Diethylnitrosamine-induced hepatic tumorigenesis in rats 1. Gross findings of livers (Diethylnitrosamine 을 투여한 rat 간장의 tumorigenesis에 관하여 1. 간장의 육안적 소견)

  • The study was designated to investigate the gross findings following treatment with diethylnitrosamine (DEN) in rats. Forty four male rats (Sprague Dawley), initially 5 to 6 weeks of age and 120 to 150gm in body weight were continuously given water containing 0.01% DEN for 13 weeks and 3~6 rats per week were randomly sacrified at intervals of a week from 8 weeks to 17 weeks. The numbers of rats died until 17 weeks were 11 rats and first death occurred at 10 weeks. Body, liver and spleen weights were weighed and the relative weight levels of the liver and spleen per body weights were increased from 4.4% and 0.5% at control groups to 15.1% and 1.1% at 17 weeks. The numbers of RBC were not varied but PCV were decreased from 44.5% in control group to 27.5% in 16 weeks. A few of greyish-white foci or nodules of tumors were developed or not on the surfaces of the livers at 8 weeks but were developed on all livers after 9 weeks. The diameter of the largest tumor from all rat livers was 35.8 mm at 12 weeks. The numbers of developed tumors per a rat liver were appeared to be about 20 in 13 weeks and about 50~60 in 17 weeks. The diameter of some larger tumors were found to be 3~9mm in 13 weeks and 7~15mm in 17 weeks and more number of tumors were developed in the visceral surfaces than in the diaphragmatic surfaces of the livers. The parenchymae of livers were fragility.

A Study on the Tonic Effects of Ginseng - Effects of Ginseng Saponins on the Rat Heart (인삼의 강장효과에 관한 연구 - 백서 심장에 대한 인삼사포닌의 효과)

  • The investigation is concerned with the action of ginseng saponin on the contractile force in the rat heart and with the elucidation of the mechanism of the action. The effect of total ginseng saponin, ginsenoside Rb$_{1}$ of protopanaxadiol derivatives and ginsenoside Re of protopanaxatriol derivatives on the contractile force in isolated spontaneously beating normal rat heart was investigated. Total ginseng saponin was obtained from white ginseng by the method of Shibata and Namba. Ginsenoside Rb$_{1}$ and ginsenoside Re were isolated by the method of and Han, respectively. Total ginseng saponin exhibited a slight increase of the contractile force. Ginsenoside Rb$_{1}$ increased markedly the contractile force and dose dependent increase in contractile force was observed. However, ginsenoside Re did not increase the contractile force, but it prevented spontaneous decrease of the contractility of the heart. The mixture of the same dose of ginsenoside Rb$_{1}$ and Re showed a slight increase in the contractile force and its effect was similar to that obtained by total ginseng saponin. Pretreatment with propranolol abolished the positive inotropic effect of ginsenoside Rb$_{1}$ and the positive inotropic effect of ginsenoside Rb$_{1}$ was not observed in a reserpinized rat heart. Pretreatment with ginsenoside Re decreased or abolished the positive inotropic effect of epinephrine. Activities of Na+, K+ -ATPase were inhibited by ginsenoside Rb$_{1}$, total ginseng saponin and ginsenoside Re and these inhibitory effects were dose dependent. The results suggest that catecholamine release or inhibition of Na+, K+ -ATPase activities may be involved in the positive inotropic effect of gindenoside Rb$_{1}$. Ginsenoside Re counteracted the positive inotropic effect of ginsenoside Rb$_{1}$.

Effect of Thyroid hormone on Lipogenesis in Rat White and Brown Adipocytes Culture System

  • Thyroid hormone(T3) stimulates hepatic lipogenesis by increasing expression of genes, indluding acetyl-CoA carboxylase and fatty acid synthase. S14 protein, which is thougth to be involved in lipid metabolism , appears to respond in parallel . Effect of T3 on lipogenesis in white and brown adipose tissue are less clear, and may be complicated by indirect effects of the hormone. We developed an adipocytes system where the indirect effects of thyroid hormone are abolished and direct effects of T3 on lipogenesis could be tested. Fat accumulation was mesured by Oil-Red O staining. Insulin clearly enhanced fat accumulation by 2-fold . Isobutylemethylxanthie(IBMX) apeared to inhibit insulin -stimulated fat accumulation. Dexamethasone increased insulin-stimulatedfat accumulation about 1.3-fold. confluent adipocytes were cultured in serum-free medium or medium containing 10% fetal calf serum or 10% fetal calf serum stripped of thyroid hormone and lipogenesis, assessed by the incorporation of 3H2O , was measured. Medium without serum or supplemented with T3-depleted serum did not amplify the stimulatory effect of T3 on lipogenesis compared to medium containing 10% fetal calf seru. Dexamethasone alone led to a decrease inlopogenesis of about 50 % in white adipocytes and 25% in brown adipocytes. However, dexamethasone amplified the lipogenic respnse to T3 by about 30% in whit eadipocytes and 60% in brown adipocytes. T3(1$\mu$M) stimulated lipogenesis and acetyl-CoA carboxylase and fatty acid syntase mRNA levels up to 2 -fold in both types of adipocytes. It seems that these adipocytes systems are as useful model to study the effects of hormones on lipogenic gene expression as well as lipogenesis.

Comparisons of Renoprotective Activities between White Ginseng Radix and Rootlet in Spontaneously Hypertensive Rats with Diabetes

  • The renoprotective activities of white ginseng radix and rootlet were compared in spontaneously hypertensive rat (SHR) with diabetes. During oral administration of white ginseng radix (Ginseng Radix Alba, GRA) and white ginseng rootlet (Ginseng Radix Palva, GRP) for four weeks, arterial blood pressure and blood glucose levels were determined at every 10 days. In both GRA- and GRP-treatment groups, arterial blood pressures started to go down after 10 days of administration and maintained throughout the study period. After four weeks administrations of GRA and GRP, diastolic blood pressures were significantly decreased with 17% and 9%, respectively. GRA treatment also decreased blood glucose levels after 10 days of administration when compared with diabetic SHR group. At the end of the experiment, serum creatinine (Scr) and blood urea nitrogen (BUN) were not significantly different between the groups, except 62% higher value of BUN in diabetic SHR group when compared with SHR group. In the diabetic SHR group, the excretion of urinary albumin was increased significantly when compared with SHR. The level of urinary albumin in GRA treated group was markedly reduced when compared with diabetic SHR group $(67.8{\pm}4.7\;vs.\;131.3{\pm}13.5\;mg/24\;h).$ To examine the effects of ginseng radices on an overt diabetic nephropathy, index of kidney hypertrophy and transforming growth $factor-{\beta}1\;(TGF-{\beta}1)$ protein levels were evaluated. The glomerular and tubular cells stained positive for $TGF-{\beta}1$ seemed to be more abundant in diabetic SHR than in those with SHR, and GRA treated rats showed somewhat less $TGF-{\beta}1$ protein in glomerular and tubular cells when compared with diabetic SHR. Our results suggest that GRA might be a useful antihypertensive and antidiabetic agent with renoprotective effect.

Effect of White and Red Panax ginseng Extract on Serum Lipids Level in High-fat-diet Fed Rats (백삼 및 홍삼추출물이 고지방 식이 급여 흰쥐의 혈청 지질 함량에 미치는 영향)

  • This study was carried out to determine the effect of white and red Panak ginseng on body weight gain, feed efficiency, epididymal fat weight, and total serum cholesterol and triglyceride levels in male rats (Wister/ST, 8-week-old, 320 g) fed high fat diet. Rats were divided largely into two groups; normal control (NC) and high-fat-diet (HFD)-fed groups. HFD-fed animals were subdivided into 2 groups; HFD-fed control (FC), and HFD- and ginseng-fed groups. HFD and ginseng fed animals were furthor subdivided into white (WG)- and red ginseng (RG)-treated groups. Ginseng-treated groups received ethanol extract at daily doses of 250, 500, 1,000 mg/kg b.w. for 4 weeks. Consequently, a total of 40 rats were divided into 8 groups. Body weight gain of WG-500 (P<0.05), WG1000 (P<0.01) and RG500 (P<0.05) was significantly lower than that of the NC. Feed efficiency showed same result. The epididymal fat weight of WG500, WG1000 and RG250 was markedly lower (P<0.05) than that in FC. Total serum cholesterol level of WG250, WG500, WG1000 and RG1000 groups were significantly lower (P<0.05) than that of FC. Decrease in the total cholesterol level was most significant in WG100 group. Treatment of ginseng significantly reduced serum free cholesterol and triglyceride levels of the HFD-fed rats irrespective of white or red ginseng. Among the test groups, administration of ginseng at daily dose of 1,000 mg/kg b.w. showed the most potent free cholesterol and triglyceride lowering activity. These results led us to the conclusion that administration of ginseng lowers serum total or free cholesterol and triglyceride levels in HFD-fed animals. Moreover the beneficial effect of white ginseng was slightly more potent than that of the red ginseng.

The Effect of Tokoro Rhizoma Pharmacopuncture at $KI_{10}$ in Lipopolysaccharide Induced Acute Nephritis in Rats (음곡에 시술한 비해약침이 Lipopolysaccharide로 유도된 흰쥐의 신장염에 미치는 영향)

  • Objectives : This study was designed to evaluate the effects of Tokoro Rhizoma pharmat $KI_{10}$ in nephritis induced by lipopolysaccharide(LPS) in rat. Methods : Rats were divided into 4 groups and 3 groups(LPS, saline, Tokoro Rhizoma pharmacopuncture(TR-P) group) were injected LPS to induce nephritis. TR-P group was treated with TR at $KI_{10}$ three times for a week, saline group with normal saline. To evaluate the effects of TR at $KI_{10}$ on nephritis in rats, white blood cell(WBC), neutrophil in blood, creatinine, cytokine-induced neutrophil chemoattractant-1(CINC-1) in serum and urinary volume, creatinine in urine, renal myeloperoxidase(MPO) were measured and renal tissue was analyzed. Results : TR-P group significantly reduced WBC, neutrophil in blood, creatinine, CINC-1 in serum, creatinine in urine and renal MPO than LPS group. TR-P group increased urinary volume but, not significant. Conclusion : TR at $KI_{10}$ has a therapeutic effect on nephritis in LPS stimulated rat. Therefore, it is suggested that TR at $KI_{10}$ may be an useful therapeutics for nephritis in clinical field after further researches.

  • Demonstration of the highly positive correlation between blood cholestrol levels and heart disease has made consumers wary of the fats in meat, milk and eggs. The egg, as perhaps the single largest common source of cholesterol, has been cited by many members of the medical and scientific world as a food contributing to heart disease. In light of decreasing per capita egg consumption and continuing dietary egg cholesterol controversy, many researchers have focused their efforts on egg nutrition. The results reported, however, are often contradictory. In spite of the disputable scientific evidence, the egg has been labelled (erroneously) as a highly cholesterogenic food. The objective of this presentation is to present a general picture of the problem and discuss our laboratory findings relevant to the problem. An isotope technique was utilized to incorporate $^{14}C$-cholesterol into egg yolk lipoproteins and study the metabolic fate of dietary ovo-cholesterol in rats. Two hundred and fifty micro-curies of 4-$^{14}C$-cholesterol, emulsified in corn oil, were orally administered to five Single Comb White Leghorn laying hens. Eggs were collected, hard-boiled, and the hot dried egg yolk powder (HEY) was prepared. Total radioactivity excreted via feces was determined. The rat groups fed egg yolk powder excreted more than 95% of the ingested ovo-cholesterol, whereas the rat chow group excreted only 47%. No difference was observed between HEY and CEY treatments. Therefore, an unknown lipid factor present in egg folk accelerates cholesterol turnover rate and excretion via feces.

The Effects of Panax gineseng on the Side Effects of Cyclophosphamide to the Spleen in the Rat (인삼추출물이 Cyclophosphamide에 손상된 흰쥐 비장조직의 회복에 미치는 영향)

  • This study were performed for the effects of Panax ginseng on the side effect of Cyclophosphamide to splenic tissue in the Rats. The experimental animals were divided into control group, test group by the way of the method treatment of the drug. Each group was sacrificed and stained in accordance with the schedule and observed under light microscope. The results of this study were as follows : 1. After the ginseng extract administration rat's weight and volume were similarly increased than normal and control group. 2. The decrease of the numbers of the splenic tissue after treatment of cyclophosphamide(CP) were recovered with prescription of the ginseng extract; the decrease of white pulp, red pulp, marginal zone were recovered. 3. Increased macrophage in red pulp of splenic tissue with administration of CP were disapeared after treatment of ginseng extract.

ELECTRON MICROSCOPIC STUDY OF THE IRRADIATION EFFECT ON THE RAT MANDIBULAR CONDYLE (방사선조사가 하악과두에 미치는 영향에 관한 전자현미경적 연구)

  • In attempt to determine radiation effect on the mandibular condyle of the growing rat, 27 white female rats (Sprague-Dawley) were divided into 3 groups and irradiated respectively 5Gy, 10Gy, 20Gy using MK Cell Irradiator. Mandibular condyles from rats on the day of 1, 7, 14 after the irradiation day were obtained, sectioned sagittally and examined by light microscopy, and thereafter middle portion through anteroposterior direction on the sagittal plane was selected to examine the ultrastructural change by transmission electron microscopy. The obtained results are followings. 1. In the proliferative zone some cells showed little organelles in case of 5Gy irradiation, in addition the number of degenerative cells increased and in case of 10Gy irradiation, and in case of 20 Gy irradiation total number of cells decreased. 2. In the hypertrophic zone, narrowing of width and partial disorder in hypertrophic process were noted in case of 5 Gy irradiation, and more prominent narrowing of width and more irregular disorder in hypertrophic process in case of both 10Gy and 20Gy irradiation. 3. In the upper hypertrophic zone some chondrocytes seemed to be dying and the polarity of nuclei could not be seen, if any. 4. The periodic observation showed the severest change at day 7 and the signs of recovery at day 14 after irradiation.

Disease vector occurrence and ecological characteristics of chiggers on the chestnut white-bellied rat Niviventer fulvescens in Southwest China between 2001 and 2019

  • Chigger mites are the vector of scrub typhus. This study estimates the infestation status and ecological characteristics of chiggers on the chestnut white-bellied rat Niviventer fulvescens in Southwest China between 2001 and 2019. Chiggers were identified under the microscope, and infestation indices were calculated. The Preston's log-normal model was used to fit the curve of species abundance distribution. A total of 6,557 chiggers were collected in 136 of 342 N. fulvescens rats, showing high overall infestation indices (prevalence=39.8%, mean abundance=19.2, mean intensity=48.2) and high species diversity (S=100, H'=3.0). Leptotrombidium cangjiangense, Neotrombicula japonica, and Ascoschoengastia sifanga were the three dominant chigger species (constituent ratio=42.9%; 2,736/6,384) and exhibited an aggregated distribution among different rat individuals. We identified 100 chigger species, with 3 of them (Leptotrombidium scutellare, Leptotrombidium wenense, and Leptotrombidium deliense) as the main vectors of scrub typhus in China and nine species as potential vectors of this disease. Disease vector occurrence on N. fulvescens may increase the risk of spreading scrub typhus from rats to humans. Chigger infestation on N. fulvescens varied significantly in different environments. The species abundance distribution showed a log-normal distribution pattern. The estimated number of chigger species on N. fulvescens was 126 species.
