The effects of explant sources, plant growth regulators on callus induction and plantlet differentiation from leaf blade, petiole, and meristem tissue of Pelalgonium citrosa were investigated under illumination or in dark condition Leaf blade explants cultured on Murashige and Skoog's medium containing 2,4-D and kinetin did not form callus or organ. But those cultured on medium with NAA and BA produced callcus and shoots. Dark condition was more effective than light condition to callus induction and showed that some of shoot were differentiated directly from leaf blade explane. Callus proliferated vigorously on meristem tissue after 7 days of culture, and multiple shoots were obtained Sum callus on medium with 0.5 mg/L NAA and BA. Roots formed readily from about 80% of the shoots cultured on medium with 1.0 mg/L NAA. Regenerated plantlets regenerated had phenotypically normal leaves and roots.

For utilizing vitrification to treat low and intermediate level waste, industrial pilot plant was designed and constructed in October 1999 at Daejon, Korea through the joint research program among NETEC, MOBIS and SGN. More than 70 tests were performed on simulated IER, DAW etc. including key nuclide surrogate(Cs, Co); this plant has been shown to vitrify the target waste effectively and safely, however, some dust are generated from the HTF(High Temperature Filter) as a secondary waste. In case of long term operation, it is also concerned that pipe plugging can be occurred due to deposited dust in cooling pipe namely, connecting pipe between CCM(Cold Crucible Melter) and HTF. In this regard, we have developed the special complementary system of the off-gas treatment system to recycle the dust from HTF to CCM and to remove the interior dust of cooling pipe. Main concept of the dust recycling is to feed the dust to the CCM as a slurry state; this system is regarded as of an important position in the viewpoint of volume reduction, waste disposal cost and glass melt control in CCM. The role of DRS(Dust Recycling System) is to recycle the major glass components and key nuclides; this system is served to lower glass viscosity and increase waste solubility by recycling B, Na, Li components into glass melt and also to re-entrain and incorporate into glass melt like Cs, Co. Therefore dust recycling is helpful to control the molten glass; it is unnecessary to consider a separate dust treatment system like a cementation equipment. The effects of Dust Cleaner are to prevent the pipe plugging due to dust and to treat the deposited dust by raking the dust into CCM. During the pilot vitrification test, overall performance assessment was successfully performed; DRS and Dust Cleaner are found to be useful and effective for recycling the dust from HTF and also removing the dust in cooling pipe. The obtained operational data and operational experiences will be used as a basis of the commercial facility.
This study was carried out to determine factors affecting on the mass propagation of Rehmannia glutinosa seedlings in bioreactor culture. Air-lift type bioreactor was more compatible to shoot formation than stirrer type. Fifty grams(90 stem explants) of inoculum in 1.5L medium was placed into 2.5L bioreactor with aeration rate of 0.5 v.v.m., which was proper for effective shoot formation. Adding MES as pH buffer to culture medium increased the numbers of shoot formation. Adding 5g/l of anti-vitrifying agent into culture medium was highly effective for diminishing the rate of vitrification in shoots formed.
In order to examine and compare the characteristics of two vitrified forms (AG8W1 and DG2) simulated for the operation of a commercial vitrification facility being constructed in Ulchin nuclear power plant, the vitrified forms were cooled by the natural cooling and annealing methods respectively. And the Product Consistency Test (PCT), compressive strength, thermal conductivity, specific heat, phase stability, softening point and Coefficient of Thermal Expansion (CTE) of the vitrified farms were experimented. Consequently, it was shown that there were no significant differences on the physiochemical properties of the vitrified forms performed the natural cooling and annealing.

Legacy waste from the decommissioned A-1 nuclear power plant in the Slovak Republic is scheduled for immobilisation within a tailored alkali borosilicate glass formulation, as part of ongoing site cleanup. The aqueous durability and characterisation of a simulant glass wasteform for Chrompik III legacy waste, was investigated, including dissolution experiments up to 112 days (90℃, ASTM Type 1 water). The wasteform was an amorphous, light green glassy product, with no observed phase separation or crystalline inclusions. Aqueous leach testing revealed a suitably durable product over the timescale investigated, comparing positively to other simulant nuclear waste glasses and vitreous products tested under similar conditions. Iron and titanium rich precipitates were observed to form at the surface of monolithic samples during leaching, with the formation of an alkali deficient alteration layer behind these at later ages. Overall this glass appears to perform well, and in line with expectations for this chemistry, although longer-term testing would be required to predict overall durability. This work will contribute to developing confidence in the disposability of vitrified Chrompik legacy wastes.

The expression profiles of low-temperature-related genes were examined in in vitro potatoes exposed to a cold condition for 1-3 days. The expression levels of PPI1 and CI21B genes were lineraly elevated from day 1 to day 3, while the opposite trend was observed for CBF4 and CI21A. In addition, the expression of the genes CI21A and CI21B varied, along with specific tissues (leaf, stem, and tuber) and the treatment periods. Therefore, potato shoot tips were cryopreserved with LS and PVS2 containing different oncentrations of AFP. It can thus be inferred that the presence of AFP in LS and PVS2 was likely to elevate expression pattern of the genes. Furthermore, the concentration of AFP (1,500 ng/ml for LS and 500 ng/mg for PVS2) was the best for the cryopreservation of potatoes.
This study was carried out to investigate the optimum composition and concentration of medium for the acclimation of the shoots derived from bioreactor culture of Scrophularia buergeriana. Rooting and growth of the shoots cultured in bioreactor for 4 weeks were better on liquid full strength MS medium while one-tenth of MS medium was good for rooting and growth of the shoots cultured in bioreactor for 6 weeks. Leaf expansion was more effective in the shoots cultured for 6 weeks on solid MS medium than in those cultured for 4 weeks. No leaf expanded in liquid medium. Vitrification rate was higher in full strength solid MS medium than in half strength MS medium. It is concluded that acclimation of the shoots is more effective on the half strength solid MS medium containing 1.2% agar.
We investigated the anatomical aspects of vitrification in peace poplar. Comparisons were made with regard to characteristics occurring between hyperhydric and normal shoots in shoot proliferation cultures on MS medium containing 0.2 mg/L BA. Compared with normal plants, hyperhydric plants had thick, curled, and dark green leaves. Hyperhydric stems were thicker and shorter than those of normal stems. When examined under the microscopes, the mesophyll palisade cells of hyperhydric leaves were vacuolated, whereas those of normal leaves contained normal and enriched vacuole with cytoplasm. Generally, the hyperhydric leaves showed poorly developed palisade parenchyma, and revealed irregular and bigger sized intercellular structures in both palisade and spongy parenchyma as well as epidermis cells compare to those of normal leaves. In addition, the hyperhydric leaves had lower stomatal density and bigger sized cell. Vascular tissues of hyperhydic stems were less differentiated because of poorly lignified xylem tissue. The greatly expanded cortical cells and pith appeared to be the main cause of thick stems as compared with normal stems.

Effects of media and plant growth regulators on the germination of somatic embryos of Angelica tree(Aralia elata Seem.) was studied for the mass production of Angelica tree through tissue culture. MS medium was found to be the most effective for the germination of somatic embryos(65% germination rate), Among the MS medium, the medium containing 25% less inorganic salts and 1% less sucrose was found to be the most effective. Gelling agent with 0.2-0.3% gelrite promoted the germination of somatic embryo$(65{\sim}70%)$ and caused good growth of shoots and roots. 0.1 mg/l of BA and kinetin treatment caused $65{\sim}70%$ germination rate of somatic embryos and good growth of shoots and roots, and resulted in high percentage of dry matter. 1mg/l or 5 mg/l treatment of putrescine, and 10 mg/l treatment of spermidine caused 90% germination rate of somatic embryos and good growth of plant organs, and inhibited vitrification of regenerated plants.
