Advanced wastewater treatment capacity and growth of Chlorella vulgaris by nitrogen and phosphorus concentrations (N, P 농도에 따른 Chlorella vulgaris의 성장 및 하수고도처리능 평가)

  • The growth and removal capacity of nitrogen and phosphorus of Chlorella vulgaris were evaluated in artificial wastewater with different nitrogen and phosphorus concentrations as element growing components for microalgae growth. The nitrogen concentration was varied in 9, 15, 30 and 60 mg-N/L with fixed phosphorus concentration of 3 mg-P/L. The growth and phosphorus removal capacity of C. vulgaris were high at initial nitrogen concentration of 15 and 30 mg-N/L, and the corresponding N/P ratios calculated were 5 and 10. In the case of varying in 1.5, 3, 6 and 10 mg-P/L of phosphorus concentration with fixed nitrogen concentration of 30 mg-N/L, the growth and removal capacity of nitrogen and phosphorus were excellent with phosphorus concentration of 3 and 6 mg-P/L. The corresponding N/P ratios were shown as 10 and 5. Therefore, the appropriate N/P ratio was concluded between 5 and 10 for wastewater treatment using C. vulgaris.

  • Verruca vulgaris is one of the most frequent lesions of the skin. Evidences support that this lesion has strong association with low-risk human papillomavirus. Unlike skin lesion, oral verrucae vulgaris are not easily encountered and often misdiagnosed as squamous papilloma. Herein, we reported eight cases of oral verruca vulgaris and analyzed their clinicopathological features.

  • The genus Proteus has been rarely isolated from cultured fish. In this study, we reported Proteus vulgaris isolated from fish skin lesion of Black-spotted porcupine fish (Diodon hystrix). Identification of f vulgaris was performed by general biological test, including API 20E and API ZYM. The sensitivity of isolated strains to different groups of antibiotics was evaluated using the disc diffusion method. It showed that the isolate is sensitive to ciprofloxacin $(5{\mu}g)$, norfloxacin $(10{\mu}g)$, enrofloxacin $(5{\mu}g)$, cefotaxime $(30{\mu}g)$.

Effects of volatile fatty acids on microalgae growth and N, P consumption in the advanced treatment process of digested food waste leachate by mixotrophic microalgae (Mixotrophic microalgae에 의한 음폐수 소화액 고도처리에 있어 유기산이 microalgae의 성장 및 질소, 인 제거에 미치는 영향)

  • Acetate, propionate, butyrate are the major soluble volatile fatty acids metabolites of fermented food waste leachates. This work investigate the effects of volatile fatty acid on the growth rate and $NH_4-N$, $PO_4-P$ removal efficiency of mixotrophic microalgae Chlorella vulgaris to treat digested food waste leachates. The results showed that acetate, propionate and butyrate were efficiently utilized by Chlorella vulgaris and microalgae growth was higher than control condition. Similar trends were observed upon $NH_4-N$ and $PO_4-P$ consumption. Volatile fatty acids promoted Chlorella vulgaris growth, and nutrient removal efficiencies were highest when acetate was used, and butyrate and propionate showed second and third. From this work it could be said that using mixotrophic microalgae, in this work Chlorella vulgaris, fermented food waste leachates can be treated with high efficiencies.

  • 한국산 Genus Vespula THOMSON 으로는 다음 6종이 알려지고 있다. Vespula austpiaca PANZER 삼성말벌 V. germanica flevicets SMITH 해첨배기말벌 V. media RETZIUS 좀말벌 V. koieensis YADOSZKOWSKI 참땅벌 V. rufa L. var. schrenckii RADOSZAKOWSKI 슈렝크말벌 V. vulgaris L. var. Lewisii CAMERON 땅벌 상기종중에서 V. austriaca, V. rufa, V. vulgaris의 종간은 각지에서 채집된 기록이 있으나 나머지 것은 한국에 산한다고만 되어있다. 채집된 상기 3종은 형태초적으로는 거의 비슷하고 반문 만을 달리하니 그 반문의 변이를 조사하는 것은 중대한 의의를 가진다. 필자는 서울 근교우이동에서 밤을 이용하여 8개의 벌집속 벌을 모조리 채집하여 그 중 2 벌집의 개체만에 관하여 그 변이를 조사한 결과를 제1보로 여기에 발표한다. 한 벌집 속 개체수는 726과 661 이었고 다라서 총개체수 1337 마리를 조사한 결과 상당히 심히 문반변이가 잇음을 알었고 더우기 V. austriaca, V. rufa, V. koreensis 와 V. vulgaris 간의 반문차는 동일종내의 변이에 불과함을 알게 되었다. 따라서, V. austriaca, V. rufa, V. koreensis는 V. vulgaris 의 synonym 임을 밝히는 바이다. 본연구에 있어서 귀중한 교\ulcorner을 빌려주신 고대이과부장 조복의교수와 재료의 채집과 조사에 협력해준 고대생물학과생 김원임, 이경\ulcorner, 이병준, 이동\ulcorner \ulcorner군에게 감사를 표하는 바이다.

Development of Microbial Fuel Cells Using Proteus vulgaris

  • Microbial fuel cells comprising the microorganism P. vulgaris, thionin as a mediator, and various mono- and disaccharides in an anodic compartment have been developed. A cathodic compartment containing a Pt electrode and Fe$(CN)_6^{3-}$ was separated from an anode by the Nafion membrane. From absorbance-time measurements, it was found that the absorbance of thionin was not altered by the addition of P. vulgaris, even in the presence of sugars. However, thionin was effectively reduced when P. vulgaris was present. These results differ substantially from the case of safranine O, a phenazine-derivative, indicating that thionin takes up electrons during the metabolic oxidation processes of carbohydrates. Maximum fuel cell efficiency was observed at 37 $^{\circ}C$, optimum temperature for the growth of P. vulgaris, and 0.5 V cell voltage was obtained, which indicates that the metabolism of the microorganism directly affects the efficiency. Thionin concentration was closely related to cell performance. When the charging-discharging characteristics were tested with glucose, galactose, sucrose, maltose, and trehalose as carbon sources, galactose was found to give the highest coulombic efficiency. Cell performance was almost fully recovered with only small degradation when glucose and sucrose were used in the repetitive operation. Current was maintained nearly twice as long for sucrose than in the case of glucose.

A Case Report of Verruca Vulgaris Patient, Treated only with Korean Medicine (한약 단독 치료를 이용한 심상성 사마귀 1례 보고)

  • Objectives : The purpose of this study is to report a case of verruca vulgaris patient, treated only with Korean medicine. Methods : A 6-years-old female patient was treated only with Korean medicine due to verruca vulgaris on left hand. Photographs and size measurements of lesions were used to evaluate the changes in symptoms. Results : After 3 months of treatment, the patient's lesions showed almost complete clearance and there were no adverse effects. Conclusions : According to the results, Korean medicine with Bojungikgi-tang Hab Saengmaek-san Gamibang could be an effective and safe treatment for verruca vulgaris in a pediatric patient.

Efficient Expression System of High Value Proteins, hGM-CSF and hEGF, using Agrobacterium-mediated Chlorella Transformation (아그로박테리움 매개 클로렐라 형질전환을 이용하여 유용 단백질인 hGM-CSF와 hEGF의 효율적인 발현 시스템)

  • Chlorella has various biotechnological applications, including in the biomedical and pharmaceutical industries, because of its advantages, including rich nutrients, fast growth rate, easy cultivation, and high biomass. We used the Agrobacterium-mediated transformation method to express human GM-CSF and EGF proteins, which are widely used in regenerative medicine, cosmetics, and pharmaceutical materials in Chlorella. The codon-optimized hGM-CSF and hEGF genes were cloned into plant binary vectors and transformed into Chlorella vulgaris using the Agrobacterium-mediated coculture transformation method. After transformation, genomic DNA PCR was performed for each C. vulgaris line that was stably subcultured on an antibiotic-resistant solid medium to confirm the insertion of hGM-CSF and hEGF into the chromosome. Furthermore, PT-PCR and protein expression of hGM-CSF and hEGF in each transformed C. vulgaris were significantly increased compared to the untransformed Chlorella. This study suggests that high-value proteins, including hGM-CSF and hEGF, which are foreign genes of C. vulgaris, can be stably expressed through the Agrobacterium-mediated Chlorella transformation system.

Utilization of Lipid-Extracted Algae Cell Residue in Cultivation of Chlorella vulgaris for Biodiesel Production

  • Lipid-extracted algae (LEA) cell residue is generated as an organic solid waste in the process of biodiesel production from microalgae, and its recycling or reuse is important in the aspect of waste minimization. In this study, the influence of the addition of LEA hydrolysate to Chlorella vulgaris (C. vulgaris) cultivation on cell growth and biodiesel production was investigated to seek a possible use of LEA as a carbon source. LEA was hydrolyzed by three different methods: acid hydrolysis, autoclave, and ultra-sonication. The resulting hydrolysates were supplemented with three background media: formulated defined medium (BG-11), organic liquid fertilizer (PAL1), and distilled water. Both cell growth and lipid production of C. vulgaris were improved under mixotrophic cultivation. By supplementing hydrolysates, biomass productivity was increased several folds because the LEA hydrolysates contained monosaccharides such as glucose and galactose. Lipid contents and biodiesel productivity were bestly increased in PAL1 medium supplemented with ultrasonication (UL) hydrolysate, from 11 to 25% after 14 days, while nitrate concentration was quickly reduced from 55 to below 10 mg/L. The suggested recycling option of the LEA to microalgae cultivation was helpful to improve biodiesel productivity as well as to reduce organic waste generation.

  • From the seeds of Zizyphus vulgaris var. spinosus, aporphine alkaloids : nuciferine, N-methylasimilobine, nornuciferine, norisocorydine, caaverine and tetrahydro benzylisoquinoline alkaloid : (+) coclaurine were isolated and identified. Zyzyphusine, a new quaternary aporphinium alkaloid form butanol soluble fraction was isolated and characterized by spectral data.
