Studies on the Manufacturing of Carbonaceous Products (Ⅰ) Effects of the Binder by Cutting Condition of Coal Tar (炭素製品 製造에 關한 硏究 (第 1 報) Coal tar의 Cutting 條件에 依한 粘結劑로서의 檢討)

  • As a practical method to produce the carbonaceous binder, the destructive distillation of coal tar has been studied. The optimum condition on binder in various temp. and cutting time were examined and the compositions of pitch were also examined in connection with ${\alpha},\;{\beta}\;and\;{\gamma}$ by solvent extraction. Coke powder was mixed with binder as treated pitch in the proportions of 77% of coke to 23% of pitch, to give a plastic mix which could then be pressed into molding. The properties of baked body after molding was tested. It was found that cutting of the coal tar at 350${\circ}C$, for 30 min., at 300${\circ}C$. for 1 hr., and at 250${\circ}C$. for 8 hrs. were good condition. Particularly, we found that the cutting of the coal tar obtained by destructive distillation at high temp. and in minimum period of time had shown the best condition for utilization of carbonaceous pitch as binder. The above mentioned cutting condition results in good quality of pitch which is available to be used as binder in carbon industry.

Separation and Concentration of L-Phenylalanine using a Supported Liquid Membrane

  • The separation and concentration of L-phenylalanine (L-Phe) using a supported liquid membrane (SLM) is investigated. A cation complex agent, di-2-ethylhexyl phosphoric acid (D2EHPA), is used as a carrier in the SLM with n-Heptane as a solvent. The reaction order and equilibrium constant in the formation reaction of L-phe-carrier complex are obtained from the extraction experiment. A mathematical model for a carrier mediated counter transport process is proposed to estimate the diffusion coefficient of L-phe-carrier complexly in the liquid membrant. Permeation experiments of L-phe using a SLM are performed under various operating conditions and optimum conditions for the transport of L-phe are obtained. Concentration of L-phe in the strip phase against its concentration is observed. Transport rate of glucose through liquid membrane is less than that of L-phe in the competitive transport of L-phe and glucose. And the existence of glucose reduced the transport rate of L-phe. The performance of separation with continuous strip phase is increased due to the dilution effect in the strip phase.

Insecticidal Activity of Coptis chinensis Extract Against Myzus persicae (Sulzer)

  • In view of the environmental and health hazards posed by synthetic insecticides, the use of plant products as botanical insecticides has gained increasing in recent years. In this study, we reported the insecticidal activity of extracts isolated from Coptis chinensis. On crude extraction, among the various solvent types tested (water, 1% (w/v) of sodium hydroxide, 70% ethanol), the 70% ethanol extract showed the best insecticidal activity (36.5%). Three different fractions (n-hexane, chloroform and ethyl acetate) were obtained from crude extract (70% ethanol) of the chloroform fraction and found to have noteworthy insecticidal activity (62.9%) by filter paper contact bioassay. Their chemical structures were identified as 2-methoxy-4-vinylphenol and aniline by head space-GC-MS analysis. Both compounds displayed a dose-dependent insecticidal activity of Myzus persicae (Sulzer). Insecticidal activity at the lowest concentration tested (500 ppm) approached 85.4% in the aniline compared with 79.9% in the 2-methoxy-4-vinylphenol. The insecticidal activity was greater for the aniline than 2-methoxy-4-vinylphenol. It is believed that the insecticidal activity is due mainly to the presence of aniline.

A Study on the Pharmaceutical Characteristics and Analysis of Natural Ginger Extract (천연 생강 추출물의 약리학적 특성 및 분석 연구)

  • Natural Ginger extract was extracted from Ginger component using ethanol as a solvent, and we tested various pharmaceutical characteristics in this extract. Characteristics experiment to use natural Ginger extract tested antimicrobial experiment using microbe, and analyzed organic and inorganic component with analysis instrument. From the result of characteristics experiment, some conclusions are obtained as follow. In this experiment, natural Ginger extract was obtained about 22.50 g-Ginger extract(extraction ratio : 1.80%) as liquid state of some viscous yellow-brown color. The result of antimicrobial experiment with staphylococcus aureus and aspergillus niger in microbe decreased more and more according to passage of reaction time. This phenomenon could know that Ginger extract has influenced to antimicrobial effect. In the result of instrumental analysis, aromatic organic components of benzimidazole(1.283), propyl isothiocyanate or isothiocyanic acid(1.477) etcs from Ginger extract were detected with GC/MS and inorganic component of Ca(27.6 ppm), K(24.08 ppm), Mg(6.03 ppm), Na(1.02 ppm), Zn(1.02 ppm), Sb(0.711 ppm) and Li(0.079 ppm) etcs from Ginger extract were detected with ICP/OES.

  • $MoO{_4}^{2-}$ is the stable chemical species of Mo(VI) in alkaline solution. In the pH range of 2 to 6, condensation polymerization between $MoO{_4}^{2-}$ and hydrogen ion results in the formation of various polyanions of Mo(VI). Polycations of Mo(VI) begin to form when solution pH is less than 2. As the concentration of inorganic acid increases, polycations of Mo(VI) can react with the anion of the inorganic acid, resulting in the formation of heteranions of Mo(VI). The distribution of Mo(VI) species at pH < 6 depends on the concentration of Mo(V) and inorganic acid. In order to analyze the solvent extraction and ion exchange data on Mo(VI) from concentrated inorganic acid solution, it is necessary to elucidate the nature of Mo(VI) complexes.

Effect of extraction conditions on chemical composition and antioxidant properties of mulberry fruit

  • Mulberry fruit (Morus alba L.) contains phytochemicals, including 1-deoxynojirimycin, quercetin-glucoside, kaempferol-glucoside, and anthocyanins, which have antioxidant effects. In this study, mulberry fruit extract was prepared at various temperatures (25-100℃) and water/ethanol solvent concentrations (0%-100% ethanol). Fourier-transform infrared spectroscopy (FT-IR) and circular dichroism (CD) data indicated that the content of bioactive compounds such as polyphenols and flavonoids was lower in 100% ethanolic extracts than in 30%-50% ethanolic extracts. Radical scavenging activity determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-di-3-ethylbenzthiazoline sulfonic acid (ABTS) assays was highly correlated with polyphenol and flavonoid content. In conclusion, 30%-50% ethanolic extracts contained the highest contents of bioactive compounds and exhibited high levels of radical scavenging activity. These findings may inform the use of mulberry fruit extract as a functional food.

  • Many therapeutic class drugs such as beta-blocker, corticosteroids, NSAIDs, etc are prohibited substances in the horse racing industry. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) technology makes it possible to isolate drugs from interference, enables various drug analyses in complex biological samples due to its sensitive sensitivity, and has been successfully applied to doping control. In this paper, we describe a rapid and sensitive method based on solid-phase extraction (SPE) using solid phase cartridge and LC-MS/MS to screen for different class's 35 drug targets in equine plasma. Plasma samples were pretreated by SPE with the NEXUS cartridge consisted non-polar carbon resin and minimum buffer solvent. Chromatographic separation of the analytes was performed on ACQUITY HSS C18 column (2.1 × 150 mm, 1.8 ㎛). The elution gradient was conducted with 5 mM ammonium formate (pH 3.0) in distilled water and 0.1% formic acid in acetonitrile at a flow rate of 0.25 mL/min. The selected reaction monitoring (SRM) mode was used for drug screening with multiple transitions in the positive ionization mode. The specificity, limit of detection, recovery, and stability was evaluated for validation. The method was found to be sensitive and reproducible for drug screening. The method was applied to plasma sample analysis for the proficiency test from the Association of Racing Chemist.

Screening of Effective Extraction Conditions for Increasing Antioxidant Activities of Licorice Extracts from Various Countries of Origin (원산지별 감초추출물의 항산화활성 증가를 위한 효율적인 추출조건 탐색)

  • In this work, licorice extracts were prepared using various extraction conditions such as extraction solvent, temperature, and time from Glycyrrhiza uralensis (G. uralensis) produced in Korea and China and Glycyrrhiza glabra (G. glabra) in Uzbekistan. The optimum extraction condition was selected from the extraction yields and antioxidative activities of extracts. Korea licorice extracts showed the highest free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity (46.05%) under the extraction condition of 85% ethanol at $60^{\circ}C$ for 6 hours. The prominent ROS (reactive oxygen species) scavenging activity using luminol-dependent chemiluminescence assay and the cellular protective effect against $^1O_2$ induced cellular membrane damage were also shown from the extracts obtained from the same condition. Especially, Korea G. uralensis extracts exhibited the higher prominent protective effect (${\tau}_{50}$ = 116.4 min) than (+)-(+)-${\alpha}$-tocopherol (${\tau}_{50}$ = 28.5 min) and the extraction yield of Korea licorice extract was 18.75%, which is 1.2 times and 2.5 times higher than that of Uzbekistan and China, respectively. These results indicate that the extraction condition of 85% ethanol at $60^{\circ}C$ for 6 hours is optimal to prepare licorice extracts, which can be applicable as anti-oxidative cosmetic materials.

Simultaneous Determination of Pesticides in Water Using a GC/MS Coupled with Micro Extraction by Packed Sorbent (MEPS-GC/MS를 이용한 농약류 동시 수질분석)

  • This study established an analytical method to simultaneously determine six organophosphorous pesticides [methyldemetone-S, diazinon, fenitrothion, parathion, phentoate, and O-ethyl O-(4-nitrophenyl) phenylphosphonothioate (EPN)] and carbaryl in water using a gas chromatography/mass spectrometry (GC/MS) system coupled with on-line micro extraction by packed sorbent (MEPS) and programmed temperature vaporizer (PTV) injector. Polystyrene divinylbenzene (PDVB) was used as a sorbent of MEPS. The effects of elution solvents, pH, elution volume and draw-eject cycles of samples on sample pretreatment process were investigated. Also, quality assurance and quality control (QA/QC) and the recovery of the pesticides in environmental samples were evaluated. The elution was performed using $30{\mu}L$ of a mixed solvent (acetone : dichloromethane = 80 : 20 (v/v)). Sample pretreatment processes were optimized with seven cycles of draw-eject of sample (1 mL) spiking an internal standard and sulfuric acid. At lower pH, the analytical sensitivity of diazinon decreased, but that of carbaryl increased. The method detection limit and the limit of quantification for this method were 0.02~0.18 and $0.08{\sim}0.59{\mu}g/L$, respectively. The method precision and accuracy were 1.5~11.5% and 83.3~129.8%, respectively, at concentrations of $0.5{\sim}5.0{\mu}g/L$. The recovery rates for all the pesticides except carbaryl in various environmental samples ranged 75.7~129.3%. The recovery rate of carbaryl in effluent sample was over 200% whereas carbaryl in drinking water, groundwater, and river water were in the acceptable range.

Extraction of Pigment from Sea Mustard ( Undaiia pinnatinda) using Supercritical Carbon Dioxide and Entrainer (초임계 이산화탄소와 보조용매를 이용한 미역으로부터 색소 추출)

  • In order to develop a new separation technology, supercritical fluid extraction process was used to produce high purity pigments and fatty acids from seaweed (Undaria pinnatifida). Supercritical carbon dioxide was used as a solvent and ethanol as an entrainer. The sample was treated by a frozen drier and experiments were conducted with a semi-batch flow system at various operating conditions (pressure range, $10.3\~17.2$ MPa; temperature range, $30\~45^{\circ}C$: particle size, $500\~1,000{\mu}m$ extraction time, 60 min). Characteristics of the recovered pigment (chlorophyll a) and fatty acids were determined by UV-spectrophotometry and gas chromatography, respectively. The highest extraction efficiency for fatty acids and pigments was achieved at 12.4 MPa, $35^{\circ}C$, $500{\mu}m$of seaweed size.
