  • To detect the genetic variations among infectious bursal disease (IBD) vaccine strains, the hypervariable region of VP2 gene of seven IBDV vaccine strains were amplified using reverse transcriptase/polymerase chain reation(RT/PCR). Ampllified PCR products of IBDV were cloned, sequenced, and compared with published sequences for IBDV. Vaccine strains (JOONG, HAN, B7, IB, BU2, G2, CIL) used in Korea and Korean field isolates (SH/92, K1, 310) had 81%(310 and HAN) ~ 98%(SH/92 and CIL) amino acid sequence similarity. Vaccine strains had 80%(HAN and IB) ~ 99%(JOONG and BU2) amino acid sequence similartiy. Intermediate plus vaccine strain, CIL was not substituted at positions 279(D $\rightarrow$ N) and 284(A $\rightarrow$ T), and conserved in serine-rich heptapeptide. At the two hydrophilic region, JOONG, IB and Bu2 strains had identical amino acid sequence comparing with STC strain. By phylogenetic analysis, JOONG and DAE strains were categorized in same group with BU2. The CIL and STC strains closely related but seperated from G2, HAN, B7 and IB strains.

  • The VP2 full gene of Korean infectious bursal disease virus(IBDV) strain, SH/92, three attenuated vaccine strains, Bur706, Bursine-2 and CEV/AC strains, were amplified by reverse transcriptase-polymerase chain reaction and sequenced and compared with published VP2 gene sequences of IBDVs. The VP2 nucleotide sequence similarity between SH/92 and three vaccine stains was 95.6~96.5% whereas the nucleic acid similarity among three vaccine strains was 97.5~98.5%. The amino acid sequence similarity of VP2 of SH/92 compared with three vaccine strains was between 94.4 and 97.6% while the amino acid similarity among three vaccine strains was between 97.4 and 98.4%. The amino acid similarity between SH/92 and classical virulent strain, 52/70 and STC strain was 96.4 and 96.5%, respectively. The serine-rich heptapeptide was conserved in CEVAC and Bursine-2 as well as SH/92 but not in Bur706. The phylogenetic tree developed from amino acid sequences showed that SH/92 was categorized with vv IBDVs(HK46, OKYM, KKI, UPM94/273, SH95) in one branch while three vaccine strains were catagorized with STC strain in the other branch.

  • Japanese encephalitis virus (JEV) is one of the main causes of viral encephalitis in human and animals. For over 30 years, a live attenuated JEV vaccine strain has been used in the veterinary field, and it is required to conduct quality evaluation studies on the commercial vaccines. For the quality control of live attenuated JEV vaccine, we investigated the nucleotide sequence similarity of prME gene derived from five JEV vaccines commercially available in pigs in Korea. The Vero cells infected with JEV vaccines showed specific cytopathic effect, which was characterized by rounding and detached cells. In the phylogenetic analysis, all of the vaccine strains showed a close relationship with the original vaccine seed strain (Anyang 300) and clustered into the genotype 3. In comparison of the nucleotide and deduced amino acid sequences of prME genes with the original strain, all JEV vaccine strains showed high amino acid similarity ranging from 98.9% to 99.5%, but had several point mutations, probably due to high mutation rates of viral RNA polymerase by several virus passages. Even though the current JEV vaccine strains have been maintained and produced for a long period of time, the genetic characterization of them have been rarely changed. However, since the mid 1990's, molecular epidemiology of JEV has been changed sharply from genotype 3 to genotype 1 in Korea, further studies on new vaccine strains to genotype 1 is required for more effective prevention in the field.

  • Biochemical and genetic analysis were carried out to investigate the potential recovery of pathogenecity or related mutations of Brucella abortus RB51 vaccine strains. RB51 strains were recovered from commercial vaccines, including related seed stocks from private companies in Republic of Korea, strain from USA, a reference strain from C university and a field isolate (Daehungjin) from aborted dairy cow after RB51 vaccination were compared with two identified virulent wild strains (S2308 and a field strain isolated from dairy cow in Korea) at the same conditions. All the strains examined, except identified pathogenic strains, revealed the identical characteristics to the original RB51 in biochemical properties, antigen and bacteriophage typing. Outer membrane protein (OMP) profiles from strains of RB51 showed the same patterns with standard RB51 in SDS-PAGE. In addition, Western blotting with the brucella specific monoclonal antibody also indicated that all the vaccine strains were completely deficient in their LPS compared to the pathogenic Br abortus strains. The differences in DNA structures among strains were also possible to detect after PCR. All vaccine strains, except S19, S1119-3, S1075, S544 and Br suis, were amplified a 178bp DNA fragment of eri-gene, and 364bp of IS711 elements. In contrast, 498bp DNA product was only found with Br abortus. Overall evidences in the present study confirmed that the RB51 strains for vaccine production in Korea did not originated from the phenomena of possible recovery of pathogenicity or related to any potential mutation event at all.

Genetic analysis of canine parvovirus vaccine strains in Korea

  • After the original identification of canine parvovirus (CPV) type 2 (CPV-2) in 1978, new antigenic variants such as CPV-2a, CPV-2b and CPV-2c have become widespread in the most countries. In this study, the genetic analysis of canine parvovirus was investigated in a total of 13 CPV vaccines, which have been licensed in Korea since late 1980s, and a field isolate of CPV from a dog with CPV infection clinical symptom. The partial VP2 gene of CPV was amplified and sequenced from 13 vaccine strains and one field isolate. The results showed that of the 13 vaccine strains, 10 strains belong to the CPV-2, 2 strains to CPV-2b, the remaining and one isolate to CPV-2a type, respectively. Several mutations of amino acids were detected at residues of the critical region of the commercial vaccine strains. These data suggest that new type of vaccines containing CPV-2a or CPV-2b/2c type may be required for the better prevention of new CPV infection in dog population in Korea, because CPV-2 contained in most licensed vaccines has been replaced by antigenic variants designated CPV-2a or CPV-2b/c in the worldwide dog population.

Multi-epitope vaccine against drug-resistant strains of Mycobacterium tuberculosis: a proteome-wide subtraction and immunoinformatics approach

  • Mycobacterium tuberculosis (Mtb) is the causative agent of tuberculosis, one of the most deadly infections in humans. The emergence of multidrug-resistant and extensively drug-resistant Mtb strains presents a global challenge. Mtb has shown resistance to many frontline antibiotics, including rifampicin, kanamycin, isoniazid, and capreomycin. The only licensed vaccine, Bacille Calmette-Guerin, does not efficiently protect against adult pulmonary tuberculosis. Therefore, it is urgently necessary to develop new vaccines to prevent infections caused by these strains. We used a subtractive proteomics approach on 23 virulent Mtb strains and identified a conserved membrane protein (MmpL4, NP_214964.1) as both a potential drug target and vaccine candidate. MmpL4 is a non-homologous essential protein in the host and is involved in the pathogen-specific pathway. Furthermore, MmpL4 shows no homology with anti-targets and has limited homology to human gut microflora, potentially reducing the likelihood of adverse effects and cross-reactivity if therapeutics specific to this protein are developed. Subsequently, we constructed a highly soluble, safe, antigenic, and stable multi-subunit vaccine from the MmpL4 protein using immunoinformatics. Molecular dynamics simulations revealed the stability of the vaccine-bound Tolllike receptor-4 complex on a nanosecond scale, and immune simulations indicated strong primary and secondary immune responses in the host. Therefore, our study identifies a new target that could expedite the design of effective therapeutics, and the designed vaccine should be validated. Future directions include an extensive molecular interaction analysis, in silico cloning, wet-lab experiments, and evaluation and comparison of the designed candidate as both a DNA vaccine and protein vaccine.

Strain differentiation of canine distemper virus by reverse transcriptase polymerase chain reaction and restriction fragment length polymorphism analysis

  • To detect CDV RNA in clinical samples and differentiate prevailing CDV virulent strains affecting susceptible animals from attenuated vaccine strains, we performed RT-PCR, RFLP, and sequencing. CDV specific primers were generated from the middle part of nucleocapsid gene. The expected size of PCR products, 519 bp, was observed in tissues of Jindo dog, poodle dog, badger, fourteen of nineteen blood samples as well as 5 vaccine strains including domestic and imported products. The PCR products obtained from tissues and PBMCs of infected animals were digested to 317- and 202-bp fragments by Bam HI, but the products obtained from four of five vaccine strains and Lederle strain were not digesed by Bam HI. Only one vaccine strain of which the PCR products were digested by Bam HI was confirmed as imported vaccine, modified Synider Hill strain. Based on seqencing data obtained from the 519-bp products, it was confirmed that Bam HI restriction site tends to be conserved in field isolates compared to the commercially available attenuated vaccine strains. Partial nucleotide sequences of CDV NP gene obtained from tissues of Jindo dog, poodle and badger shared 100% homology each other, whereas the nucleotide sequences showed 96.3, 96.5, 93.6 and 93.4% homology with Yanaka (virulent), Han95 (virulent), Lederle (attenuated) and Onderstepoort (attenuated) strain, respectively.

Evaluation of the antigen relatedness and efficacy of a single vaccination with different infectious bronchitis virus strains against a challenge with Malaysian variant and QX-like IBV strains

  • Background: The predominant infectious bronchitis virus (IBV) strains detected in chickens in Malaysia are the Malaysian variant (MV) and QX-like, which are associated with respiratory distress, nephropathy, and high mortality. On the other hand, the antigenic relatedness and efficacy of IBV vaccines against these 2 field IBV strains are not well characterized. Objectives: This study aimed to determine the antigen relatedness and efficacy of different IB vaccine strains against a challenge with MV and QX-like strains. Methods: The antigen relatedness and the ability of different IB vaccine strains in conferring protection against MV and QX-like were assessed based on the clinical signs, macroscopic lesions, and ciliary activity. Results: The MV strain IBS037A/2014 showed minor antigenic subtype differences with the vaccine virus Mass H120 and 4/91 strains but showed major antigenic subtype differences with the K2 strain. The Malaysian QX-like strain IBS130/2015 showed major antigenic subtype differences with the MV strain IBS037A/2014 and the vaccine strains except for K2. Chickens vaccinated once with Mass (H120) or with non-Mass (4/91 and K2) developed antibody responses with the highest antibody titer detected in the groups vaccinated with H120 and 4/91. The mean ciliary activities of the vaccinated chickens were between 56 to 59% and 48 to 52% in chickens challenged with IBS037A/2014 and IBS130/2015, respectively. The vaccinated and challenged birds showed mild to severe lesions in the lungs and kidneys. Conclusions: Despite the minor antigenic subtype differences, a single inoculation with Mass or non-Mass vaccines could not protect against the MV IBS037A/2014 and QX-like IBS130/2015.

Preparation of a Vibrio vulnificus Vaccine with Immunogenicity and Protective Efficacy

  • Vibrio vulnificus is a halophilic gram-negative human pathogen, which affects people with underlying liver diseases or a suppressed immune system, often leading to primary septicemia with a mortality rate of higher than 60%. In an effort to develop an oral vaccine against V. vulnificus infection, we prepared a whole cell killed vaccine of V. vulnificus on a large scale and compared the immunogenicity and protective efficacy of the vaccine administered in three formulation forms in rabbits. Since V. vulnificus O-antigen serotypes 1, 2, 3, 4, 5, and 7 account for more than 95% of clinical isolates, we prepared cell lysates from these six serotype strains and mixed in equal amounts for a vaccine. The vaccine was administered to rabbits intramuscularly (i.m.), orally as granules or as enteric-coated granules. In rabbits, all three formulation forms elicited a high level of serum IgG antibody reactive not only to the six strains but also to other O-antigen serotypes 6, 8 and 9, indicating cross-reactivities among the strains. Immunotherapeutic efficacy of the antisera was also evaluated by a passive immunization assay, which revealed that the orally immunized antisera as well as the i.m. immunized antisera was protective against a subsequent lethal challenge of V. vulnificus. These data demonstrate that oral immunization with a V. vulnificus whole cell lysate vaccine induced a systemic immune response and suggest the feasibility of development of this vaccine preparation as an oral vaccine.

국내 시판 Salmonella gallinarum 9R vaccine의 안전성 및 면역원성 비교 (Comparison of the safety and immunogenicity of commercial S. gallinarum 9R vaccine)

  • Salmonella enterica subsp. enterica serovar gallinarum (S. gallinarum) is the agent of fowl typhoid, and the 9R vaccine is a commercial live vaccine for the prevention of fowl typhoid. The aim of this study was to assess the safety and immunogenicity of different brands of S. gallinarum 9R vaccine used in commercial laying chickens in Korea. All 9R strains originated from three different brands showed the same pattern in the biochemical and serological properties, and pulsed field gel electrophoresis (PFGE) profile. But, there was a difference in rhamnose fermentaion, agglutination with Salmonella group $D_1$ antiserum and PFGE pattern between 9R vaccine strain and field S. gallinarum isolates. In laboratory and field trials for assesment of safety and immunogenicity of 9R vaccine, all of the three 9R vaccines showed the same safety in commercial laying chickens. In addition, there was a significant difference between the vaccinated and unvaccinated control groups in mortality and the re-isolation rate of the challenge strain from the tissues (p < 0.05), and no difference by the brands of 9R vaccine. The results from this study indicated that all three different brands of S. gallinarum 9R vaccine showed highly protection against mortality and organ invasion in commercial laying chickens exposed to virulent strains of S. gallinarum.
