Comparative Study on Acute Toxicity of Treated Effluent Containing Salt using Daphnia magna and Vibrio fischeri (염 함유 폐수처리수에 대한 Daphnia magna 및 Vibrio fischeri 급성독성 비교연구)

  • This paper aims to evaluate the results of acute toxicity testing with Daphnia mag$Na^+$ and Vibrio fischeri and characteristics of ionic substance of treated effluent which contained salt. Acute toxicity with Daphnia mag$Na^+$ and Vibrio fischeri and salinity of 19 samples (4 business categories) were a$Na^+$lysed. Salinity of effluent could explain the fluctuation of toxicity with D. mag$Na^+$ about 66% ~ 91% ($r^2=0.66{\sim}0.91$). The results of acute toxicity testing with V. fischeri of treated effluent (aggregate manufacture facilities) did not indicate toxicity (TU = 0), whereas that of chemical manufacture facilities indicated toxicity. V. fischeri, a candidate test organism, seemed suitable test organism for acute toxicity testing of effluent except high salinity (above 65‰ ~ 70‰) in aggregate manufacture facilities (nonmetalic minerals facilities). The performance of ion composition about treated effluent of surveyed facilities indicated that ion concentration of $Na^+$ (5,740 mg/L) and $Cl^-$ (9,727 mg/L) showed high level among 6 major ions ($Na^+$, $K^+$, $Ca^{2+}$, $Mg^{2+}$, $SO_4{^{2-}}$, $Cl^-$) in effluent of nonmetalic minerals facilities. In addition, Clion seemed to influence the D. magna survival rather than $Na^+$ ion.

  • The toxicity of heavy metals (Zn, Pb) and diesel, in single and binary solution was investigated using the photobacterium Vibrio fischeri (Microtox test) as a test organism. In this experiment, the concentration of water soluble fraction of diesel was based on the total petroleum hydrocarbon (TPH). The toxicity of each single compound showed the following $EC_{50}$ (15min): Zn 1.90 mg/L, Pb 0.31 mg/L, TPH 2.09 mg/L. The observed toxicity of binary mixtures increased, depending on the concentration of the mixed substance. The effects were defined as synergistic, antagonistic, or additive, in accordance with the sign of difference between the predicted and observed toxicity at binary mixtures. The interactive effects between zinc and lead were synergistic, on the other hand, antagonistic and additive effects were found in each metal and TPH mixtures on the bioluminescence of V. fischeri.

  • The light-organ symbiont of pine cone fish, Vibrio fischeri, senses its presence in the host and responds to environmental changes by differentially expressing its symbiosis-related luminescence genes. The V. fischeri luminescence genes are activated by LuxR protein in the presence of an autoinducer. In an effort to elucidate the mechanism of regulation of luxR, a plasmid containing luxR was mutagenized in vitro with hydroxylamine and a luxR mutant plasmid was isolated by its ability to activate luminescence genes cloned in E. coli in the absence of the autoinducer. The specific base change identified by DNA sequencing was only single base transition at +78 from the transcriptional start of luxR. Based on a Western immunoblot analysis, the nucleotide change directed the synthesis of much higher level of LuxR protein without any amino acid substitutions. The results suggest that the region including the +78th base is presumably internal operator required for autorepression of luxR, and the increased cellular level of LuxR results in activation of luminescence genes by autoinducer independent fashion.

Evaluation of Toxicity of Heavy Metals and Surfactants Using Vibrio Fischeri and Daphnia Magna (발광박테리아(Vibrio fischeri)와 물벼룩(Daphnia magna)을 이용한 중금속 및 계면활성제의 혼합독성 평가)

  • In this study, the mixed toxicity of heavy metals (Cu, Pb, Hg) and surfactants (SLS, ALS) was evaluated by using Vibrio fischeri and Daphnia magna. The sensitivity of ecotoxicity to heavy metals was sensitive to daphnia but the case of surfactants were more sensitive to Vibrio fischeri. Experimental results of Vibrio fischeri show that the toxicity value P(O) was lower than the predicted value P(E), and the antagonistic effect was observed when the heavy metal and the surfactant were mixed. It seems that SLS and ALS, which are anionic surfactants, have anionic form on the hydrophilic head, so that they have an antagonistic effect that they are bonded with heavy metal ion which is a cation type and the actual toxicity is lowered. In Daphnia magna, the results showed that antagonistic, additive and synergistic effects were in order as concentrations increased. As the concentration increases, Daphnia magna, which is highly sensitive to heavy metals, seems to have a synergistic effect with a rapid increase in mortality.

  • Cyanides and dichlorophenols were important pollutants in industrial effluents of steel, petroleum, plastics, pesticides, synthetic dye and/or fiber manufacturing. The toxic effects of cyanide and 3, 5-dichlorophenol in the unary and binary solutions to Vibrio fischeri were determined using the newly developed $N-tox^{(R)}$ bioassay system. This bioassay system relies upon the attenuation of light intensity emitted by Vibrio fischeri exposed to various pollutants including metals and organic compounds. Most of studies dealing with toxicity of pollutants concerned single chemical species, while the organisms were typically exposed to pollutant mixtures. The present study showed that the toxicity of some binary combinations of cyanide and 3, 5-dichlorophenol significantly was lower than the predicted toxicity from the addicted model. This antagonistic interaction was well explained by chemical interaction model presented in this study.

A Effect of Heavy Metal to Toxicity of Triclosan Focused on Vibrio fischeri Assay (Triclosan의 독성에 중금속이 미치는 영향 - V. fischeri Assay 관련 내용 중심으로 -)

  • The purpose of this study is to evaluate effect of heavy metals (i.e., $Cu^{2+}$, $Zn^{2+}$, $Cr^{6+}$, $Cd^{2+}$, $Hg^{2+}$, and $Pb^{2+}$) to toxicity of Triclosan as binary mixture. The individual toxicity and combined toxic effects of Triclosan with heavy metals were evaluated by Vibrio fischeri assay. In individual toxicity, the $Hg^{2+}$ was found to be most toxic followed by Triclosan, $Pb^{2+}$, $Cr^{6+}$, $Cu^{2+}$, $Zn^{2+}$, and $Cd^{2+}$, respectively. To evaluate combined toxic effect, correlation analysis of 'predicted value' calculated by Concentration addition (CA) model and Independent action (IA) model with 'experimental value' were performed based on the toxicity of individual compound. As a result, all of the combinations showed that IA model were more correlated with experimental value than CA model. On the basis of the median effect concentration of combination ($EC_{50mix}$) predicted by IA model, experimental $EC_{50mix}$ of Triclosan + Cu, Triclosan + Zn, Triclosan + Pb, Triclosan + Hg, Triclosan + Cd, and Triclosan + Cr were 191%, 226%, 138%, 137%, 209%, and 138% of $EC_{50mix}$ predicted by IA model, respectively, indicating that all of the combinations produced antagonistic effect.

Evaluation of Vibrio Fischeri Toxicity for Biological Treatment of TNT-contaminated Soil (TNT 오염토양의 생물학적 처리에 대한 Vibrio Fischeri 독성 평가)

  • This research was performed to evaluate Vibrio fischeri toxicity for TNT-contaminated soils treated by composting and slurry phase bioreactor, which were operated for 45 and 200 days, respectively. In case of composting, the GL value of acetone-added soil was 40, which was lower than th at of glucose-added soil after treatment. In case of slurry phase bioreactors under anaerobic, anaerobic/aerobic, and aerobic regimes, they showed the GL values of 6, 8, and 4 after treatment, respectively. It was evaluated that the toxicity of all slurry phase bioreactors was reduced significantly to detoxification. The relationships between GL value and the number of S. typhimurium in both composting and slurry phase bioreactor were developed as the first order equations with high correlation coefficient (r > 0.8890).

  • A comparison of promoter fusions with the luxCDABE genes from Vibrio fischeri and Photorhabdus luminescens was made using promoters from several genes (katG, sodA, and pqi-5) of E. coli that are responsive to oxidative damage. The respective characteristics, such as the basal and maximum bioluminescence and the relative bioluminescence, were compared. E. coli strains carrying fusions of the promoters to P. luminescens lux showed higher basal and maximally induced bioluminescent levels than strains carrying the same promoter fused to the luxCDABE genes from V. fischeri. The sensitivities between the strains were similar, regardless of the luciferase used, but lower response ratios were seen from strains harboring the P. luminescens lux fusions. Furthermore, using the two katG::lux fusion strains, the bioluminescence from the P. luminescens lux fusion strain, DK1, was stable after reaching a maximum, while that of strain DPD2511 decreased very rapidly due to substrate limitation.

Toxic Interactions of Perfluorinated Compounds (PFCs) with Heavy Metals Using Vibrio fischeri (발광박테리아 Vibrio fischeri를 이용한 과불화합물과 중금속의 복합독성평가)

  • The object of this study was to evaluate the combined toxic interactions of the perfluorooctanoic acid (PFOA) or perfluorooctane sulfonate (PFOS) with six heavy metals (Cu, Zn, Cr, Cd, Hg, and Pb). The individual and combined toxic effects were assessed using the Vibrio fischeri assay. In case of the individual toxicity, PFOA was higher toxic than PFOS and toxicity of PFOA and PFOS were lower than heavy metal. In the toxicity of heavy metals, the $Hg^{2+}$ was found to be most toxic followed by $Pb^{2+}$, $Cr^{6+}$, $Cu^{2+}$, $Zn^{2+}$, and $Cd^{2+}$. The combined toxicity of PFOA or PFOS with $Cr^{6+}$ were synergistic effect because the $EC_{50}$ mix values were less than 1 TU. PFOA + $Zn^{2+}$, PFOS + $Zn^{2+}$, PFOA + $Cd^{2+}$ and PFOS + $Cd^{2+}$ produced addictive effect. Except in these case, all of binary mixtures show antagonistic effect. This study proved potential risk of coexistent with perfluorinated compounds and heavy metals in water environment.

  • To investigate bioassay for toxic level evaluation of wastewater, toxic levels were checked influents and effluents of 6 wastewater discharge facilities with Daphnia magna and Vibrio fischeri. In view of test duration, D. magna is preferred at 48 hours. And it was judged to efficient that one of the two was choosen for toxicity test method (Daphnia test and Vibrio test). Analysis data for wastewater is average toxic level for influent more higher than effluent. And effluent toxic level is sharp decrease than effluents.
