Development of an Analytical Model of BOD(AMB) for Total Pollution Load Regulation (오염총량관리를 위한 분석적 BOD모델(AMB) 개발)

  • Water quality models can be applied to manage the regional water quality problems and to estimate the target and allowable pollution load in watershed effectively. Since the models such as QUAL2E, W ASP5 and HSPF need many data and are not easily applied in real systems, the water quality model, which would be simple and easily applicable, has been required. Thus, an Analytical Model of BOD(AMB) considering travel time of pollutant was developed for the total pollution load regulation in drainage basin. It was found that the main stream length of a sub-basin in the AMB should be shorter than 7km and the length of distributed pollutant load should also be shorter than 3.5km in a sub-basin. The basin in the AMB could be divided into sub-basins with almost same hydraulic characteristics and reaction rate constant satisfying the proposed stream length. The running results of the AMB in a small stream were very close to the results of QUAL2E, which is widely used one in the world. Therefore, the AlVIB can be used to regulate the total pollution load in drainage district by local government.rnment.

Ecological Characteristics and Storage Condition of Diglyphus isaea(Walker) (Hymenoptera: Eulophidae), an Ectoparasite of Liriomyza trifolii(Burgess) (Diptera: Agromyzidae) (아메리카잎굴파리 (Liriomyza trifolii)의 외부기생봉 굴파리좀벌 (Diglyphus isaea)의 생태적 특성 및 저장 조건 구명)

  • Experiments have been performed to investigate biological characteristics of Diglyphus isaea (Walker), an ectoparasite of Liriomyza trifolii (Burgess). At $25^{\circ}C$, development time of D. isaea was estimated as 1.3 days for eggs, 4.5 days for larvae and 6.3 days for pupae. Adult longevity was 25.1 days, and a female adult could lay 305.3 eggs throughout its lifetime. Number of L. trifolii parasitized or killed by the wasp was 962.2 individuals/adult. Most of all parasitized or killed larvae were identified as third instar. With 20% honeyed water as food source, half of the adults could survive 180 days at $5^{\circ}C$, 150 days at $10^{\circ}C$, and 90 days at $15^{\circ}C$. After storing at $10^{\circ}C$ with absolute honey for 30 days, 70 days and 140 days, a female adult in room temperature laid eggs 190.8 individuals, 104.0 individuals and 89.4 individuals, respectively.

  • The Wiskott-Aldrich Syndrome (WAS) is an inherited immunodeficiency caused by a variety of mutations in the gene encoding the WAS protein (WASp). First described in 1937 by Wiskott, the incidence of WAS has so far been estimated at 4 in 106 live births. The Wiskott-Aldrich Syndrome is an X-linked condition characterized by 1) an increased tendency to bleed caused by a reduced number of platelets, 2) recurrent bacterial, viral and fungal infections, and 3) eczema of the skin. The purpose of this report is to present cases highlighting the clinical features of the syndrome and the required considerations in the treatment of patients. The report consists of two particular cases: a 2-year-11-month-old boy seen for a routine oral examination prior to his bone marrow transplantation and a 2-year-6-month-old boy with herpes gingivostomatitis and teeth discoloration.

1SWASP J093010.78+533859.5: A Possible Hierarchical Quintuple System

  • Among quadruples or higher multiplicity stars, only a few binary systems have been discovered. They are important targets to understand the formation and evolution of multiple stellar systems because we can obtain accurate stellar parameters from photometric and spectroscopic studies. We present the observational results of this kind of rare object 1SWASP J093010.78+533859.5, for which the doubly eclipsing feature had been detected previously from the SuperWASP photometric archive. Individual PSF photometry for two objects with a separation of about 1.9 arcsec was performed for the first time in this study. Our time-series photometric data show that the brighter object A is an Algol-type detached eclipsing binary with an orbital period of 1.3 days and the fainter B is a W UMa-type contact eclipsing binary with a period of 0.23 days. Using the high-resolution optical spectra, we obtained well-defined radial velocity variations of the system A. Furthermore, stationary spectral lines were detected and should have originated from the other stellar component, which was confirmed by the third object contribution from the light curve analysis. No spectral feature of the system B was detected, probably due to its faintness. We obtained the binary parameters and the absolute dimensions from each light curve synthesis. The primary and secondary components of the system A have a spectral type of K1 and K5 main sequences, respectively. Two components of system B have nearly the same type of K3 main sequence. Light variations at out of eclipses were appeared in both systems, interpreting as the effect of stellar spots on these late spectral type stars. We estimated the distances to the systems A and B individually. They may have similar distances of about 70 pc and seem to be gravitationally bound with a separation of about 130 AU. In conclusion, we suggest that 1SWASP J093010.78+533859.5 is a quintuple stellar system with a hierarchical structure of a triple system A(ab)c and a binary system B(ab).

  • 1992년 한강수계법이 제정되면서 우리나라에 도입된 오염총량관리제는 현재 2차총량관리 시행계획수립 단계에 이르렀다. 오염총량제에서 수질모델은 수계구간별로 설정된 기준유량과 목표수질 조건을 달성하는 지를 판단할 수 있는 도구로 사용되며, 다양한 모델들이 사용되고 있다. 그 중 하천수질모형으로는 주로 QUAL2E, QUALKO, QUALKO2 모형으로 압축할 수 있다. QUAL2E 모형은 1980년대에 개발되어 국내외로 널리 이용하고 있으나 SOD를 0차나 일정량으로 처리하였고, 부착조류에 의한 용존산소 변화와 부유 조류 사멸시 발생하는 유기물이 고려되지 않았다. 또한 용존산소가 부족한 상태에서 반응이 활발한 탈질화과정이 포함되지 않아 이들 반응에 의해 수질이 영향을 받는 하천에 적용하기에는 한계가 있었다. 그리고 QUAL2E 모델은 여러 개의 지류를 가진 대형 하천에는 적용하기 어려운 단점이 있다. 국내에서는 1999년 QUAL2E 모델에 WASP5의 장점을 접목시켜 QUALKO 모델을 개발하였다. 이 모델은 QUAL2E에 부유성 조류의 사멸로 인한 유기물의 내부증가, 탈질화 반응 및 부착식물의 광합성 호흡 과정을 추가한 것이다. 또한 QUAL2E 모델에서 BOD는 CBOD로 입력되고 모의되므로 bottle BOD의 개념이 결여되어 있으므로 이러한 문제점을 보완하고, 조류의 생산 및 사멸에 의한 내부생산 유기물 증가와 탈질화 반응 과정을 추가한 것이다. 우리나라에서 진행되고 있는 총량관리 대상물질은 2010년까지는 $BOD_5$이며, 2011년부터는 일부 지역에 총인이 포함될 예정이다. 2007년에 실험실에서 측정하는 BOD5나 유기성 질소 또는 유기성 인을 그대로 입력하여 계산되고 출력할 수 있으며, 향후 오염총량제의 관리대상항목으로 논의되고 있는 TOC를 모의할 수 있는 QUALKO2가 개발되었다. 이에 본 연구에서는 향후 활용도가 클 것으로 기대되는 QUALKO2 모형에 기존 QUAL2E-UNCAS 모형에 서 수행할 수 있는 불확실성 해석 기법인 Monte Carlo 모의를 가능하도록 모형을 수정하고자 한다. 실제 하천에서의 수질해석에 대한 단순한 표현인 수학적 모형은 불확실성을 내포하고 있으며, Monte Carlo 해석을 사용하여 모형의 불확실도 정량화와 매개변수의 불확실성을 통계학적으로 기술할 수 있다. 또한 각 지점에 대한 계산결과치들에 대해 빈도 및 누가빈도분포 값을 제시함으로서 모형 예측치들의 전반적인 분포경향을 평가할 수 있으며, 하천수질에 대해서 환경기준치를 위배할 가능성을 산정하는데 활용할 수 있다. 우리나라 실정에 맞는 QUALKO2 모형에 Monte Carlo 모의를 통해 신뢰도 기반의 수질해석을 수행하게 된다면 수질정책의 기초자료 제공에 기여할 것으로 판단된다.

Pest control managements for preservation of wooden cultural properties (목조문화재의 원형보존을 위한 충해 방제방안)

  • The cultural properties are damaged by various causes according to the characteristics of material, the condition of preservation, and the period of time. Especially, biodeterioration makes lots of damages in organic properties than inorganic ones. The damages of wooden cultural properties by insects usually are caused by the three orders; Isoptera, Coleoptera, and Hymenoptera. As the result of investigation on the state of 141 buildings of wooden cultural properties in 1999, some of them were damaged by many kinds off actors; wasp, powder post beetle, cigarette beetle, termite, decay, and physical cracking. And it was found that the patterns of damages were related to species-specific habits of insects. There are several methods of pest control for the prevention of wooden cultural properties from damages caused by insects. Those are as follows; physical control, chemical control, biological control, and integrated pest management. When insects and fungi were detected at the wooden buildings, the fumigation is best treatment to stop biodeterioration. And then, wood materials also need to be treated with insecticidal and antiseptic chemicals to avoid a reinfestation, because the fumigant is volatile. The six commercial chemicals which are applied to the insecticidal and antiseptic treatment of wooden cultural properties were purchased to test their abilities. According to the comparative results of efficacy of them in laboratory, chemical D showed excellent efficacy in all items, including antiseptic and termiticidal items. The goal of these pest controls is to protect wooden buildings from insects and microorganisms. The most effective method used currently is chemical control(fumigation, insecticidal and anticeptic chemical treatment), but it has to be treated periodically to control pest effectively. Recently environmentally-friendly control methods such as bait system or biological treatments are replacing traditional barrier treatments using large amounts of chemicals. Especially, termite is a social insect which makes a colony. Although a building with fumigation treatment is safe for a while, once attacked building has a risk of damage by reinfestation of termite. Therefore, to control termites from damaged building, the entire colony including reproductives(queen and king) and larvae around buildings must beeliminated. Bait system can be used as a preventive measure in early detection of them through termites colony monitoring and baiting. It would be the most effective for termite control if bait system would be used together with the chemical controls.

Insect Resistance of Tobacco Plant Expressing CpBV-ELP1 Derived from a Polydnavirus (폴리드나바이러스 유래 CpBV-ELP1 발현 담배의 내충성)

  • Polydnaviruses (PDVs) are a group of double-stranded DNA viruses symbiotic to some endoparasitoid wasps. Cotesia plutellae bracovirus (CpBV) is a PDV symbiotic to an endoparasitoid wasp, C. plutellae, parasitizing young larvae of Plutella xylostella. An early expressed gene, CpBV-ELP1, plays an important role in the parasitism by suppressing host cellular immunity by its cytotoxic activity against hemocytes. This study aimed to test its oral toxicity against insect pest by expressing it in a recombinant tobacco plant. A recombinant CpBV-ELP1 protein was produced using a baculovirus expression system and secreted to cell culture medium. The cell cultured media were used to purify CpBV-ELP1 by a sequential array of purification steps: ammonium sulfate fractionation, size exclusion chromatography, and ion exchange chromatography. Purified rCpBV-ELP1 exhibited a significant cytotoxicity against Spodoptera exigua hemocytes. CpBV-ELP1 was highly toxic to the fifth instar larvae of S. exigua by injection to hemocoel. It also showed a significant oral toxicity to fifth instar larvae of S. exigua by a leaf-dipping assay. CpBV-ELP1 was cloned into pBI121 vector under CaMV 35S promoter with opaline synthase terminator. Resulting recombinant vector (pBI121-ELP1) was used to transform Agrobacterium tumefaciens LBA4404. The recombinant bacteria were then used to induce callus of a tobacco (Nicotiana tabacum Xanthi) leaves and subsequent generation (T1) plants were selected. T1 generation tobacco plants expressing CpBV-ELP1 gave significant insecticidal activities against S. exigua larvae. These results suggest that CpBV-ELP1 gene can be used to control insect pests by constructing transgenic crops.

Inhibitory Effect of Cotesia plutellae Bracovirus (CpBV) on Development of a Non-natural Host, Spodoptera exigua (프루텔고치벌(Cotesia plutellae) 유래 폴리드나바이러스의 비자연 기주 파밤나방(Spodoptera exigua)에 대한 발육 억제 효과)

  • Polydnavirus is a symbiotic virus of some endoparasitic wasps and plays crucial roles in inhibiting immune responses and retarding development of the parasitized hosts. Cotesia plutellae bracovirus (CpBV) is a polydnavirus suggesting a major causative to change developmental physiology of the parasitized host. Here, we investigated whether CpBV can interrupt development of non-natural host. Beet armyworm, Spodoptera exigua, is used as a non-permissible host for parasitization of C. plutellae. Extract from the calyx region of C. plutellae contained CpBV, which was confirmed by immunoblotting with a polyclonal antibody raised against CpBV. One female equivalent of CpBV extract was injected into hemocoel of late 4th instar larvae of S. exigua. The injected larvae showed delayed larval period, decrease of body weight gain, and inability of pupal metamorphosis. These inhibitory effect of the CpBV extract was rescued by injection along with CpBV antibody, though the antibody itself did not give any effect on development of the larvae. This result clearly shows that CpBV can interrupt developmental physiology of a non-natural host for its symbiotic wasp.

Polydnavirus Replication and Ovipositional Habit of Cotesia plutellae (프루텔고치벌(Cotesia plutellae) 폴리드나바이러스 복제와 산란 습성)

  • An endoparasitoid wasp, Cotesia plutellae, has been used for a biological control agent against the diamondback moth, Plutellae xylostella. It has a symbiotic polydnavirus in their reproductive tract, which is required for its successful parasitization. Here, we measured a specific replication time of the polydnavirus during female development of C. plutellae. We, also, analyzed the reproductive potentials of female C. plutellae under mating or different host conditions. At $25^{\circ}C$, pupal C. plutellae began to develop adult tissues such as compound eyes and wings since day 2. At day 5, all adult tissues including antennae were developed and were ready to emerge. With polyclonal antibody raised against C. plutellae polydnavirus, an immunobloting could confirm virus replication at day 4 during pupal stage. Virus particles could be visualized by transmission electron microscope in the oviduct lumen of day 5 pupae. After adult eclosion, venom gland and ovarian calyx increased in size, though ovarioles did not. Mated females layed large number of eggs (over $60\%$) at first 4 days during their mean longevity of ca. 8 days at $25^{\circ}C$. Unmated females showed less active ovipositional behavior, where all the eggs developed into males. C. Plutellae parasitized both P. xylostella and fall webworm, Hyphantria cunea. However, C. Plutellae developed faster and showed higher successful paarasitization in P. xylostella than in H. cunea.

Influence of Aggregation Pheromone of Riptortus clavatus (Heteroptera: Alydidae) on the Occurrence of Egg Parasitoid, Gryon japonicum (Hymenoptera: Scelionidae) (톱다리개미허리노린재의 집합페로몬이 노린재검정알벌의 발생에 미치는 영향)

  • The kairomonal activity of the aggregation pheromones of Riptortus clavatus for the egg parasitoid, Gryon japonicum, were investigated in soybean field. The parasitism rate of G. japonicum gradually decreased when the distances between pheromone traps and egg sites were increased from 0 to 15 m. The active distance of pheromone source for parasitoid attraction was estimated to be ca. 15 m. Under field conditions, the parasitoid wasp population peaked during the late August and early September, and the parasitism rate increased when the number of pheromone traps was increased from one to three per $165\;m^2$. However, insecticide-treated plots, the level of parasitism recorded is relatively low. Sex ratios in field populations of G. japonicum were female-biased.
