Immunosuppressive effect of Cryptosporidium bnileyi infection on vaccination against avian infectious bronchitis in chicks (닭에 있어서 닭와포자충 감염이 닭전염성기관지염 예방접종에 대한 면역억제 효과)

  • Two-day-old commercial chicks were inoculated orally with 2 × 106 oocysts of Cwptosporinium bailevi and vaccinated with 103.5 EID50/head of a commercially available avian infectious bronchitis (IB) live virus vaccine at 4 and 14 days following inoculation. Chicks infected with C. baileyi were shown to have an immunosuppressive effect on IB virus. It is concluded that infection with the protozoon in early life may increase their susceptibility to IB.

Risk factors of African swine fever virus in suspected infected pigs in smallholder farming systems in South-Kivu province, Democratic Republic of Congo

  • Background: African swine fever (ASF) is an infectious viral disease of domestic pigs that presents as a hemorrhagic fever, and for which no effective vaccine is available. The disease has a serious negative social and economic impact on pig keepers. There is limited information on the potential risk factors responsible for the spread of ASF in South Kivu. Objective: The aim of this study was to determine the potential risk factors associated with ASF infection in suspected ASF virus (ASFV)-infected pigs. Methods: We sampled whole blood from 391 pigs. Additionally, 300 pig farmers were interviewed using a structured questionnaire. Viral DNA was detected by using the real-time polymerase chain reaction technique. Results: The majority of pigs sampled, 78% (95% confidence interval [CI], 74.4-82.6), were of local breeds. Over half, 60.4% (95% CI, 55.5-65.2), were female, and most of them, 90.5% (95% CI, 87.6-93.4), were adult pigs (> 1 year old). Viral DNA was detected in 72 of the 391 sampled pigs, indicating an overall infection rate of 18.4% (95% CI, 14.5-22.4). Multivariable logistic regression analysis revealed several risk factors positively associated with ASFV infection: feeding with swill in pen (odds ratio [OR], 3.8; 95% CI, 2.12-6.77); mixed ages of pigs in the same pen (OR, 3.3; 95% CI, 1.99-5.57); introduction of new animals to the farm (OR, 5.4; 95% CI, 1.91-15.28). The risk factors that were negatively (protective) correlated with ASFV positivity were the presence of male animals and the use of an in-pen breeding system. Conclusion: Local pig farmers should be encouraged to adopt proper husbandry and feeding practices in order to increase the number of ASF-free farms.

  • In this study, we investigated safety and efficacy of a low temperature immunization protocol with NNV in red spotted grouper, Epinephelus akaara and long tooth grouper, Epinephelus bruneus and seven band grouper, Hyporthodus septemfasciatus. Further, growth rate between immunized and naïve fish was evaluated during the experiment to check side effect of immunization. Three grouper species were immunized by immersion method with live NNV at 105.0 TCID50/mL at 16.5℃ for 30 min and reared for 120 days at natural sea water temperature. To evaluate growth rate, total length and wet weight was measured 7 times after immunization. Immunized three grouper species were challenged by intramuscular inoculation with NNV at 104.2 TCID50/100 µL/fish. Immunization at low temperature with live NNV did not show any clinical symptoms of infection, mortality and inhibition of growth. After challenge, cumulative mortality of naïve seven band grouper, red spotted grouper, long tooth grouper were 45, 10, 20 %, respectively. However no mortality was observed at immunized groupers. Thus, it was demonstrated that immunization at low temperature with live NNV are able to protect three different species of groupers without inhibition of growth.

Studies on the Immunization Against New-castle Disease I. A Survey on the Immune Status of Poultry Flocks Againt New-castle Disease in the Epizootic Area (뉴캣슬병 면역에 대한 검토 I. 뉴캣슬병 유행지역에서의 계군면역에 대한 실태조사)

  • A study was conducted to determine whether the vaccination programs for the control of New castle disease (ND) would affect the immune status of birds against the disease. Twenty-six poultry flocks in the epizootic area of ND were surveyed to investigate the level of urn antibody against ND virus and the programs used for the vaccination of birds. The mortality rates and vaccination status of birds during the epizootic of ND were also examined in the infected poultry flocks to elucidate the immune effect against the epizootic with particular regard to various vaccination programs used in the field. The results obtained are summerized as follows: 1. Of 26 poultry flocks investigated, 22 flocks were immunized with live and killed vaccines, their haemagglutination-inhibition (HI) antibody titer being 146 and 50, respectively. Among 22 farms using live and killed vaccines two flocks which showed the lowest HI titer of 10 and 23 had the disease later on. However, no cases of ND were recorded in the killed vaccine groups, although their HI titers were in the range of 38 to 64. 2. Of 14 infected flecks, one flock was not vaccmated against ND while all the remaining 13 flocks were vaccinated against the disease, of which 8 flocks were vaccinated with live vaccine only and the other 5 flocks with both live and killed vaccines. The mortality rate of 8 infected flocks which had been vaccinated with only live vaccine was as high as 32.5% while that of 5 flocks with both live and killed vaccines was as low as 5.1%. 3. It was found that in majority of flocks B$_1$vaccine was used via drinking water and in a few flocks the vaccine was administered via intramuscular route or method of dipping mouth, nose and eye of birds into vaccine solution.

Distribution of Human Rotavirus Genotypes in a Tertiary Hospital, Seoul, Korea During 2009-2013 (2009년부터 2013년까지 서울의 일개 대학병원에서 동정된 로타바이러스 유전형의 분포)

  • Purpose: Group A rotavirus (RV) is most common etiologic agent of acute gastroenteritis (AGE) in children worldwide. Recently, vaccination has been introduced in several countries to reduce the disease burden caused by RV infections, but continuous surveillance of RV strains is necessary to detect the emergence of potential variants induced by vaccine-immune pressure. This study aimed to investigate the changing pattern of RV genotypes in children with AGE, following the introduction of vaccination in Korea. Methods: Genotyping of RVs by RT-PCR on the basis of VP7 and VP4 gene segment sequence was carried out on 201 rotavirus-positive stool samples, from children hospitalized with AGE between August 2009 and June 2013. We have directly sequenced PCR products and analyzed the phylogenetic tree. Results: The most prevalent G genotype was G9 (33.3%), followed by G1 (22.4%), G3 (15.9%), G2 (6.0%), G4 (3.0%), G10 (1.5%), and mixed G-type (15.4%), with some nontypeable cases (2.5%). The detected P genotypes were P[4] (45.3%), P[8] (43.8%), mixed P-type (10.4 %), and P[2] (0.5%). The G9P[4] genotype was predominantly observed in hospitalized cases in Seoul in 2010/2011, however G1P[8] has been re-emerged as the predominant genotype in the following season (P =0.004). Conclusions: It seems that the periodic fluctuation in predominance of the G1, G3, and G9 strains occurred in Korea during 2009-2013, following the introduction of RV vaccination.

Introduction of Hog Cholera Virus Gene into Potato Plants by Agrobacterium-mediated Transformation and the Analysis of Its Expression

  • The HCV gene was expressed in potato plants under the control of the constitutive CaMV 355 promoter or tuber-specific patatin promoter. Solanum tuberosum plants carrying a plant expression vector harboring the encoding region of HCV gene were generated by Agrobacterium tumefaciens-mediated in vitro transformation methods. The presence of HCV gene in the plant genome was detected by PCR and DNA hybridization experiments. We obtained the 5 lines of transgenic potato with the pMBPHCV construct and 4 lines of transgenic potato with the pATHCV construct. The HCV transgenic stably integrated into the potato genome, as well as their transcription. HCV mRNA was identified in leaf and tuber tissues of transgenic plants by Northern blot analysis. The transgenic potato plants produced the expected transcript, and the corresponding HCV protein accumulated in individual transgenic plants.

A Case of Thickened Gallbladder Wall and Pneumonia in a Child with Infectious Mononucleosis (담낭염과 폐렴을 동반한 전염성 단핵구증 1례)

  • Acute infectious mononucleosis, caused by Epstein-Barr virus(EBV), is a self limited lymphoproliferative illness that is common in adolescents and young adults. It shows many complications in multiple organ systems, but the hepatobiliary and the respiratory complication is uncommon. We report a case with thickened gallbladder wall and pneumonia as complications of acute infectious mononucleosis in a child. Also the related literature were reviewed. A 4 year old boy presented with a history of high fever, cough, and abdominal distension for 20days. Physical Examination revealed audible crackles in whole lung field and gross hepatomegaly. Chest X-ray showed pneumonia and liver function tests were abnormal. Ultrasonography and computed tomography revealed a thickened gallbladder wall and hepatosplenomegaly. The diagnosis of primary Epstein-Barr viral infection was eventually made by specific serologic tests. The patients's fever subsided 6 weeks later and pneumonia was recovered around this time. Liver function tests returned near normal 2 months later and ultrasonography of gallbladder was normal at this time.

  • Varicella-zoster virus is a human herpesvirus that causes chickenpox (varicella), becomes latent in cranial nerve and dorsal root ganglia, and frequently reactivates to produce shingles (zoster) and postherpetic neuralgia. Varicella zoster meningitis is a rare complication after varicella zoster infection. It usually affects a patient of immunocompromised or impaired cellular immunity, is rare in a immunocompetent child. We report two cases of aseptic meningitis in association with varicella zoster, not having any complication in the immunocompetent children.

Correlation between Disease Prevalence and Production Performance in Korean Swine Farms (양돈 생산성에 따른 주요 질병 분포 조사)

  • Currently, various diseases reside in Korean swine farms and affect production performance of the farms greatly. These damages from disease are further aggravated by the concurrent infection of other disease. In this study, y investigating the distribution of porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2), Salmonella spp. in farms, correlation between the damage and the prevalence of disease was analyzed. Ten selected Korean swine farms that uses PCV2 vaccine were tested for presence of antibody and antigen of PRRSV, PCV2, Salmonella spp. per ages of pigs, 4weeks, 7weeks, 11weeks and 17weeks, respectively. The results were analyzed by dividing the farms in to groups with MSY above 19, and that with MSY below 19. Then calculating the distribution of disease each ages of pigs. Farms with MSY below 19 showed high prevalence of disease by PRRSV, PCV2 and Salmonella spp.. In this group, the detection rate of PCV2 and Salmonella spp. was increased by the activation/viremia of PRRSV in the young ages of pigs. The results are proved that the correlation between disease prevalence and production performance in Korean swine farms were very significant. The prevalence of PRRSV is more important index which influence to the productivity in current prevalence of diseases.

Isolation and identification of canine parvovirus type 2b in Korean dogs

  • Canine parvovirus (CPV) is a major diarrhea-causing agent in puppies. Since CPV type 2 (CPV-2) emerged in 1978, new antigenic variants including CPV-2a, CPV-2b, and CPV-2c have been identified in many countries. Two puppies died suddenly at a veterinary clinic in Gyeonggi province, South Korea. Two viruses were isolated in A72 cells, confirmed as CPV strains based on a CPV rapid kit and an indirect fluorescence test and designated QIACP1403 and QIACP1404. The nucleotide sequences of complete VP2 genes of QIACP1403 and QIACP1404 were determined, and the corresponding amino acid sequences were deduced. Molecular analyses revealed that the QIACP1403 and QIACP1404 isolates were type CPV-2b. Several mutated amino acids were detected on VP2 gene residues of the two isolates. Phylogenetic analyses showed that the two isolates were most closely related to strain CPV-BM11, which was isolated from Chinese dogs in 2011. Our results suggest that these isolates may be a candidate for a vaccine to prevent CPV infection in dogs after conducting passages of the isolates in an in vitro culture system.
