만성적인 에탄올 섭취 시 수종 천연 추출물 투여가 흰쥐의 혈청과 간의 지질 함량에 미치는 영향 (Effects of Herbal Extracts on Serum and Liver Lipid Levels in Chronic Ethanol Administered Rats)

  • This study had been done for the investigation of the effect of Vitis vinifera extract (VV), Schisandra chinensis extract (SC), Taraxacum officinale extract (TO), Gardeniajasminoides extract (GJ), Angelica acutiloba extract (AA) and Paeonia japonica extract (PJ) on fatty liver and hepatotoxicity which was induced by Lieber-DeCarli ethanol liquid diet. Male Sprague-Dawley rats were randomly divided into eight groups: ethanol diet (ED), normal diet (ND), ED+VV (100mg/kg), ED+SC, ED+TO, ED+GJ, ED+AA, and ED+PJ (300mg/kg/day). Rats fed liquid diets for 6 weeks showed remarkable increase in serum and hepatic lipids indicating the onset of alcoholic fatty liver. The increasing levels of GPT, ALP activities in serum were observed in the groups fed with alcohol-containing diets compared to those of the ND group. The VV, SC, TO, GJ, AA and PJ groups were decreased the levels of triglyceride, free fatty acid and total cholesterol in serum and liver and GPT, ALP activities in serum. Therefore, they can be utiliaed as a health functional food or new drug candidate for fatty liver and hepatotoxicity which was induced by chronic alcohol consumption.

포도의 품종별 휘발성 향기성분 분석 (Volatile Flavor Components in Various Varieties of Grape(Vitis vinifera L.))

  • SDE 방법으로 추출한 블랙올림피아, 캠벨, 델라웨어의 향기성분을 분석하여 각각 77종, 72종, 74종의 휘발성 향기성분을 분리.동정하였다. 블랙올림피아의 주요향기성분으로 (E)-2-hexenal(20.36%), diethylacetal(18.03%), hexanal, ethyl acetate가 다량 함유되어 있었다. 캠벨에서는 에스터류로써 30.81%를 나타낸 ethyl acetate가 가장 높은 함량을 보였으며, ethyl (E,E)-2,4-decadienoate와 ethyl butanoate가 각각 7.96%, 2.59%를 차지하였고, 알코올류와 알데이드류는 주로 $C_{6}$ 화합물이 다량 함유되었다. 델라웨어에서는 블랙올림피아나 켐벨과는 달리 알코올류가 높은 함량을 나타내었으며, 주요 성분은 (E)-2-hexenal (21.07%)과 (E)-2-hexen-1-ol(19.43%) 이었다. 세 종류의 품종에서 공통적으로 hexanal, (E)-2-hexenal, hexanol, (E)-2-hexen-1-ol, ethyal acetate 둥이 다량 동정되어 $C_{6}$-화합물이 포도의 주된 향기성분으로 확인되었으나 테르펜올류는 델라웨어에서만 미량으로 검출되었다. 품종별 휘발성 향기성분의 총함량으로 블랙올림피아, 캠벨, 델라웨어에서 각각 13.7 mg/kg, 48.5 mg/kg,15.3 mg/kg이 정량분석 되었다.

Anthocyanins in 'Cabernet Gernischet' (Vitis vinifera L. cv.) Aged Red Wine and Their Color in Aqueous Solution Analyzed by Partial Least Square Regression

  • Anthocyanins are considered one of the main color determinants in aged red wine. The anthocyanins in aged red wine made from 'Cabernet Gernischet' (Vitis vinifera L. cv.) grape were investigated by high performance liquid chromatography- electronic spray ionization- mass spectrometry (HPLC-ESI-MS) and their color presented in aqueous solution were evaluated using partial least square regression (PLS). The results showed that there were 37 anthocyanins identified in this wine, including 22 pyranoanthocyanins. The analysis of PLS indicated that different anthocyanins showed distinct color values: malvidin 3-O-(6-O-acetyl)-glucoside-4-vinylguaiacol (Mv3-acet-glu-vg) presented the highest color values, while malvidin 3-O-glucoside (Mv3-glu) showed least. Among the free non-acylated anthocyanins, peonidin 3-O-oglucoside (Pn3-glu) showed the highest color values; the coumarylated anthocyanins presented higher color values than their corresponding acetylated anthocyanins and parent anthocyanins; pyranoanthocyanins presented also higher color values than their original anthocyanins; the color of anthocyanins depended on their structure. This work will be helpful to reveal evolution in aged red wine.

Ethyl oleate 처리에 의한 'Merlot' 포도 과피의 안토시아닌 함량과 해부학적 변화 (Anatomical Changes and Anthocyanin Contents of the Exocarp by Ethyl Oleate Treatment on 'Merlot' Grapes)

  • 'Merlot' 포도 품종의 수확전 ethyl oleate 처리는 외표피와 아표피의 두께를 감소시키며 착색을 현저히 향상시킨다. 무처리구의 과피 두께는 $126-189{\mu}m$인데 반하여 처리 과실의 과피두께는 $90-107{\mu}m$로 조사되었으며, 이러한 외표피와 아표피 층의 두께 감소는 처리 후 외표피와 아표피를 구성하는 세포의 빠른 노화에 의하여 세포가 죽거나 탈수가 나타나기 때문인 것으로 조사되었다. 처리 후에 빠르게 과피 표면의 왁스층이 녹은 듯한 형태를 보이며 이는 착색을 증진시키는 것으로 관찰되었으며, 전체적인 안토시아닌 함량 역시 증가하는 것으로 조사되었다. 반면에 각각의 안토시아닌 함량에 있어서는 메톡시기와 결합하거나 유기산과 결합한 안토시아닌이 증가한 반면 수산기와 결합한 안토시아닌은 감소하는 경향이었다.

  • This study had been done for the investigation of the effect of Vitis vinifera extract(V), Schisandra chinensis extract (S), Taraxacum officinale extract (T), Gardenia jasminoides extract (G), Angelica acutiloba extract (A) and Paeonia japonica extract (P), and their mixtures on the antioxidant and ethanol oxidation system which was induced by Lieber-DeCarli ethanol liquid diet. Male Sprague-Dawley rats were randomly divided into eight groups: ethanol diet (ED), normal diet (ND), ED+V (100 mg/kg/day), ED+S, ED+T, ED+G, ED+A and ED+P (300 mg/kg/day). We studied the effect on alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) after herbal extracts administration for 6 weeks in rats induced by Lieber-DeCarli ethanol liquid diet. The differences in ADH and ALDH activity of the rats treated with herbal extracts and ED group were not significant. Phase I enzyme activity was found to be significantly higher in the ED+V than the ED group. Phase II enzymes (glutathione-S-transferase, phenol sulfatransferase) activities were found to be higher in the herbal extracts than the ED group. Herbal extracts not only reduced ethanol-induced elevation of level malondialdehyde but also protected against ethanol-induced decrease of reduced glutathione, gluthione reducatse, glutathione peroxidase, catalase and superoxide dismutase activities. Therefore, they can be utilized as a health functional food or new drug candidate for fatty liver and hepatotoxicity which was induced by chronic alcohol consumption.

Transcriptome profiling and identification of functional genes involved in H2S response in grapevine tissue cultured plantlets

  • Hydrogen sulfide ($H_2S$), a small bioactive gas, has been proved functioning in plant growth and development as well as alleviation of abiotic stresses, which including promoting seed germination, accelerating embryonic root growth, regulating flower senescence, inducing stomatal closure, and defending drought, heat, heavy metals and osmotic stresses etc. However, the molecular functioning mechanism of $H_2S$ was still unclear. The primary objective of this research was to analyze the transcriptional differences and functional genes involved in the $H_2S$ responses. In details, 4-week-old plantlets in tissue culture of grapevine (Vitis vinifera L.) cultivar 'Zuoyouhong' were sprayed with 0.1 mM NaHS for 12 h, and then transcriptome sequencing and qRT-PCR analysis were used to study the transcriptional differences and functional genes involved in the $H_2S$ responses. Our results indicated that 650 genes were differentially expressed after $H_2S$ treatment, in which 224 genes were up-regulated and 426 genes were down-regulated. The GO enrichment analysis and KEGG enrichment analysis results indicated that the up-regulated genes after $H_2S$ treatment focused on carbon metabolism, biosynthesis of amino acids, and glycolysis/gluconeogenesis, and the down-regulated genes were mainly in metabolic pathways, biosynthesis of secondary metabolites, and plant hormone signal transduction. Analyzing the transcription factor coding genes in details, it was indicated that 10 AP2/EREBPs, 5 NACs, 3 WRKYs, 3 MYBs, and 2 bHLHs etc. transcription factor coding genes were up-regulated, while 4 MYBs, 3 OFPs, 3 bHLHs, 2 AP2/EREBPs, 2 HBs etc. transcription factor coding genes were down-regulated. Taken together, $H_2S$ increased the productions in secondary metabolites and a variety of defensive compounds to improve plant development and abiotic resistance, and extend fruits postharvest shelf life by regulating the expression of AP2/EREBPs, WRKYs, MYBs, CABs, GRIP22, FERRITINs, TPSs, UGTs, and GHs etc.

Entelon150® (Vitis vinifera Seed Extract) Attenuates Degenerative Changes in Intravascular Valve Prostheses in Rabbits

  • Background and Objectives: The therapeutic strategy for inflammation and degenerative calcification is of utmost importance for bioprosthetic heart valve (BHV) implanted patients. The purpose of this study was to compare the anti-inflammatory and anti-calcification effects of Entelon150® (grape seed extract), losartan, and rosuvastatin, in a rabbit model of intravascular BHV leaflet implantation in bovine pericardium. Methods: A total of 28 rabbits were implanted with BHV leaflet in the external jugular veins. The Entelon150® group was administered 7.7 mg/kg Entelon150® twice daily for 6 weeks after surgery. The losartan and rosuvastatin groups received 5.14 mg/kg and 1 mg/kg, respectively, once per day. The control group received 1 ml of saline once daily. And then, calcium concentration was measured in the implanted BHV, and histological and molecular analyses were performed on the surrounding tissues. Results: The calcium content of the implanted tissue in the Entelon150® group (0.013±0.004 mg/g) was lower than that in the control group (0.066±0.039 mg/g) (p=0.008). The losartan (0.024±0.016 mg/g, p=0.032) and rosuvastatin (0.022±0.011 mg/g, p=0.032) groups had lower calcium content than the control group, and higher tendency than the Entelon150® group. Immunohistochemistry revealed that the expressions of bone morphogenic protein 2 (BMP2), S-100, and angiotensin II type 1 receptor in the Entelon150® group showed lower tendency than those in the control group. The protein expression levels of BMP2 were reduced in the Entelon150® group compared with those in the control group. Conclusions: Entelon150® exhibited a significant effect, similar to other drugs, in reducing calcification and inflammation in the intravascular bovine pericardium.

Rhizopus stolonifer에 의한 포도 무름병 (Rhizopus Soft Rot on Grape Caused by Rhizopus stolonifer in Korea)

  • 2003년부터 2006년까지 4년 동안 경남농업기술원 과수포장과 진주시 농산물 도매시장에서 유통 판매중인 포도에서 이상증상이 발생하였다. 병징은 상처나 열과된 과실이 수침상으로 물러지면서 빠르게 부패되었다. 균총은 처음 흰색에서 연한 갈색으로 되며 검은색의 포자낭을 많이 형성한다. 균사생육 적온은 $25^{\circ}C$였다. 포자낭경은 처음 흰색에서 회색을 나타내며 크기는 $850{\sim}1,800{\mu}m$이며 폭은 $12{\sim}23{\mu}m$이었다. 포자낭은 처음에 흰색에서 검은색으로 되며 모양은 반구형으로 크기는 $80{\sim}190{\mu}m$이었다. 주축의 모양은 반구형으로 크기는 $70{\sim}95{\mu}m$이었다. 포자낭포자는 담갈색으로 단포이며 구형 또는 타원형으로 불규칙한 것이 많으며 크기는 $8{\sim}22{\times}6{\sim}10{\mu}m$이었다. 이상과 같이 병원균의 균학적 특징과 병원성을 검정한 결과, 본 병해를 Rhizopus stolonifer (Ehrenberg ex. Fr.) Lind에 의한 포도 무름병으로 명명할 것을 제안한다.

$SO^2$ 발생 패드처리가 포도 과실의 저장성에 미치는 영향 (Influence of $SO^2$ generating pad treatment on storage of grape berries)

  • The occurrence of grape berry shattering and decay is influenced by the pre- and/or postharvest infection of decay organisms and pest. Postharvest infection is associated with physical damage due to rough handling. To control quality loss during storage of table grapes, the effects of slow releasing $SO^2$ pad on storability of 'Rosario Bianco' (Vitis vinifera) and 'Campbell Early' (Vitis labruscana) grapes were examined, respectively. The $SO^2$ concentration in package of tables grapes reached to at about 40 $ug{\cdot}L^{-1}$ after 40 days of treatment and remained more than 80 days above 15 $ug{\cdot}L^{-1}$. Decay was found in untreated 'Rosario Bianco' and 'Campbell Early' grapes at each for 44 and 85 days after storage, but not in $SO^2$ pad treated grapes. The storage potential of grape berries at $0^{\circ}C$ increased 2~3 times depending on cultivar by $SO^2$ pad treatment. The application of $SO^2$ pad is confirmed to be effective on the increase of market potential with minimizing quality loss such as berry shattering and decay.

다양한 과일나무에서 유래된 추출물의 농업해충 및 저장물해충에 대한 살충활성 (Toxicity of various fruit tree extracts to five agricultural and four stored-product anthropod pests)

  • 17과일의 43가지 부위의 메탄을 추출물을 대상으로 5종의 주요 농업해충 및 4종의 저장물 해충에 대한 살충효과를 조사한 효과는 과일류의 종류 및 해충의 종류에 따라 커다란 차이를 보였다. 5,000 ppm 농도에서 80% 이상의 살충효과를 나타내는 10종의 시료를 2,500 ppm에서 살충효과를 검정하였다. 모과와 석류씨가 벼멸구, 포도씨가 복숭아혹진딧물, 유자, 자몽, 메론, 감, 사과, 복숭아, 포도씨가 배추좀나방, 모과씨가 담배거세미나방, 모과와 포도 추출물이 점박이응애에 대하여 80% 이상의 살충효과를 나타냈다. 4종의 저장물해충에 대한 살충효과는 50 ppm의 농도로 처리할 때 모과 및 포도 추출물이 쌀바구미, 모과, 유자, 감 및 포도 추출물이 팥바구미에 대하여 80% 이상의 살충효과를 나타냈다. 그러나 화랑곡나방와 궐련벌레에 대하여는 사용된 과일류의 추출물이 활성을 나타내지 않았다. 이상의 결과로부터 복숭아혹진딧물, 벼멸구, 배추좀나방 담배거세미나방 및 점박이응애에 높은 방제효과를 보인 상기과일류 추출물들은 농업해충 방제제로서 사용 가능성이 예상되었으며, 또한 쌀바구미와 팥바구미에 강한 살충효과를 보인 추출물은 저장물해충방제에 이용할 수 있을 것으로 기대된다.
