세심탕(洗心湯) 열수추출물, 초미세분말제형이 Alzheimer's Disease 병태 모델에 미치는 영향 (Effects of Sesim-tang Hot Water Extract & Ultra-fine Powder on the Alzheimer's Disease Model)

  • This experiment was designed to investigate the effect of the SST hot water extract & ultra-fine Powder on Alzheimer's Disease Model Induced by ${\beta}$A. The effects of the SST hot water extract on expression of IL-1${\beta}$, IL-6, TNF-${\alpha}$, NOS-II, COX-2 mRNA and production of IL-l${\beta}$, IL-6, TNF-${\alpha}$, NO in BV2 microglial cell line treated by lipopolysacchaide(LPS). The effects of the SST hot water extract & ultra-fine powder on (1) the behavior (2) expression of IL-1${\beta}$, TNF-${\alpha}$, MDA, (3) Glucose, AChE in serum (4) the infarction area of the hippocampus, and brain tissue injury in Alzheimer's diseased mice induced with ${\beta}$A were investigated. The SST hot water extract suppressed the expression of IL-1${\beta}$, IL-6 and TNF-a mRNA ${\alpha}$in BV2 microglia cell line treated with LPS. The SST hot water extract suppressed the production of IL-1${\beta}$, IL-6, TNF-${\alpha}$, NO in BV2 microglial cell line treated with LPS. The SST hot water extract & ultra-fine powder a significant inhibitory effect on the memory deficit was shown for the mice with Alzheimer's disease induced by ${\beta}$A in the Morris water maze experiment, which measured stop-through latency. The SST ultra-fine powder suppressed the expression of TNF-a protein significantly in the microglial cell of mice with Alzheimer's disease induced by ${\beta}$A. The SST hot water extract & ultra-fine powder reduced the MDA and suppressed the over-expression of CD68, CD11b in the mice with Alzheimer's disease induced by ${\beta}$A. The SST hot water extract & ultra-fine powder decreased AChE significantly in the serum of the mice with Alzheimer's disease induced by ${\beta}$A. The SST hot water extract & ultra-fine powder reduced infarction area of hippocampus, and controlled the injury of brain tissue in the mice with Alzheimer's disease induced by ${\beta}$A. The SST hot water extract & ultra-fine powder reduced the tau protein, GFAP, and presenilin1, 2 of hippocampus in the mice with Alzheimer's disease induced by ${\beta}$A. These results suggest that the SST hot water extract & ultra-fine powder may be effective for the prevention and treatment of A1zheimer's disease. Investigation into the clinical use of the SST hot water extract & ultra-fine powder for Alzheimer's disease is suggested for future research.

  • Objective : This experiment was designed to investigate the effect of the CBD hot water extract & ultra-fine Powder on Alzheimer's Disease Model Induced by ${\beta}A$. Method : The effects of the CBD hot water extract on expression of interleukin-1 beta($IL-1{\beta}$), $TNF-{\alpha}$ mRNA and production of IL-6, $TNF-{\alpha}$ in BV2 microglial cell line treated by lipopolysacchaide(LPS). The effects of the CBD hot water extract & ultra-fine powder on (1) the behavior (2) expression of $IL-1{\beta}$, tumor necrosis factor-alpha($TNF-{\alpha}$), (3) the infarction area of the hippocampus in Alzheimer's diseased mice induced with ${\beta}A$ were investigated. Result : The CBD hot water extract suppressed the expression of $IL-1{\beta}$, $TNF-{\alpha}$ mRNA in BV2 microglia cell line treated with LPS. The CBD hot water extract significantly suppressed the production of $IL-1{\beta}$, $TNF-{\alpha}$ in BV2 microglial cell line treated with LPS. The CBD hot water extract & ultra-fine powder a significant inhibitory effect on the memory deficit was shown for the mice with Alzheimer's disease induced by ${\beta}A$ in the Morris water maze experiment, which measured step-through latency and distance movement-through latency. The CBD hot water extract & ultra-fine powder significantly suppressed the expression of $IL-l{\beta}$ and $TNF-{\alpha}$ protein in the microglial cell of mice with Alzheimer's disease induced by ${\beta}A$. The CBD hot water extract & ultra-fine powder suppressed the over-expression of AChE activity in the serum of the mice with Alzheimer's disease induced by ${\beta}A$. The CBD hot water extract & ultra-fine powder reduced infarction area of hippocampus, in the mice with Alzheimer's disease induced by ${\beta}A$. Conclusions : These results suggest that the CBD hot water extract & ultra-fine powder may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the CBD hot water extract & ultra-fine powder for Alzheimer's disease is suggested for future research.

약용식물 물 추출물이 항당뇨 효소의 유전자 발현에 미치는 영향 (Effects of Medicinal Plant Water Extracts on Expression of Anti-diabetic Enzymes mRNA)

  • 본 연구에서는 동충하초, 자소엽, 단삼, 전칠, 명월초 물 추출물이 당대사 관련 효소인 GCK(glucokinase), PDH(pyruvate dehydrogenase) 및 ACC(acetyl CoA carboxylase) mRNA 발현 정도에 미치는 영향을 검토하였다. GCK mRNA 발현은 동충하초 물 추출물을 250 ppm로 처리했을때 181%로 가장 높게 증가되었다. PDH mRNA 발현은 100 ppm에서 명월초(215%)> 단삼(193%)> 전칠(125%)> 동충하초(111%)의 순으로 증가되었으며, 250 및 500 ppm에서는 각각 명월초(251%)와 동충하초(210%) 물 추출물에서 높게 나타났다. ACC mRNA 발현은 자소엽, 단삼, 전칠의 경우 대조구보다 낮거나 유사한 수준을 나타내었지만, 동충하초 물 추출물은 대조구와 비교하여 100 ppm에서 141%, 250 ppm에서 284%로 크게 증가되었다. 명월초 물 추출물은 187~199% 범위에서 ACC mRNA 발현을 크게 증가시켰다. 결과적으로 동충하초와 명월초 물 추출물이 다른 소재들에 비해 간의 GCK, PDH, ACC mRNA 발현을 증가시킴으로써 혈당강하 작용을 나타낼 것으로 판단하였다.

녹용 물 추출물의 파골세포 분화 억제효과 (Effect of Water Extract of Deer Antler in Osteoclast Differentiation)

  • Natural substances have recently received much attention as therapeutic drugs to prevent many diseases in humans because they avoid the many side effects of treatment with chemical compounds. We examined the effect of water extract of deer antler in RANKL-induced osteoclast differentiation. The effects of water extract of deer antler in osteoclast differentiation were determined by culture of bone marrow macrophages (BMMs). The mRNA expression levels of c-Fos, NFATc1, TRAP, and GAPDH in BMMs were analyzed by RT-PCR. Cell lysates were obtained from the treated cells, the expression levels of c-Fos and NFATc1 were determined by western blotting with antibodies for c-Fos and NFATc1. Water extract of deer antler greatly inhibited RANKL-mediated osteoclast differentiation in osteoclast precursors without cytotoxicity. Water extract of deer antler inhibited the expression of c-Fos and NFATc1 in BMMs treated with RANKL. Our findings suggest that water extract of deer antler inhibited osteoclast differentiation by suppressing c-Fos and NFATc1 expression in response to RANKL. These results demonstrate that water extract of deer antler may be a useful the treatment of bone-related disease such as osteoporosis.

Autonomic Neural Regulation of Sodium Transporters and Water Channels in Rat Submandibular Gland

  • The present study was undertaken to explore the role of autonomic nerves in the regulation of sodium transporters and water channels in the salivary gland. Rats were denervated of their sympathetic or parasympathetic nerves to the submandibular gland. One week later, the expression of Na,K-ATPase, epithelial sodium channels (ENaC), and aquaporins (AQP) was examined in the denervated and contralateral glands. The sympathetic denervation slightly but significantly decreased the expression of ${\alpha}1$ subunit of Na,K-ATPase, whereas the parasympathetic denervation increased it. The expression of ${\alpha}$-subunit of ENaC was significantly increased in both the denervated and contralateral glands either by the sympathetic or parasympathetic denervation. The sympathetic denervation significantly increased the expression of AQP5 in both the denervated and contralateral glands, whereas the parasympathetic denervation decreased it. It is suggested that the autonomic nerves have a tonic effect on the regulation of sodium transporters and AQP water channels in the salivary gland.

흰쥐 악하선에서 교감신경과 부교감신경에 의한 나트륨 운반체 및 수분 통로 조절 (ALTERED EXPRESSION OF SODIUM TRANSPORTERS AND WATER CHANNELS FOLLOWING SYMPATHETIC AND PARASYMPATHETIC DENERVATION IN RAT SUBMANDIBULAR GLAND)

  • 타액선에서 교감 신경과 부교감 신경이 나트륨 운반체와 수분 통로의 조절에 어떠한 기능을 하는지 알아 보고자 흰쥐 악하선을 지배하는 교감 신경과 부교감 신경을 절제하고 나서 타액선내 나트륨 운반체와 수분 통로의 발현을 조사하여 다음과 같은 성적을 얻었다. 1. Na,K-ATPase의 ${\alpha}1,\;{\beta}1$ 소단위는 교감 신경 절제에 의해 크게 영향받지 않았으나 부교감 신경 절제에 의해서는 두 소단위의 발현이 모두 감소되었다. 2. ENaC ${\alpha}-,\;{\beta}-,\;{\gamma}-$ 소단위는 그 발현이 교감 신경 절제에 의해 영향 받지 않았으나 부교감 신경 절제에 의해서 도리어 증가하였다. 3. NHE3는 교감 신경 및 부교감 신경 절제에 의해 모두 크게 감소했다. 4. 교감 신경 절제시 AQP1의 발현이 크게 증가했으나, 부교감 신경 절제시 영향 받지 않았다. 5. 교감 신경 절제 및 부교감 신경 절제는 AQP4 발현을 크게 증가시켰다. 6. AQP5는 교감 신경 절제시 영향을 받지 않았으나, 부교감 신경 절제시 크게 감소되었다. 이상의 실험성적을 요약할 때 악하선을 지배하는 교감 신경 및 부교감 신경은 선의 나트륨 운반체 및 수분 통로 발현에 긴장성 조절을 영위함으로써 타액의 전해질 및 수분 조성을 결정하는 데 공헌함을 알 수 있다. 그리고 이 신경의 절제시 보이는 나트륨 운반체와 수분 통로의 변화는 기능적으로 신경 절제 타액선에서의 타액 분비량과 타액의 무기질 조성 변화의 원인이 될 것이라 생각된다.

Chronic Exposure of Nicotine Modulates the Expressions of the Cerebellar Glial Glutamate Transporters in Rats

  • Rats were given nicotine (25 ppm) in their drinking water at the start of their mating period in order to study the expressions of glutamate transporter subtypes in cerebellar astrocytes following the chronic exposure of nicotine after mating. After the offspring were delivered, each group was divided into two subgroups. One group, the control group, was given distilled water only and the other group, the experimental group, was given distilled water containing nicotine. The cerebellar astrocytes were prepared from 7 day-old pups at each group. Ten days after the cells were cultured, the expression of the glutamate transporter subtypes (GLAST and GLT-1) was determined using immunochemistry and immunoblotting. During the continuous treatments, the developmental expression patterns of the GLAST and GLT-1 in the cerebellum were also determined from 2, 4 and 8 week-old rats. The expression levels of GLAST in cultured astrocytes of both the pre- or post-natally exposed groups were higher than those of the control group. However, these expression levels of the continuously exposed group were lower than those of the control group. Compared to those of the control group, the GLT-1 expression levels of all the nicotine-treated groups were higher, particularly in the continuously treated group.. According to the results from the immochemistry procedure, the cerebellar GLAST and GLT-1 expression levels of all nicotine-treated groups were lower than those of the control group at each age. However, the immunoblotting procedure showed that the cerebellar GLT-1 expression levels of all the nicotine-treated groups were higher than those of the control group, except for the rats that were continuously exposed for 8 weeks using immunoblotting. These results suggest that the expression of the glial GLAST and GLT-1 are altered differently depending on the initial exposure time and the partcicular period of nicotine exposure. In addition, nicotine exposure during gestation has persistent effects on glial cells.

Responses of HSP Gene Expressions to Elevated Water Temperature in the Nile tilapia Oreochromis niloticus

  • Water temperature influences on various key biological events in fish, but the internal pathway of the temperature effects are not well understood. Heat shock proteins (HSPs), known to respond in the level of cells to many environmental factors including temperature, could improve our understanding on the pathway. Some biological processes such as gonadal development and sex differentiation in the Nile tilapia Oreochromis niloticus is particularly sensitive to water temperature. In this study, we have investigated the expressions of HSP70 and HSP90 genes in young tilapia at an ordinary temperature ($28^{\circ}C$) and elevated water temperature ($36^{\circ}C$). The distribution of the expressions of HSP70 and HSP90 mRNA in this species were found to be almost ubiquitous, being detected in all tissues studied here (brain, gonad, liver and muscle), suggesting the house keeping functions of these genes. Heat shock by elevating temperature from $28^{\circ}C$ to $36^{\circ}C$ significantly increased the expression of HSP70 mRNA in the gonad, liver and muscle for several hours (P<0.05) (brain tissue was not examined for this). The increased level of HSP70 gene expression recovered to the level at control temperature ($28^{\circ}C$) when fish were kept continuously at high temperature ($36^{\circ}C$) for 24 hours. Contrary to this, expression of HSP90 mRNA did not show significant increase in the gonad and muscle by the same heat shock (P>0.05), except in the liver where the expression of HSP90 mRNA increased continuously for 24 hours at $36^{\circ}C$. The results obtained in this study suggest that response to temperature change in different tissue or organ may utilize different heat shock proteins, and that HSP70 may have some importance in temperature-sensitive gonadal event in the Nile tilapia.
