Expression of Lily Chloroplastic Cu,Zn Superoxide Dismutase Enhances Resistance to Erwinia carotovora in Potatoes

  • Previously, a chloroplast-localized Cu,Zn superoxide dismutase (chCu,ZnSOD) was isolated from lily and the sense- and antisense- sequences of the lily chCu,ZnSOD were used to transform potato plants. Two selected lines, the sense- and anti-sense strand of transgenic plants, were further characterized for resistance to Erwinia carotovora, which is a severe pathogen affecting potato plants. Only the sense-strand transgenic potato, which contained less $O_2^{.-}$ and more $H_2O_2$ than wild-type and antisense-strand transgenic plants, showed increased resistance to E. carotovora. Additional studies using $O_2^{.-}$ or $H_2O_2$ scavengers in wild-type, sense-strand, and antisense-strand transgenic plants suggest that resistance to E. carotovora is induced by reduced $O_2^{.-}$ and is not influenced by $H_2O_2$. To the best of our knowledge, this report is the first study suggesting that resistance to E. carotovora is enhanced by reduced $O_2^{.-}$, and not by increased amounts of $H_2O_2$.

Iron fortification of grains by introducing a recombinant gene of ferritin with seed promoters in rice (종자 특이 프로모터와 대두 Ferritin 유전자에 의한 벼 종실의 철분강화)

  • The recombinant DNAs, pGBF, pGTF, and pZ4F, using soybean ferritin gene have constructed with the promoters derived from seed proteins, glutelin, globulin, and zein. The recombinant ferritin genes were transformed into rice plant by Agrobacterium-mediated transformation. Iron contents and agronomic traits have been evaluated in the transgenic progenies. The embryogenic calli survived from second selection medium were regenerated at the rates of 19.2% with pGBF, 15.0% with pGTF, and 18.4% with pZ4F in Donganbyeo and 6.7% with pGBF, 11.7% with pGTF, and 3.4% with pZ4F in Hwashinbyeo. The introduction of ferritin gene in putative transgenic rice plants was confirmed by PCR and Southern blot analysis and also the expression of ferritin gene was identified by Northern blot and Western blot analysis. The iron accumulation in transgenic rice grains of the transgenic rice plant, T1-2, with zein promoter and ferritin gene contained 171.4 ppm showing 6.4 times higher than 26.7 ppm of Hwashinbyeo seed as wild type rice, but the transgenic plants with globulin and glutelin showed a bit higher iron contents with a range from 2.1 to 3.0 times compare to wild type grain. The growth responses of transgenic plants showed the large variances in plant height and number of tillers. However, there were some transgenic plants having similar phenotype to wild type plants. In the T1 generation of transgenic plants, plant height, culm length, panicle length, and number of tillers were similar to those of wild type plants, but ripened grain ratio ranged from 53.3% to 82.2% with relatively high variation. The transgenic rice plants would be useful for developing rice varieties with high iron content in rice grains.

Complete Genome Sequence and Analysis of Carnation Italian Ringspot Virus from Erigeron annuus (L.) Pers. in Korea

  • In this study, we aimed to study carnation italian ringspot virus (CIRV) in Erigeron annuus (L.) Pers. in Bonghwa County, Korea. The collected samples showed mosaic and malformation symptoms. To identify the virus species, we performed high-throughput sequencing, reverse transcription polymerase chain reaction, and cloning. The virus was confirmed to be an unreported species, and therefore we performed genome sequencing of the samples. The complete genome was 4,746 nucleotides in length. The CIRV contained five open reading frames (ORFs), and it showed the typical features of members of the genus Tombusvirus. Phylogenetic analyses revealed that ClRV isolates had the highest nucleotide identities with the CZ isolate (95.89%) from Korea. In recent years, these viruses have sporadically been reported in floral scent and medicinal plants. This research found the first natural host infected with CIRV, and provides baseline information to determine the correlation between weeds and crops.

Overexpression of Cotton Glutathione S-Transferase (GST) cDNA and Increase of low Temperature and Salt Tolerance in Plants

  • Cotton Glutathione S-Transferase(GST: EC 2.5.1.18) was cloned and Gh-5 cDNA was overexpressed in tobacco (Nicotiana tabacum) plants. The transformation of cotton GST in tobacco plant was confirmed by northern blot analysis. Type I and Type II transcript patterns were identified in Gh-5 transgenic tobacco plants. Type I transcripts was only discussed in this paper. Glutathione and 1-chloro-2,4-dinitrobenzene (CDNB) were used as the substrates, and the activity of GST in the type I transgenic plants was about 2.5-fold higher than the non-expressers and wild type tobacco plants. The expression of cotton GST in tobacco plants proved that Gh-5 could be translated into functional protein. Type I transgenic plants produced functional GST in the cells. Type I showed higher GST specific activity than Type II in the transgenic plants. Control and transgenic seedlings were grown in the growth chamber and under the light at 15$^{\circ}C$, and the effects of cotton GST in the seedlings was evaluated. The growth rate of Gh-5 overexpressors was better than the control and non-transgenic tobacco plants. Salinity tolerance was also analyzed on the seeds of transgenic plants. Seeds of Gh-5 overexpressors and the wild type tobacco seedlings were germinated and grown at 0, 50, 100, 150, and 200 mM NaCl solution. Gh-5 transgenic seedlings showed higher growth rate over control seedlings at both 50 and 100 mM NaCl solution. But at 0, 150, and 200 mM NaCl concentration, the difference in growth rate was not detected.

CaPUB1, a Hot Pepper U-box E3 Ubiquitin Ligase, Confers Enhanced Cold Stress Tolerance and Decreased Drought Stress Tolerance in Transgenic Rice (Oryza sativa L.)

  • Abiotic stresses such as drought and low temperature critically restrict plant growth, reproduction, and productivity. Higher plants have developed various defense strategies against these unfavorable conditions. CaPUB1 (Capsicum annuum Putative U-box protein 1) is a hot pepper U-box E3 Ub ligase. Transgenic Arabidopsis plants that constitutively expressed CaPUB1 exhibited drought-sensitive phenotypes, suggesting that it functions as a negative regulator of the drought stress response. In this study, CaPUB1 was over-expressed in rice (Oryza sativa L.), and the phenotypic properties of transgenic rice plants were examined in terms of their drought and cold stress tolerance. Ubi:CaPUB1 T3 transgenic rice plants displayed phenotypes hypersensitive to dehydration, suggesting that its role in the negative regulation of drought stress response is conserved in dicot Arabidopsis and monocot rice plants. In contrast, Ubi:CaPUB1 progeny exhibited phenotypes markedly tolerant to prolonged low temperature ($4^{\circ}C$) treatment, compared to those of wild-type plants, as determined by survival rates, electrolyte leakage, and total chlorophyll content. Cold stress-induced marker genes, including DREB1A, DREB1B, DREB1C, and Cytochrome P450, were more up-regulated by cold treatment in Ubi:CaPUB1 plants than in wild-type plants. These results suggest that CaPUB1 serves as both a negative regulator of the drought stress response and a positive regulator of the cold stress response in transgenic rice plants. This raises the possibility that CaPUB1 participates in the cross-talk between drought and low-temperature signaling pathways.

Characteristics of Naturalized Plants in the Gwangyang Steel Works (광양제철소 내의 귀화식물상의 특성)

  • The naturalized plants in the Gwangyang Steel Works were listed 70 taxa; 18 families, 51 genus, 66 species, 3 varieties and 1 form. by field survey. The naturalized plants divided into pattern by survey of annual plants ratio is 31 taxa (44.3%) by life form spectrum and perennials is 23 taxa (32.9%), biennials is 13 taxa (18.5%), two trees (Robinia pseudoacacia, Alianthus altissima) and one shrub (Amorpha furticosa) were founded. According to analysis results form place of origin, Europe covered 26 taxa (37.1%), North America covered 23 taxa (32.8%). Naturalized degree 3 plants, as common but not abundant, founded as 24 taxa (35.0%) were hold most highly ratio, naturalized degree 1 plants founded as 3 taxa (4.0%) were hold lowest. Introduction period 1 covered 31 taxa (44.3%) have had highest score and period 2 covered 11 taxa (15.7%) have had lowest scores. In addition, the urbanization index based on 271 taxa was 25.8% and 2 taxa (Solanum carolinense, Ambrosia artemisiaefolia) is growing in the Gwangyang Steel Works by ecosystem disturbing wild plants. Meanwhile, Ambrosia artemisiaefolia have confirmed into several tens~hundred in the Gwangyang Steel Works were damage the human body of plants develop an allergy to pollen. Solanium carolinense have composed several large communities about $10m{\times}10m$ ($100m^2$) and hold a dominant position, so management plan of the sequel of monitoring them might be required.

  • The purpose of this study was to understand the geographical and environmental distribution of wild animals in Baekdudaegan region using field survey and GIS data. Crucial factors were selected and analyzed to understand the distributional characteristics of wild animals (7 species in 6 families) and rare endemic plants (14 species in 11 families). These crucial factors include stand factors (forest type, DBH class, and crown density), soil factors (bed rock, soil texture, and organic matter), geographical factors (elevation, slope, aspect) and climatic factors (temperature, rain fall, humidity). Finally, 10 factors were selected by statistical analysis and categorized for analyzing geographical and environmental features. Four families such as Muridae, Sciuridae, Suidae and Talpidea in wild animal showed the similar habitat characteristics with the 10 factors.

  • Mitochondrial DNA restriction fragment length polymorphisms are convenient markers for identifying cytoplasmic variation among plants. We have collected 212 wild soybeans (Glycine soja Sieb. et Zucc) from all over Korea, and classified mitochondrial genome types based on hybridization patterns in DNA gel-blot analyses using two mitochondrial DNA clones, cox2 and atp6, as probes. Korean wild soybean was classified with eight-mtDNA types, and some of the mtDNAs showed geographical clines among the regions. The diversity index of the mtDNA was much higher in the western and southern regions than in the eastern and northern regions of Korea, respectively. Dissemination and distributive characteristics of wild soybeans in Korea were discussed.

Characterization of in vitro Growth and Differentiation of an Albino Mutant of Nicotiana tobacum L. (Albino 담배 변이체의 기내 생장과 기내 분화의 특성)

  • The albino plants of tobacco (Nicotiana tobacum L. cv. BY-4) were isolated from seed populations that were induced by heavy-ion ($^{14}N$) beam irradiation to proembryo and the in vitro growth and differentiation have been characterized. The in vitro cultured albino plants showed significant reduction of chlorophyll content and possessed larger number of stomata on both upper and lower epidermis than that of wild-type plants. Stem growth of the mutants remained dwarfed, however, the internode recovered its normal length after GA$_3$ treatment (10.0mg/L) on the MS medium containing sucrose under continuous light. When explants of leaf blades of albino plants were cultured, multiple shoots formed directly on MS medium containing 1.0mg/L of BAP or kinetin and a large number of calli were induced on the MS medium containing 1.0mg/L NAA or 1.0 mg/L 2,4-D. The albino calli regenerated multiple albino plantlets in the MS medium containing 0.1mg/L NAA + 1.0 mg/L BAP. No significant differences between the wild-type and albino plants were detected in the multiple shoot induction, callus formation from the explants and the plantlets regeneration from calli. In addition, albino plants have a similar organogenesis Pattern to that of the wild-type in the media with different combinations of NAA (0 to 5.0mg/L) and BAP (0 to 5.0mg/L) treatment. These results indicate that the albino mutant has the same normal regeneration ability as that of wild-type, although the mutant has lost functions in photosynthesis, such as pigmentation.

Antioxidant Activity of Solvent Fractions from Cultivated and Wild Gromwell (재배와 야생 지치의 추출물과 용매별 분획물의 항산화효과)

  • In order to set up an accurate quality criteria for the Boraginaceae that have been traditionally used for medical purposes and food colorant, and to assess its viability as functional food ingredient, antioxidant tests were conducted on the wild and cultivated plants. Variety of indicators including total contents of phenol, DPPH, SOD-liked effect, hydroxy radical-scavenging effect, lecithin oxidation inhibitory effect, etc were analyzed. Wild and cultivated gromwell's total contents of phenol in their methanol extracts were 0.14% and 0.13%, while they were most active in ethyl acetate extracts and n-hexane extracts, respectively. $IC_{50}$ values of methanol extract of the wild and cultivated plants were 794.41 ${\mu}g$/mL and 971.86 ${\mu}g$/mL, indicating that the wild plant is more responsive (p<0.05) to low concentration. Also the wild and cultivated plants were most active in ethyl acetate fraction and n-hexane extracts when their $IC_{50}$ values were measured by each solvent extracts. SOD-liked effects of both plants were concentration dependent while methanol extracts were more active (p<0.05) in 500 ${\mu}g$/mL than other solvent extracts. Hydroxy radical-scavenging effect of both plants showed less than 50% activity in concentration of 1,000 ${\mu}g$/mL except in chloroform fraction and n-hexane fraction. Lecithin oxidation inhibitory effects of the wild and cultivated plants were active in methanol and solvent extracts of 200~1000 ${\mu}g$/mL. Especially it showed 90% of high inhibitory effect in 1,000 ${\mu}g$/mL of chloroform fraction. Hence, both wild and cultivated Boraginaceae were analyzed to be viable as functional food ingredient.
