Effects of NaCl Concentrations on Production and Yields of Fruiting Body of Oyster Mushrooms, Pleurotus spp. (NaCl의 농도가 느타리버섯 자실체 발생 및 수량에 미치는 영향)

  • This studies investigated the effect of concentrations of sodium chloride (NaCl) on occurrence and growth of fruitbody in oyster mushrooms, Pleurotus spp. Our experiments divided into two parts. When the water contents in substrate were added with sodium chloride solution in cotton waste box cultivation as a first experiment, the growth of mushroom was damaged as the concentration was increased, even though there was a little difference according to the strains. The yield in 1.0% NaCl solution was decreased to 72% compared to non-treated plot while that in 3.0% solution was only 2% of the non-treated plot. Morphological characteristics of mushrooms cultivated in substrate with the different concentration of the solution showed different results. For example, the size and thickness of pilei were not influenced by NaCl concentration, but the length of stipes and individual weight were much influenced. In plastic box cultivation filled with cotton waste, watering treatment with the different concentrations of sodium chloride solution, the second experiment, did not show any difference according to the concentration until 1.0% solution but there was a little difference according to the strains. The productivity of fruitbody started to decrease at 2.0% of the solution and the yield and quality of mushroom in 3.0% solution treatment were generally low. After the second flush, days for mushroom sprouting were generally prolonged in proportion to the solution concentration. Taken altogether, the second experiment did not show a clear effect as the case of the first experiment.

Removal Properties of Methylene Blue using Biochar Prepared from Street Tree Pruning Branches and Household Wood Waste (가로수 전정가지 및 생활계 폐목재를 이용하여 제조한 바이오차의 Methylene Blue 흡착특성)

  • In order to improve water quality of the water system contaminated with dyes, biochars prepared using discarded waste resources were applied in this study. Biochars with a large specific surface area were manufactured using street tree pruning products or waste wood, and were applied to remove an organic dye in synthetic water. Biochars were made by pyrolysis of typical street tree porch products (Platanas, Ginkgo, Aak) and waste wood under air-controlled conditions. Methylene blue (MB), which is widely used in phosphofibers, paper, leather, and cotton media, was selected in this study. The adsorption capacity of Platanas for MB was the highest and the qmax value obtained using the Langmuir model equation was 78.47 mg/g. In addition, the adsorption energy (E) (kJ/mol) of MB using the Dubinin-Radushkevich (D-R) model equation was 4.891 kJ/mol which was less than 8 kJ/mol (a criteria distinguishing physical adsorption from chemical adsorption). This result suggests a physical adsorption with weak interactions such as van der Waals force between the biochar and MB. In addition, the physical adsorption may resulted from that Platanas-based biohar has the largest specific surface area and pore volume. The ∆G value obtained through the adsorption experiment according to temperature variation was -3.67 to -7.68, which also suggests a physical adsorption. Considering these adsorption results, the adsorption of MB onto Platanas-based biochar seems to occur through physical adsorption. Overall, it was possible to suggest that adsorption capacity of the biochr prepared from this study was equal to or greater than that of commercial activated carbon reported in other studies.

Evaluation of Microbially Ensiled Spent Mushroom (Pleurotus osteratus) Substrates (Bed-Type Cultivation) as a Roughage for Ruminants (균상재배 느타리버섯부산물 생균발효사료의 반추동물 조사료원으로서의 가치 평가)

  • An in situ ruminal disappearance trial and an in vivo sheep metabolism trial were conducted to evaluate the nutritional value of spent mushroom substrate (SMS, originated from Pleurotus osteratus bed-type cultivation). The raw SMS was ensiled (ESMS) for 30 days with 5% (w/w, DM basis) molasses, 0.5% (v/w) yeast (Saccharomyces cerevisiae) and 0.5% (v/w) lactic acid bacteria (Lactobacillus plantarum). Two ruminally cannulated Holsteins (average BW 620 kg) were used to evaluate in situ disappearance. Six sheeps (average BW 48 kg) were fed, in $3{\times}3$ Latin square design, rice straw alone (Control), 25% (ESMS-25) and 50% (ESMS-50) of rice straw were replaced with ensiled SMS. For an in situ trial, ruminal DM and neutral detergent fiber (NDF) disappearance of SMS were increased after ensiling (P<0.05). For a sheep trial, protein and fiber (NDF, acid detergent fiber, crude fiber) digestibilities were decreased (P<0.05), crude ash digestibility was increased (P<0.001), and nitrogen retention was not affected (P>0.05) as rice straw was replaced with ensiled SMS. Ruminating time was decreased by an average of 28% by feeding ensiled SMS (P<0.05). Ensiled SMS (Bed-type cultivation) had 76% of energy value of rice straw. Consequently, ensiled SMS (Bed-type cultivation, 100% cotton waste) could be used as a roughage source appropriate for maintenance type rations for ruminants.

Biological Control of Cucumber Powdery Mildew by Bacillus amyloliquefaciens M27 (Bacillus amyloliquefaciens M27에 의한 오이 흰가루병의 생물적 방제)

  • Bacillus amyloliquefaciens M27 was isolated from the cotton-waste compost for cultivation of oyster mushroom (Pleurotus ostreatus). B. amyloliquefaciens M27 is a biocontrol agent with antagonistic activities against a wide range of fungal pathogens. The aim of this work was to evaluate the possibility of exploiting antagonistic bacteria, B. amyloliquefaciens M27, in the biological control of the cucumber powdery mildew fungus, Podosphaera fusca. In greenhouse tests, the isolate was found to be very effective to control powdery mildew on cucumber leaves showing 4.0% diseased area, whereas diseased area in the control was 80.5%. The filtrate of the isolate cultured on MH and LB media were more effective for control of the disease than those cultured on TSB, NB, and KB media. When two, five, ten, 20, 50 and 100-fold diluted culture broth of isolate on LB media were treated, disease areas were 0%, 0%, 0%, 1.3%, 3.1% and 5.0%, respectively, whereas diseased area in the control was 60.0%. The filtrate of the isolate cultured on LB media was treated to cucumber plants on July, October and December just before the outbreak of the powdery mildew occurred. When 10-fold diluted filtrate of the isolate was treated, control efficacy was 88.9~98.9% in the treated seasons. The results showed that the culture filtrate of B. amyloliquefaciens M27 was very effective to control powdery mildew of cucumber.

  • The cellulolytic activities of Aspergillus fumigatus KC-1 was investigated, which showed the most active producer of cellulase among the 256 strains of cellulose-decomposing microorganisms screened in our laboratory. All the examined cellulolytic activities (filter paper-, Avicel-, cotton-, CMC-, salicin- and xylansaccharifying activity) in a culture of A. fumigatus KC-1 grown on 1% popular sawdust pretreated with peroxide alkaline reached a maximum within 4-5 days. The optimum pH and temperature for the enzymatic activity was found to be pH 4.5 and $60^{\circ}C$ respectively. The sawdust of poplar wood delignified with 1% NaOH and 20% peracetic acid succesively recorded the highest hydrolysis rate in the tests of enzymatic saccharification. The major end product of hydrolysis of poplar wood with the cellulolytic enzymes obtained from A. fumigatus KC-1 was glucose with small amount of cellobiose and xylose. It can be concluded from these results that A. fumigatus KC-1 is an advantagous source of a cellulase that is capable of hydrolyzing cellulose to glucose rapidly. The influence of degree of delignification, substrate size and its concentration on the rate of hydrolysis of poplar wood was also discussed.

Characteristics of a new cultivar 'Hwaseong 5ho' in Pleurotus ostreatus (신품종 느타리버섯 '화성5호'의 특성)

  • "Hwaseong 5ho" was developed by the method of Di-mon mating between monokaryotic strains derived from "Hwaseong 1ho" and dikaryotic strain "PSC109". The color of pileus was dark grayish brown, the shape of pileus was convex or infundibuliform. The length of stipe was longer and the thickness of stipe was some thinner than Suhan 1ho. Material properties of stipe of "Hwaseong 5ho" was higher in strength, hardness, chewingness and brittleness than Suhan 1ho, but similar in elasticity and cohesion. RAPD using URP-primer showed not the same between two strains. Days of primordia formation period were 22-27 days after spawning, that was a little later than Suhan 1ho. In the trial using culture box containing composted cotton waste, yield index of 'Hwaseong 5ho' was 16.6% higher than Suhan 1ho. The farm field trial were showed stable productivity in each different growing conditions.

A Short Composting Method by the Single Phase Composter for the Production of Oyster Mushroom (느타리버섯 배지 제조기를 이용한 배지의 제조 연구)

  • A single phase composter was constructed by modifying the conventional mixer of sawdust for the cultivation of oyster mushroom Pleurotus ostreatus. The machine was designed on the basis of 3-phase-1 system which was controlled in prewetting, pasteurization and fermentation processes. In composting 200 kg of straw and cotton waste in the machine, it took 20 minutes in prewetting step and also to hours at $65^{\circ}C$ in pasteurization process. Postfermentation by aerothermophiles was completed by treating the compost at $45^{\circ}C-50^{\circ}C$ for 48 hours which was shorten 24 hours from the conventional method. In the postfermentation at high temperature, forced aeration and/or vigorous mixing process(es) played a great role in the improvement of spawn quality. The growth of mycelium of oyster mushroom was excellent in the culture combinated with 3 parts of surface inoculation and 7 parts of mechanical mixing.

Cultivation of Oyster Mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at Medium with Ca(OH)2 Treatment (수산화칼슘[Ca(OH)2] 처리 배지를 이용한 느타리버섯의 재배)

  • This research is aimed to investigate the cultivation method of oyster mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at the non-sterilized medium with $Ca(OH)_2$ treatment. Therefore, experiments were carried out to develop non-sterilization method of medium by addition of $Ca(OH)_2$ for omission of heat sterilization progress of medium. General components, minerals and amino acid in Jiri wild type No. 1 (Pleurotus ostreatus) and production cost were analyzed. For the purpose of omission of heat sterilization progress, treatment ratio of $Ca(OH)_2$ (purity 95%) was 5%(w/w) of dry medium. Initial pH of this medium was 11 and then the pH was changed by 9 after the uniform mixing of the medium for half an hour. The various germs occurred 50% and 100% at pH 8 and pH 7 of the non-sterilized medium, respectively. Production of oyster mushroom increased by $2,030\;ton\;ha^{-1}$ when the main raw material used corn pith instead of waste cotton. The time required of mycelium culture was 30 days when hypha was cultured at the non-sterilized medium, and pinhead occurred when 2 or 3 days was passed after the time required of mycelium culture. Occurrence of pinhead was most rapid at the condition of $22{\sim}26^{\circ}C$, 65% humidity and pH $6.5{\sim}7.0$ and required of $22{\sim}28$ days at $70{\sim}80\;mm$ thickness of non-sterilized medium. Ca content in 1st harvest oyster mushroom was higher than that in 2nd harvest one, and its difference was $30.3\;mg\;kg^{-1}$. Amino acid content by stipe thickness of oyster mushroom was ranged from 411.2 to $343.9\;mg\;kg^{-1}$ both in a pileus and a stipe of 1st harvest mushroom, and from 402.4 to $498.2\;mg\;kg^{-1}$ and from 442.6 to $470.4\;mg\;kg^{-1}$ in those of 2nd harvest one, respectively. The results of the present study suggest that the non-sterilization medium by addition of $Ca(OH)_2$ is usable with the cultivation of oyster mushroom.
