Gene Expression Analysis of Immune Cell Activation Markers in Extracts of Platycodon grandiflorum Containing Medicinal Herbs (도라지 추출액과 한약재 함유 도라지 추출액에 의한 면역세포 활성 표지유전자 발현 분석)

  • Extracts of Platycodon grandiflorum have been reported to show anti-inflammatory, antioxidant, anti-metastatic, and hepato-protective effects. This study was designed to evaluate T-cell activation and M1/M2 differential macrophage activation by extracts of P. grandiflorum or P. grandiflorum containing various medicinal herbs. Using real-time RT-PCR, we analyzed expression levels of c-fos, and CD40L (T-cell activation markers) in splenocytes and iNOS, Ym1, and ARG1 in RAW 246.7 cells after treatment of CC (hot water extract of P. grandiflorum), MAEK (hot water extract of P. grandiflorum [82%] and six different plants), and HWAL (hot water extract of P. grandiflorum [7%] and eight different plants. The results showed that MAEK significantly elevated the expression of T-cell activation markers of splenocytes, with the c-fos gene activated more than 10-fold and the CD40L gene activated more than 6-fold. Although CD40L was significantly increased by CC and HWAL, the increase was only about 2-fold. In addition, CC and HWAL did not significantly activate the expression of the c-fos gene. On the other hand, CC elevated the M1 activation marker iNOS, and HWAL elevated the M2 activation marker Ym1 and ARG1 gene expression. In conclusion, MAEK could be used as an immune stimulant because of its ability to activate T cells (elicited c-fos and CD40L gene expression), whereas HWAL could serve as an anti-inflammatory agent because of its differential activation of M2 macrophages.

A Study on Dielectrical Constant under Ground Conditions (지반조건에 따른 유전상수 변화에 관한 연구)

  • In this study, dielectrical constant of the ground was measured using TDR method and correlated with water contents and density of ground. In order to evaluate the applicability as a cavity exploration, model experiments were carried out to analyze the effects of cavity size on the dielectrical constant. Test result indicated that dielectrical constant of the ground tended to linearly increase with the increase in water contents and density, which can be represented in a certain relational expression. Also, the dielectrical constant of ground varied sensitively with the cavity size of ground. The results conclude that the dielectrical constant, water contents and density of the ground proved to have a correlation among them, and the dielectrical constant is expected to be a basic data on cavity exploration.

  • Antiallergic and antipsoriatic effects of korean Red Ginseng (KRG, steamed root of panax ginseng C.A. Meyer, Family Araliaceae) were measured. Orally administered KRG water extract potently inhibited passive cutaneous anaphylaxis (PCA). KRG water extract also showed the potent inhibition in oxazolone-induced mouse dermatitis, and suppressed mouse ear swelling by $39\%$ at 16 days at a dose of $0.1\%$. KRG water extract reduced the levels of mRNA of cyclooxygenase (COX)-2, $IL-1\beta$, $TNF-\alpha$ and $INF-\gamma$ increased in oxazolone-applied mouse ears, however, did not inhibit that of IL-4. KRG water extract also inhibited iNOS and COX-2 mRNA expression level of RAW264.7 cell induced by lipopolysaccharide. Based on these findings, we suggest that KRG can improve atopic and contact dermatitis by the regulation of $ IL-1\beta$ and $TNF-\alpha$ produced by macrophage cells and $interferon-\gamma$ produced by Th1 cells.

Anti-rheumatiod Arthritis Effects of I-Myo-San Water Extract (이묘산(二妙散)의 항(抗)류마티즘 효능(效能)에 대한 연구(硏究))

  • Rheumatoid arthritis is characterized by focal loss of cartilage due to an up-regulation of inflammatory pathways, which produce pro-inflammatory mediators, such as interleukin-1(IL-1), tumour necrosis factor alpha($TNF-\alpha$), prostaglantin, and nitric oxide(NO). We investigated the anti-arthritic effects of water extracts from Pellodendri cortex and Atractylodis rhizoma in vitro and in vivo. Each extract suppressed the production of inflammatory mediators(NO, $IL-1\beta$, $TNF-\alpha$, and prostaglandin $E_2$) and the expression of inducible NO synthase and cyclooxygenase-2 in lipopolysaccharide-stimulated macrophages in a dose-dependent manner. These suppressive effects were synergistically increased by their combination. The same results were also observed in the rat osteoblast sarcoma cell line ROS17/2.8 stimulated with $IL-1\beta$, $IFN-\gamma,$ and $TNF-\alpha$. Moreover, the combination of these water extracts significantly suppressed collagen-induced mouse arthritis. These results suggest that the combination of water-extractable components of Pellodendri cortex and Atractylodis rhizoma may be useful for therapeutic drugs against rheumatoid arthritis, probably by suppressing the production of inflammatory mediators.

Study on the Mechanical Extraction Properties of Tobacco Stem Biomass (담배 주맥 바이오매스의 압착추출특성 연구)

  • This work evaluated the extractability of tobacco stem biomass for the papermaking type Reconstituted Tobacco Sheet(RTS). The effects of the soaking conditions on the hydration of stem biomass and the effects of the hydrated state on the mechanical extraction were investigated. In order to simulate the mechanical expression process of a papermaking type RTS mill, for example, the screw press process, the novel mechanical pressing analyzer was developed for this study. The hydration of stem biomass by soaking process was greatly affected by the soaking time and the soaking temperature. The longer soaking time and the higher soaking temperature resulted in the higher hydrated stem biomass. Since the higher hydrated stem had more combined water in the inner structure and resulted in the more flexible structure, the higher hydrated stem leaded to the more compressed filter cake and the higher water contents in the filter cake after the mechanical pressing. The pilot pulping experiments showed the difference in hydration and extractability between burley and bright tobacco stem. The bulkier structure of the burley stem resulted in the faster hydration by pilot pulping and leaded to the larger reduction in water soluble components. And the hydration process showed the major influence on the separation efficiency of water soluble components.

Effect of Ammonium Chloride on the Mixed Methanotrophs Species Composition and Methanol Metabolism (염화암모늄 영향에 따른 혼합종 메탄산화균의 종조성 변화 및 메탄올 대사 특성)

  • This study analyzed the utility of ammonium chloride ($NH_4Cl$) as a nitrogen source for methanotroph communities. When cultured in nitrate mineral salt (NMS) medium, the methanotroph community we identified four families, seven genera, and 16 type I and type II species of methanotrophs. Among species in the Methylobacter genus, Methylobacter marinus could be actively cultured in NMS medium without NaCl addition. Following the addition of 25 mM $NH_4Cl$, the numbers of the type I genera Methylomonas, Methylococcus, and Methylobacter were increased, whereas the numbers of the type II genera Methylocystis and Methylosinus were decreased after 5 days. In methanotroph communities, certain concentrations of $NH_4Cl$ affected methane consumption and growth of methanotrophs at the community level. $NH_4Cl$ caused a considerable decrease in the methane consumption rate and the expression of soluble methane monooxygenases (sMMOs) but did not inhibit the growth of Methylomonas methanica expressing sMMO. These results could be attributed to competitive antagonism of MMOs due to their direct involvement in ammonia oxidation.

Random dynamic analysis for simplified vehicle model based on explicit time-domain method

  • On the basis of the explicit time-domain method, an investigation is performed on the influence of the rotational stiffness and rotational damping of the vehicle body and front-rear bogies on the dynamic responses of the vehicle-bridge coupled systems. The equation of motion for the vehicle subsystem is derived employing rigid dynamical theories without considering the rotational stiffness and rotational damping of the vehicle body, as well as the front-rear bogies. The explicit expressions for the dynamic responses of the vehicle and bridge subsystems to contact forces are generated utilizing the explicit time-domain method. Due to the compact wheel-rail model, which reflects the compatibility requirement of the two subsystems, the explicit expression of the evolutionary statistical moment for the contact forces may be performed with relative ease. Then, the evolutionary statistical moments for the respective responses of the two subsystems can be determined. The numerical results indicate that the simplification of vehicle model has little effect on the responses of the bridge subsystem and the vehicle body, except for the responses of the rotational degrees of freedom for the vehicle subsystem, regardless of whether deterministic or random analyses are performed.

Effect of Padina arborescens on $H_2O_2$-induced Oxidative Stress in Human Endothelial Cell line, ECV304 cells (부챗말 추출액이 $H_2O_2$에 의한 혈관내피세포주인 ECV304세포의 산화적 스트레스에 미치는 영향)

  • The marine algae, Padina arborescens, have been used traditionally for treatment of various brain diseases. However, the molecular studies on the effect of Padina arborescens have not been carried out. In the present study, the protective effect of the water extract of Padina arborescens (PAWE) was researched in $H_2O_2$-treated human vascular endothelial cells, ECV304. ECV304 cells were pre-incubated with PAWE (0, 400, 800, 1,200 and $1,600{\mu}g/m{\ell}$) for 12 h and treated with 500 uM $H_2O_2$ for 12 h, and then the protective effects of PAWE were determined. PAWE recovered the $H_2O_2$-induced cell damage and decreased ROS production in ECV304 cells. Moreover, PAWE increased ERK expression and inhibited p38 and JNK expression. Furthermore, PAWE dosedependently increased the expression of heme oxygenase-1 (HO-1) and the HO-1 expression was reduced by ERK inhibitor treatment in $H_2O_2$-treated EVC304 cells. These results suggested that protective effect of PAWE on $H_2O_2$-induced oxidative stress in ECV304 cells might be associated with the production of HO-1 through the ERK signal pathway.

Inhibitory effect of Mojaban (Sargassum fulvellum) extract on ultraviolet B-induced photoaging in hairless mouse skin (무모 마우스 피부에서 모자반 추출물의 자외선비(ultraviolet B)에 의해 유도된 광노화 억제 효과)

  • Ultraviolet radiation is a major cause of photoaging, characterized by wrinkle formation and skin dehydration. Mojaban (Sargassum fulvellum), belonging to the brown algae of the Sargassaceae family, exerts anti-inflammatory, antioxidant, and anti-atopic activities. Particularly, Mojaban extract alleviated ultraviolet B (UVB)-induced inflammatory responses in human keratinocytes and BALB/c mice. Here, the dorsal skin of hairless mice was exposed to UVB for photoaging. The mice were administrated 300 mg/kg Mojaban extract (S. fulvellum extract; SFE) daily. Phenotypically, SFE reduced wrinkle formation and epidermal thickness and increased skin elasticity. SFE increased hydroxyproline content by upregulating the expression of collagen-related genes and downregulating matrix metalloproteinase expression. Furthermore, SFE increased skin moisture content and inhibited transepidermal water loss. SFE upregulated the expression of involucrin, loricrin, and transglutaminase for cornified envelope formation as well as the expression of filaggrin and caspase-14 for natural moisturizing factors. Collectively, SFE may be employed as a nutraceutical ingredient for anti-photoaging.

Protective Effects of Folium Artemisiae Argyi Herbal Acupuncture on Transient Forebrain Ischemic Injury in Rats (흰쥐의 일과성 전뇌 허혈 손상에 대한 애엽 약침의 신경보호 작용)

  • Objectives : Recently, the new therapeutic tool, that is herbal acupuncture, has been developed and applied to various diseases including the cerebrovascular accident. The main characteristics of herbal acupuncture are a combination of acupuncture and herbal medicine. It was not well known the therapeutic effect and the mechanism of herbal acupuncture on transient forebrain ischemic injury, although it has been used frequently in clinics. The objective was to determine the effect of folium artemisiae argyi (艾葉) herbal acupuncture on the trasient forebrain ischemic injured rat. Methods : In this study, the effects of folium artemisiae argyi (艾葉) herbal acupuncture on the $LR_3$ named Taechung, on neuroprotection after the transient forebrain ischemia were investigated in Sprague-Dawely rats. Expressions of cFos, FosB and BDNF protein in the hippocampus and cortex were observed at 2 hrs and 48 hrs after transient forebrain ischemia by immunohistochemistry and ELISA technique. Results : Expression of cFos protein was increased slightly in the hippocampus and cortex at 2 hrs after transient forebrain ischemia, but FosB protein was increased highly comparing to cFos protein. However, pretreatment with folium artemisiae argyi' herbal acupuncture on $LR_3$ significantly increased expression of cFos protein and significantly decreased expression of FosB protein compared to control group. These features were observed in the retrosplenial granular cortex as well as the hippocampus. Also, pretreatment with folium artemisiae argyi' herbal acupuncture on $LR_3$ significantly increased the expression of BDNF protein in the hippocampus ($263.26{\pm}44.56{\;}pg/ml$ extracted by water, $275.35{\pm}51.47{\;}pg/ml$ extracted by moxa tar)and the cortex ($102.33{\pm}13.65{\;}pg/ml$ extracted by water, $109.54{\pm}9.37{\;}pg/ml$ extracted by moxa tar) compared to the hippocampus ($134.07{\pm}2.96{\;}pg/ml$) and the cortex ($61.16{\pm}4.11{\;}pg/ml$) in control group at 48 hrs after transient forebrain ischemia. Conclusions : These results suggest that pretreatment with folium artemisiae argyi' herbal acupuncture on $LR_3$ has neuroprotective effect on transient forebrain ischemia and the herbal acupunture on $LR_3$ may be related to antioxidative function of folium artemisiae argyi.
