Effects of Aqueous Extracts from Angelica dahurica Benth. on ${\alpha}-Melanocyte$ Stimulating Hormone-induced Melanogenesis in Bl6 Mouse Melanoma Cell (백지(白芷)의 ${\alpha}-Melanocyte$ Stimulating Hormone에 의해 유도된 Bl6 흑색종 세포의 멜라닌 형성에 미치는 영향)

  • Melanin determines phenotypic appearance and its election-opaque property protects cells from physical, including ultraviolet (UV) radiation, and chemical stimuli such as free radicals. However hyper-pigmentation is associated with various skin diseases such as keloid scar. The aim of present study was to investigate the effects of aqueous extracts from Angelica dahurica Benth. (AEAD) on ${\alpha}-Melanocyte$ stimulating hormone $({\alpha}-MSH)-induced$ melanogenesis in B16 mouse melanoma cell. Relative high doses ($5\;mg/m{\ell}$) of AEAD could inhibit melanin formation without apoptotic death in cells treated with ${\alpha}-MSH$. And also, ${\alpha}-MSH-induced$ activation of tyrosinase was inhibited in cells treated with AEAD. These results suggest that AEAD inhibit melanogenesis through inhibiting tyrosinase activity, and also, AEAD may apply to develop whitening drugs and cosmetics.

A Development and Validation of Cosmetic Container Based on L-Ascorbic Acid Oxidation Property (L-Ascorbic Acid의 산화특성에 따른 화장품 용기 개발 및 유효성 분석)

  • L-ascorbic acid, the representative antioxidants, has a great effect on skin whitening, collagen synthesis, and anti-aging, but has low oxidative stability during storage. Therefore, in this study, thermal and oxidation properties of L-ascorbic acid under various storage conditions (powder, aqueous phase, changes of temperature, UV-irradiation, and inflow of external air etc.) were investigated. And the storage stability of ingredient was validated in the double-spaced pouch by analysing oxidation properties under each storage conditions (powder phase and blended with essence). In oder to analyze the thermal properties, TGA, DSC, and FT-IR analysis were carried out and UV-visible spectrophotometer & redox titration were used in parallel for oxidation property analyses. From the result of experiment, L-ascorbic acid was oxidized fast when it contained lots of metallic ion, hydroxy ion in aqueous solution under high temperature, UV-irradiation & inflow external air, whereas it was not oxidized for a long time when it was stored as pure powder although it has same condition as heating up, UV-irradiation & inflow external air. Based on this result, retention period of cosmetics which is using L-ascorbic acid, less stable material in oxidation can be innovatively increased when using double-spaced pouch that is designed and produced for separating storage of active ingredients.

  • We propose two feedback interference cancellers(FICs) to improve the performance of the equalization digital on-channel repeater(EDOCR) with the FIC for the ATSC DTV broadcasting system. The FIC estimates the feedback channel between Tx. and Rx. antennas of the repeater by cross-correlating the reference signal and the feedback signal. Since there is a DC pilot which ruins the white property of the ATSC DTV signals, the FIC cannot estimate the feedback channel accurately. To solve the problem, the structural method which uses an additional DC pilot free reference for feedback channel estimation and the algorithmic method based on the digital signal processing which whitens the ATSC DTV signals and performs the feedback cancellation in the whitened signal domain. Simulation results show that the proposed two FICs show better feedback cancellation performance than the conventional FIC.

  • The aqueous extract from Asiasari radix (AEAR) was used to investigate the effect of ${\alpha}$-melanocyte stimulating hormone induced melanogenesis in B16F10 mouse melnoma cells. The treatment with AEAR at the 1.0 and 2.0 mg/ml level significantly inhibited the biosynthesis of melanin without changes of cell growth and morphology compared with untreated control. The AEAR-treated cells at the 2.0 mg/ml level were more efficient than commercial arbutin at 0.1 mg/ml. The tyrosinase activity also significantly decreased in AEAR-treated cells at the 1.0 and 2.0 mg/ml level. The Western analyses confirmed the slightly decreased expression of tyrosinase by AEAR treatment. These results indicate that AEAR may contribute to the inhibition of melanin biosynthesis through regulating tyrosinase activity and expression and serve as a new candidate in the design of new skin-whitening or therapeutic agents.

Identification of Pigment Separated from Cockle Shell (새조개 연체부위의 색소 동정)

  • Physicochemical characteristics of pigment separated from cockle shell were studied. Pigment on the cockle shell was peeled with distilled water, and soaked in 4% TCA solution for 30 minutes and filtered. Filtrate was washed 3 times with distilled water and Iyophilized. Dried pigment was dissolved with 2N NaOH solution and preparative TLC with 2N NaOH : acetone(1 : 1). UV/vis spectrum of pigment separated from cockle shell and melanin as standard was same one peak at UV area and λ$_{max}$was 226nm. Dissolved property of pigment was same as that of melanin. Pigment was not ommochrome but like melanin, because it was insoluble in formic acid, whitening in $H_2O_2, $ and pattern of UV-spectrum was same as melanin. Pigment separated from cockle shell was stable at high temperatures. In thermal treatment at $70^{\circ}C, $ $80^{\circ}C, $ $90^{\circ}C$ and $100^{\circ}C$ for 8 hours, retention ratio of pigment separated from cockle shell were 95.0%, 93.3%, 90.8% and 87.6%, respectively.

Consideration of preservation methods for plant genetic resources in natural monument - Focusing on preparation for becoming effective of Nagoya Protocol - (천연기념물 식물유전자원의 보존방안 고찰 - 나고야의정서 발효 대비 중심으로 -)

  • Natural Monument is a designated cultural property as part of the country. According to Article 2 of the Cultural Properties Protection Act, a national, ethnic and global heritage artificially or naturally formed, with a great historical, artistic, scientific and landscape significance is defined as a cultural heritage. Animals, plants, topography, geology, minerals, caves, biological products and special natural phenomena, having a great of historic, scenic and scientific value, are defined as the monument. According to Article 3 of Cultural Properties Protection Act, the conservation, management and utilization of National Heritage should be kept intact in its original form. So, Natural monuments are managing as retained its original form under the Basic Principles of current law. The highest population of coniferous tree in natural monument plant is ginkgo tree including 22 objects, followed by pines, junipers that order. And in case of broadleaf tree, there are zelkova trees, retusa fringe trees, pagoda trees, cork oaks, silver magnolias and etc. There are many of reported efficacy in available natural monument plants. The efficacy of plant species on pharmaceutical like anti-cancer, anti-diabetic, anti-obesity, antioxidant activity, neuroprotective, improves cholesterol, anti-inflammatory, liver protection and anti-bacterial efficacy, on cosmetics and beauty like the inhibiting formation of skin wrinkles, whitening effect, variety of materials and the efficacy of the proposed utilization of its various papers and etc have been widely reported. Before the Nagoya Protocol enters into force, the future role of the National Research Institute for Cultural Properties Administration of Cultural Heritage should be obtain a legal right to manage the social, cultural and national natural monument with emotional value to the plant genetic resource as a natural monument efficient ways to study and preserve traditional knowledge biological resources by securing a claim to the sovereignty of the material will be ready.
