A rainwater harvesting device was developed for runoff flow harvesting in a small stream or channel and its performance was evaluated in small fields. The rainwater harvesting device has slits on its side of cylindrical volume in 15 cm diameter and 70 cm length, which is designed to increase its water flow harvesting capacity. The maximum collectable water quantity was about 0.0022 ton/sec (130 L/min). Rainwater harvesting device were installed in two locations (P1, P2). P1 is a point for rainfall runoff flow harvesting. P2 is a point for ordinary flow harvesting. During this study, total rainfall depth was 334.5 mm. Runoff of 1,722 ton and 7,984 ton occurred in P1 and P2, and 273 ton and 125 ton were collected by this rainwater harvesting device. Harvesting efficiency was calculated as 15.85 % and 1.57 % in P1 and P2. Clogging of screen filter media in the cylinder due to soil and suspended solids has lowered the harvesting efficiency. However, it was possible to harvest 30 ton/month of rainwater harvesting and it is expected that it will help to solve short-term water shortage.
Escherichia coli (E. coli) heat-labile enterotoxin B subunit (LTB) was regarded as one of the most powerful mucosal immunoadjuvants eliciting strong immunoresponse to coadministered antigens. In the research, the high-level secretory expression of functional LTB was achieved in P. pastoris through high-density fermentation in a 5-1 fermentor. Meanwhile, the protein was expressed in E. coli by the way of inclusion body, although the gene was cloned from E. coli. Some positive yeast and E. coli transformants were obtained respectively by a series of screenings and identifications. Fusion proteins LTB-6$\times$His could be secreted into the supernatant of the medium after the recombinant P. pastoris was induced by 0.5% (v/v) methanol at $30^{\circ}C$, whereas E. coli transformants expressed target protein in inclusion body after being induced by 1 mM IPTG at $37^{\circ}C$. The expression level increased dramatically to 250-300 mg/l supernatant of fermentation in the former and 80-100 mg/l in the latter. The LTB-6$\times$His were purified to 95% purity by affinity chromatography and characterized by SDS-PAGE and Western blot. Adjuvant activity of target protein was analyzed by binding ability with GMI gangliosides. The MW of LTB-6$\times$His expressed in P. pastoris was greater than that in E. coli, which was equal to the expected 11 kDa, possibly resulted from glycosylation by P. pastoris that would enhance the immunogenicity of co-administered antigens. These data demonstrated that P. pastoris producing heterologous LTB has significant advantages in higher expression level and in adjuvant activity compared with the homologous E. coli system.
To know the role of 'the solid compressive pressure to the first solid layer of a cake, $p_d$' in the 'unified theory on solid-liquid separation', we analyzed extremely compressible cake formed with floc for the following three operations with our new concepts. First, the role of $p_d$ was studied in calculating the sediment thickness of floc, and in calculating the cake thickness in cake filtration performed with floc sediment. Second, we calculated the expression procedure using $p_d$. Finally, the influence of $p_d$ on cake thickness in hindered sedimentation, and on the calculation of the procedure of hindered sedimentation $p_d$ was verified. Thus the possibility of application of 'unified theory on solid-liquid separation' and the importance of $p_d$ was verified. Through these processes, new theoretical definitions of the cake filtration, expression and hindered sedimentation was established.
A lab-on-a-chip (LoC) was designed for simultaneous monitoring of microorganisms, antibiotic residues, somatic cells, and pH in raw milk. The LoC was fabricated from polydimethylsiloxane (PDMS) using microelectromechanical system (MEMS) technology, which consisted of two parts; a protein array and microchannel. The protein array was fabricated by immobilizing five types of antibodies corresponding to two microorganisms, two antibiotic residues, and somatic cells. A sol-gel film was deposited on a glass substrate to immobilize the antibodies. The target analytes in raw milk could be bound with the corresponding antibody by an immunoreaction, and the antigen-antibody complex was detected using fluorescence microscopy. SNARF-dextran was used as a pH indicator, and the SNARF-entrapped hydrogel was attached to the microchannel in the chip. After injecting the milk sample into the channel, the pH was measured by monitoring the change in fluorescence intensity by fluorescence microscopy. The on-chip simultaneous assay of two microorganisms (E. coli O157:H7 and Streptococcus agalactiae), two antibiotic residues (penicillin G and dihydrostreptomycin), and neutrophils was successfully accomplished using the proposed LoC system.
The objective of this study was to develop a differential medium with improved selectivity for the isolation of Bacillus cereus. Mannitol egg yolk polymyxin medium supplemented with D-galactose allowed the differentiation of diarrheal- and emetic-type B. cereus through pH monitoring. The pH of the medium decreased significantly when incubating the emetic-type B. cereus, whereas the pH change was not significant when incubating the diarrheal-type. The addition of pH indicators, such as methyl red and phenol red, to the medium allowed visual differentiation between diarrheal- and emetic-type B. cereus. A solid agar medium was also developed by optimizing the concentrations of medium components such as monosaccharides, agar, egg yolk enrichment, pH indicators, and antibiotics. This study indicates the possibility of applying selective media for the differentiation of diarrheal- and emetic-type B. cereus.

The leaching behavior of heavy metal ions with pH and colloid/interface property was analyzed by ICP and SEM. The heavy metals in EAF dust are 'amphoteric metal' and the heavy metal ions leached a little at pH 10. And the leaching concentrations of heavy metals at pH 12 were higher than the that at pH 8. The leaching concentrations of heavy metal ion were decreased with adding the clay to the EAF dust. Especially, the leaching concentrations of heavy metal ion were effectively decreased at pH 12. The observation of colloid/interface properties shows that the soluble silicon hydroxide from clay at pH 12 was precipitated at the surface of the heavy metal and clay particles. This silicon hydroxide precipitates were named the PSHP. The leaching concentrations of heavy metal ion were effectively decreased by the formation of PSHP when adding the clay to the EAF dust and controlling the pH of the slurry at 12.

In this study, we synthesized p-hexyloxybenzaldehyde(HBA) by using p-hydroxybenzaldehyde and n-hexylchloride. p-dodecylaniline(DDA) prepared by nitration and reduction of dodecylbenzene. The schiff base, p-dodecylbenzylidene-$p^\prime$-hexyloxyaniline (DBHA) was synthesized by reaction of HBA and DDA. The color of synthetic compound was pale-brown and the yield was 62%. Photosynthetic parameters of Scendesmus quadricauda, such as the maximum photosynthetic rate ($P_{max}$), photosynthetic efficiency (α) and the initial saturation intensity of irradiance for photosynthesis ($I_K$) were obtained using photosynthesis-irradiance (P-I) curve in a phosphorus-limited chemostat. S. quadricauda exhibitied no photoinhibition until at 200 μmol·$m^{-2}$ . $P_{max}$ (r=0.963, P=0.002) and $I_K$(r=0.904, P=0.013) showed linear relationships with growth rate. Chlorophyll-α concentration and cell dry weight decreased at higher growth rates, ut chlorophyll-α content per cell dry weight increased. The increase in photosynthetic rates at higher growth rates was due to the increase of $P_{max}$ and $I_K$ which was caused mainly by the increase in the absolute amount of chlorophyll-α rather than the increased photosynthetic efficiency of individual chlorphyll-α. The α did not show a significant relationship with growth rate (r=0.714, P=0.111). The cell quota of carbon (r=0.554, P=0.254) was not correlated with growth rate, but cell quota of nitrogen (r=0.818, P=0.047) and phosphorus (r=0.855, P=0.030) exhibited linear correlations with growth rate.

The rare actinomycete Pseudonocardia autotrophica was previously shown to produce a solubility-improved toxicity-reduced novel polyene compound named $\underline{N}ystatin$-like $\underline{P}seudonocardia$$\underline{P}olyene$ (NPP). The low productivity of NPP in P. autotrophica implies that its biosynthetic pathway is tightly regulated. In this study, $wblA_{pau}$ was isolated and identified as a novel negative regulatory gene for NPP production in P. autotrophica, which showed approximately 49% amino acid identity with a global antibiotic down-regulatory gene, wblA, identified from various Streptomycetes species. Although no significant difference in NPP production was observed between P. autotrophica harboring empty vector and the S. coelicolor wblA under its native promoter, approximately 12% less NPP was produced in P. autotrophica expressing the wblA gene under the strong constitutive $ermE^*$ promoter. Furthermore, disruption of the $wblA_{pau}$ gene from P. autotrophica resulted in an approximately 80% increase in NPP productivity. These results strongly suggest that identification and inactivation of the global antibiotic down-regulatory gene wblA ortholog are a critical strategy for improving secondary metabolite overproduction in not only Streptomyces but also non-Streptomyces rare actinomycete species.
The effects of water dropwort on the physical characteristics of tofu were investigated and consumer acceptance test was conducted. pH decreased significantly with increasing water dropwort concentration from 0 to 3%, whereas titratable acidity showed the reverse trend (p<0.05). The amount of water dropwort did not significantly influence the moisture content up to 2% substitution (p>0.05). Turbidity increased significantly with increasing water dropwort concentration (p<0.05). A significantly lower moisture content in 3% water dropwort tofu may be attributed to the denser structure and hardness of that was significantly higher than those of the other samples (p<0.05). Even though $a^*$-values of 3% water dropwort tofu was lower than that of control, $a^*$-values increased significantly with increasing water dropwort content (p<0.05). On the contrary, $b^*$-values decreased significantly with increasing water dropwort content (p<0.05). 2% water dropwort tofu was found to be the best with respect to its overall acceptability score.
