Analysis of Vicamine Using High Performance Liquid Chromatography and Antioxidant Activity of Vincaminor Extract (High performance liquid chromatography를 이용한 빈카민 분석 및 빈카마이너의 항산화능 측정)

  • Vincamine, one of the major indole alkaloids in vincaminor (Vinca minor L.) is commonly used for treating cerebrovascular diseases. The antioxidant activity of vincaminor extracts and vincamine were measured by 1.1-diphenyl-2-picrylhydrazyl (DPPH) and lipid malonaldehyde (MA) assay. Vincaminor leaves were pulverized and extracted with various solvents such as water, methanol, and ethanol. The antioxidant activities of the extracts varied in accordance with solvents and assays. In DPPH assay, the water extract showed the highest antioxidant activity. In lipid MA assay, However, the ethanol extract inhibited MA formation from cod liver oil by 82% at the level of 5,000 ${\mu}g/mL$. Vincamine in the extract was analyzed by high-performance liquid chromatogram and the concentration of vincamine was 0.419$\pm$0.005 ${\mu}g/mL$.

Antimicrobial and Immunological activities of Vinca minor Extracts (빈카 마이너 추출물의 항균 및 면역활성 연구)

  • Vinca alkaloid compounds including vincamine from Vinca minor L. have been extracted by ethanol and hot water extraction methods. Antimicrobial properties of those extracts have been investigated against dandruff causing microorganism Malassezia furfur, yeast, Gram positive and negative bacteria. Vincamine standard and ethanol extract showed no sign of antimicrobial activity, whereas hot-water extract had the activity against M. furfur. Furthermore hot water extract had antimicrobial activity against Gram positive Bacillus sp. and Gram negative Escherichia coli. Cytotoxic properties of those extracts have also been investigated with HaCaT cell (human keratinocyte), HT-29 cell (human colorectal adenocarcinoma cell) and Raw cell, showing no significant cytotoxic effects. We also measured the ROS using dichlorofluorescein diacetate (DCFDA), a popular fluorescence-based probe for reactive oxygen species detection. The result showed the increasement of reactive oxygen species formation (20%) in HaCaT and HT-29 cell lines.

Antioxidant Effects and Spectrophotometry Analysis of Alkaloids from Vinca minor (빈카 마이너 추출물에 함유된 알칼로이드들의 분광학적 분석 및 항산화 효능 연구)

  • Alkaloids of Vinca minor L. were characterized by HPLC equipped with the photo diode array detector and compared their UV/vis absorption spectra with an index material such as vincamine. UV/vis absorption spectra of more than 9 compounds belong to various classes such as flavonoids were shown and characterized. Extracts with ethanol or hot water were tested against the DPPH (2,2-diphenyl-1-picrylhydrazyl) free-radical to investigate their antioxidant activity. Based on the results, those extracts show about 14-15% of antioxidant activity of quercetine and vitamin C used as standards.

Biological Effects of Vinca minor extract; Tyrosinase inhibition, stimulation of ROS generation and increasement of cell migration activity in keratinocytes

  • Vinca alkaloids from plant Vinca minor have been investigated for their effects of tyrosinase inhibition, stimulation of ROS generation and increasement of cell migration activity. The methanolic crude extract and the water-soluble fraction exhibited $IC_{50}$ value of 3.1 mg/mL and 2.1 mg/mL. Vinca minor extract treatment significantly increased ROS levels in HaCaT cells, in a concentration-dependent manner. Treatments of Vinca minor extract led to increase wound closure when compared with non-treatment. Low dose (0.1% or 0.3%) of extracts have not significantly affected, compared with that in controls. By contrast, 0.5% extract have dramatic effect on wound healing activity of keratinocytes. Effects of Vinca minor extract in a filter-based cell mobility assay appear similar to that of wound closure assay, which suggests that the Vinca minor extract have wound healing effects on skin.
