함정 신호해석 연구에 필요한 해양기상환경 자료의 표본추출에 관한 연구 (A Study on the Sampling of Ocean Meteorological Data to Analyze Signature of Naval Ships)

  • 본 연구에서는 함정 신호해석 연구에 필요한 해양기상환경 자료의 표본추출에 관한 연구를 수행하였다. 이를 위해 한국 기상청으로부터 품질검사가 완료된 최신의 해양 기상자료를 수집하고, 측정 자료의 가용 구간을 설정하여 이상치를 제거하였다. 품질검사와 이상치를 제거한 자료는 함정 신호의 확률론적 해석을 위해 지정학적 위협과 부이의 유효면적을 고려하여 단순임의추출법을 통해 축소되었다. 이후 표본의 크기는 모집단의 크기와 신뢰수준 95%를 고려하여 100, 200 그리고 400개로 설정하였으며, 상관관계가 높은 수온과 기온을 기준으로 하는 2차원 층화추출법으로 최종표본을 추출하였다. 추출 표본은 제곱오차의 합과 신뢰구간을 추정하고 이를 비교하여 함정의 확률론적 신호해석을 위해 필요한 해양기상조건의 크기를 제시하였다.

추출 시간을 달리하여 제조한 대구 육수의 품질 특성 - 고압 가열 추출 방식으로 - (Quality Characteristics of Cod Stock with Different Extraction Time - Using High Pressure Extraction Time -)

  • In this study, cod bone were used as the main ingredient of a stock that can be eaten with Korean dried noodles. The stock was produced with heating for 30, 40, 50, 60, or 70 minutes using high-pressure extraction methods. Moisture content, chromaticity, pH, sugar content, salinity, mineral contents, quantitative analysis, and overall acceptance were studied to determine the optimal heating time. In our result, S1 the showed highest water contents 98.10%, while S5 showed the lowest moisture content (97.47%). There were no significant difference in pH between samples, while salinity and sugar content increased with increases in heating time. In terms of mineral contents, sodium and potassium showed proportional difference with increases in heating time, however, magnesium, calcium, and iron did not change. The results from a quantitative analysis test showed that transparency, fishy smell, delicate flavor, savory flavor, salt taste, and umami taste became stronger with increases in heating time increase. From the above results, the overall taste of the stock increases with increases in high-pressure heating time. Specifically, S4 high-pressure heating scored the highest on appearance, flavor, taste, after-taste, and overall acceptance. Thus, 60 minutes of high-pressure heating time is most desirable when producing stock with cod bones as a main ingredient.

HPLC를 이용한 정공등의 다성분 동시함량분석 (Simultaneous Quantification Analysis of Multi-components on Erycibae Caulis by HPLC)

  • In this study, we developed and validated the HPLC method using the isolated components from Erycibae caulis. Their structures were elucidated by spectroscopic methods including UV, $^1H$-NMR, $^{13}C$-NMR, FAB-Mass and ESI-Mass as Compound 1 (crypto-chlorogenic acid), Compound 2 (scopolin), Compound 3 (neochlorogenic acid) and Compound 4 (3,4-di-O-caffeoylquinic acid). Major three compounds and scopoletin were decided as representative components of Erycibae caulis. We established HPLC analytical method by using the representative components and 20 commercial samples which were collected considering to various cultivated area. The HPLC fingerprinting was successfully achieved with an AKZO NOBEL Kromasil 100-5C18 column. The mobile phase consisted of 0.5% acetic acid in water (A) and methanol (B) using gradient method of 85(A) to 50(A) for 35min. The fingerprints of chromatograms were recorded at an optimized wavelength of 330 nm. This developed analytical method was validated with specificity, selectivity, accuracy and precision. And it is suggested that scopolin, scopoletin, neochlorogenic acid, 3,4-di-O-caffeoylquinic acid were more than 0.162%, 0.133%, 0.057%, 0.044%, respectively. In addition, principal component analysis (PCA) was performed on the analytical data of 20 different Erycibae caulis samples in order to classify samples collected from different regions. We hope that this assay can be readily utilized as quality control method for Erycibae caulis.

SECOND ATLAS DOMESTIC STANDARD PROBLEM (DSP-02) FOR A CODE ASSESSMENT

  • KAERI (Korea Atomic Energy Research Institute) has been operating an integral effect test facility, the Advanced Thermal-Hydraulic Test Loop for Accident Simulation (ATLAS), for transient and accident simulations of advanced pressurized water reactors (PWRs). Using ATLAS, a high-quality integral effect test database has been established for major design basis accidents of the APR1400 plant. A Domestic Standard Problem (DSP) exercise using the ATLAS database was promoted to transfer the database to domestic nuclear industries and contribute to improving a safety analysis methodology for PWRs. This $2^{nd}$ ATLAS DSP (DSP-02) exercise aims at an effective utilization of an integral effect database obtained from ATLAS, the establishment of a cooperation framework among the domestic nuclear industry, a better understanding of the thermal hydraulic phenomena, and an investigation into the possible limitation of the existing best-estimate safety analysis codes. A small break loss of coolant accident with a 6-inch break at the cold leg was determined as a target scenario by considering its technical importance and by incorporating interests from participants. This DSP exercise was performed in an open calculation environment where the integral effect test data was open to participants prior to the code calculations. This paper includes major information of the DSP-02 exercise as well as comparison results between the calculations and the experimental data.

안동・임하호 저니토의 토양 성분과 중금속 용출 특성 (Soil Components and Elution Characteristics of Heavy Metals in Sediments of Andong and Imha Reservoir)

  • 본 연구는 안동지역에 소재하는 안동 임하호 저니토가 호소수에 미치는 영향을 조사하기 위하여 안동호와 임하호 6개 지역에서 채취한 저니토의 토양성분과 중금속 용출 특성을 분석하였다. 대부분의 저니토는 illite(I), kaolinite(Ka), quartz(Q) 및 feldspar(F)로 구성되어 있다. 저니토에 포함된 중금속 중 아연(Zn), 비소(As), 크롬(Cr), 구리(Cu), 납(Pb)의 농도는 임하호에 비하여 안동호에서 상대적으로 높게 나타났다. 특히 안동호의 저니토에서는 다른 중금속들에 비해 비소(As)와 아연(Zn)의 용출이 크게 나타났다. 저니토를 이용한 흡착실험에서는 아연(Zn), 카드늄(Cd) 및 구리(Cu)의 흡착은 적게 일어난 반면 납(Pb)과 크롬(Cr)은 높은 흡착력을 보여주었다.

Trend analysis of rainfall characteristics and its impact on stormwater runoff quality from urban and agricultural catchment

  • Climate change has significantly affected the rainfall characteristics which can influence the pollutant build-up and wash-off patterns from the catchment. Therefore, this study explored the influence of varying rainfall characteristics on urban and agricultural runoff pollutant export using statistical approaches. For this purpose, Mann-Kendall and Pettitt's test were applied to detect the trend and breakpoint in rainfall characteristics time series. In addition, double mass curve and correlation analysis were used to drive the relationship between rainfall-runoff and pollutant exports from both catchments. The results indicate a significant decreased in total rainfall and average rainfall intensity, while a significant increased trend for antecedents dry days and total storm duration over the study periods. The breakpoint was determined to be 2013 which shows remarkable trend shifts for total rainfall, average rainfall intensity and antecedents dry days except total duration. Double mass curve exhibited a straight line with significant rainfall-runoff relationship indicates a climate change effect on both sites. Overall, higher pollutant exports were observed at both sites during the baseline period as compared to change periods. In agricultural site, most of the pollutants exhibited significant (p< 0.05) association with total rainfall, average rainfall intensity and total storm duration. In contrast, pollutants from urban site significantly correlated with antecedent dry days and average rainfall intensity. Thus, total rainfall, average rainfall intensity and total duration were the significant factors for the agricultural catchment while, antecedents dry days and average rainfall intensity were key factors in build-up and wash-off from the urban catchment.

Optimization of fish oil extraction from Lophius litulon liver and fatty acid composition analysis

  • The Lophius litulon liver was used as raw material for the extraction of fish oil via various extraction methods. The extraction rate by water extraction, potassium hydroxide (KOH) hydrolysis and protease hydrolysis were compared and the results revealed the protease hydrolysis extraction had a higher extraction rate with good protein-lipid separation as observed by optical microscope. Furthermore, subsequent experiments determined neutrase to be the best hydrolytic enzyme in terms of extraction rate and cost. The extraction conditions of neutrase hydrolysis were optimized by single-factor experiment and response surface analysis, and the optimal extraction rate was 58.40 ± 0.25% with the following conditions: enzyme concentration 2,000 IU/g, extraction time 1.0 h, liquid-solid ratio 1.95:1, extraction temperature 40.5℃ and pH 6.5. The fatty acids composition in fish oil from optimized extraction condition was composed of 19.75% saturated fatty acids and 80.25% unsaturated fatty acids. The content of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were 8.06% and 1.19%, respectively, with the ratio (6.77:1) surpassed to the recommendation in current researches (5:1). The results in this study suggest protease treatment is an efficient method for high-quality fish oil extraction from Lophius litulon liver with a satisfactory ratio of DHA and EPA.

Development of dry milling suitable rice cultivar to invigorate rice processing products

  • Rice consumption has been continuously decreasing as the eating habits of Koreans have become westernized and diversified. The per capita annual rice consumption in Korea has dropped sharply from 136.4 kg in 1970 to 61.9 kg in 2016. The Korean government, therefore, has been trying to promote rice consumption by invigorating the processed food industry using rice flour. To facilitate the market for processed rice foods, it is essential to develop proper milling technology in terms of flour particle size and damaged starch content to produce high quality rice flour at competitive cost. Dry milling and wet milling are the two major processes used to produce rice flour. Although the dry milling process is relatively simple with a lower production cost, damaged starch content increases because of the high grain hardness of rice. In wet milling, the quality of rice flour is improved by reducing flour particle size as well as damaged starch content through soaking procedures. However, the production costs are high because of the additional expenses associated with the disposal of waste water, sterilization and drying of the wet flour. Recently developed technologies such as jet milling and cryogenic milling also require expensive investment and production. Therefore, developing new rice cultivars with dry milling adaptability as well as good processing properties is an important goal of rice breeding in Korea. 'Suweon 542' is a floury endosperm mutant line derived from sodium azide treatment on a high-yield, early maturing, and non-glutinous japonica rice cultivar, 'Namil'. Compared with the wild type, after dry milling process, the grain hardness of 'Suweon 542' was significantly lower because of its round and loosely packed starch granules. Also, the flour of 'Suweon 542' had significantly smaller particles and less damaged starch than 'Namil' and other rice cultivars and its particle size distribution was similar to a commercial wheat cultivar. Recently, through collaborations with nine universities and food companies, a total of 21 kinds of processed prototypes, using the dry milling flour of 'Suweon 542', were evaluated. In the production of major rice processing products, there was no significant quality difference between the flours prepared by wet milling and dry milling. Although the amount of water added to the dough was slightly increased, it was confirmed that the recipe applying the wet flour could be used without significant change. To efficiently transfer the floury endosperm characteristics of 'Suweon 542' to other commercial rice cultivars, it is essential to develop DNA marker tightly linked to the target gene. Association analysis using 70 genome-wide SSR markers and 94 F2 plants derived from 'Suweon 542'/'Milyang 23' showed that markers on chromosome 5 explained a large portion of the variation in floury grains percentage (FGP). Further analysis with an increased number of SSR markers revealed that the floury endosperm of 'Suweon 542' was directed by a major recessive locus, flo7(t), located in the 19.33-19.86 Mbp region of chromosome 5, with RM18639 explaining 92.2% of FGP variation in the F2 population. Through further physical mapping, a co-segregate and co-dominant DNA marker with the locus, flo7(t) was successfully developed, by which, thereby, breeding efficiency of rice cultivars having proper dry milling adaptability with high yield potential or useful functional materials would be improved. 'Suweon 542' maintained the early maturity of the wild type, Namil, which can be used in rice-wheat double cropping systems in Korea not only for improved arable land but also for sharing flour production facilities. In addition to the high susceptibility against major rice diseases, nevertheless, another possible drawback of 'Suweon 542' is the high rate of viviparous under prolonged rainfall during the harvesting season. To overcome susceptibility and vivipary of 'Suweon 542', the progeny lines, derived from the crosses 'Suweon 542' and 'Jopyeong', an early maturing rice cultivar with multiple resistance against rice blast, bacterial blight, and rice strip virus, and 'Heugjinju', a anthocyanin pigment containing black rice cultivar, were intensively evaluated. As the outputs, three dry milling suitable rice elite lines, 'Jeonju614', 'Jeonju615', and 'Jeonju616' were developed.

섬진강상류 유량측정지점의 유수단면적과 유량변화에 따른 상관관계 분석 (Analysis of Correlation with Cross Sectional Area of Flow and Flow Rate Variation of Discharge Measurement Point in the Upper Stream of Seomjin River)

  • 본 연구는 단면변화를 고려한 수위-단면적 변화 및 평균 유속-단면적 간의 상관관계를 분석하여 유량 산정 및 하천관리에 활용하고자 하였다. 수위에 따른 단면적 변화는 SB-A 지점은 1.0 m, 1.9 m, SB-C 지점은 0.6 m, 1.8 m, CL-A 지점은 1.0 m, 1.8 m, OS-A 지점은 0.6 m, 2.0m에서 발생되었다. 이 중 첫 번째 변화는 평 저수기에 해당되고, 두 번째 변화는 홍수기 및 하천 좌 우 안에 인위적 자연적으로 형성된 둔치 등으로 판단된다. 수위-단면적 변화에 따른 관계식의 기울기는 지수형 0.5539~1.9013, 선형 9.040~52.544의 범위를 가진다. 기울기는 두 곡선 모두 고수위로 갈수록 증가하는 경향을 보인다. 평균유속-단면적 변화의 관계는 지수와 직선의 방정식으로 지수형 기울기와 상관계수는 각각 0.1182~0.8734, 0.22~0.86이며, 선형의 기울기와 상관계수는 0.0028~0.1032, 0.20~0.87로 분석되었다. SB-A, SB-C 지점의 저수위는 다른 수위보다 상관관계가 높게 산정되었는데, 이는 수위구간이 좁고, 하천 단면적의 변화가 크지 않기 때문으로 판단된다. CL-A, OS-A 지점은 월류보의 영향으로 저수위일 경우에 상관관계가 낮았다. 수위-단면적, 평균유속-단면적의 상관관계 및 곡선식 등을 이용하여 하천 정비계획 등의 수립에 활용할 수 있으며, 제외지의 단면적이 변하는 지점의 유량 변화 등의 예측에 활용할 수 있을 것이다.

홍수 규모별 대청호에 유입하는 하천 밀도류의 특성 변화 (Variation of Inflow Density Currents with Different Flood Magnitude in Daecheong Reservoir)

  • 여름철 홍수시 성층화된 저수지로 유입하는 하천수는 저수지 표층수에 비해 낮은 수온과 높은 부유고형물질(SS) 농도를 가지므로 주변수에 비해 상대적으로 높은 밀도를 가지게 된다. 이러한 밀도차로 인해 형성된 밀도류의 저수지 내 진행과정은 수질과 수생태계에 큰 영향을 주게 된다. 따라서 하천수 밀도류의 거동분석은 저수지 수질관리를 위한 현장조사의 최적화, SS와 영양염류 등 오염물질의 이송 확산 해석에 중요한 요소이다. 본 연구의 목적은 기존 연구를 통해 검증된 2차원 수치모델을 이용하여 다양한 홍수규모에서 대청호로 유입하는 하천 밀도류의 거동 특성인 침강점 수심($d_p$)과 거리($X_p$), 분리점 수심($d_s$), 중층류 관입두께($h_i$), 댐축 도달시간($t_a$), 감소율(${\beta}$)을 분석함으로써 저수지 수질관리를 위한 기초정보를 제공하는데 있다. 모의조건은 평수년이었던 2004년 6월 13일부터 7월 3일 동안 발생한 홍수사상의 수문곡선을 기준으로 유입 유량의 규모를 10개의 등급으로 나누었으며, 초기 성층조건은 탁수가 유입되기 전의 발달된 성층구조를 적용하였다. 유입수와 저수지 성층구조의 특성치는 밀도 Froude 수(Fri)로 나타내었으며, 10개의 $Fr_i$ 조건별로 $d_p,\;X_p,\;d_s,\;h_i,\;t_a$, SS의 ${\beta}$값 등을 산정하였다. 연구결과 $d_p,\;X_p,\;d_s,\;h_i$는 대체로 $Fr_i$ 값과 비례하여 증가하였으며 중층류의 진행속도도 빨라지는 경향을 나타내었으나, 저수지 지형변화에 큰 영향을 받는 것으로 나타났다. 정상상태를 가정하는 Hebbert 식은 저수지 수위변화와 지형변화를 고려하지 못하기 때문에 수치모델 보다 $d_p$값을 과대 산정하였다. 유입 SS 농도의 감소율(${\beta}$)은 $Fr_i$가 클수록 작아지는 경향을 보였으나, $Fr_i$>9.0에서는 난류혼합효과 때문에 다시 증가하였다. 연구결과는 저수지운영 실무자들이 홍수규모별로 탁수의 초기 거동특성을 간단히 예측하는 목적으로 사용할 수 있다.
