Occurrence of Lepidopteran Insect Pests and Injury Aspects in Adzuki Bean Fields (팥 포장에서 나비목 해충의 발생과 피해 양상)

  • Three lepidopteran insect pests of the legume pod borer, Maruca vitrata (Lepidoptera: Pyralidae), the soybean pod worm, Matsumuraeses phaseoli (Lepidoptera: Tortricidae) and an Ostrinia spp. (Lepidoptera: Pyralidae) attacking adzuki bean, Vigna angularis, were confirmed as the major insect pests during the reproductive developmental stage of adzuki bean, and M. vitrata existed dominantly. Almost all plants in the adzuki bean field were injured by at least one of the three species, and $15{\sim}60%$ of reproductive organs were injured. Unbloomed inflorescence occurred continuously through the reproductive stage of adzuki bean. While the flower was many at the early stage and its number soon decreased, the pod was few at the early stage, but its number increased soon and maintained at a constant level. The results suggested that the adzuki bean injured by insect pests compensate the flower loss by developing new inflorescence, but the compensation do not cause the development of new pods. While M. vitrata and M. phaseoli were observed in flowers, pods and stem mainly during the first half of reproductive stage of adzuki bean, Ostrinia spp. was observed only in pods and stem during the second half. In addition, while all instars of larvae of M. vitrata were observed, larvae between the third and fifth instars for M. pahseoli and Ostrinia spp. were observed.

Studies on tile Resources for the Artificial Diet and Feeding Response of the Silkworm, Bombyx mori (가잠의 인공사료자원과 섭식성에 관한 연구)

  • Forty-nine plant species as additives to silkworm artificial diet and 5 species as cellulose sources for artificial diet were screened for their economic values as feed-resources for the silkworm. Feeding response to artificial diet was tested on 82 silkworm strains. The effect of rearing conditions on feeding response and enzyme activities in the silkworm was investigated. The results were summarized as follows. 1. Seven species out of 49, Vigna sinensis ENDL, Ipomoea vatatas Lamarck, Cyperus anuricus Var. Laxus, Alnus japonica Stendel, Trifolium repens L, Prunus serrulata Lindley. Var, Glycine max L increased feeding response, compared with the basic formula of artificial diet. 2. The economic values of Vigna sinensis ENDL, Ipomoea vatatas Lamarck, Cyperus anuricus Var. Laxus, Ainus japonica Stendel, Cassia tera L, Erigeron canedensis L as feed-resources for artificiale diet were recognized, through feeding experiment during the entire larval stage. 3. Mulberry cellulose showed the best results in rearing and cocoon characteristics. 4. The extent of feeding response varied according to strains and varieties. Varieties in japanese strains showed higher feeding response than those in chinese and european varieties, with considerable variations among a varieties in strains. 5. The begining of 4th instar seems to be a proper time to convert from mulberry to artificial diet, or artificial diet to mulberry, however the middle of 3rd instar seems acceptable. 6. The optimum temperature for artificial diet rearing is 30$^{\circ}C$ during the period of 1st-3rd instar and 28$^{\circ}C$ for 4th-5th instar. 7. Electrophoretic isozyme patterns of esterase and acid phosphatase on agarose gel, as affected by strain. rearing temperature and feed-resources, were observed as follow. (1) Isozyme patterns of mid-gut esterase varied, depending on instar. One or two more isozyme bands were observed in the larvae than feed on the mulberry fed for the artificial diet. (2) A strain, chinese-15 with a higher feeding response, had 1∼2 more bands than chinese-60 with a lower feeding response. (3) Five bands of mid-gut esterase in 3rd and 4th instar larvae reared at 28$^{\circ}C$. and 4 for 3rd instar and 6∼7 for 4th instar larvae at 35$^{\circ}C$ were observed. (4) No similar esterase bands could be found among mid-gut, blood and silkgland. There are five esterase bands in the midgut, one in blood and three in silkgland. (5) There was rather small digerence in acid phosphatase types of mid-gut and blood according to varieties and rearing temperature. No active band was shown in silkgland. In midgut, there was one acid phosphatase band at 3rd instar, two at 4th instar and three at 5th instar. In blood, one active band at 3rd or 4th instar and three bands at 5th inster wire detected.

Studies on the Efficiency of the Artificial Diet for the Silkworm, Bombyx mori L. (누에인공사료의 사료효율증진에 관한 연구)

  • Silkworm(Bombix mori) were reared with modified artificial diets which were mixed with, as additives, leaf powders of Erigeron canadensis L., Cassia tora L., Cyperus anuricus Var.Laxus and Vigna Sinensis NEDL. The effects of additives on silkworm characteristisc of tested plants were summarized as follows ; 1. About 2-5% addition on dry weight base of leaf powders of E. canadensis, C. tora, C. anuricus or V.sinensis to the basic artificial diet promoted feeding response and digestion and resulted in good practical silkworm characteristics. The addition of V.sinensis and C.anuricus showed especially good effects. 2. The syneristic effect between different plant species was not recognized based on the feeding response and digestion of silkworm reared with various combinations of 2-4 different plant additives. 3. Electrophoretic zymograms of estrase, protease and phosphatase on haemolymph, intestine and silkgland were significantly different among treatments. In general, 1 or 2 more electrophoretic bands were detected when feeding response and digestion were promoted. 4. Contents of starch, crude fat, crude protein and inorganic base were apparently higher in the tested plants than in mulberry leaves. However, no volatile ingredent which is directly realted with feeding response was identified.

Characterization of Cucumber mosaic virus Isolated from Trifolium repens in Korea (국내 토끼풀에서 분리한 Cucumber mosaic virus의 특성)

  • A Cucumber mosaic virus (named CMV-Tr1) isolated from the white clover (Trifolium repens) showing mosaic and malformation that found in a pepper field. Cucumber mosaic virus was identified through confirmation with PT-PCR, PCR-restriction fragment length polymorphism, and sequence analysis of coat protein (CP) gene. CMV-Tr1 mosaic symptom on the upper leaves of five tobacco species including Nicotiana benthamiana, Cucumis sativus, Physalis angulata, and Solanum lycopersicon. In Chenopodium quinoa and Vigna unguiculata the isolate showed local lesions in inoculated leaves. CMV-Tr1 compared with CMV-As in the sequence identity of CP gene. CMV-Tr1 showed 98.9% and 99.5% homologies at nucleotide and amino acid levels, respectively. Phylogenetic analysis of the CP gene indicated that CMV-Tr1 belongs to the CMV subgroup IB base on the CP. To our knowledge, this is the first report of CMV in T. repens in Korea.

Can Maruca vitrata (Lepidoptera: Crambidae) Over-winter in Suwon Area? (콩명나방(Maruca vitrata)은 수원지방에서 월동할 수 있는가?)

  • Maruca vitrata is a main insect pest against crops of Vigna species (V. angularis and V. radiata) and Sesbania sesban in Fabaceae, but the life cycle of the insect is unclear in Korea. In order to know over-wintering possibility, its stage of the insect, over-wintering entry season, and the first adult emergence season of the next year, we investigated over-wintering rates of the insect in outdoor conditions in Suwon ($37^{\circ}16^{\prime}N$ $126^{\circ}59^{\prime}E$ 35ASL). In all colonies which the rearing for larvae, pupae and eggs started after mid-September, adults did not emerge within the year, and all insects died before next June. In several trials for over-wintering of larvae and pupae in soil, all insects died, too. Larvae stored during specified periods at low temperatures (10 and $13^{\circ}C$) did not emerge. From the results, it was postulated that M. vitrata does not have an adaptability to temperature below zero, and cannot over-winter in Suwon area.

Identification of Major Broad Leaved Weed Seedlings Based on Morphological Characteristics (주요 광엽잡초 유묘의 형태적 특성을 기초로 한 간이 식별법)

  • This study was conducted to develope identification method with major broad leaved weed seedlings. Characteristics of weed seedlings were investigated at two to four true leaf stage. Qualitative characteristics were more useful to identification of weed species than quantitative characteristics at the seedling stage. Many species were elliptic and broadly elliptic type in their cotyledon shape. Cotyledon shape of Cassia nomame and Calystegia sepium were broadly oblong. Four species were transverse broadly elliptic in their cotyledon shape. Rumex japonicus, Persicaria hydropiper, Euphorbia supina and Acalypha australis have three or more types in their cotyledon shape. Cotyledons of Vigna angularis var. nipponensis remained below ground after its germination. Many species were round in apex and were cuneate in base of cotyledon. Humulus japonicus and Polygonum aviculare were sessile in cotyledon base. The second true leaf shape of some species changed after first leaf stages. Glycine soja, V. angularis var. nipponensis and Kummerowia striata were broadly ovate, heart-shaped, orbicular or broadly obovate in their first true leaf, respectively, but the second true leaf shape of them were tripalmately compound. Aeschynomene indica and Cassia nomame were paripinnately compound leaf in their first and second true leaves. Margin of true leaves were entire in many species but six species including Xanthium occidentale were dentate in their true leaf margins. Margin of Euphorbia supina was entire in first true leaf and dentate in second true leaf. Margin of Chenopodium album and C. serotinum were entire in their first true leaf stage and erose in their second true leaf stage. The key for weed seedling identification of major weed species were made based on quantitative and qualitative morphological characteristics.

Characterization of Cucumver mosaic virus Isolated from Hydrangea macrophylla for. otaksa (Sieb. et Zucc) Wils. (수국에서 분리한 Cucumber mosaic virus의 특성)

  • An isolate of Cucumber mosaic cucumovirus(CMV) was isolated from Hydrangea macrophylla for. otaksa(Sieb. et Zucc. ) Wils. showing mosaic symptoms, and designated as Hm-CMV. Hm-CMV was characterized by the tests of host range, physical properties, serological properties, RNA and coat protein compositions, and reverse transcription and polymerase chain reaction (RT-PCR) analysis. Twelve species in 4 families were used in the host range test of Hm-CMV and could be differentiated from Y-CMV used as a control CMV by the ringspot and line pattern on inoculated leaves of several tobacco plants. Thevirus produced local lesions on inoculated leaves of Chenopodium amarticolor, C. quinoa and Vigna unguiculata. The physical properties of the virus were as follows; thermal inactivation point(TIP) was 60$\^{C}$, dilution end point (DEP) was 10$\^$-3/, and longevity in vitro (LIP) was 3∼4 days. Hm-CMV was serologically identical to Y-CMV. SDS-polyaciylamide gel electrophoresis(SDS-PAGE) showed one major protein band of about 28 kDa. In RNA or dsRNA analysis, Hm-CMV consisted of four RNA or dsRNA species, but satellite RNA was not detected. In RT-PCR using CMV-common primer and CMV subgroup I-specific primer, bothe amplified expected size of about 490 bp and 200 bp DNA fragments from Hm-CMV, respectively. Restriction enzyme analysis of the 490 bp RT-PCR products using EcoR I and Msp I showed that Hm-CMV belonged to CMV subgroup I. However, Hm-CMV could be differentiated from other CMV subgroup I isolates by RNA fingerprinting by arbitrarily primed polymerase chain reaction (RAP-PCR).

Fungicidal activities of leguminous seed extracts toward phytopathogenic fungi (콩과식물 종실 추출물의 살균활성)

  • Methanol extracts from 25 leguminous seeds were tested for their fungicidal activities toward six phytopathogenic fungi, using whole plant test in a greenhouse. The efficacy varied with both the plant pathogen and legume species used. At 5 mg/pot, potent fungicidal activities were produced from extracts of Cassia obtusifolia, Glycine max var. solitae, G. max var. yagkong, G. max var. hooktae, Phaseolus multiflorus, P. radiatus var. aurea, and Vigna sinensis against Botrytis cinerea, Puccinia recondita, and Erysiphe graminis. These seed extracts were highly effective against three B. cinerea strains resistant to carbendazim, procymidone, and diethofencarb. All leguminous seed extracts revealed weak or no fungicidal effect against Rhizoctonia solani, Pyricularia grisea, and Phytophthora infestans. As a naturally occurring fungicide, leguminous seed-derived materials described could be useful as new fungicidal products against various plant diseases induced by phytopathogenic fungi.

Effects of Scarification and Water Soaking Treatment on Germination of Hard-Seeded Legumes (두과 작물의 경실종자 발아촉진에 대한 종피연화처리의 효과)

  • The crops showing hard seed character have high seed viability after seed storage for long period. The low germination rate due to hard seed coat, however, cause a problem of low seedling establishment in field condition. Three legumes used in the experiment, lablab bean (Dolichos lablab L.), asparagus bean (Vigna sesquipedalis L. Fruwirth), and soybean (Glycine max L. Merr.), showed low germination rate (26, 17, and 5%, respectively) due to thick and hard seed coat. In this study artificial treatment for breaking dormancy was tested in hard seeds. The effect of proper treatment was various depending on species. The germination rate of lablab bean was highly improved up to 94% by soaking into water for 24 hours. In the case of asparagus bean, the rate was increased up to 90% by soaking for eight hours near boiling water until it cools. The germination rate of small hard seed soybean was increase to 96% by soaking into concentrated sulfuric acid for 10 minutes. Ultrastructural change revealed by scanning electron microscope (SEM) reflects that the structure of micropyle was changed and water uptake was facilitated with all treatments tested in the experiment. Especially, sulfuric acid treatment resulted in the degradation of micropylar tissue. These results demonstrate that the artificial treatment including sulfuric acid and (hot) water soaking treatment for promoting water uptake can be applied to improve seed germination in legume seed with thick and hard seed coat.

  • The cowpea aphid Aphis craccivora Koch (Hemiptera: Aphididae) is a polyphagous species with a worldwide distribution. We investigated the temperature effects on development periods of nymphs, and the longevity and fecundity of apterous female of A. craccivora. The study was conducted at six constant temperatures of 10.0, 15.0, 20.0, 25, 30.0, and $32.5^{\circ}C$. A. craccivora developed successfully from nymph to adult stage at all temperatures subjected. The developmental rate of A. craccivora increased as temperature increased. The lower developmental threshold (LT) and thermal constant (K) of A. craccivora nymph stage were estimated by linear regression as $5.3^{\circ}C$ and 128.4 degree-days (DD), respectively. Lower and higher threshold temperatures (TL, TH and TH-TL, respectively) were calculated by the Sharpe_Schoolfield_Ikemoto (SSI) model as $17.0^{\circ}C$, $34.6^{\circ}C$ and $17.5^{\circ}C$. Developmental completion of nymph stages was described using a three-parameter Weibull function. Life table parameters were estimated. The intrinsic rate of increase was highest at $25^{\circ}C$, while the net reproductive rate was highest at $20^{\circ}C$. Biological characteristics of A. craccivora populations from different geographic areas were discussed.
