Epigenetic Regulation of Fungal Development and Pathogenesis in the Rice Blast Fungus

  • Fungal pathogens have huge impact on health and economic wellbeing of human by causing life-threatening mycoses in immune-compromised patients or by destroying crop plants. A key determinant of fungal pathogenesis is their ability to undergo developmental change in response to host or environmental factors. Genetic pathways that regulate such morphological transitions and adaptation are therefore extensively studied during the last few decades. Given that epigenetic as well as genetic components play pivotal roles in development of plants and mammals, contribution of microbial epigenetic counterparts to this morphogenetic process is intriguing yet nearly unappreciated question to date. To bridge this gap in our knowledge, we set out to investigate histone modifications among epigenetic mechanisms that possibly regulate fungal adaptation and processes involved in pathogenesis of a model plant pathogenic fungus, Magnaporthe oryzae. M. oryzae is a causal agent of rice blast disease, which destroys 10 to 30% of the rice crop annually. Since the rice is the staple food for more than half of human population, the disease is a major threat to global food security. In addition to the socioeconomic impact of the disease it causes, the fungus is genetically tractable and can undergo well-defined morphological transitions including asexual spore production and appressorium (a specialized infection structure) formation in vitro, making it a model to study fungal development and pathogenicity. For functional and comparative analysis of histone modifications, a web-based database (dbHiMo) was constructed to archive and analyze histone modifying enzymes from eukaryotic species whose genome sequences are available. Histone modifying enzymes were identified applying a search pipeline built upon profile hidden Markov model (HMM) to proteomes. The database incorporates 22,169 histone-modifying enzymes identified from 342 species including 214 fungal, 33 plants, and 77 metazoan species. The dbHiMo provides users with web-based personalized data browsing and analysis tools, supporting comparative and evolutionary genomics. Based on the database entries, functional analysis of genes encoding histone acetyltransferases and histone demethylases is under way. Here I provide examples of such analyses that show how histone acetylation and methylation is implicated in regulating important aspects of fungal pathogenesis. Current analysis of histone modifying enzymes will be followed by ChIP-Seq and RNA-seq experiments to pinpoint the genes that are controlled by particular histone modifications. We anticipate that our work will provide not only the significant advances in our understanding of epigenetic mechanisms operating in microbial eukaryotes but also basis to expand our perspective on regulation of development in fungal pathogens.

A Content Analysis on Career Education in Home Economics Textbook of the Secondary School developed by the 7th Educational Process (제7차 교육과정의 중등학교 가정 교과서에 나타난 진로교육 내용분석)

  • The purpose of this study was to find out how much home economics(HE) textbook was focused on what kind of factors in career education, by analyzing the HE textbooks from the 7th grade(1st grade of the middle school) to the 10th grade(1st grade of high school), which were developed by applying the 7th educational process. 12 textbooks which were published by three publishers were randomly selected by grade. This study used the instrument developed by Korea Research Institute for Vacational Education & Training(2001) as an analysis frame, along with a content analysis system. The results of this study were as follows. The publishers of HE textbook stressed similar points in each section of career education, evenly reflecting the objectives and contents of career education. Three kinds of publishers also placed the same weight on the factors of career education in each unit, and suggested the contents related to career education in the whole units. HE textbook was designed to lead learners to understand self and have interest in varied occupations. Additionally, it provided the learners with a chance to consider their career and occupation by means of detailed job information, and its contents including learning of occupational ability for job selection were useful to plan and prepare for career. Accordingly, as HE textbook is most effective to promote connection between work and learning and contains proper factors of career guidance, it is one of the most appropriate textbooks to deal with career education.

MODFLOW or FEFLOW: A Case Study of Groundwater Model Selection for the Upper Waikato Catchment, New Zealand

  • Groundwater in the Waikatoregion is a valuable resource for agriculture, water supply, forestry and industries. The 434,000 ha study area comprises the upper Waikato River catchment from the outflow of Lake Taupo (New Zealand's largest lake) through to Lake Karapiro (a man-made hydro lake with high recreational value) (Figure 1). Water quality in the area is naturally high. However, there are indications that this quality is deteriorating as a result of land use intensification and deforestation. Compounding this concern for decision makers is the lag time between land use changes and the realisation of effects on groundwater and surface water quality. It is expected that the effects of land use changes have not yet fully manifested, and additional intensification may take decadesto fully develop, further compounding the deterioration. Consequently, Environment Waikato (EW) have proposed a programme of work to develop a groundwater model to assist managing water quality and appropriate policy development within the catchment. One of the most important and critical decisions of any modelling exercise is the choice of the modelling platform to be used. It must not inhibit future decision making and scenario exploration and needs to allow as accurate representation of reality as feasible. With this in mind, EW requested that two modelling platforms, MODFLOW/MT3DMS and FEFLOW, be assessed for their ability to deliver the long-term modelling objectives for this project. The two platforms were compared alongside various selection criteria including complexity of model set-up and development, computational burden, ease and accuracy of representing surface water-groundwater interactions, precision in predictive scenarios and ease with which the model input and output files could be interrogated. This latter criteria is essential for the thorough assessment of predictive uncertainty with third-party software, such as PEST. This paper will focus on the attributes of each modelling platform and the comparison of the two approaches against the key criteria in the selection process. Primarily due to the ease of handling and developing input files and interrogating output files, MODFLOW/MT3DMS was selected as the preferred platform. Other advantages and disadvantages of the two modelling platforms were somewhat balanced. A preliminary regional groundwater numerical model of the study area was subsequently constructed. The model simulates steady state groundwater and surface water flows using MODFLOW and transient contaminant transport with MT3DMS, focussing on nitrate nitrogen (as a conservative solute). A Study of the Experiences and Changes of the Self-Sufficient Centers that Converted the Centre Corporation into Social Cooperative (지역자활센터의 사회적협동조합 전환과정과 변화에 대한 연구)

  • The pilot project for type diversification of self-sufficient support center that convert the centre corporation into social cooperative was introduced in 2015. That aimed to increase the number of participants of self-sufficient programmes and to create more jobs for the working poor through expanding the center's autonomy of usage of budget and arrangement of self-sufficient programmes. This study analyzed the organization transmitting processes and changes of the centers took part in the pilot project, then aimed to suggest the improvement schemes to be helpful for reinforcing positive efforts and removing obstacles in the process converting. The results show that converting into social cooperative is a choice for the sustainability of the centers led by surroundings rather than self-initiated plan. There are some positive changes that are increasing the enterprising spirit and obligation of the center staff. Further more, the cooperation with other community organizations is enhanced based on achieving regional recognition, and support of local governments and the opportunity for new businesses are expanded. However, these are very limited in the regions where social economy is undeveloped. Eventually, we should seek the development schemes of the pilot project considering the reduction of the number of participants in self-sufficient programmes and decreasing of their ability to work, even if some positive aspects are revealed.

A Study on Influence of Naeil Cheum Deduction System on Job Satisfaction of small and medium-sized enterprises in Daegu and Gyeongbuk region (대구.경북지역 중소기업의 내일채움공제 제도가 직무만족도에 미치는 영향에 관한 연구)

  • The purpose of this study is to analyze the effect of the Naeil Chaeum deduction system on job satisfaction. The results of the study are summarized as follows. A total of 159 small to medium-sized enterprise workers participated in the deduction for Naeil Chaeum. The data were analyzed using SPSS Statistics 20, which included frequency analysis, factor analysis, reliability analysis, correlation analysis, regression analysis, and post analysis. First, the suitability of the method of the Naeil Chaeum deduction system has the highest positive effect on the job satisfaction. Second, the correlation of job satisfaction after filling in Naeil Chaeum deductions showed that fitness (p <.01, ${\beta}=.307$) and efficiency (p <.01, ${\beta}=.296$) had a significant effect on job satisfaction. Third, the analysis of the difference of job satisfaction according to the academic ability of the workers who were enrolled in Naeil Chaeum deduction system was analyzed that professional college graduates (2.08) were higher than job college graduates (1.99).The purpose of this study is to analyze the impact of the Fill - Down System on the job satisfaction by analyzing the importance of quality of life, job satisfaction, and muscle strength, analyzing the difference according to the demographic characteristics of the workers, And helping to stabilize the work life, and to verify the effectiveness of the study.

Platycarya strobilacea S. et Z. Extract Has a High Antioxidant Capacity and Exhibits Hair Growth-promoting Effects in Male C57BL/6 Mice

This study was conducted to evaluate the effects of Platycarya strobilacea S. et Z. (PSE) extract on mouse hair growth and to determine the mechanism of action of PSE. PSE was purchased and its antioxidant activities, such as electron donating ability, total polyphenol content, and flavonoid content were tested. Toxicity during topical treatment was determined by the CCK-8 assay, a cell viability test. Fifteen 4-week-old male C57BL/6 mice were assigned to receive one of three treatments: dimethyl sulfoxide (negative control), minoxidil (positive control) or PSE. Test materials were topically applied to the shaved dorsal skin of each mouse daily for 3 weeks. After 21 days, we observed skin tissue hair follicle morphology and length, mast cell number, and stem cell factor (SCF) expression using hematoxylin and eosin (H&E), toluidine blue, and immunohistochemical staining, respectively. Furthermore, the expression of cytokines involved in hair growth [i.e., insulin-like growth factor (IGF)-1, keratinocyte growth factor (KGF), and transforming growth factor (TGF)-${\beta}1$] was determined by PCR. PSE was found to have very high antioxidant activity. The cell viability rate of PSE-treated mice was markedly higher than that of mice in the control group. We also observed an increase in hair follicle length, strong SCF staining, and a decrease in mast cell number in the PSE group. In addition, PSE-treated mice had higher IGF-1 and KGF expression and lower TGF-${\beta}1$ expression than mice in the minoxidil-treated group. These results suggest that topical application of PSE promotes hair growth by intensifying SCF, suppressing mast cell production, and increasing hair growth-promoting cytokine expression.

Characteristics of Fermentation and Aging by Different Adding of Brewing Water in Korean Traditional Cheongju (가수량을 달리한 전통 청주의 발효 및 숙성 특성 연구)

  • This present work was aimed to analyse characteristics of fermentation and aging of Korean traditional cheongju prepared by adding of different brewing water. The three different mashes were used for fermentation and maturation. The cheongju(A) was prepared without adding of brewing water in two stage mashing. Cheongju(B) and cheongju(C) were made with adding of 1 liter and 2 liter brewing water in two stage mashing respectively whereas the cheongju(B) and cheongju(C) showed higher fermentation ability than that of cheongju(A) regarding to pH, ethanol, specific gravity and sugar concentration(brix). In terms of volatile compounds content after six months aging, cheongju(A and B) prepared by adding brewing water showed the higher content of ethyl acetate than that of cheong(C) and the concentrations were maintained during the aging period. In addition, the content of fusel oil(n-propanol, i-butanol, n-butanol, i-bmyl alcohol, n-bmyl alcohol)indicated similar pattern as ethyl acetate. The organic acid of cheongju B(4873.46 mg/l) and C(4963.12 mg/l) also indicated the higher content than that of cheongju A(4661.47 mg/l). In conclusion, cheongju B and C prepared by adding brewing water in two stage mashing showed better quality and taste than that of cheongju A.

  • Objectives: This study, as for activating measures for capable medical physicians to penetrate into the public health sector, is to provide a basic informations which are used for the enlargement of human resources of physicians in the public health sector, by investigating the perception of physicians, who are now working in the public health center, on the training and development of physicians in the public health sector. Methods: The subjects of this study were 126 individuals. The data was analyzed by frequency analysis using SPSS ver. 17.0K. Results: According to the investigation of 'how to support physicians in public health sector', the necessity of almost questions is considered to be important. Especially, regarding to investigation on 'obstacles of physicians' entrance to public health sector', 'relatively low salary' and 'lack of promotion chances' were thought to be considerable. The most significant education programs to work for public health sector is to improve the ability of health administration planning and service performance. Conclusions: The important methods to reinforce and easily obtain the human resources of physicians in public health sector are not only to improve the penetration of physicians to public health sector, but also to enhance the ability of present physicians, even though sufficient recruitment of physicians is essential.

  • This study was done to investigate the situation of classes-related stress in creative's of korean advertising industry. Though creative's are carrying out the most important roles in advertising agency, there has not been much study on them. In this point view, this study approached by examining prior studies on classes-related stress. The findings of this study were as followings ; In all classes, client related stress were the highest. The classes of the directors of bureaus have the highest stress on the average of five high items in the stress detail items. The classes of the staffs have higher stress, and the next is the classes of the heads of departments. The classes of the directors have higher stress about themselves especially. The classes of heads have higher stress for family complain about holiday work than any other classes relatively. The classes of the staffs have higher stress for heavy duty than any other classes. The classes of the directors of bureaus have higher stress in the situation change like presentation than any other classes. There can be many conditions about advertisement creative team members‘stress. I have made dear the internal stress structure according to the classes in the thesis partially. The stress for the classes‘structure can not be solved easily, so it will hare an effect on all members normally if the stress exists for a long time. This study should be continued by reasonable and systematic method. A successful creative's come out from an advertising agency's organizational system, but the organization, after all, is made up of creative's ability and endeavor.

Establishment of Porcine Embryonic Stem Cells by Aggregation of Parthenogenetic Embryos

  • The pig has been considered to serve as an appropriate model of human disease. Therefore, establishment of porcine embryonic stem cell lines is important. The purpose of the present study was to further work in this direction. We produced porcine parthenogenetic embryos, and separately aggregated two of each of two-cell ($2{\times}2$), four-cell ($2{\times}4$), and eight-cell ($2{\times}8$) embryos derived by parthenogenesis. After culture for 4 days, the developmental ability of the aggregates and total blastocyst cell numbers were evaluated. The percentage of blastocysts was significantly higher in both $2{\times}4$- and $2{\times}8$-aggregated embryos ($58.3{\pm}1.9%$ and $37.2{\pm}2.8%$, respectively) than in the control or $2{\times}2$-aggregated embryos ($23.6{\pm}1.1%$ and $12.5{\pm}2.4%$, respectively). Total blastocyst cell numbers were increased in the $2{\times}4$- and $2{\times}8$-aggregated embryos (by $44{\pm}3.0%$ and $45{\pm}3.3%$, respectively) compared with those of control or $2{\times}2$-aggregated embryos ($30.5{\pm}2.1%$ and $30.7{\pm}2.6%$, respectively; p<0.05). The levels of mRNA encoding Oct-4 were higher in both the $2{\times}4$- and $2{\times}8$-aggregated embryos than in the control. When blastocysts derived from $2{\times}4$- aggregated embryos or intact normal embryos were cultured on mouse embryonic fibroblast feeder cells to obtain porcine stem cells, blastocysts from aggregated embryos formed colonies that were better in shape compared with those derived from intact blastocysts. Together, the data show that aggregation of porcine embryos not only improves blastocyst quality but also serves as an efficient procedure by which porcine embryonic stem cells can become established.
