Variation of Characteristics of Nonstoichiometric Apatite Induced by Sodium Salt (나트륨염에 의한 비양론적 인회석의 특성 및 SaOS-2 세포반응에 미치는 영향에 관한 연구)

  • The purpose of this work is to develope sodium-containing nonstoichiometric apatitic coatings on solid substrate. The apatitic coatings prepared at different concentrations of sodium salt indicated that the presence of sodium ions exerted significant effects on the physicochemical properties of the apatitic coating including surface morphology, chemical state, and Ca/P ratio. The variation of these properties was sustained up to 0.01 mM of sodium ion concentration. The ratio of calcium to phosphorus was varied from 2.18 to 2.03 which indicated the apatitic coating prepared in this study was calcium-rich nonstoichiometric apatite. The structure of all the samples appeared to be low crystalline. In the presence of sodium ion within the apaptitic coating, the adhesion of human osteoblast-like SaOS-2 cells was significantly promoted. On the other hand, the proliferation of the cells on the apatitic coatings was decreased with the increase of sodium ions. This reverse response of SaOS-2 cells indicates that the interaction between SaOS-2 and apatitic surface triggered considerable changes in intracellular mechanisms including cellular signal transductions.

Induction of Effective Osteogenesis by Mesenchymal Stem Cells from the Human Subchondral Bone (사람 연골하골 중간엽 줄기세포의 효율적인 골형성 유도)

  • Background : Mesenchymal stem cells (MSCs) are present in most of the tissue matrix, taking part in their regeneration when injury or damage occurs. The aim of this study was to investigate the presence of cells with pluripotential characteristics in human subchondral bone and the capacity of these cells to differentiate to osteoblast. Methods : Human subchondral bone were digested with collagenase. Isolated cells were cultured with a-MEM, 15% FBS, 10-8M dexamethasone and 50 ng/mL ascoric acid. Cells from 0 day(isolated cells), 7 day (first subculture) and 14 days (third subculture) were used to carry out phenotypic characterization experiments flowcytometry analysis with 11 monoclonal antibodies) and osteogenic differentiation experiments. Osteogenic differentiation of cells was assessment by quantification of bone extracellular matrix components by following analysis: alkaline phosphatase(ALP) stains to detect ALP activity, RT-PCR and western blot to detect osteocalcin (OCN), osteopontin (OPN) and type I collagen(Col I), and Alizarin red stains to detect calcium deposition. Results : Flowcytometry analyses showed that in our population more than 98% of cells were positive for MSC markers: SH-2(CD105, 99%), CD29 (95%), CD73 (95%). Cells were negative for hematopoietic markers (CD11b, CD34, and CD45). Furthermore, cells showed positive stain to multipotent markers such as CDl17 (c-kit) (15.1%), and CD166 (74.9%), and cell adhesion molecules such as CD54 (78.1%) and CD106 (63.5%). The osteogenic specific marker analyses showed that the culture of these cells for 7 and 14 days stimulates ALP, OCN, OPN and Col I synthesis by RT-PCR and Western blot analysis. Also, after 14 days in the culture of MSCs induces mineralization by Arizarin red stain. Conclusion : In this work, we demonstrated a new and efficient method for osteoblastic differentiation of human subchondral bone stem cells. As MSCs takes part in reparative processes of adult tissues, these cells could play an important role in osteogenesis.

  • Injectable RGD-bioconjugated Mussel Adhesive Proteins (RGD-MAPs) composite hydroxypropyl methylcellulose (HPMC) hydrogels provide local periodontal tissue for bone filling in periodontal surgery. Previously we developed a novel type of injectable self-supported hydrogel (2 mg/ml of RGD-MAPs/HPMC) based porcine nano hydroxyapatite (MPH) for dental graft, which could good handling property, biodegradation or biocompatibility with the hydrogel disassembly and provided efficient cell adhesion activity and no inflammatory responses. Herein, the aim of this work was to evaluate bone formation following implantation of MPH and collagen membrane in rabbit calvarial defects. Eight male New Zealand rabbits were used and four circular calvarial defects were created on each animal. Defects were filled with different graft materials: 1) collagen membrane, 2) collagen membrane with MPH, 3) collagen membrane with bovine bone hydroxyapatite (BBH), and 4) control. The animals were sacrificed after 2 and 8 weeks of healing periods for histologic analysis. Both sites receiving MPH and BBH showed statistically increased augmented volume and new bone formation (p < 0.05). However, there was no statistical difference in new bone formation between the MPH, BBH and collagen membrane group at all healing periods. Within the limits of this study, collagen membrane with MPH was an effective material for bone formation and space maintaining in rabbit calvarial defects.

  • Commercially pure titanium (cp-Ti) and Ti alloys (typically Ti-6Al-4V) display excellent corrosion resistance and biocompatibility. Although the chemical composition and topography are considered important, the mechanical properties of the material and the loading conditions in the host have, conventionally. Ti and its alloys are not bioactive. Therefore, they do not chemically bond to the bone, whereas they physically bond with bone tissue. The electrochemical deposition process provides an effective surface for biocompatibility because large surface area can be served to cell proliferation. Electrochemical deposition method is an attractive technique for the deposition of hydroxyapatite (HAp). However, the adhesions of these coatings to the Ti surface needs to be improved for clinical used. Plasma electrolyte oxidation (PEO) enables control in the chemical com position, porous structure, and thickness of the $TiO_2$ layer on Ti surface. In addition, previous studies h ave concluded that the presence of $Ca^{+2}$ and ${PO_4}^{3-}$ ion coating on porous $TiO_2$ surface induced adhesion strength between HAp and Ti surface during electrochemical deposition. Silicon (Si) in particular has been found to be essential for normal bone and cartilage growth and development. Zinc (Zn) plays very important roles in bone formation and immune system regulation, and is also the most abundant trace element in bone. The objective of this work was to study electrochemical characteristcs of Zn and Si coating on Ti-6Al-4V by PEO treatment. The coating process involves two steps: 1) formation of porous $TiO_2$ on Ti-6Al-4V at high potential. A pulsed DC power supply was employed. 2) Electrochemical tests were carried out using potentiodynamic and AC impedance methoeds. The morphology, the chemical composition, and the micro-structure an alysis of the sample were examined using FE-SEM, EDS, and XRD. The enhancements of the HAp forming ability arise from $Si/Zn-TiO_2$ surface, which has formed the reduction of the Si/Zn ions. The promising results successfully demonstrate the immense potential of $Si/Zn-TiO_2$ coatings in dental and biomaterials applications.

  • This study was initiated to solve the difficulties of aged and female workers in agriculture society due to aging and demise of young people. In the case of the conventional elevated lift, the risk of exposure to uneven road or work environment, not the difficulty of professional qualification and operation, and the risk of exposure to the uneven road or working environment, were also studied based on previous researches so that women could easily and efficiently perform productive agriculture. First, the simulation was carried out through the prediction model of traction performance using the object of agricultural forklift, and the soil of the Kimhae city in Gyeongnam (34.125kPa, internal friction angle 35.294deg, external friction angle 13.620deg, Adhesion force 5.750 kPa, average cone index 0-15 cm cl, 1001.8 kPa). In the case of the forklift for simulation, the driving force and the kinetic resistance prediction modeling of the agricultural electric forklift are modeled. Based on this model, the motor control drive adopts the 1232E model, which is a drive dedicated to AC motor, and divides the two drivers into master and slave And the model for the simulation was designed to control motor drive, hydraulic drive, and various outputs on the main PCB. The simulation model is undergoing continuous simulation, modification and supplementation. Based on this research, we will continue research for development of safer and more efficient agricultural electric forklift.

Characteristic of Ru Thin Film Deposited by ALD

  • Recently, many platinoid metals like platinum and ruthenium have been used as an electrode of microelectronic devices because of their low resistivity and high work-function. However the material cost of Ru is very expensive and it usually takes long initial nucleation time on SiO2 during chemical deposition. Therefore many researchers have focused on how to enhance the initial growth rate on SiO2 surface. There are two methods to deposit Ru film with atomic layer deposition (ALD); the one is thermal ALD using dilute oxygen gas as a reactant, and the other is plasma enhanced ALD (PEALD) using NH3 plasma as a reactant. Generally, the film roughness of Ru film deposited by PEALD is smoother than that deposited by thermal ALD. However, the plasma is not favorable in the application of high aspect ratio structure. In this study, we used a bis(ethylcyclopentadienyl)ruthenium [Ru(EtCp)2] as a metal organic precursor for both thermal and plasma enhanced ALDs. In order to reduce initial nucleation time, we use several methods such as Ar plasma pre-treatment for PEALD and usage of sacrificial RuO2 under layer for thermal ALD. In case of PEALD, some of surface hydroxyls were removed from SiO2 substrate during the Ar plasma treatment. And relatively high surface nitrogen concentration after first NH3 plasma exposure step in ALD process was observed with in-situ Auger electron spectroscopy (AES). This means that surface amine filled the hydroxyl removed sites by the NH3 plasma. Surface amine played a role as a reduction site but not a nucleation site. Therefore, the precursor reduction was enhanced but the adhesion property was degraded. In case of thermal ALD, a Ru film was deposited from Ru precursors on the surface of RuO2 and the RuO2 film was reduced from RuO2/SiO2 interface to Ru during the deposition. The reduction process was controlled by oxygen partial pressure in ambient. Under high oxygen partial pressure, RuO2 was deposited on RuO2/SiO2, and under medium oxygen partial pressure, RuO2 was partially reduced and oxygen concentration in RuO2 film was decreased. Under low oxygen partial pressure, finally RuO2 was disappeared and about 3% of oxygen was remained. Usually rough surface was observed with longer initial nucleation time. However, the Ru deposited with reduction of RuO2 exhibits smooth surface and was deposited quickly because the sacrificial RuO2 has no initial nucleation time on SiO2 and played a role as a buffer layer between Ru and SiO2.

Synthesis and performance assessment of modified epoxy resins containing fatty acid (지방산 변성 에폭시수지 합성과 성능평가)

  • In this work, modified epoxy resins which were well melted in mild solvent were synthesized and solubility assessment was carried out for synthesized epoxy resins. Bisphenol-A type, phenol novolac type and ortho-cresol novolac type epoxy resins were used and fatty acid, dodecyl phenol (DP) and toluene diisocyanate (TDI) were added for synthesis of modified epoxy resins containing fatty acid (MEFA). Composition was epoxy resin/fatty acid = 1.0/0.5 and fatty acid/DP = 0.25/0.25 by equivalent weight and twelve MEFAs were synthesized according to epoxy resins. Viscosity and solubility were measured for twelve MEFAs. As a result, solubility of MEFA was excellent for mild solvent according to increasement of contents of benzene ring, glycidyl group and carbon number of alkyl group. And physical properties were measured for each coating of paints after preparing transparent paints of MEFA to melt well in mild solvent among twelve MEFAs. As a result, they showed an optimal performance on conditions of equivalent ratio of bisphenol-A type epoxy resin/fatty acid/DP/TDI; 1.0/0.25/0.25/0.5 and equivalent ratio of phenol novolac type epoxy resin/fatty acid/DP; 1.0/0.25/0.25 for drying time, adhesion, hardness, impact resistance and alkali resistance.

Formation of a MnSixOy barrier with Cu-Mn alloy film deposited using PEALD

  • With the scaling down of ultra large integrated circuits (ULSI) to the sub-50 nm technology node, the need for an ultra-thin, continuous and conformal diffusion barrier and Cu seed layer is increasing. However, diffusion barrier and Cu seed layer formation with a physical vapor deposition (PVD) method has become difficult as the technology node is reduced to 30 nm and beyond. Recent work on self-forming barrier processes using PVD Cu alloys have attracted great attention due to the capability of conformal ultra-thin barrier formation using a simple technique. However, as in the case of the conventional barrier and Cu seed layer, PVD of the Cu alloy seed layer will eventually encounter the difficulty in conformal deposition in narrow line trenches and via holes. Atomic layer deposition (ALD) has been known for its good step coverage and precise thickness control, and is a candidate technique for the formation of a thin conformal barrier layer and Cu seed layer. Conformal Cu-Mn seed layers were deposited by plasma enhanced atomic layer deposition (PEALD) at low temperature ($120^{\circ}C$), and the Mn content in the Cu-Mn alloys were controlled form 0 to approximately 10 atomic percent with various Mn precursor feeding times. Resistivity of the Cu-Mn alloy films decreased by annealing due to out-diffusion of Mn atoms. Out-diffused Mn atoms were segregated to the surface of the film and interface between a Cu-Mn alloy and $SiO_2$, resulting in self-formed $MnO_x$ and $MnSi_xO_y$, respectively. No inter-diffusion was observed between Cu and $SiO_2$ after annealing at $500^{\circ}C$ for 12 h, indicating an excellent diffusion barrier property of the $MnSi_xO_y$. The adhesion between Cu and $SiO_2$ was enhanced by the formation of $MnSi_xO_y$. Continuous and conductive Cu-Mn seed layers were deposited with PEALD into 32 nm $SiO_2$ trench, enabling a low temperature process, and the trench was perfectly filled using electrochemical plating (ECD) under conventional conditions. Thus, it is the resultant self-forming barrier process with PEALD Cu-Mn alloy film as a seed layer for plating Cu that has further potential to meet the requirement of the smaller than 30 nm node.

Experimental Study on Adfreeze Bond Strength Between Frozen Sand and Aluminium with Varying Freezing Temperature and Vertical Confining Pressure (동결온도와 수직구속응력 변화에 따른 모래와 알루미늄 재료의 접촉면에서 작용하는 동착강도 실험 연구)

  • Bearing capacity of pile foundations in cold region is dominated by adfreeze bond strength between surrounding soil and pile perimeter. Adfreeze bond strength is considered to be the most important design parameter for foundations in cold region. Many studies in last 50 years have been conducted to analyze characteristics of adfreeze bond strength. However, most studies have been performed under constant temperature and normal stress conditions in order to analyze affecting factors like soil type, pile material, loading speed, etc. In this study, both freezing temperature and normal stress acting on pile surface were considered to be primary factors affecting adfreeze bond strength, while other factors such as soil type, pile material and loading speed were predefined. Direct shear box was used to measure adfreeze bond strength between Joomoonjin sand and aluminium because it is easy to work for various roughness. Test was performed with temperatures of > $0^{\circ}C$, $-1^{\circ}C$, $-2^{\circ}C$, $-5^{\circ}C$, and $-10^{\circ}C$ and vertical confining pressures of 1atm, 2atm, and 3atm. Based on the test results, the effects of temperature and vertical stress on adfreeze bond strength were analyzed. The test results showed that adfreeze bond strength increases with decreased temperature and increased vertical stress. It was also noted that two types of distinct sections exist, owing to the rate of increase of adfreeze bond strength along the change of freezing temperature: 1)rapidly increasing section and 2)gradually decreasing section. In addition, the results showed that a main factor affecting adfreeze bond strength switches from friction angle to adhesion as freezing temperature decreases.

Interfacial and Mechanical Properties of MGF Reinforced p-DCPD Composites with Surface Treatments (MGF 표면처리에 따른 p-DCPD 복합재료의 계면 및 기계적 특성 연구)

  • p-DCPD (poly dicyclopentadiene) is the resin that the versatile mechanical properties can be changeable via the control of inner monomer and catalysts. In this work, to improve the strength of composites, surface treated MGF (milled glass fiber) was used as an reinforcement in p-DCPD by molybdenum (Mo) catalyst matrix. The optimum concentration of surface treatment was obtained and the cohesion of MGF themselves increased with concentration. In case of 0.2 wt% silane concentration, the maximized mechanical properties of MGF/p-DCPD composite exhibited because of minimized MGF cohesion. When butyl silane showing minimizing cohesion was used as the optimized alkyl length, high tensile and flexure strength exhibited due to the steric hindrance effect among MGFs. Mechanical and their fractured surfaces of MGF/p-DCPD composites was compared for 4 different chemical functional groups. Norbornene functional groups containing similar chemical structure to DCPD matrix exhibited higher interfacial adhesion between MGFs and DCPD matrix.
