Molecular Structure of the PHA Synthesis Gene Cluster from New mcl-PHA Producer Pseudomonas putida KCTC1639

  • Pseudomonas putida KCTC 1639 was newly identified as a potential producer of biodegradable medium chain length polyhydroxyalkanoates. It exhibited a carbon assimilation pattern quite different from other known P. putida strains, but a more similar pattern with P. oleovorans, which assimilates the carbon sources mainly through ${\beta}$-oxidation rather than the fatty acid biosynthesis pathway. The PHA synthesis gene cluster from P. putida KCTC1639 was composed of two gene loci; the PHA synthase gene locus and granule-associated gene locus, which were cloned and deposited in the GenBank under accession numbers AY286491 and AY750858 as a new nucleotide sequence, respectively. The molecular structure and amino acid homology of the new gene cluster were compared with those from Pseudomonas species, including other P. putida strains and P. oleovorans, and a higher than $90\%$ homology was observed.

Effect of Ultrasonic Process of Electroless Ni-P-Al2O3 Composite Coatings

  • In general, surface treatments of electroless Ni-P coating are extensively applied in the industry due to their excellent properties for considerable wear resistance, hardness, corrosion resistance. This study aims to determine the effect of ultrasonic conditions on the morphology, alumina content, roughness, hardness, and corrosion resistance of electroless Ni-P-Al2O3 composite coatings. The characteristics were analyzed by Energy-dispersive X-ray spectroscopy (EDX), x-ray diffractions (XRD), and atomic force microscopy (AFM), etc. In this study, the effect of ultrasonic condition uniformly distributed alumina within Ni-P solution resulting in a smoother surface, lower surface roughness. Furthermore, the corrosion resistance behavior of the coating was analyzed using tafel polarization curves in a 3.5 wt.% NaCl solution at 25 ℃. Under ultrasonic, Al2O3 content in Ni-P composite solution increased from 0.5 to 5.0 g/L, Al2O3 content at 3.0 g/L was showed a significantly enhanced corrosion resistance. These results suggested that ultrasonic condition was an effective method to improve the properties of the composite coating.

  • In this work, we developed a selective etching process for GaN that is a key process in p-GaN/AlGaN/GaN enhancement-mode (E-mode) power switching field-effect transistor (FET) fabrication. In order to achieve a high current density of p-GaN/AlGaN/GaN E-mode FET, the p-GaN layer beside the gate region must be selectively etched whereas the underneath AlGaN layer should be maintained. A selective etching process was implemented by oxidizing the surface of the AlGaN layer and the GaN layer by adding O2 gas to Cl2/N2 gas which is generally used for GaN etching. A selective etching process was optimized using Cl2/N2/O2 gas mixture and a high selectivity of 53:1 (= GaN/AlGaN) was achieved.

Determination of the pH of Iso-Selectivity of the Interfacial Diffusion Layer of Fe

  • Passive metal forms an interfacial diffuse layer on the surface of passive film by its reaction with $H^+$ or $OH^-$ ions in solution depending on solution pH. There is a critical pH, called pH point of iso-selectivity ($pH_{pis}$) at which the nature of the diffuse layer is changed from the anion-permeable at pH<$pH_{pis}$ to the cation-permeable at pH>$pH_{pis}$. The $pH_{pis}$ for a passivated Fe was determined by examining the effects of pH on the thickness of passive film and on the dissolution reaction occurring on the passive film under a gavanostatic reduction in borate-phosphate buffer solutions at various pH of 7~11. The steady-state thickness of passive film formed on Fe showed the maximum at pH 8.5~9, and further the nature of film dissolution reaction was changed from a reaction producing $Fe^{3+}$ ion at $pH\leq8.5$ to that producing $FeO_2{^-}$ at $pH\geq9$, suggesting that the $pH_{pis}$ of Fe is about pH 8.5~9. In addition, the passive film formed at pH 8.5~9, $pH_{pis}$, was found to be the most protective with the lowest defect density as confirmed by the Mott-Schottky analysis. Pitting potential was decreased with increasing $Cl^-$ concentration at $pH\leq8.5$ due probably to the formation of anion permeable diffuse layer, but it was almost constant at $pH\geq9$ irrespective of $Cl^-$ concentration due primarily to the formation of cation permeable diffuse layer on the film, confirming again that $pH_{pis}$ of Fe is 8.5~9.

퇴적물에서의 인의 용출특성과 존재형태에 관한 연구 (A study on release characteristics and forms of phosphorus in sediments)

  • This study was carried out to investigate release characteristics of phosphorus with DO, pH and temperature, to suppose its behaviour with time using mathematical model, and to under-stand its forms with pH. Released SRP was in inversely proportional to DO and it did few release in initial aerobic conditions, but it did actively with decreasing DO concentration. Also, its release was increased with increasing pH and temperature. It was found that relation between time and released SRP concentration was zero order reaction. As compared with k values in various pH and temp., they was $k_{15}>k_{25}$ in pH 6 but was $k_{15} in pH 7 and 8. Considering forms of phosphorus with pH, Resdi.-P & NAI-P increased but Ads.-P & Apt.-P decreased with increasing pH.

Material and rheological properties of (glycidoxypropyl) trimethoxysilane modified colloidal silica coatings

  • Colloidal coating solution was prepared to enhance the hydrophilic property of the film surface. Water and ethanol were used as the dispersion media and (glycidoxypropyl) trimethoxysilane (GPS) as a binder in the colloidal silica coatings. Ethylene diamine was added to the colloidal silica solution as the curing agent. The colloidal silica solution was regarded as a hard-sphere suspension model with low volume fraction of the silica particles. Rheological properties of the silica suspensions modified with GPS have been investigated as a function of pH and concentration. The acidic solution showed high viscosity change by fast hydrolysis reaction and adsorption of the organic binders on the surface of silica particles. However, the hydrolysis was slow at the basic condition and the binders combined with themselves by condensation. The viscosity change was smallest at pH 7. The viscosity increased with the curing time after adding ethylenediamine, and the increase of viscosity at low pH was higher than that at high pH. The hydrophilic properties of the coating film were investigated by the contact angle of water and film surface. The smallest contact angle was shown under the strong acidic condition of pH 2.

강원도 지역의 용도별 토양의 수소이온농도 및 중금속 오염 평가 (A Study on the Characteristical Evaluation of pH and Heavy Metals Concentrations of Soil in the Gangwon-do)

  • 본 연구는 강원도 지역의 토양환경을 평가하기 위하여 측정망 167개 지점의 토양시료를 분석하여 pH와 중금속에 관한 오염도 조사를 수행하였다. 전체 토양의 pH의 범위는 4.3~8.4로 나타났으며, 중금속의 경우 Cd 0.089 mg/kg (0.000~3.493 mg/kg), Cu 3.093 mg/kg (0.078~60.263 mg/kg), Pb 4.74 mg/kg (0.010~38.080 mg/kg), Hg 0.054 mg/kg (0.002~1.050 mg/kg), As 0.971 mg/kg (0.031~77.051 mg/kg)으로 나타나으며,$Cr^{6+}$은 검출되지 않았다. 중금속 오염에 의한 토양의 상태를 종합적으로 판단할 때 토양의 우려기준에 크게 못 미침으로 강원도의 토양은 비교적 청정한 것으로 나타났다.

  • 용액의 pH, 주입방법, 온도, 유량의 변화를 주면서 질산알루미늄($Al_{3}(NO_{3})_3{\cdot}9H_{2}O$)을 암모니아수($NH_{4}OH$)로 침전시켜 베마이트(Boehmite)를 합성하였다. 베마이트 상이 형성되는 pH 범위와 입자의 형상과 기공특성에 미치는 합성 조건의 영향을 조사하였다. 베마이트는 반응 용액의 pH가 7.5~9일 때 형성되었고, P2jet 주입방법은 침전이 일어나는 동안 pH를 일정하게 유지할 수 있어 용액내의 반응에 참여하는 이온의 농도가 일정하게 유지되어 균일한 크기의 입자와 기공을 형성할 수 있게 하였다. 따라서 비표면적과 기공부피 두 가지 동시에 향상되었다. 온도가 올라갈수록, 유량이 감소할수록 비표면적과 기공부피가 증가함을 보이고, $60^{\circ}C$ 이상에서는 미세섬유모양의 입자를 얻을 수 있었다. 최적의 조건은 pH 9에서 P2jet 방법으로 주입하고 반응온도 $90^{\circ}C$와 유량 2.5 mL/min을 유지하였을 경우로 비표면적은 $385.46m^2/g$이고 기공 부피는 1.0252 mL/g을 가지는 평균 10 nm의 기공이 형성된 다공성 베마이트를 얻을 수 있었다.

Effects of Ionic Speciation of Lysine on Its Adsorption and Desorption Through a Sulfone-type Ion-Exchange Column

  • Lysine produced during microbial fermentation is usually recovered by an ion-exchange process, in which lysine is first converted to the cationic form (by lowering the pH to less than 2.0 with sulfuric acid) and then fed to a cationexchange column containing an exchanger that has a sulfone group with a weak counterion such as NH;. Ammonia water with a pH above 11 is then supplied to the column to displace the purified lysine from the column and allow its recovery. To enhance the adsorption capacity and for a possible reduction in chemical consumption, monovalent lysine fed at pH 4 was investigated in comparison with conventional divalent lysine fed at pH 1.5. The adsorption capacity increased by more than 70% on a mass basis using pH 4 feeding compared with pH 1.5 feeding. Lysine adsorbed at pH 4 started to elute earlier than that adsorbed at pH 1.5 when ammonia water was used as the eluant solution, and the extent of early elution became more notable at lower concentrations of ammonia. Moreover, the elution of monovalent lysine fed at pH 4 displayed a stiffer front boundary and higher peak concentration. However, when the ammonium concentration was greater than 2.0 N, complete saturation of the bed was delayed during adsorption and the percent recovery yield from elution was lowered., both drawbacks that were considered inevitable features originating from the increased adsorption of monovalent lysine.

물유리를 이용한 나노실리카 제조 시 pH가 미치는 영향 (The Effect of pH on Synthesis of Nano-Silica Using Water Glass)

  • Synthesis of nano-silica using water glass in a Sol-Gel process is one of several methods to manufacture nano-silica. In nano-silica synthesized from water glass, there are various metal impurities. However, synthesis of nano-silica using water glass in a Sol-Gel process is an interesting method because it is relatively simple and cheap. In this study, nano-silica was synthesized from water glass; we investigated the effect of pH on the synthesis of nano-silica. The morphology of the nanosilica with pH 2 was flat, but the surface of the nano-silica with pH 10 had holes similar to small craters. As a result of ICP-OES analysis, the amount of Na in the nano-silica with pH 2 was found to be 170 mg/kg. On the other hand, the amount of Na in the nano-silica with pH 10 was found to be 56,930 mg/kg. After calcination, the crystal structure of the nano-silica with pH 2 was amorphous. The crystal structure of the nano-silica with pH 10 transformed from amorphous to tridymite. This is because elemental Na in the nano-silica had the effect of decreasing the phase transformation temperature.
