Morphological Changes of Uterine and Vaginal Epithelium in Postpartum Korean Native Goats (韓國 在來山羊에 있어서 分娩後 子宮 및 膣上皮細胞의 形態學的 變化)

  • This present study was designed to investigate by light microscope the morphologic changes in the uterine and vaginal epithelium of postpartum Korean native goats. Tissues were obtained for study on days 1, 3, 10 and 21 postpartum. The results obtained in this study were as followed; 1. Light microscopically, the height of the uterine epithelium was gradually decreased with the intervals and secretory profiles of apocrine were observed at between 1 and 10 days postpartum. The frequency of the light and dark cells with Masson's trichrom stain was high at 1 day postpartum an low at 21 days. A few of PAS positive cells were generally observed at 1 day postpartum, while PAS positive cells were not seen at 21 days. Numerous globule leucocytes were found between the epithelium and in the subepithelium at 1 day and thereafter moderate globule leucocytes were also found in the other periods. The intraepithelial vacuoles with crystalline structure appeared at 10 days postpartum. 2. The height of the vaginal mucosa was gradually decreased with the intervals but the highest layer was found at 3 days postpartum. The frequency of the mast cells was increased with times. At 3 and 10 days postpartum the shape of the surface-epithelium was cuboidal and the vacuolation of the epithelium was observed at 10 and 21 days.

Development of Somatic Cell Nuclear Transfer Bovine Embryos following Activation Time of Recipient Cytoplasm (수핵란의 활성화 시간에 따른 소 체세포 핵이식란의 발육)

  • This study was conducted to examine the effect of pre activation treatment and activation time of recipient cytoplasm on the development of bovine somatic cell nuclear transfer(NT) embryos. Donor cells were transferred and electrofused to enucleated oocytes before(pre-AC) or after activation(post-AC). Activation was induced with a combination of $Ca^{2+}$-ionophore(A23187) and DMAP. NT embryos were cultured in CR1aa containing 3 mg/ml BSA for 9 days. Some NT embryos were fixed at 0.5 to 2.5 hr after fusion(for post-AC) or activation(for pre-AC) for confocal microscopy. Developmental rate to the blastocyst stage was slightly high in the post-AC group(20.6%) compared to that of pre-AC group(15.3%). However, developmental speed of embryos in the pre-AC group was faster than that of embryos in the post-AC group. Development rates to the blastocyst stage were similar among different activation time before fusion(0.5,2 and 4 hr). The result of the present study suggests that development and nuclear morphology are affected f the activation status of the recipient cytoplasm before fusion.

Effect of Mechanical Enucleation Methods on Development of Bovine Nuclear Transfer Embryos (물리적 탈핵 방법이 소 복제수정란의 발달 능력에 미치는 영향)

  • The aim of this study was to examine the effect of mechanical enucleation methods, aspiration and squeezing, on the developmental ability of nuclear transfer bovine embryos. Enucleated oocytes made by both enucleation methods were fused to adult ear skin cells. After 7 days of culture, developmental ability up to blastocyst stage was similar in both squeezing($33.6{\pm}15.7%$) and aspiration enucleation methods($31.9{\pm}13.4%$). The proportion of blastocysts at Day 8 of culture was also similar between the aspiration($37.8{\pm}10.4%$) and squeezing enucleatign s($35.3{\pm}15.1%$). The mean cell number in Day 7 blastocysts was also similar between the both groups(aspiration: $110.3{\pm}39.2$ vs. squeezing: $103.7{\pm}42.8$). The ratio of apoptotic cells was also found to be not significant different between the both groups(aspiration: $2.8{\pm}2.6%$ vs. squeezing: $4.3{\pm}4.4%$). These results suggest that aspiration and squeezing methods, as mechanical enucleation technique, are both useful for the production of bovine somatic cell nuclear transfer embryos.

A Proposal for Promotion of Research Activities by Analysis of KOSEF's Basic Research Supports in Animal Resources Science Field (한국과학재단의 동물자원과학 분야의 기초연구지원 추이분석을 통한 연구활동지원 활성화 제언)

  • This research was conducted to make suggestions for the promotion of research activities in the field of animal resources sciences, and we evaluated the research funding trend and the present status of research funding offered by KOSEF in this field. Comparative portion of the number of research projects and grants in this field to other fields has a tendency to be decreased year by year except recent few years. Researchers in this field have received more research funding from the group-based program than from the individual-based program. Also, they have received less money(per project) than did researchers in the fields of general agricultural sciences and other science and technology. Researchers in this field ranges from 43 to 51 years of age and showed 48 years of average age. It was found that researchers who has been funded in the field of animal resources sciences have showed tendency of publication of more articles to SCI journals in recent 5 years. The strong points of the animal resources of agricultural sciences field in South Korea include: lots of researchers, the establishment of research infra-structure, the excellence in research competitiveness and technology level. However, its weaknesses are: a lack of leadership in relevant societies and institutes, a predicted shortfall of researchers in the next generation and insufficient research productivity. The opportunities include: increasing the importance of the biotechnology industry, activating international cooperation researches and exploring the multitude of possible research areas to be studied. However, some concerns still exist, such as threats from developed countries for the government to open the agricultural market, the reduction of the number of full-time farms and intensification of needs for economic and social effects. The diverse actions and systems based upon the strongpoint, weakness, opportunity and threats above-mentioned are required to encourage research activities in the field of animal resources of agricultural sciences in Korea In addition, researchers in this fields would make an effort to keep pace with international society as well as domestic demands.

Production and Characterization of vitellogenin monoclonal antibody on the Scorpion fish Sebastiscus marmoratus (쏨뱅이, Sebastiscus marmoratus의 vitellogenin 단클론 항체생산 및 특성에 관한 연구)

  • In order to establish bio-marker systems for the screening of endocrine-disrupting chemicals contaminated in various environment, Vitellogenin(Vtg) bio-marker have been developed to detect Scorpion fish's(Sebastiscus marmoratus) Vtg. Vtg has been induced by administration of estradiol into S. marmoratus, and purified by gel filtration and ion-exchange chromatography from serum of the fish. After immunization of the purified Vtg into BALB/c mouse, hybridomas secreting anti-Vtg antibodies have been produced. The size of induced Vtg in the serum was about 440 kDa by gel filtration using Sepharose CL-6B. By SDS-PAGE analysis, the main band of Vtg, however, was at 175 kDa, and several minor bands have been detected with the main band. Eight different monoclonal antibodies have been produced from established hybridomas and the antibodies did not cross-react with sera from different species of fishes tested in this study except with that of Sebastes hubbsi. These results suggested that the monoclonal antibody of S28 and S15 can used as capture and tracer antibodies for ELISA and ICG assays. The detection systems developed in this study can be used as Bio-marker assays to check endocrine disrupting activity of various chemicals as well as to detect known endocrine disrupting chemicals contaminated in environment.

Effects of Extract from Fermented Flower-buds of Panax ginseng C.A. Meyer on Mouse Cytokine IL-6, TNF-α Production (발효 인삼꽃 추출물의 경구 투여가 마우스 사이토카인 IL-6, TNF-α의 생성에 미치는 영향)

  • Panax ginseng C.A. Meyer has been used as a traditional medicinal ingredient and the ginseng flower-buds also proved to have good medicinal properties. In this study, in order to enhance immune activities of ginseng flower-bud, the ginseng flower-bud extract was being fermented by Bacillus subtilis KCTC 1022 (BS), Lactobacillus plantarum KCTC3 and Saccharomyces cerevisiae strain CHY1011 (SC). Mice were orally administered daily for two weeks at two different concentrations (100 and 200 mg/kg B.W.). Treatment samples were water extracts of ginseng flower-buds (FD), water extracts of fermented ginseng flower-buds (FM) and controls for saline solution. Cytokine production (IL-6, TNF-${\alpha}$) either stimulated with LPS or not stimulated with LPS was detected by the ELISA assay when using the cytokine kit. Cytokine was statistically increased at supplemented groups with LPS in both the 100 and the 200 mg/kg B.W. and treatment with FM significantly decreased the LPS-induced TNF-${\alpha}$ and IL-6 production more than the treatment with FD. The results of this study may suggest that supplementation with FM increases the immune function by regulating cytokine production capacity for activated macrophages.

Development and Performance Evaluation of Hydroxyl Radical Generator using Electron Emission Type High Voltage and Low Current Discharger (전자방사식 고압 저전력 방전을 이용한 OH radical 발생기의 개발과 성능 평가)

  • In this study, we developed an electron-emission OH radical generator for waste water treatment. The stability of the circuitry was ensured by implementing stable pulse waves with a MOSFET and reducing the momentary current rise. The OH radical generator uses a high-voltage and low-current discharger. The performance of the device was evaluated experimentally, which showed that it is possible to produce a stable and uniform pulse waveform for the drain current of the power MOSFET, which is connected to the input side of an AC multiplying converter through negative feedback circuitry with CR-snubber architecture. It was also possible to reduce the excitation current of the converter and improve the stability of the oscillation circuit. In addition, the generator can generate hydroxyl radicals stably. The bactericidal activities were also evaluated, and the germicidal power for E. coli, S. aureus, and S. flexneriwas improved by 99.9% or more after 60 minutes.

  • The foot and mouth disease(FMD) was occurred from Andong city on November 23, 2010 and spread out the whole country except Jeju island and Jeolla-do. About 3.4 million livestock such as cow and pig was buired at 4,200 sites during preventive measures of FMD. Government did not effectively respond to the FMD crisis management so FMD spread out the whole country. To Prevent the spread FMD, Farms have to fast approaching and respond directly to smartphones and Tablet PC applications. Resolve the difficulties of using smart devices and easy to operate for the effective utilization of the development of simple applications. This application of FMD, developed for the prevention and alarm applications, foot and mouth disease will be caused, farmers around the farm in case of risk and the seriousness of the FMD will notify smartphone, FMD prevent additional damage due to be interested in preventing further that allows your application is for development purposes.

Stage specific transcriptome analysis of liver tissue from a crossbred Korean Native Pig (KNP × Yorkshire)

  • Korean Native Pig (KNP) has a uniform black coat color, excellent meat quality, white colored fat, solid fat structure and good marbling. However, its growth performance is low, while the western origin Yorkshire pig has high growth performance. To take advantage of the unique performance of the two pig breeds, we raised crossbreeds (KNP ${\times}$ Yorkshire to make use of the heterotic effect. We then analyzed the liver transcriptome as it plays an important role in fat metabolism. We sampled at two stages: 10 weeks and at 26 weeks. The stages were chosen to correspond to the change in feeding system. A total of 16 pigs (8 from each stage) were sampled and RNA sequencing was performed. The reads were mapped to the reference genome and differential expression analysis was performed with edgeR package. A total of 324 genes were found to be significantly differentially expressed (${\left|log2FC\right|}$ > 1 & q < 0.01), out of which 180 genes were up-regulated and 144 genes were down-regulated. Principal Component Analysis (PCA) showed that the samples clustered according to stages. Functional annotation of significant DEGs (differentially expressed genes) showed that GO terms such as DNA replication, cell division, protein phosphorylation, regulation of signal transduction by p53 class mediator, ribosome, focal adhesion, DNA helicase activity, protein kinase activity etc. were enriched. KEGG pathway analysis showed that the DEGs functioned in cell cycle, Ras signaling pathway, p53 signaling pathway, MAPK signaling pathway etc. Twenty-nine transcripts were also part of the DEGs, these were predominantly Cys2His2-like fold group (C2H2) family of zinc fingers. A protein-protein interaction (PPI) network analysis showed that there were three highly interconnected clusters, suggesting an enrichment of genes with similar biological function. This study presents the first report of liver tissue specific gene regulation in a cross-bred Korean pig.

The Signal Transduciton of Ginsenosides, Active Ingredients of Panax ginseng, in Xenopus oocyte: A Model System for Ginseng Study

  • Recently, we have provided evidence that ginsenosides, the active components of Panax ginseng, utilize pertussis toxin (PTX)-insensitive $G{\alpha}_{q/11}-phospholipase\;C-{\beta}3(PLC-{\beta}3)$ signal transduction pathway for the enhancement of $Ca^{2+}-activated\;Cl^{-}$ current in the Xenopus oocyte (British J. Pharmacol. 132, 641-647, 2001; JBC 276, 48797-48802, 2001). Other investigators have shown that stimulation of receptors linked to $G{\alpha}-PLC$ pathway inhibits the activity of G proteincoupled inwardly rectifying $K^+$ (GIRK) channel. In the present study, we sought to determine whether ginsenosides influenced the activity of GIRK 1 and GIRK 4 (GIRK 1/4) channels expressed in the Xenopus oocyte, and if so, the underlying signal transduction mechanism. In oocyte injected with GIRK 1/4 channel cRNAs, bath-applied ginsenosides inhibited high potassium (HK) solution-elicited GIRK current $(EC_{50}:4.9{\pm}4.3\;{\mu}g/ml).$ Pretreatment of the oocyte with PTX reduced the HK solution-elicited GIRK current by $49\%,$ but it did not alter the inhibitory ginsenoside effect on GIRK current. Prior intraoocyte injection of cRNA(s) coding $G{\alpha}_q,\;G{\alpha}_{11}\;or\;G{\alpha}_q/G{\alpha}_{11},\;but\;not\;G{\alpha}_{i2}\;or\;G{\alpha}_{oA}$ attenuated the inhibitory ginsenoside effect. Injection of cRNAs coding $G{\beta}_{1{\gamma}2}$ also attenuated the ginsenoside effect. Similarly, injection of the cRNAs coding regulators of G protein signaling 1, 2 and 4 (RGS1, RGS2 and RGS4), which interact with $G{\alpha}_i\;and/or\;G{\alpha}_{q/11}$ and stimulates the hydrolysis of GTP to GDP in active GTP-bound $G{\alpha}$ subunit, resulted in a significant reduction of ginsenoside effect on GIRK current. Preincubation of GIRK channel-expressing oocyte in PLC inhibitor (U73122) or protein kinase C (PKC) inhibitor (staurosporine or chelerythrine) blocked the inhibitory ginsenoside effect on GIRK current. On the other hand, intraoocyte injection of BAPTA, a free $Ca^{2+}$ chelator, had no significant effect on the ginsenoside action. Taken together, these results suggest that ginsenosides inhibit the activity of GIRK 1/4 channel expressed in the Xenopus oocyte through a PTX-insensitive and $G{\alpha}_{q/11}$-,PLC-and PKC-mediated signal transduction pathway.
