Temporal Changes of the Calcium-binding Proteins in the Medial Vestibular Nucleus following Unilateral Labyrinthectomy in Rats

  • Calcium ($Ca^{2+}$) is an intracellular second messenger associated with neuronal plasticity of the central nervous system. The calcium-binding proteins regulate the $Ca^{2+}$-mediated signals in the cytoplasm and buffer the calcium concentration. This study examined temporal changes of three calcium-binding proteins (calretinin, calbindin and parvalbumin) in the medial vestibular nucleus (MVN) during vestibular compensation after unilateral labyrinthectomy (UL) in rats. Rats underwent UL, and the changes in the expression of these proteins at 2, 6, 12, 24, 48, and 72 h were examined by immuno-fluorescence staining. The expression levels of all three proteins increased immediately after UL and returned to the control level by 48 h. However, the level of calretinin showed changes different from the other two proteins, being expressed at significantly higher level in the contralateral MVN than in the ipsilateral MVN 2 h after UL, whereas the other two proteins showed similar expression levels in both the ipsilateral and contralateral MVN. These results suggest that the calcium binding proteins have some protective activity against the increased $Ca^{2+}$ levels in the MVN. In particular, calretinin might be more responsive to neuronal activity than calbindin or parvalbumin.

안뜰눈반사를 통한 노인 균형 연구 (Review of Balance in Elderly by Vestivulo-ocular Reflex)

  • 이 논문은 노인의 균형문제를 해결하기 위해 안뜰눈반사와 안뜰재활운동에 대해 고찰하고자 한다. 낙상은 노화와 관련된 주요한 문제이다. 낙상과 결과적으로 일어나는 일은 가장 흔하면서 심각한 문제이다. 균형의 결함은 낙상의 가장 높은 위험 요소 중 하나이다. 균형을 조절하고 협응하는데 사용되고 통합되는데 세 가지 신경계가 필요하다. 정상적인 이동과 머리운동을 하는 동안, 안뜰눈반사는 주시를 안정화시키고 망막의 상을 유지하도록 돕는다. 안뜰눈반사의 결함과 낙상간에 직접적인 관계가 있다. 노인들은 안뜰눈반사를 억제하고 강화시키는 능력이 감소된다. 안뜰재활운동은 중추신경계 보상을 목적으로 시작한 운동에 근거한 단체 치료법이다. 안뜰재활운동은 머리와 눈의 반복적인 운동을 사용해서 안뜰눈반사의 이득을 회복시키고 오류를 줄인다. 많은 연구들이 안뜰재활운동을 한 단체가 균형과 보행 검사에서 월등함을 보였다. 이 논문은 노인의 낙상을 방지하기 위한 운동법으로 안뜰재활운동을이 도움이 될 것으로 사료된다.

마우스 공간지각과 기억 형성에 미치는 전정 유래 정보의 규명 (Identification of Vestibular Organ Originated Information on Spatial Memory in Mice)

  • Objectives: We aimed to study the role of vestibular input on spatial memory performance in mice that had undergone bilateral surgical labyrinthectomy, semicircular canal (SCC) occlusion and 4G hypergravity exposure. Methods: Twelve to 16 weeks old ICR mice (n=30) were used for the experiment. The experimental group divided into 3 groups. One group had undergone bilateral chemical labyrinthectomy, and the other group had performed SCC occlusion surgery, and the last group was exposed to 4G hypergravity for 2 weeks. The movement of mice was recorded using camera in Y maze which had 3 radial arms (35 cm long, 7 cm high, 10 cm wide). We counted the number of visiting arms and analyzed the information of arm selection using program we developed before and after procedure. Results: The bilateral labyrinthectomy group which semicircular canal and otolithic function was impaired showed low behavioral performance and spacial memory. The semicircular canal occlusion with $CO_2$ laser group which only semicircular canal function was impaired showed no difference in performance activity and spatial memory. However the hypergravity exposure group in which only otolithic function impaired showed spatial memory function was affected but the behavioral performance was spared. The impairment of spatial memory recovered after a few days after exposure in hypergravity group. Conclusions: This spatial memory function was affected by bilateral vestibular loss. Space-related information processing seems to be determined by otolithic organ information rather than semicircular canals. Due to otolithic function impairment, spatial learning was impaired after exposure to gravity changes in animals and this impaired performance was compensated after normal gravity exposure.

급성저혈압에 의한 내측전정신경핵 신경세포의 흥분성 변화를 분석하기 위한 테트로드 기법의 적용 (Application of Tetrode Technology for Analysis of Changes in Neural Excitability of Medial Vestibular Nucleus by Acute Arterial Hypotension)

  • Objectives: Excitability o medial vestibular nucleus (MVN) in the brainstem can be affected by changes in the arterial blood pressure. Several animal studies have demonstrated that acute hypotension results in the alteration of multiunit activities and expression of cFos protein in the MVN. In the field of extracellular electrophysiological recording, tetrode technology and spike sorting algorithms can easily identify single unit activity from multiunit activities in the brain. However, detailed properties of electrophysiological changes in single unit of the MVN during acute hypotension have been unknown. Methods: Therefore, we applied tetrode techniques and electrophysiological characterization methods to know the effect of acute hypotension on single unit activities of the MVN of rats. Results: Two or 3 types of unit could be classified according to the morphology of spikes and firing properties of neurons. Acute hypotension elicited 4 types of changes in spontaneous firing of single unit in the MVN. Most of these neurons showed excitatory responses for about within 1 minute after the induction of acute hypotension and then returned to the baseline activity 10 minutes after the injection of sodium nitroprusside. There was also gradual increase in spontaneous firing in some units. In contrast small proportion of units showed rapid reduction of firing rate just after acute hypotension. Conclusions: Therefore, application of tetrode technology and spike sorting algorithms is another method for the monitoring of electrical activity of vestibular nuclear during acute hypotension.

Effects of Caffeine on Auditory- and Vestibular-Evoked Potentials in Healthy Individuals: A Double-Blind Placebo-Controlled Study

  • Background and Objectives:The blockage of adenosine receptors by caffeine changes the levels of neurotransmitters. These receptors are present in all parts of the body, including the auditory and vestibular systems. This study aimed to evaluate the effect of caffeine on evoked potentials using auditory brainstem responses (ABRs) and cervical vestibular-evoked myogenic potentials (cVEMPs) in a double-blind placebo-controlled study. Subjects and Methods: Forty individuals (20 females and 20 males; aged 18-25 years) were randomly assigned to two groups: the test group (consuming 3 mg/kg pure caffeine powder with little sugar and dry milk in 100 mL of water), and the placebo group (consuming only sugar and dry milk in 100 mL water as placebo). The cVEMPs and ABRs were recorded before and after caffeine or placebo intake. Results: A significant difference was observed in the absolute latencies of I and III (p<0.010), and V (p<0.001) and in the inter-peak latencies of III-V and I-V (p<0.001) of ABRs wave. In contrast, no significant difference was found in cVEMP parameters (P13 and N23 latency, threshold, P13-N23 amplitude, and amplitude ratio). The mean amplitudes of P13-N23 showed an increase after caffeine ingestion. However, this was not significant compared with the placebo group (p>0.050). Conclusions: It seems that the extent of caffeine's effects varies for differently evoked potentials. Latency reduction in ABRs indicates that caffeine improves transmission in the central brain auditory pathways. However, different effects of caffeine on auditory- and vestibular-evoked potentials could be attributed to the differences in sensitivities of the ABR and cVEMP tests.

영상 안진기를 이용한 전정 기능 소실 환자에서의 Galvanic 전정 자극 반응의 관찰 (Response of Galvanic Vestibular Stimulation in the Vestibular Loss Using Videooculograph)

  • 본 연구에서는 전기 전정 자극(GVS, Galvanic vestibular stimulation)이 일측 전정 기능 소실 환자에게서 정상인과 어떠한 특이성을 보이는가에 대하여 관찰하였다. 실험대상은 14 명(남:5, 여:9)의 일측 전정 기능 소실 환자(경와우절제술 12 례, 전정신경 절제술 2 례)로 자극 방법은 bipolar 로 하여 전류 세기를 1mA에서 3mA 까지 달리하며 실험하였다. GVS 에 대한 반응은 영상 안진기(VOG, Videooculograrh)를 이용하여 관찰하였다. 정상 기능이 있는 측면으로의 자극에 대해서는 정상인과 동일한 반응을 보였으나 일측 전정 기능 소실(unilateral loss)을 갖는 환자의 경우 병변이 있는 쪽에 음의 자극, 없는 쪽에 양의 자극을 가하였을 경우에는 대상자 모두에게서 어떠한 안진 반응도 일어나지 않았다. 자극 후 안진의 경우 정상인은 일반적으로 나타나지 않으나 피검자들에게서는 나타나는 특성을 보였다.

평형감각 측정기술의 개발 (Development for a method of measurement in equilibrium sense)

  • The study was performed to evaluate the sense of equilibrium by means of rotary chair system in middle school girls with dance learning. Control of equilibrium, which is the most important function in dancing, is maintained by vestibular, visual, and proprioceptive inputs. Experimental groups were divided into trained group with dance (experimental group, 13~14 ys, n=15) and untrained group (control group, 13~14 ys, n=15). experimental group was trained by programmed dancing for 12 weeks, 45 min/day and 5 day/week. Gain of eye movement was measured for vestibulocular reflex (VOR), visual vestibular stimulation (VVOR), optokinetic stimulation(OKN), and visual fixation (VFX). The gain of eye movement at higher than 0.08 hz was significant decrease in experimental group, and VVOR showed the similar fashion to VOR. The gain in OKN and VFX was not significant difference between experimental and control groups. These results suggest that programmed training of dancing improves control of balance by adaptation of the vestibular function, and rotary chair system would be useful to evaluate the sense of equilibrium ability.

Reflex Action of the Semicircular Canals on Cervical Extensor and Flexor Muscles in Cats

  • In urethane anesthetized cats, each vestibular semicircular canal nerve was electrically stimulated, and reflex responses of the cervical extensor and flexor (the splenius capitis and sternomastoid muscles) were recorded by means of electromyography. Stimulation of a unilateral (anterior, horizontal or posterior) canal nerve elicited excitation of the contralateral cervical muscles and inhibition of the ipsilateral ones; during the canal nerve stimulation, the two muscles in one side of the neck revealed synergistic responses. Based on these experimental results, we formulated a diagram showing the functional connections between the vestibular semicircular canals and the cervical muscles in the vestibulocollic reflex.

기훈 환자의 보허음 투여 후 호전 1례 (An Improved Case after Treating Chi-Vertigo Patient with Boheo-eum)

  • The subject was a 46-year-old woman who was suffering from severe vertigo. Her major symptoms were vertigo, headache, neck pain, feverishness, and her minor symptoms were intermittent nausea and tinnitus, mild chest discomfort, facial flushing and sweating. Her symptoms were diagnosed as Chi-Vertigo and the patient was treated with Boheo-eum(Buxu-yin). Next, the patient's vertigo was evaluated with Vestibular disorders activities of Daily Living scale and Dix-Hallpike maneuver per 3days. An improved state in vestibular disorders activities of daily living scale, a frequency and duration of Dix-Hallpike maneuver were observed. In this case Boheo-eum(Buxu-yin) has an effect on a patient with vertigo due to Chi-Vertigo.
