  • Drug delivery to the brain is facilitated by the synthesis of chimeric peptides, where in neuropharmaceuticals are linked to a vector such as an antibody to the transferrin receptor that mediates transcytosis through the blood-brain barrier (BBB). When disulfide linkers are used in the chimeric peptide, it is crucial that the S-S bridge is stable during transit and that cleavage does not occur prematurely within endothelial cells, as the peptide drug moiety would then be sequestered by the BBB instead of passing through it. The present study addressed that problem. As a model drug a metabolically stable opioid peptide, [$^3$H]DALDA (Tyr-dArg-Phe-Lys-NH$_2$), was used. It was monobiotinylated with NHS-SS-biotin to yield bio-[$^3$H]DALDA. The biotinylated peptide was bound to the vector OX26-SA which is a covalent conjugate of OX26 and streptavidin (molar ratio = 1: 1). In vitro treatment of the chimeric peptide, bio-[$^3$H]DALDA/OX26-SA, with a reducing agent, dithiothreitol, released the labeled peptide from the vector by conversion of bio-[$^3$H]DALDA to the desbiotinylated derivative, desbio-[$^3$H]DALDA.

Efficient Sound Processing and Synthesis in VR Environment Using Curl Vector of Obstacle Object (장애물 객체의 회전 벡터를 이용한 VR 환경에서의 효율적인 음향 처리 및 합성)

  • 본 논문에서는 장애물 객체의 회전 벡터를 이용하여 VR 환경에서의 효율적으로 음향 처리 및 합성하는 방법을 제안한다. 현실에서 소리와 장애물이 있을 때, 소리는 장애물의 형태에 따라 퍼지면서 전파되는 형태를 보여준다. 이 같은 특징을 가상현실 환경에 유사하게 음향 처리하고자 하며 이를 위해 장애물 객체의 위치와 소리의 근원지 위치를 입력으로 소리의 전파 형태를 근사한다. 이때 모서리 부근에서 표현되는 소리의 회전을 계산하기 위해 장애물의 회전벡터(Curl vector)를 기반으로 소리의 회전을 추출하였으며, 장애물 형태를 컨볼루션(Convolution)하여 소리가 바깥 방향으로 전파되는 형태를 모델링한다. 또한, 장애물과 소리 벡터 사이의 거리, 소리 근원지와 소리 벡터 사이의 거리를 계산하여 소리의 크기를 감쇠 시켜 주며, 최종적으로 장애물 주변으로 퍼지는 벡터 모양인 외부벡터를 합성하여 장애물로부터 외부로 퍼지는 벡터의 방향을 설정한다. 본 논문에서 제안하는 방법을 이용한 소리는 장애물과의 거리와 형태를 고려하여 퍼지는 사운드 벡터 형태를 보여주며, 소리 위치에 따라 소리 감소 패턴이 변경되고, 장애물 모양에 따라 흐름이 조절되는 결과를 보여준다. 이 같은 실험은 실제 현실에서 소리가 장애물의 모양에 따라 나타나는 소리의 변화 및 패턴을 거의 유사하게 표현할 수 있다.

Low Rate Speech Coding Using the Harmonic Coding Combined with CELP Coding (하모닉 코딩과 CELP방법을 이용한 저 전송률 음성 부호화 방법)

  • In this paper, we propose a 4kbps speech coder that combines the harmonic vector excitation coding with time-separated transition coding. The harmonic vector excitation coding uses the harmonic excitation coding in the voiced frame and uses the vector excitation coding with the structure of analysis-by-synthesis in the unvoiced frame, respectively. But two mode coding method is not effective for transition frame mixed in voiced and unvoiced signal and a new method beyond using unvoiced/voiced mode coding is needed. Thus, we designed a time-separated transition coding method for transition frame in which a voiced/unvoiced decision algorithm separates unvoiced and voiced duration in a frame, and harmonic-harmonic excitation coding and vector-harmonic excitation coding method is selectively used depending on the previous frame U/V decision. In the decoder, the voiced excitation signals are generated efficiently through the inverse FFT of harmonic magnitudes and the unvoiced excitation signals are made by the inverse vector quantization. The reconstructed speech signal are synthesized by the Overlap/Add method.

Unified Design Methodology and Verification Platform for Giga-scale System on Chip (기가 스케일 SoC를 위한 통합 설계 방법론 및 검증 플랫폼)

  • We proposed an unified design methodology and verification platform for giga-scale System on Chip (SoC). According to the growth of VLSI integration, the existing RTL design methodology has a limitation of a production gap because a design complexity increases. A verification methodology need an evolution to overcome a verification gap. The proposed platform includes a high level synthesis, and we develop a power-aware verification platform for low power design and verification automation using it's results. We developed a verification automation and power-aware verification methodology based on control and data flow graph (CDFG) and an abstract level language and RTL. The verification platform includes self-checking and the coverage driven verification methodology. Especially, the number of the random vector decreases minimum 5.75 times with the constrained random vector algorithm which is developed for the power-aware verification. This platform can verify a low power design with a general logic simulator using a power and power cell modeling method. This unified design and verification platform allow automatically to verify, design and synthesis the giga-scale design from the system level to RTL level in the whole design flow.

A Simplification of Linear System via Frequency Transfer Function Synthesis (주파수 전달함수 합성법에 의한 선형시스템의 간소화)

  • This paper presents an approximation method for simplifying a high-order transfer function to a low-order transfer function. A model reduction is based on minimizing the error function weighted by the numerator polynomial of reduced systems. The proposed methods provide better low frequency fit and a computer aided algorithm that estimates the coefficients vector for the numerator and denominator polynomial on the simplified systems from an overdetermined linear system constructed by frequency responses of the original systems. Two examples are given to illustrate the feasibilities of the suggested schemes.

  • GlcNAc 2-epimerase gene from human was cloned. However GIcNAc 2-epimerase was expressed in E. coli as inclusion body formation. Several approaches were tried such as expression in low temperature and low concentration of IPTG. With these treatments production of active form of human GIcNAc 2-epimerase ι ,vas enhanced. For the direct synthesis of NeuAc from GlcNAc and pyruvate, NALase and GlcNAc 2-epimerase were characterized in terms of temperature effect on activity. equilibrium and stability, inhibition by pyruvate etc. For cheap and ease preparation of both the NALase and GlcNAc 2-epimerase, pEN24ma vector was made. which express both the NALasc and GIcNAc 2-epimerase simultaneously. In addition, E. coli BL21(DE3) harboring two plasmids was also made. Of the two systems, the latter was better for the expression of both enzymes.

Synthesis and High Expression of Chitin Deacetylase from Colletotrichum lindemuthianum in Pichia pastoris GS115

  • A gene, ClCDA, encoding chitin deacetylase from Colletotrichum lindemuthianum, was optimized according to the codon usage bias of Pichia pastoris and synthesized in vitro by overlap extension PCR. It was secretorily expressed in P. pastoris GS115 using the constitutive expression vector pHMB905A. The expression level reached the highest with 110 mg/l culture supernatant after 72 h of methanol induction, which comprised 77.27 U/mg chitin deacetylase activity. SDS-PAGE, mass spectrometry, and deglycosylation assays demonstrated that partial recombinant protein was glycosylated with an apparent molecular mass of 33 kDa. The amino acid sequences of recombinant proteins were confirmed by mass spectrometry.

  • Peptide deformylase (PDF) is essential and unique to bacteria for cytoplasmic protein synthesis, but not required in eukaryotes, thus making it an attractive target for the discovery of novel antibacterial drugs. Protein synthesis in eubacteria, under normal conditions, is initiated by formyl-methionyl-tRNA. PDF removes the formyl-group of N- formylmethionine of newly synthesized polypeptides to produce a mature protein. In this study, a pdf gene from Staphylococcus aureus 6538p was cloned in pET-14b vector and transformed in Escherichia coli BL21 (DE3). (omitted)

Expression of Recombinant Human Growth Hormone in a Soluble Form in Escherichia coli by Slowing Down the Protein Synthesis Rate

  • Formation of inclusion bodies is usually observed when foreign proteins are overexpressed in E. coli. The formation of inclusion bodies might be prevented by lowering the rate of protein synthesis, and appropriate regulation of the protein expression rate may lead to the soluble expression. In this study, human growth hormone (rhGH) was expressed in a soluble form by slowing down the protein synthesis rate, which was controlled in the transcriptional and translational levels. The transcriptional level was controlled by the regulation of the amount of RNA polymerase specific to the promoter in front of the rhGH gene. For lowering the rate of translation, the T7 transcription terminator-deleted vector was used to synthesize the longer mRNA of the target gene because the longer mRNA is expected to reduce the availability of tree ribosomes. In both methods, the percentage of soluble expression increased when the expression rate slowed down, and more than 93% of rhGH expressed was a soluble form in the T7 transcription terminator-deleted expression system.

  • This paper proposes a frequency transfer function synthesis for approximating a high-order model with resonance to a low-order model in the frequency domain. The presented model approximation method is based on minimizing the error function weighted by the numerator polynomial of approximated models, which is used of the RLS(Recursive Least Square) technique to estimate the coefficient vector of approximated models. The proposed method provides better fitting in a low frequency and peak resonance. And an example is given to illustrate feasibilities of the suggested schemes.
