deaminase, guanine deaminase, and purine uncleoside phosphorylase, among all the test group. But the activitis of liver xanthine oxidase were recovered to the normal level in ethanol +FXH treated group comparing to the ethanol treated group. Furthermore, ethanol+FXH treated rats showed the similar pattern in the levels of blood uric acid and urinary allantoin with normal group. These results indicate that the decreased blood uric acid by the FXH treatment of the alcohol induced hyperuricemia rats may result from decreased activity of hepatic xanthine oxidase.

  • Cervus elaphus for herb-acupuncture solution (CEHAS) was tested for the effects of free radical generating enzyme and lipid peroxidation in rat's kidney. In vitro, levels of lipid peroxide in tissues of kidney were proportionally decreased to concentration of extracts prepared from CEHAS. They were much more decreased, when lipid peroxidation was induced with ferrous iron (FeII). Also, enzyme activities of xinthine oxidase were decreased. The ratio of type conversion of xanthine oxidase was lowered, two. But, it was not seen changes on enzyme activities of aldehyde oxidase. These results suggest that CEHAS decrease the activities of free radical generating enzymes such as xanthine oxidase which form lipid peroxide.

Effects of Sopunghwalhyul-tang Water Extract against Xanthine Oxidase / Hypoxanthine(XO/HX)-Induced Neurotoxicity in the Cultured Mouse Spinal Sensory Neurons (소풍활혈탕 열탕액이 XO/HX에 의해 손상된 배양 척수감각신경세포에 미치는 영향)

  • In order to elucidate the toxic mechanism of oxygen radicals in cultured mouse spinal sensory neurons, cytotoxic effect of oxygen radicals was evaluated by M1T assay and NR assay. In addition, protective effect of Sopunghwalhyultang(SPHHT) water extract on oxidant-induced neurotoxicity was investigated on these cultures. Spinal sensory neurons derived from mice were cultured in mediums containing various concentrations of Xanthine Oxidase / Hypoxanthine(XO/HX). Cell viability was measured by MTT assay and NR assay. XO/HX-mediated oxygen radicals remarkably decreased cell viability of cultured spinal sensory neurons in a dose-and time-dependent manner. And also, SPHHT blocked XO/HX-induced neurotoxicity in these cultures. These results suggest that oxygen radicals are toxic and SPHHT are effective in blocking against the oxidant-induced neurotoxicity in cultures of spinal sensory neurons of mice.

Effects of Jangwon-hwan(Zhuangyuan-wan) Water Extract against Xanthine Oxidase / Hypoxanthine-induced Neurotoxicity in the Cultured Mouse Cerebral Cortical Neurons (장원환이 XO/HX에 의해 손상된 대뇌피질 신경세포에 미치는 영향)

  • In order to elucidate the toxic mechanism of neurotoxical damage and neuroprotective effect of Jangwon-hwan(Zhuangyuan-wan) water extract, this experiment was performed. Neurotoxic effects of xanthine oxidase/hypoxanthine(XO/HX) were examined by MTT and NR assay, neuroprotective effects of Jangwon-hwan(Zhuangyuan-wan) water extract were examined by neurofilament enzymeimmuno assay(EIA). XO/HX induced an increase in cell viability, and a decrease in the amount of neurofilament on cultured mouse cerebral cortical neurons in dose-dependent manner. In neuroprotective effect of herb medicine, Jangwon-hwan(Zhuangyuan-wan) water extract increased the amount of neurofilament on cultured mouse cerebral cortical neurons damaged by XO/HX. From the results, it is suggested that XO/HX showed toxic effect in cultured mouse cerebral cortical Neurons and Jangwon-hwan(Zhuangyuan-wan) water extract is very effective in the prevention of neurotoxicity induced by XO/HX.

  • In the screening of those components with comparatively strong xanthine oxidase inhibition activities from Panax ginseng folium. trifolin. panasenoside. kaempferol and so far unconfirmed phenol whose chemical structure still without elucidation have been discovered as such components. Specific inhibitors of xanthine oxidase are thought to be therapeutically useful for the treatment of gout. Such opinion is considered to be useful to support the thought that ginseng folium is effective for the adjustment of gout. Although trifolin. panasenoside. and kaempferol are well known flavonoids. further studies on unidentified new phenols should be continued.

Use of a Xanthine-Xanthine Oxidase System on In Vitro Maturation and Fertilization in Pig

  • This study was undertaken to evaluate the effects of catalase using xanthine (X) - xanthine oxidase (XO) system on in vitro maturation and fertilization in pig. When follicular oocytes were cultured in maturation medium with X and/or XO, the maturation rates were not significantly different between in medium with and without catalase despite of different culture periods. However, significantly (P<0.05) higher maturation rates were obrained in culture with X-XO system. The rates of degenerated oocytes were increased with culture periods prolonged, and were significantly (P<0.05) higher in medium without than with catalase at 120 h of culture. On the other hand, the parthenogenetic oocytes were observed with high proportions at 72 h of culture, hut were not different in medium with and without catalase at various times of culture. In another experiment, the frozen-thawed boar spermatozoa treated with X-XO system for in vitro fertilization. The penetration rates were higher in medium with that than without catalase during the in vitro fertilization with, none (P<0.05), XO and X+XO. On the other hand, when sperm were treated with none, X, XO and X+XO, lipid peroxidation were higher in medium without that than with catalase. However, the changes in sperm penetration and lipid peroxidation showed opposite patterns. The sperm suspensions were also treated with X and/or XO for assay of sulfhydryl (-SH) group content. Under the above all conditions, sperm-SH group were higher detected In medium with that than without catalase. The activity of sperm binding to zona pellucida was also evaluated through binding to salt-stored porcine oocytes. In control group, sperm binding to zona pellucida were higher than in medium with X, XO and X+XO groups. No significant differences, however, were observed between medium with and without catalase. In conclusion, the exposure of follicular oocytes and spermatozoa to X-XO system may be caused stimulating in vitro maturation and fertilization in pig.
