A Novel Medium for the Enhanced Production of Cyclosporin A by Tolypocladium inflatum MTCC 557 Using Solid State Fermentation

  • Cyclosporin A (CyA) produced by Tolypocladium inflatum is a promising drug owing to its immunosuppressive and antifungal activities. From an industrial point of view, the necessity to obtain a suitable and economic medium for higher production of CyA was the aim of this work. The present study evaluated the effect of different fermentation parameters in solid state fermentation, such as selection of solid substrate, hydrolysis of substrates, initial moisture content, supplementation of salts, additional carbon, and nitrogen sources, as well as the inoculum age and size, on production of CyA by Tolypocladium inflatum MTCC 557. The fermentation was carried out at $25{\pm}2^{\circ}C$ for 9 days. A combination of hydrolyzed wheat bran flour and coconut oil cake (1:1) at 70% initial moisture content supported a maximum production of $3,872{\pm}156\;mg$ CyA/kg substrate as compared with $792{\pm}33\;mg/kg$ substrate before optimization. Furthermore, supplementation of salts, glycerol (1% w/w), and ammonium sulfate (1% w/w) increased the production of CyA to $5,454{\pm}75\;mg/kg$ substrate. Inoculation of 5 g of solid substrate with 6 ml of 72-h-old seed culture resulted in a maximum production of $6,480{\pm}95\;mg$ CyA/kg substrate.

  • Addition of the treated dietary fiber sources to wheat flour were generally decreased at Amylograph viscosity as the mixing ratio increased. Addition of the treated dietary fiber sources on the preparation of noodles increased weight and volume of cooked noodles but decreased extention force as the mixing ratio increased. The sensory test of the dietary fiber sources mixed noodles supplemented by treated soymilk residue 5% was excellent sensual properties. The properties of the dietary fiber sources mixed noodles supplemented by treated soymilk residue 10% and treated apple pomace 5% were nearly the same in the texture organoleptic properties compared with those of wheat flour noodle.

Apparent Digestibility Coefficients of Various Feed Ingredients for Olive Flounder, Paralichthys olivaceus (사료원료에 대한 넙치 Paralichthys olivaceus 소화율 평가)

  • The apparent digestibility coefficients of dry matter, crude protein and energy in white fishmeal, herring meal, anchovy meal, salmon meal, sardine meal, mackerel meal, squid meal, soybean meal, corn gluten meal and wheat flour were determined for olive flounder. Digestibility coefficients were determined using a reference diet and test diets that contained 70% of the reference diet mixture and 30% test ingredients. All diets contained 0.5% chromic oxide as a digestibility indicator. The fish averaging 220 g were held in 500 L tanks at a density of 20 fish per tank. Feces were collected from three replicated groups of fish using a fecal collection column attached to a fish-rearing tank. The apparent dry matter, crude protein and energy digestibility coefficient values observed were in the ranges 29-79%, 59-95% and 45-91%, respectively, for various test ingredients. The apparent dry matter, crude protein and energy digestibility of white fishmeal, herring meal, anchovy meal, salmon meal, sardine meal, mackerel meal and squid meal were significantly higher than those of soybean meal, corn gluten meal and wheat flour.

  • The result of analysis of ingredient of Kochujang is as follows. The experiments were prepared with wheat flour (WF) and glutinous rice(GR) ; the experimental A type is made of full WF, B type, WF 75% and GR 25%, C type, WF 50% and GR 50% and D type, only full GR. The N content of crude protein and amino type N is higher in the order of A, B, C, and D types, ethyl alcohol is higher in the order of D, C, B, and A types in aging process. The pH is some what higher in A type but in moisture and NaCl, not much difference were shown in the experimental types. The isolated sugars of Kochujang ripened for 90 days analyzed out glucose, fructose, maltose and rhamnose with glucose being the largest in quantity. Glucose is higher in A type, fructose in B type. The alcohols of the ripened Kochujang analyzed out n-propyl, iso-butyl, and iso-amyl alcohol, the content of which is below 3.2mg% and did not show much difference in each experimental types.

Changes in Physical and Microbial Properties of Starchy Pastes Added Kimchi during Fermentation (전분질첨가 김치의 숙성 중 물리적 및 미생물학적 특성의 변화)

  • Wheat flour and glutinous rice pastes added Kimchi were fermented at 10$^{\circ}C$ for 14 days. The numbers of total viable microorganisms and lactics were counted. Textural property by compression force and relevant changes in pectic composition were determined. Other physical properties such as color and viscosity of Kimchi juices were evaluated. Regarding the microorganism counts, the total cell counts of all Kimchi samples increased until 5th days but thereafter all decreased during fermentation. The number of lactics showed same tendency in all Kimchi samples. The compression force of control, wheat flour paste added Kimchi (WHFP-Kimchi) and glutinous rice paste added Kimchi (GLRP-Kimchi) decreased up to 72.0%, 77.0%, 66.5% respectively. During fermentation, hot water soluble pectin (HWSP) increased, whereas sodium hexametaphosphate soluble pectin (HXSP) and HCI soluble pectin (HCISP) decreased. The red chromaticity of all Kimchi juices decreased. The viscosity of all Kimchi samples decreased in the order of GLRP-Kimchi, WHFP-Kimchi and control.

Quality Characteristics of Castella with Chungkukjang (청국장을 첨가한 카스테라의 품질 특성)

  • Castella sponge cake was prepared by partially replacing wheat flour with Chungkukjang, a Korean fermented soybean. The physico-chemical, textural and sensory characteristics were then investigated. Specific volume and expansion ratio decreased with increasing Chungkukjang content. The addition of Chungkukjang decreased the moisture content and increased the protein content of castella. Color determination showed addition of Chungkukjang darkened the internal color of castella, probably due to browning caused by the Maillard reaction. Castella with 20 or 30% Chungkukjang had a higher textural hardness than other castellas. A sensory panel perceived that the external and internal color of castella darkened with Chungkukjang substitution. When Chungkukjang was added at or above 20%, castella had a weaker sweet flavor, a stronger roasted flavor, and less lightness and softness than other castellas as perceived by the sensory panels. Up to 30% of the wheat flour could be replaced by Chungkukjang without diminishing acceptability.

  • The optimum cultural conditions for production of yellow pigment by Monascus purpureus MMK2 were investigated in liquid culture. Monascus purpureus MMK2 was shown to give the maximum production of yellow pigment in the medium containing of 3.0% wheat flour, 0.15% $NaNO_3$, 0.25% $Na_2HPO_4\;12H_2O\;and\;0.15%\;MgSO_4\;7H_2O$. The optimal culture conditions for temperature and initial pH were $30^{\circ}C$ and 6.5, respectively. The yellow pigment production reached a maximum level at the 7th day of cultivation.

  • This study determined the optimal mixing ratio of functional materials to enhance the functionality of batter for fish cutlets. An optimal mixing ratio of the hiziki Hizikia fusiformis, fig fruit Ficus carica L. and pear Pyrus pyrifolia Niitaka extracts (HFP extract) of 3:1:1 was found. The physicochemical properties of the mixture were as follows: total phelonic contents of $1187.29{\pm}41.55mg/L$, 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity of $61.50{\pm}2.33%$, ascorbic acid content of $23.68{\pm}0.02g/100g$, ${\beta}-carotene$ content of $51{\pm}3.92{\mu}g/100g$, and sugar content of $12.7{\pm}0.1Brix^{\circ}$. A mixture of the extract, Korean herbal powder, and wheat flour was prepared as the fish cutlet batter. The fish cutlet prepared with the batter had a hardness of $166-202g/cm^2$ and Hunter's color scale values L of $31.14{\pm}2.1$ to $34.62{\pm}1.3$, a of $-4.89{\pm}0.21$ to $-2.26{\pm}0.19$ and b of $-20.65{\pm}2.65$ to $28.93{\pm}3.14$. The FRAP (ferric reducing antioxidant power) and VBN (volatile basic nitrogen) of the fish cutlets were measured to evaluate functionality and determine the optimal mixing ratio for cold storage at $-12^{\circ}C$, $-18^{\circ}C$, and $-24^{\circ}C$. The optimum mixture ratio of extracts /Korean herbal powder/ wheat flour was 1:6:3.

  • The optimum cultural conditions for production of Monascus natural pigment by Monascus purpureus MK2 were investigated in submerged culture. This strain was showed the maximum production of monascus natural pigment in the optimal medium of 3.0% wheat flour, 0.15% $NaNO_3$, 0.2% $K_{2}HPO_4$ and 0.05% $MgSO_4{\cdot}7H_{2}O$, at pH 7.0. The maximum production of this pigment was achieved at $30^{\circ}C$ for 7 day cultivation under 130 rpm shaking. At optimal condition, Monascus purpureus MK2 produced 29.10, 36.84 and 48.92 units of yellow, orange and red pigment, respectively.
