The Effect of Lactobacillus plantarumCLP-1 on the Swine Viruses (Lactobacillus plantarumCLP-1이 돼지바이러스에 미치는 효과)

  • To isolate Lactic acid bacteria for animals, we have screened from Kim-chi, swine intestine, swine feces, and dairy products by random selection and anti-viral, antipathogenic bacteria test. Among them, CLP-1 shown that inhibitory effect against rotavirus, porcine epidemic diarrhea (PED) virus, Salmonella sp, and E.coli. By examining biological property, API-ZYM and identified Lactobacillus plantarum by 16S rDNAgene sequence. CLP-1 determined resistance to low pH and bile salt. Futhermore, the cell body of CLP-1 adhered to the intestinal epithelium tissue of swine and Caco-2 cell. CLP-1 was examined on cell immune system modulating activity in vitro. The whole cell and cell culture supernatant was increasing of interferon-${\beta}$ activity. And then, CLP-1 increased prevention effect by Salmonella enteritidis infection in SPF chickens. And we determined similar result in pigs.

  • Partially purified water-soluble extract mixture from Ricini and Coptidis (named as RIC) showed to be a potent inhibitor of human immunodeficiency virus-1 (HIV-1) replication. RIC was evaluated for in vitro anti-HIV activity using SupTl and H9 cells infected by a recombinant virus (pSVCAT) containing chloramphenicol acetyltransferase (CAT) gene substituted for nef gene in the HIV-1 genome. RIC inhibited syncytiaformation of SupTl cells with a half maximal effective concentration, $IC_{50}$/, of 2.5 $\mu\textrm{g}$/mι and showed marked inhibition of CAT activity in the infected H9 cells and also suppressed reverse transcriptase (RT) activity in the supernatant of the infected H9 culture. However, RIC did not inhibit the activity of reverse transcriptase directly when it was mixed with the enzyme or with viral particles. Berberine, one of components of RIC, also showed similar anti-HIV activity as RIC did. The data suggest that there are active ingredients which mediate anti-HIV activity in RIC.

The rats were supplied by 2 different production colonies located in the eastern and western areas of the US. Disinfection effect of corona discharged plasma water on fish pathogens (코로나 방전 플라즈마 처리수에 의한 어류 병원체 소독 효과)

  • Fish culture is constantly threatened by various infectious diseases which are largely transmitted by water. Plasma technology is being used to sterilize polluted water in many industries. In this study, two bacterial pathogens Aeromonas salmonicida and Streptococcus iniae, and a virus (viral hemorrhagic septicemia virus, VHSV) were subjected to plasma water that was produced by a corona discharge system. Growth of A. salmonicida was greatly inhibited from 105.61 CFU/ml in positive control to 103.51 CFU/ml in treated group by only 60 sec contact with plasma water. Similarly, S. iniae was inhibited from 105.85 CFU/ml to 103.40 CFU/ml. VHSV titer also decreased from 104.1 TCID50/ml to 101.45 TCID50/ml by the same treatment. Activation of water by the plasma was confirmed by the existence of ozone in the plasma water. These results suggest that plasma water could efficiently disinfect fish pathogens, possibly by the action of reactive oxygen species contained in the plasma water.

Comparison for Hepatitis B Knowledge, Self Care Practice and Quality of Life according to the Disease Activity among Patients with the Hepatitis B Virus (B형간염 바이러스 보유 환자의 질환 활성도에 따른 B형간염 지식, 자가간호수행 및 삶의 질 비교)

  • Purpose: The hepatitis B virus is a major cause of chronic liver disease. The clinical guidelines recommend that inactive chronic hepatitis (ICH) patients also check their liver function every 6 to 12 months and manage the potential risks. This study compared the hepatitis B knowledge, self-care practice, and quality of life in patients with HBV according to the disease activity. Methods: This study was conducted in a university hospital and surveyed on 65 ICH patients and 68 progressive chronic liver disease (PCLD) patients from November in 2012 to September in 2013. Results: The knowledge of hepatitis B was lower in the group of a lately perceived HBV infection and ICH. Self-care practice was lower in the male and the patients group with a perceived HBV infection within 5 years. The "taking regular liver function test" score was lower in the ICH. Eight out of 12 Liver Disease Quality of Life instrument (LDQOL) subscales were lower in PCLD. Conclusion: The hepatitis B knowledge and self-care practice are relatively lacking in ICH and the patients group with a perceived HBV infection within 5 years. More effective education programs will be necessary to enhance the hepatitis B knowledge and self-care for patients with HBV and even for ICH.

Detection of Coinfection and Persistent Infection of Adenovirus and Varicella-Zoster Virus in Synovial Fluids from Synovitis Patients by Nested-PCR (Nested 중합효소연쇄반응을 이용하여 활막염 환자의 관절액으로부터 아데노바이러스와 대상포진바이러스의 이중감염과 지속감염의 검출)

  • The etiology of rheumatic arthritis (RA) is associated with a number of genetic and environmental factors, but is not definitively elucidated. Recently, more attention has been paid to the possibility of microbial etiology in the pathogenesis of RA, because many different infectious agents have been reported to precede the onset or exacerbation of RA. Adenovirus (ADV) may be one cause of persistent or recurrent inflammatory arthritis. Varicella zoster virus (VZV) arthritis is detected frequently in RA patients treated with low dose methotrexate. The demonstration of simultaneous presence of both viral agents of specific viral nucleic acid in synovial fluids from synovitis patients would provide more direct evidence for arthritis etiological relationship, but there are no confirmed results. Therefore, we studied the ability of adenovirus and VZV to establish coinfection and persistent infection in synovial fluid from synovitis patients. The presence of viral agents in the synovial fluid demonstrated by isolation of cell culture, enzyme immunoassay and nested-PCR. The synovial fluids were also investgated for the presence of viral nucleic acid by nested-PCR using specific primer. ADV produced 220 bp and VZV produced 447 bp by each nested-PCR with specific primers. We detected 4/6 cases (66.7%) with persistent infection of ADV and 5/6 cases (83.3%) of VZV with 13 synovial fluids (between 7 to 52 day intervals) from synovitis patients by monoclonal ErA and nested-PCR. 21/28 cases (75%) with coinfection of adenovirus and VZV with synovial fluids from synovitis patients by nested-PCR. ADV and VZV coinfection and persistent infection of synovial fluids may provide a chronic antigenic stimuli to the immune system therefore provoking a continuing inflammatory response and caused the possibility of synovitis and arthritis.

  • Nuclear polyhedrosis virus was successfully infected the continuous Sf cell line. At 12hrs post-infectio(P.I), the cell lost the motility and the nuclei of the cells were hypertrophied. At 24hrs P.I, the cells were somewhat abnormal form and PIB formation was observed. At 48hrs, the PIBs formed in all cells. PIBs in the nuclei were released in the culture media at 72hrs P.I. By the observation of NPV morphogenesis by electron microscopy at 13hrs P. I, the virogenic stroma formed in the nucleus, and nucleocapsids formed. At 48hrs P.I, many nucleocapsids were bundled and then occluded in PIB, and PIBs were matured. PIB shapes were mostly tetragonal and a polyhedron was about $3{\sim}10{\mu}m$ in size. Virions were rod shape. nucleocapsids ranging in size $30{\sim}40{\times}300{\sim}400nm$.

Induction of Systemic Resistance against Cucumber mosaic virus in Arabidopsis thaliana by Trichoderma asperellum SKT-1

  • Trichoderma asperellum SKT-1 is a microbial pesticide that is very effective against various diseases. Our study was undertaken to evaluate T. asperellum SKT-1 for induction of resistance against yellow strain of Cucumber mosaic virus (CMV-Y) in Arabidopsis plants. Disease severity was rated at 2 weeks post inoculation (WPI). CMV titre in Arabidopsis leaves was determined by indirect enzyme-linked immunosorbent assay (ELISA) at 2 WPI. Our results demonstrated that among all Arabidopsis plants treated with barley grain inoculum (BGI) of SKT-1 NahG and npr1 plants showed no significant reduction in disease severity and CMV titre as compared with control plants. In contrast, disease severity and CMV titre were significantly reduced in all Arabidopsis plants treated with culture filtrate (CF) of SKT-1 as compared with control plants. RT-PCR results showed increased expression levels of SA-inducible genes, but not JA/ET-inducible genes, in leaves of BGI treated plants. Moreover, expression levels of SA- and JA/ET-inducible genes were increased in leaves of CF treated plants. In conclusion, BGI treatment induced systemic resistance against CMV through SA signaling cascade in Arabidopsis plants. While, treatment with CF of SKT-1 mediated the expression of a majority of the various pathogen related genes, which led to the increased defense mechanism against CMV infection.

  • Leaf segments of the potato (Solanum tuberosum L.) genotypes, ND860-2, Norchip, Russet Norkotah, Goldrush, and Norqueen Russet were transformed with the coat protein gene of potato virus Y (PVY). The white-skinned genotypes, ND860-2 and Norchip, were easily transformed and regenerated into shoots, whereas the three russet-skinned genotypes had low frequencies of regeneration. Transformed shoots were generally recovered in four to six weeks. Antibody to PVY coat protein detected a single band of 30 kD in western blots of transgenic plants. Transformed plants had a normal phenotype in the greenhouse and many showed a delayed buildup of PVY following inoculation. Several transgenic lines had negative ELISA readings 85 days after inoculation. Transgenic lines which did not show detectable levels of PVY antigen will be further tested for resistance to PVY.

Enteric Virus Detection from Environmental Sample by Oligonucleotide DNA Chip (올리고뉴클레오티드 DNA Chip을 이용한 환경시료에서의 장관계바이러스 검출)

  • The usefulness of oligonucleotide DNA chip was evaluated for detection and primary level identification of major waterborne viruses in environmental samples. The enteric waterborne viruses included enterovirus, adenovirus, and rotavirus. Total intracellular RNA of 10 BGM cell plates showing virus-specific cytopathic effects was extracted at the third day after inoculation. The intracellular RNA was then subjected to either enterovirus-specific RT-PCR followed by sequencing analysis, or the DNA chip. Seven out of 10 positive samples in cell culture were positive but the other three sample were turned out to be negative by both RT-PCR and DNA chip analyses. Nucleotide sequencing results and the DNA chip hybridization results of the RT-PCR product were in complete agreement in the identification of the 7 positive samples as enteroviruses. Using the DNA chip, it took only 3∼4 hr to complete detection and primary level identification of target viruses and additional procedures such as gel electrophoresis or nucleotide sequencing were not necessary. We believe that the DNA chip system can be employed as a highly effective and new detection methodology for environmental viruses.
