Effect of Medium Composition on Phalaenopsis Micropropagation Using Lateral Buds From Flower Stalks (액아배양에 의한 호접란 대량번식시 배지조성의 영향)

  • The effect of medium composition on PLB formation and multiplication and shoot regeneration was studied to establish the micropropagation system of the tropical orchid Phalaenopsis. The highest frequence of PLB formation resulted from the VW medium with 1.2 times ion concentration, 1% sucrose, 1.5g/L PVP or 2.5g/L active charcoal, apple and potato extract and 4g/L gellan gum. The highest ratio of PLB multiplication was obtained from the VW medium with 2% sucrose, apple and potato extract and cotton plate. The shoot regeneration was the most effective with the hyponex medium with 3% apple, 3% potato and 4% banana extract.

  • This study was conducted to develop the clonal propagation technique through in vitro culture using by leaf sheath explants of Phalaenopsis grown in vitro. The highest frequency of protocorm-like body (PLB) formation was obtained when explants of leaf sheath were cultured on VW medium containing 30g/L sucrose, 500 mg/L activated charcoal, 150 mVL coconut water, 1 mg/L NAA, 1 mg/L 2ip and 2.5 g/L gelrite. The PLB formation rate of VW medium was highest followed by modified Hyponex medium, and lowest in MS medium. Plantlets induced from PLBs transferred to plastic pots including spagnum moss were well developed.

  • This study was conducted to develop the clonal propagation technique through in vitro culture using basal stem explants in Phalaenopsis hybrid grown in vitro. The highest frequency of protocorm-like body (PLB) formation was obtained when basal stem explants were cultured on VW medium containing 30g/L sucrose, 500 mg/L activated charcoal, 150 ml/L coconut water, 1 mg/L NAA, 5 mg/L 2iP and 2.5g/L gel rite. PLBs transferred to Hyponex medium were regenerated to plantlets. Plantlets transferred to plastic pots containing spagnum moss were developed and successfully acclimatized under greenhouse. The flower was bloomingly opened in plants regenerated from basal stem explants. The flower was not different from both mother plant and plant induced through clonal propagation of Phalaenopsis hybrid.

Establishment of proliferation and regeneration system of PLBs in Phalaenopsis by treatments of a variety of types of medium, sucrose concentrations and anti-browning agents (다양한 배지종류, sucrose 농도 및 갈변억제물질 처리에 의한 팔레놉시스 PLB 증식 및 재분화 체계확립)

  • To establish an efficient proliferation and regeneration of PLBs (protocorm-like bodies) of Phalaenopsis plants, a variety of propagation medium types, various concentraions of sucrose as well as liquid and solid type were tested in this study. Further, activated charcoal, citric acid and ascorbic acid were compared whether these agents are suppose to reduce the browning in culture process using PLBs of Phalanopsis plants. With regard to the proper propagation medium, VW medium showed 1.3 ~ 2 times highr than those of other medium in an index of increasing for fresh weight and 50% higher than those of other medium in the frequency of shoot regeneration. However, regarding liquid and solid types of culture, there were no significant differences in the proliferation of PLBs and regeneration of shoots from PLBs. In the experiment for a variety of sucrose concentrations (0 ~ 50 g/l), 10 g of sucrose showed 30 ~ 50% higher than other concentrations in increasing index and 10 ~ 50% higher in the regeneration of shoots from PLBs. Regarding the reduction of browning in tissue culture via PLBs of Phalaenopsis plants, 1 g of activated charcoal showed only 1.5% browning of PLBs cultured. Whereas, other treatments including citric acid and ascorbic acid showed 6 ~ 16% of browning of PLBs. Therefore, activated charcoal was selected as an efficient anti-browning agents for the culture of PLBs in Phalaenopsis plants. Using above-described results can be contibuted to the establishment of mass propagation system using PLBs of Phalaenopsis plants in the future.

  • Our study was carried out for plant regeneration via somatic embryogenesis from immature seeds of Bletilla striata. The highest frequency of embryogenic callus formation was obtained from the immature seeds (at 150 days after pollination) cultured on Hyponex and VW medium supplemented with 3 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 1 mg/l kinetin under the dark condition. Multiple somatic embryos were induced when embryogenic callus was transferred to VW medium without growth regulators under continued illumination. Somatic embryos were observed histologically with scanning electron microscopy. Regeneration of Bletilla striata was obtained from somatic embryos with a well-defined scutellum and coleoptile as well as with one or more shoot primordia and root primordia. We think that these methods for orchid multiplication must be useful to access clonal propagation of orchids.

  • Cymbidium aloifolium (L). Sw. is an exquisite epiphytic orchid of the Kolli Hills (Eastern Ghats) of Tamil Nadu in Southern India. It is fast disappearing from its natural habitats due to deforestation and low germination rate in natural habitat. In the present study, an attempt was made to germinate the seeds from un-dehisced capsule of Cymbidium aloifolium (L). Sw under in vitro condition. The seed germination and protocorm development were recorded in three different well known media namely Knudson C (KC), Half strength Murashige & Skoog (1/2 MS) and Vacin & Went (VW) media. The highest seed germination of 90% was observed KC basal media after $30^{th}$ days whereas germination percentages were 40% and 30% on 1/2 MS and VW media respectively. The well-developed protocorm were transferred to KC media supplemented with 6-Benzyl Amino Purine (BAP) and Naphthalene acetic acid (NAA) where BAP (1.0 mg/l) and NAA (1.0 mg/l) together were found to be optimum for the highest shoot formation. About 90% of the shoots found to be well rooted after transfer to the KC medium differently supplemented with 1.5 mg/l Indole-3-acetic acid (IAA) and 1.0 mg/l Indole-3-butyric acid (IBA). Though rooting also took place in the two basic media but the duration was longer when compared with the hormone-supplemented media. The rooted plantlets were hardened and kept under greenhouse conditions which can be relocated in natural habitats.

Effect of Culture Medium, Temperature, and Light Intensity on PLB Propagation of Phalaenopsis (팔레높시스의 PLB 증식에 미치는 배지와 배양온도 및 광도의 영향)

  • This study was conducted to investigate the effect of culture media and environment on PLB proliferation by using PLBs produced from leaf segments excised from shoot of Phalanopsis flower stalk. The fresh weight of PLBs propagated was higher in MS medium than in NDM (New Dogashima medium) or VW, but the condition of PLB was better in NDM medium. Natural additives of Coconut water, potato and apple were absolutely required for the PLB propagation. PLB propagation was better in solid medium than in liquid medium including cotton as support. Optimal sucrose concentration for proliferation was 10 g/L. PLB proliferation was very effective condition 14.3 $\mu$mol.s$^{-1}$ m$^{-2}$ in PPFD and $25^{\circ}C$.

  • This study was conducted to find out the commercial in vitro propagation methods through protocom-like bodies (PLB) induced from shoot tip culture of Miltonia spp. Among several culture media for induction PLB from shoot tip in Miltonia spp., MS basal medium was better than Hyponex, Vacin & Went basal medium and other media supplemented with natural additives. PLB's proliferation and differentiation in Hyponex medium including $20g{\cdot}L^{-1}$ banana + $20g{\cdot}L^{-1}$ sucrose(pH 5.2) was better than in MS medium. It was tendency that solid media showed higher PLB fresh weight than liquid medium or other cotton bridge culture. The dark culture for 1~2 weeks and adding $10{\sim}20g{\cdot}L^{-1}$ sucrose on hyponex basal medium was the most effective to increase the PLBs growth and shoot number.

  • The objective of the study is to propose the catch-up strategy of Hyundai Kia to become the globally leading company in the automobile industry. For this, we try to compare Hyundai Kia with three leading automobile companies: Toyota, VW, and GM by using the Generalized Double Diamond Model. As a result, we present three ways that Hyundai Kia can become a "First Mover." The three strategies are : 1) controlling increase of wage, raising productivity, and increasing R&D investment 2) increasing overseas investment of the small and medium sized automobile factories 3) pursuing the two tracks strategy of a public car and a prestige car more aggressively. This study is significant in the fact that it presents the catch-up strategy of Hyundai Kia in the automobile industry, of which the leaders can not be changed easily, by the Generalized Double Diamond Model approach.

Substituent Effect on the Fungicidal Activity of New N-substituted Benzotriazol-1-yl Derivatives (새로운 N-치환 benzotriazol-1-yl유도체의 항균활성에 미치는 치환기 효과)

  • Series of new chiral N-substituted benzotriazol-1-yl derivatives were synthesized and their fungicidal activities in vitro against gray mold(Botrytis cinerea), black spot(Alternaria kikuchiana) and phytophthora blight(Phytophthora capsici) were measured by the agar medium dilution method. The substituents effects between the fungicidal activities (obs. $pI_{50}$) and a various physicochemical parameters of phenoxy or thiophenoxy group(X) & alkyl or phenyl group(Y) were analyzed by the multiple regression technique. From the analyzed substituent effects, the structure-activity relationship(SAR) equations shows that the antifungal activities depend on the parameters for the optimal molecular hydrophobicity($({\Sigma}logP)_{opt}$), Van der Waals (${\Sigma}Vw$>0) volume(${\AA}^3$) and inductive constant with electron withdrawing group(${\sigma}_I$,Y>0). The activity in affected by the inductive effect (${\sigma}_I$,Y>${\sigma}_g$X) of Y-group rather than the X-group. The phenoxy substituents, 1, showed higher antifungal activity tn the thiophenoxy substituents, 2. For 1, polar substituent constant(${\sigma}^*$) was an important factor in determining the activity. And the tribromomethyl substituent, 1g showed the highest activity against the tee fungi.
