Sulfate Reduction in the Marine Environments: Its Controlling Factors and Relative Significance in Mineralization of Organic Matter (해양환경의 황산염 환원율 조절요인 및 유기물 분해에 있어 황산염 환원의 중요성)

  • Sulfate reduction is a microbiological process which occurs ubiquitously in anaerobic marine environment. Sulfate reducing bacteria play a significant role in anaerobic decomposition of organic matter and regeneration of inorganic nutrients which supports the primary production in the water column (i.e., benthic-pelagic coupling) and, in special case, could be responsible for the harmful algal bloom in the coastal marine environment. Summary of the sulfate reduction rates reported in various marine sedimentary environments revealed that supply of organic substrates and presence of various electron acceptors (i.e., $O_2$, NO$_{3}$$^{[-10]}$ , Fe(III) and Mn(IV), etc.) for other aerobic and anaerobic respiration directly affect the sulfate reduction rate and relative significance of sulfate reduction in organic matter mineralization. Significance of temperature, macrophytes and bioturbation is discussed as factors controlling supply of organic substrates and distribution of electron acceptors. Finally, we suggest studies on the anaerobic microbiological processes associated with biogeochemical element cycles in the coastal environments of Korea where massive operation of organic enriched fish cage farm, frequent occurrence of toxic algal bloom and hypoxia and conservation of tidal flat are of major environmental issues.

Development of Analytical Method for Sodium Alginate in Foods (식품 중 알긴산나트륨의 분석법 개발)

  • Method of analyzing sodium alginate in foods was developed using high performance liquid chromatography (HPLC). HPLC conditions for sodium alginate were: column, MCI GEL $(8\;mm\;i.d{\times}300\;mm)$; mobile phase, deionized water; detector, refractive index detection (sensitivity = 16). Separation of sodium alginate was achieved within 15 min. Sodium alginate showed good linear relationship at 0.1-2.0% range. Correlation coefficient of calibration curve for sodium alginate exceeded 0.999, and detection limit was 0.005%. Recovery rate of sodium alginate in wheat flour dough was 106.67%. This method was successfully applied to analyses of cereals, saccharides, and ice cream, etc. Sodium alginate was detected in chocolate, noodles, and kelp at 0-44.8% range.

Studies on the Determination Method of Hydrogen Peroxide in Foods (식품 중 과산화수소의 분석법에 관한 연구)

  • Micro-LC method for the quantitative determination of hydrogen peroxide in foods has been established. This method was carried out on cation-exchange resin gel column using distilled water as mobile phase with 50 mM sodium sulfate as electrolyte. The detection was performed with an electrochemical detector (ECD) at 0.6 voltage. Under this analytical condition, the recovery rates of hydrogen peroxide in tomato and lemon were 98.3 and 97.4%, respectively. Among 28 food types, hydrogen peroxide concentrations were 0.6, 0.5, 1.9, 0.9, 0.5, 0.6, 0.9, 0.8, and 0.4 ppm in banana, peach, orange, strawberry, pepper, onion, cucumber, burdock, and egg plant, respectively, Whereas none was detected in remaing 19 samples.

  • The present study was conducted to prepare seaweed extracts suppressing mutagenic activity of 2-amino-1-methyl-6-phenylimidazo[4,5-b] pyridine ( PhIP ) ana 2-amino-3,8-dimethylmidazo[4,5-f]quinoxaline (MeIQx) derived from cooked meat produce. The tumor initiation activity of PhIP and MeIQx was assayed with Ames method using Salmonella typhimurium TA98 in the presence of S-9 mixtures before and after addition of methanol-solubles of seaweed, such as, Phaeophyta; Undaria pinnatifida, Ecklonia stolonifera Ecklonia cava, Laminaia japonica Sargassum fulvellum, Sargassum hornezi, Sargassum miyabei, Sargassum thunbergii, Agarum cribrosum and Hizikia fusifomis, Rhodophpei Porphyra yezoensis, Grateloupia eiliptiut Lomentraria catenata, Ploemium telfairiae and Glarilaria verrucosa, Chlorophyta; Codium fragile, Enteromorpha compresa and Ulva pertusa. Among seaweed tested, Phaeophyta was shown the higher desmutagenic activity than Rhodophyta and Chlorophyta. E. stolonifera, E. cava and S. miyabei, among Phaeophyta exerted the stronger desmutagenic activity (above $90\%$/2 mg). The ethyl acetate, diethyl ether and chlomform extracts except water extracts from E. stolonifera exhibited a high desmutagenic activity. The ethyl acetate extract of E. stolonifera which showed highest activity was fractionated with Sephadex LH20 column chromatography to give active fraction A-7, which showed desmutagenicity of $90\%$/mg against PhIP and $80\%$/mg against MeIQx. The active fraction had the absorbance at 207.7 and 232nm.

  • Green mustard leves were found to effectively prevent lipid peroxidation of bovine-brain tissue by ascor-bate/Fe system, The 50% methanol extracts mustard leaves were separated into four solvent faction using n-hexane,. EtOAc, n-BiOH and water. Then n-BiOH fraction exclusively exhibited the antioxidative activities at concentration above 100 $\mu\textrm{g}$/mL/ The n-BuOH fraction was further isolated to a single compound using TLC analysis and silica gel chromatography. The active antiodidative compounds were identified as sinapic acid methyl ester and ferulic acid methyl ester by $^{1}$H-NMR and $^{13}$ C-NMR, The sinapic acid methyl ester and ferulic acid methyl ester were prepared by methylating of sinapic acid and ferulic acid with diazomethane. The results strongly suggested that sinapic acid and ferulic acid could be emplyed as a potential antioxiative agents for preventing the bovine brain lipid peroxidation. lipid peroxidation.

Recovery of Aglycone of Daidzein and Genistein in Soybeans (콩에 포함된 비배당체 다이드제인과 제니스테인의 회수)

  • Using aqueous solution of 60% ethanol, the isoflavones contained in the Soybean (Chungsun, Korea), Tawon (Suwon, Korea), Jackbean (Hongsung, Korea), Soybean (Jilin, China), Black Soybean (Jilin, China), Kidneybean (Jilin, China) and Soybean (California, U.S.A.) were extracted. In this work, the pretreatment step was composed of agitation extraction, filtration, concentration, ultracentrifuge, and membrane filtration. The analysis by $C_{18}$ column was performed, and the mobile phase applied was linearly changed with A/B of 85/15-65/35 vol% for 50 min (A water/acetic acid, 99.9/0.1 vol%, B acetonitrile/acetic acid, 99.9/0.1 vol%). Among the soybeans tested, the total amounts of the four isoflavones (daidzin, genistin, daidzein, and genistein) extracted by the soybeans from Korea, China and U.S.A. were 1.37, 1.60, and 2.25 mg/g, respectively. It was also found that the total amount of aglycone of daidzein and genistein from soybean (American California) was $144{\mu}g/g$, which was the largest for the soybeans experimented.

Structure Determination of the Extractives from the Taxus Cuspidata Fruits (주목열매 추출물 구조분석)

  • The fruits of Taxus cuspidata were collected, divided into seeds and fruits, and extracted with 95% EtOH. The extracts were evaporated under the reduced vacuum pressure, concentrated, then successively fractionated with a series of n-hexane, dichloromethane, ethyl acetate and water on a separatory funnel to get some freeze dried samples. A portion of the EtOAc (arils:1.65 g, seeds:1.04 g) and $H_2O$ (arils:7 g, seeds:10 g) soluble samples were chromatographed on a Sephadex column using MeOH-$H_2O$ (1:1, 1:3, 1:5, v/v), EtOH-hexane (3:1, v/v) mixture and 100% $H_2O$ as eluting solvents to isolate pure compounds from the fractions. The isolates were developed by cellulose TLC using t-BuOH-HOAc-$H_2O$ (TBA; 3:1:1, v/v/v) and 6% aqueous HOAc. Visualization was done under ultraviolet light and by spraying the vanillin-HCl-EtOH reagent (4.8:12:480, v/v/v). followed by heating. The structures of the isolates were characterized by $^1H$- and $^{13}C$-NMR, DEPT, 2D-NMR, LC/MS and EI-MS spectra. In addition to the NMR and MS spectra, acid hydrolysis and permethylation were used to determine the correct structure of the isolated sugar compound. Their structures were elucidated as (+)-catechin (1), (-)-epicatechin (2), (+)-gallocatechin (3), (-)-epigallocatechin (4) and ${\beta}$-D-fructofuranose-($2{\rightarrow}4$)-O-${\beta}$-D-glucopyranose($1{\rightarrow}4$)-O-${\alpha}$-D-glucopyranose ($1{\rightarrow}2$)-O-${\beta}$-D-fructofuranose (5) on the basis of the above experimental evidences.

Simulation of TOA Visible Radiance for the Ocean Target and its Possible use for Satellite Sensor Calibration (해양 표적을 이용한 대기 상단 가시영역에서의 복사휘도 모의와 위성 센서 검보정에의 활용 가능성 연구)

  • Vicarious calibration for the satellite sensor relies on simulated TOA (Top-of-Atmosphere) radiances over various targets. In this study, TOA visible radiance was calculated over ocean targets which are located in five different regions over the Indian and Pacific ocean, and its possible use for the satellite sensor calibration was examined. TOA radiances are simulated with the 6S radiative transfer model for the comparison with MODIS/Terra and SeaWiFS measurements. Geometric angles and sensor characteristics of the reference satellites were taken into account for the simulation. AOT (Aerosol Optical Thickness) from MODIS/Terra, pigment concentrations from Sea WiFS, and ozone amount from OMI measurements were used as inputs to the model. Other atmospheric input parameters such as surface wind and total column water vapor were taken from NCEP/NCAR reanalysis data. The 5-day averaged radiances over all targets show that the percent differences between simulated and observed radiances are within about ${\pm}5%$ in year 2005, indicating that the calculated radiances are in good agreement with satellite measurements. It has also been shown that the algorithm can produce the SeaWiFS radiances within about ${\pm}5%$ uncertainty range. It has been suggested that the algorithm can be used as a tool for calibrating the VIS bands within about 5% uncertainty range.

Growth of Creeping Bentgrass on Bottom Ash and Dredged up Sand with Four Organic Matter Amendment Rates Under Saline Irrigation Condition (염해 조건에서 유기물이 첨가된 준설모래와 석탄회 토양이 크리핑 벤트그래스의 생육에 미치는 영향)

  • This study was carried out to check the possibility of substituting bottom ash from the Seosan power plant for sand as growing media for creeping bentgrass (Agrostis stolonifera L.) under saline irrigation condition. Characteristics of growing media were evaluated by using column and leaching method. Creeping bentgrass cv. Pen-A1 was grown in pots with dredged up sand (DS) and bottom ash (BA) media those were amended using 1%, 2%, and 3 % OM rates in a green house. The plants were irrigated with 1.5 $dSm^{-1}$ saline water. Results showed that visual quality, plant height and shoot dry weight from DS treatment were higher than those of BA treatment. Even though BA contained more salts, repeated leaching could decrease ECe efficiently. In case of no OM amendment, the visual quality, plant height and shoot dry weight were similar between in BA and DS. Amendment of 2% OM increased the height of creeping bentgrass in DS, while decreased the plant growth in BA.

Isolation and Identification of Antimicrobial Active Substances from Rhodiola sachlinensis (홍경천(Rhodiola sachlinensis)에서 항균성 물질의 분리 및 동정)

  • The antimicrobial substances from Rhodiola sachalinensis were extncted, isolated and identified. The highest level of antimicrobial activity and its yield were obtained in methanol extract. The minimum inhibition concentrations of the methanol extract were 500 $\mu\textrm{g}$mL on agar plate and 100 $\mu\textrm{g}$mL in broth media for four gram positive and four gram negative microbials. The methanol extract was fractionated by n-hexane, chloroform, ethyl ether, ethyl acetate, and butanol, orderly. The separate was developed on the TLC plate with different solvent system ratio of chloroform and methanol. Nine substances were isolated from chloroform and methanol mixture(9:1, v/v). Among them, three isolates showed antimicrobial activity. Three substances separated by HPLC were identified by GC/MS(EI) spectrum and $^1$H, /sup13/C-NMR spectrum. They were gallic acid, (-)-epicatechin and kaempferol. The antimicrobial activities of each substances were shown gallic acid, (-)-epicatechin, kaempferol orderly.
