Effect of Concentration Methods on the Quality of Single and Blended Juice Concentrates

  • Clarified apple, carrot and orange juices were prepared using ultrafiltration and their single and blend juices were further concentrated using ultrafiltration, freeze-drying, and rotary evaporation. Effect of concentration methods on the quality of concentrated single juices and juice blends was investigated. Turbidity values of samples concentrated by evaporation were significantly higher than those prepared by ultrafiltration and vacuum freezing regardless of juice source (i.e., apple, orange or carrot) or blending (p<0.05). The highest soluble solids contents were obtained for the samples concentrated by evaporation process. Concentrated apple juice contained significantly higher amount of vitamin C and soluble solids than concentrated orange and carrot juices regardless of concentration methods (p<0.05). For blended samples, no direct relationships between blend ratio and total amount of vitamin C were found; however, samples contained more apple juice showed the highest value of soluble solids regardless of concentration methods.

  • 국립광주박물관은 전남문화재연구원에서 발굴 조사한 완도 보길도 윤선도 유적에서 출토된 나막신의 보존처리를 실시하였다. 나막신의 수종은 소나무속 경송류(Pinus sp. (hard pine))로 식별되었으며 처리방법은 t-butanol 치환 후 PEG-진공동결건조법을 이용하여 처리하였다.

  • One of the major problems of biological ToF-SIMS imaging is the lack of protein and peptide imaging. Most of biological story telling is mianly based on proteins. The biological implication of lipid ToF-SIMS imaging would be much higher if protein imaging is provided together. Utilizing high secondary ion yields of metals, proteins can be ToF-SIMS imaged with nanoparticle tagged proteins. Nanoparticles such as Fe3O4, SiO2, PbS were used for imaing NeuN, MCH, Orexin A, ${\alpha}$ synucline, TH(Tryosine Hydroxylase) in mouse tissues with a spatial resolution of ${\sim}2{\mu}m$ using a TOF-SIMS. Lipids and neurotransmitters images obtained simultaneously with protein images were overlayed for more deeper understanding of neurobiology, which is not allowed by any other bioimaging technqiues. The protein images from TOF-SIMS were compared with confocal fluorescence microscopy and NanoSIMS images. A new sample preparation method for imaging single cell membranes in a tissue using the vibrotome technique to prepare a tissue slice without any fixation and freeze drying will be also presented briefly for Hippocampus and Hypothalamus tissues.

Improving the Viability of Freeze-dried Probiotics Using a Lysine-based Rehydration Mixture

  • The probiotic market is constantly continuing to grow, concomitantly with a widening in the range and diversity of probiotic products. Probiotics are defined as live microorganisms that provide a benefit to the host when consumed at a proper dose; the viability of a probiotic is therefore of crucial importance for its efficacy. Many products undergo lyophilization for maintaining their shelf-life. Unfortunately, this procedure may damage the integrity of the cells due to stress conditions during both the freezing and (vacuum-) drying process, thereby impacting their functionality. We propose a lysine-based mixture for rehydration of freeze-dried probiotics for improving their viability during in vitro simulated gastric and duodenum stress conditions. Measurement of the zeta potential served as an indicator of cell integrity and efficacy of this mixture, while functionality was estimated by adhesion to a human enterocyte-like Caco-2 cell-line. The freeze-dried bacteria exhibited a significantly different zeta potential compared to fresh cultures; however, this condition could be restored by rehydration with the lysine mixture. Recovery of the surface charge was found to influence adhesion ability to the Caco-2 cell-line. The optimum lysine concentration of the formulation, designated "Zeta-bio", was found to be 0.03 M for improving the viability of Lactiplantibacillus plantarum Lp-115 by up to 13.86% and a 7-strain mixture (400B) to 41.99% compared to the control rehydrated with distilled water. In addition, the lysine Zeta-bio formulation notably increased the adherence ability of lyophilized Lp-115 to the Caco-2 cell-line after subjected to the in vitro stress conditions of the simulated gastrointestinal tract passage.

경산 임당유적 목제품 보존 - 갑옷틀 및 칠기 - (Conservation for Wooden Objects Excavated From Imdang, KyungSan - Wooden frame of Armor and Lacquer Wares -)

  • 경산 임당저습지유적에서 출토된 목제 갑옷틀과 다양한 칠기유물들은 재질이 매우 취약하고 옻칠이 되어 있어 공기 중에 노출되면 건조가 진행되어 곧바로 수축·변형이 일어나기 쉽다. 목제 갑옷틀의 경우 대형으로 동결건조과정 중 융해의 발생우려가 있고 칠기의 경우 목재표면에 옻칠이 되어있어 약제가 잘 침투되지 않고 처리 중 칠막이 부풀거나 탈락될 우려가 높다. 따라서 목제 갑옷틀은 진공 동결건조과정 중 융해방지를 위하여 t-butanol로 치환한 후 최종 PEG#3,350 43%의 t-butanol용액에 함침하고, 칠기는 최종 PEG#3,350 40%의 수용액(水溶液)에 실온에서 함침처리 후 동결 건조하여 보존처리 하였다. 칠기의 칠 도막 분석결과 뚜껑 및 고배는 초칠을 한 후 밑층에 흑색안료(그을음)와 옻칠을 혼합하여 바른 후 2-3회 생칠을 하였고 주칠 컵형목기는 밑층에 흑색안료와 옻칠을 혼합하여 바른 후 그 위에 1회 중칠을 하였으며, 산화철(Fe2O3: 석간주)이 붉은 칠의 발색안료로 쓰였을 가능성을 보여준다.

동결건조 어류 알 농축물의 식품기능성 및 생리활성 (Food Functionality and Bioactivity of Vacuum Freeze-dried Fish Roe Concentrates)

  • The purpose of this study was to evaluate the food functional properties and in vitro bioactivity of vacuum freeze-dried fish roe concentrates (FRCs) prepared from Alaska pollock Theragra chalcogramma (AP), bastard halibut Paralichthys olivaceus (BH) and skipjack tuna Katsuwonus pelamis (ST). All three species showed better buffering capacity on the alkaline side (pH 10-12) than on the acidic side. The water-holding capacities of the FRCs were 3.5, 8.5 and 4.2 g/g protein for AP, BH and ST, respectively, and were significantly higher than that of commercial egg white. The protein solubilities of the FRCs were 42.5% (AP), 50.0% (BH) and 13.9% (ST). The foaming capacities of the FRCs were not significantly different among the species (128.0% for AP, 128.3% for BH, and 143.3% for ST; P>0.05), and their foam stability was maintained at 53.0-74.2% for 60 minutes. The oil-in-water emulsifying activity indexes of AP and BH (19.5 and 20.2 ㎡/g protein, respectively) were significantly superior to that of ST (P<0.05). The 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis-3-ethylbenzothia-zoline-6-sulfonic acid radical-scavenging activities (IC50, mg/mL) of the FRCs were in the ranges of 1.05-3.26 and 0.13-0.18 mg/mL, respectively, and the angiotensin I converting enzyme inhibitory activity was in the range of 0.97-1.89 mg/mL.

건조방법을 달리한 보리 잎의 생리활성 (Physiology Activity of Barley Leaf Using Different Drying Methods)

  • 다양한 방법으로 건조한 보리 잎의 생리활성을 분석한 결과, 보리 잎의 추출수율은 건조방법 간에 유의적인 차이를 보이지 않았다. 총 페놀함량은 건조방법별 함량에 유의적인 차이가 없었으나, 총 플라보노이드 함량은 열처리 후 건조한 보리 잎에서 생잎 다음으로 높게 나타났고, 음건, 진공 동결 건조, 전자레인지 후 건조한 보리 잎에서는 유의성을 보이지 않았다. SOD 유사활성은 생잎 $95.69{\pm}0.33%$, 열처리 후 건조한 보리 잎 $94.96{\pm}0.77%$, 음건 $92.04{\pm}1.35%$, 진공 동결건조 $91.53{\pm}0.40%$, 전자레인지 건조 $92.58{\pm}0.27%$로 90% 이상의 높은 활성을 보였으며, 대조군인 항산화제 sesamol $88.42{\pm}0.80%$, tocopherol $88.86{\pm}0.61%$, BHT $86.62{\pm}0.89%$에 비해 유의적으로 높은 활성을 나타냈다. 또한 생잎과 전자레인지 건조한 보리 잎을 제외하고 80% 이상의 peroxyl radical 소거능 및 90% 이상의 높은 xanthine oxidase 저해능을 보였다. 본 연구결과, 건조방법별 보리 잎의 총 페놀, 총 플라보노이드 함량은 건조방법에 따라 차이를 보였으나, 항산화 능력에는 크게 영향을 미치지 않는 것으로 사료된다.

이성산성 출토 목제칠기 보존 (Conservation for Wooden Lacquer Ware Excavated from Isong Sansung Fortress)

  • 이성산성 저수지안에서 출토된 목제칠기는 이미 국립문화재연구소에서 t-부탄올로 치환하여 PEG함침처리 후 진공동결건조법을 적용하여 보존처리되었다. 당시 이루어지지 않은 목제칠기의 수종 및 칠조사를 통해 칠기의 제작기법을 규명하고 여러개의 편으로 보관되어 온 칠기편을 전시 및 보관을 위하여 원형복원하였다. 그 결과 오리나무속(Alnus)중(中) 오리나무류를 사용하여 목제칠기를 제작하였으며 칠은 밑층에 토분과 칠을 혼합하여 칠하였고 칠에 흑색안료를 섞은 흑색칠과 순수한 칠을 2번 교대로 칠하여 총 5회 칠을 올려 제작한 것으로 확인되었다.

  • For thestudying and utilizing the thorn sea cucumber synthetically, the method preparing polysaccharide and polypeptide from sea cucumber was studied. The comparatively rational craft for preparing crude polysaccharide is fresh raw materials deal with oar form, $60\%$ of the ethanol precipitate after hydrolyzed by pretense and vibrated with ultrasonic wave auxiliary. The purification of polysaccharide and removal of protein fromcrude polysaccharide were made through precipitation method using the acetate. The polypeptide is obtained by concentrating in vacuum, freeze-drying after mixing precipitations of two times. Polysaccharide acute poisoning experiment indicate there is no bad reaction when LD50>5000g/kg, there strain rate of liver tumor H11 approach present generally acknowledged tumor treat medicine cyclophosphamide, and there is no side effect. The fatigue resistance function of polypeptide experiment also indicates that the fatigue resistance ability of mouse which fed on added sea cucumber polypeptide has a great improvement.

통발용 인공미끼의 개발 (The Improved Artificial Trap Baits)

  • The aim of this study was to develop the artificial trap baits being able to replace the natural anchovy or sardine baits. The fishing trials for evaluating the fishing efficiencies of the artificial baits were conducted with commercial traps at coastal areas near Tongyoung, southern Korea. The preferable artificial baits which were made of minced anchovy, sardine, mackerel, or fish surplus products mixed with wheat flour into dry or wet pelleted or crumbled types, were showed some merits as the following. \circled1 Fishing efficiencies of the artificial baits were same or little higher level than that of the natural bait anchovy; \circled2 bait cost were same or little lower level; \circled3 dry artificial baits stocked in the room condition showed good fishing efficiencies; \circled4 making of artificial baits were very simple; \circled5 there were no harm to the fish and little pollution to the environment; \circled6 the releasing rate of attractants in the artificial or natural bait showed similar patterns. The artificial baits using the fish by-products which were vacuum freeze-drying extract of boiled anchovy or tunas extract, showed lower catches in the conger eel, Coner miriaster, but similar catches in the crabs, mainly Charybdis japonica, to the those of natural anchovy bait.
